Nucleotide-binding oligomerization domain-like receptors (NLRs) play a key role in the innate immune response as pattern-recognition receptors. However, the role of NLRC5, which is a member of the NLR family, in NF-κB activation and MHC-I expression remains debatable. Infection with the J group avian leukosis virus (ALV-J) can result in immunosuppression and a subsequent increase in susceptibility to secondary infection. This results in huge economic losses to the poultry industry worldwide. Using quantitative real-time polymerase chain reaction (qRT-PCR), we investigated the