Background: Escherichia coli (E. coli) is an enteropathogen that commonly causes diarrhea in calves. However, not all E. coli isolates are pathogenic. The aim of this study was to identify E. coli virulence factors derived from fecal samples collected in the southern region of Rio Grande do Sul state (RS) from calves with and without diarrhea, as well as investigate the antimicrobial susceptibility of E. coli isolates from calves with diarrhea.Materials, Methods & Results: Forty stool samples were collected in 12 farms, each one from calves having one day to six months of age, with and without diarrhea. The total DNA of from these isolates was extracted and a PCR using primers specific for the virulence factors Stx1, Eae, F41, F5 and STa was conducted. The susceptibility testing used the disk diffusion method and the susceptibility profile was evaluated against the following antimicrobials: ampicillin, penicillin, chloramphenicol, enrofloxacin, gentamicin, trimethoprim, sulfonamide, tetracycline and streptomycin. The canine monocytic ehrlichiosis, caused by Ehrlichia canis, is endemic in several regions of Brazil. Some serological diagnostic techniques using immunodominant proteins of E. canis as antigens are available, but their specificities and sensitivities are questionable. Based on this, the objective of this study was to test the antigenic potential of the recombinant gp19 protein (rGP19) for subsequent use in diagnostic tests. The rGP19 expressed in the Escherichia coli strain BL21 (DE3) C41 was recognized in the sera from experimentally infected dogs using ELISA and Western blotting. Thus, it was possible to obtain a promising antigen with the ability to differentiate between E. canis-positive and -negative animals, even 1 week after infection.


A erliquiose monocítica canina, causada por Ehrlichia canis, é uma doença endêmica em diversas regiões do Brasil. Algumas técnicas de diagnóstico sorológico, utilizando proteínas imunodominantes de E. canis como antígenos, estão disponíveis, porém suas especificidades e sensibilidades são questionáveis. Com base nesse fato, o objetivo deste trabalho foi testar o potencial antigênico da proteína GP19 recombinante (rGP19) para posterior utilização em testes diagnósticos. A rGP19, expressa em E. coli cepa BL21 (DE3) C41, foi reconhecida por soros de cães experimentalmente infectados pelas técnicas de ELISA e Western blotting. Cattle ticks are responsible for great economic losses in cattle farming worldwide, and their main control method, chemicals, has been showing problems, whether resulting from the development of resistant strains of ticks or environmental contamination. Research studies directed toward developing vaccines against ticks are emerging. One way to evaluate those vaccines is to calculate the percentage of efficacy. The aim of this study was to analyze scientific publications archived in PubMed that used this method of assessment and discuss the main factors that may affect its calculation. Thus, 25 articles addressing this subject were selected. The percentage of efficacy was usually calculated in one of two ways, with one considering the reduced fertility of eggs and the other not. The latter method may underestimate the vaccine efficacy, and the most complete formula for calculating the efficacy reflects how much the vaccine actually affects the infestation. In our view, the use of the complete formula for calculating the percentage of efficacy is broader and more representative of the vaccine effect on the tick population.


Carrapatos de bovinos são responsáveis por grandes perdas econômicas para a pecuária bovina mundial e seu principal método de controle, o químico, vem apresentando problemas, seja pelo desenvolvimento de amostras de carrapatos resistentes ou pela contaminação ambiental. Na tentativa de diminuir a utilização dos acaricidas, surgem pesquisas direcionadas ao desenvolvimento de vacinas contra carrapatos. Uma maneira de avaliar essas vacinas é pelo cálculo de percentagem de eficácia. O objetivo deste trabalho foi analisar as publicações científicas indexadas no PubMed que utilizaram este método de avaliação e discutir os principais fatores que podem interferir no seu cálculo. Dessa maneira, selecionaram-se 25 artigos que tratavam desse assunto. A percentagem de eficácia apareceu sendo calculada de duas formas, uma considerando a redução da fertilidade dos ovos e a outra não. Essa última pode subestimar a eficiência da vacina, e a fórmula de cálculo da eficácia mais completa representa o quanto da infestação a vacina realmente reduziu. Em nosso entendimento, a utilização da fórmula completa para o cálculo da percentagem de eficácia é mais abrangente e representativa do efeito da vacina na população de carrapatos.

Herein, we report tick species found on wild and domestic animals and in the environment during a one-year sampling period at the Brazilian Farming Research Company beef cattle unit (Embrapa Beef Cattle), which is located within the urban area of Campo Grande, Mato Grosso do Sul, Brazil. From 55 wild hosts including six different species (Nasua nasua, Cebus spp., Cerdocyon thous,Myrmecophaga tridactyla, Tamandua tetradactyla and Dasyprocta aguti), 323 ticks were collected. Amblyomma ovale ticks were found solely on coatis, and Amblyomma nodosum was identified solely on anteaters. No ticks were found on capuchin monkeys. However, Amblyomma cajennense was found on all parasitized host species with the exception of capuchin monkeys. Giant anteaters displayed the highest infestation abundance, with a mean of 53 ticksanimal. Environmental sampling yielded 166 adult A. cajennense ticks. The tick species found on domestic animals (Rhipicephalus(Boophilus) microplus,R. sanguineus, Dermacentor nitens andA. cajennense) were those typically found on these hosts in Brazil. The most prevalent tick species, A. cajennense, was found on both wild and domestic animals and was also prevalent in the environment. Thus, this tick species is the primary vector that allows pathogens to bridge wild and domestic animals in the Cerrado.


Neste trabalho são descritas as espécies de carrapatos de animais selvagens e domésticos e do ambiente coletados por um ano na EMBRAPA Gado de Corte localizado na área urbana de Campo Grande, Mato Grosso do Sul, Brasil. Dos 55 hospedeiros selvagens de seis espécies diferentes (Nasua nasua, Cebus spp.,Cerdocyon thous, Myrmecophaga tridactyla,Tamandua tetradactyla e Dasyprocta aguti) foram coletados 323 carrapatos. Amblyomma ovale foi encontrado apenas em quatis e Amblyomma nodosum apenas sobre tamanduás. Nenhum carrapato foi encontrado sobre macacos-prego. Por outro lado, Amblyomma cajennense foi encontrado em todos os hospedeiros com exceção dos macacos-pregos. A maior abundancia de infestação foi aquela em tamanduás-bandeira com média de 53 carrapatosanimal. No ambiente foram capturados 166 carrapatos, todos da espécie A. cajennense. As espécies de carrapatos em animais domésticos (Rhipicephalus(Boophilus) microplus,Rhipicephalus sanguineus, Dermacentor nitens e A. cajennense) foram aquelas características nestes hospedeiros no Brasil. De forma geral a espécie de carrapato A. cajennense foi a mais prevalente sendo encontrada em animais domésticos e selvagens bem como no ambiente. Portanto, esta é a principal espécie de vetor a estabelecer uma ponte para bioagentes patogênicos entre animais domésticos e selvagens.

The cattle tick Rhipicephalus (Boophilus) microplus is responsible for great economic losses. It is mainly controlled chemically, with limitations regarding development of resistance to the chemicals. Vaccines may help control this parasite, thereby reducing tick pesticide use. In this light, we performed subcloning of the gene of the protein Bm86-GC, the homologue protein that currently forms the basis of vaccines (GavacTM and TickGardPLUS) that have been developed against cattle ticks. The subcloning was done in the pPIC9 expression vector, for transformation in the yeast Pichia pastoris. This protein was characterized by expression of the recombinant Mut+ strain, which expressed greater quantities of protein. The expressed protein (rBm86-CG) was recognized in the Western-blot assay using anti-Gavac, anti-TickGard, anti-larval extract and anti-rBm86-CG polyclonal sera. The serum produced in cattle vaccinated with the antigen CG rBm86 presented high antibody titers and recognized the native protein. The rBm86-GC has potential relevance as an immunogen for vaccine formulation against cattle ticks.


O carrapato-do-boi Rhipicephalus (Boophilus) microplus é responsável por grandes perdas econômicas. Seu controle é principalmente químico e apresenta limitações quanto ao desenvolvimento de resistência aos princípios ativos. As vacinas podem auxiliar no controle deste parasita diminuindo as aplicações de carrapaticidas. Considerando isso, foi realizada a subclonagem do gene da proteína Bm86-CG, proteína homologa a que atualmente é a base das vacinas desenvolvidas (GavacTM e TickGardPLUS) contra o carrapato-do-boi, no vetor de expressão pPIC9, para ser transformado em levedura, Pichia pastoris. Esta proteína foi caracterizada pela expressão da cepa recombinante Mut+ que expressou maior quantidade de proteína. A proteína expressa, rBm86-CG, foi reconhecida no ensaio de Western-blot pelos soros policlonais anti-Gavac, anti-TickGard, anti-Extrato de larva e anti-rBm86-CG. O soro produzido em bovinos vacinados com o antígeno rBm86-CG apresentou altos títulos de anticorpo e reconheceu a proteína nativa. A rBm86-CG possui potencial relevância como imunógeno para formulação vacinal contra o carrapato de bovinos.

Neospora caninum is an obligate intracellular parasite that can infect domestic and wild canids, ruminants, and horses. The purpose of this study was to evaluate the association between the presence of antibodies to N. caninum and reproductive loss in beef heifers in the South Pantanal region of Mato Grosso, Brazil. A total of 1098 heifers were evaluated from breeding to calving with regard to reproductive performance, and serology for neosporosis was assayed by indirect immunofluorescence reaction. The prevalence of N. caninum in cows that had failed to conceive was 22.13% higher than in those that had conceived (p 0.0001), indicating a significant correlation between non-conception and presence of anti-N. caninum antibodies. The extraction rates for seropositive and seronegative heifers were 28.24 and 50.12%, respectively, with a statistically significant value (p 0.0001) that showed a decrease of 44% in the extraction rate of seropositive animals. The results emphasize the importance of diagnosis for monitoring of neosporosis in primiparous cows, and indicators of seropositive females as a factor in strategies for culling the beef cattle herd.


Neospora caninum é um parasita intracelular obrigatório que pode infectar canídeos domésticos e selvagens, ruminantes e equídeos. Neste trabalho estudou-se a associação entre soropositividade para Neospora caninum e perda reprodutiva em novilhas de corte na região do Pantanal Sul-Mato-Grossense. Foram avaliadas 1098 novilhas da estação de reprodução ao nascimento dos bezerros com relação ao desempenho reprodutivo e realizada a sorologia para N. caninum através da reação de imunofluorescência indireta. A soroprevalência de N. caninum nas vacas que falhara na concepção foi 22,13% maior que nas que conceberam (p 0,0001), indicando que há uma correlação significativa entre a não concepção e a presença de anticorpos anti-N. caninum. As taxas de desfrute para as novilhas soropositivas e soronegativas para neosporose foram de 28,24 e 50,12%, respectivamente, mostrando uma redução de 44% na taxa de desfrute de novilhas soropositivas para N. caninum. Ressalta-se a importância do monitoramento do diagnóstico da neosporose em primíparas, e o indicador de fêmeas soropositivas como um fator nas estratégias de descarte no rebanho bovino de corte.
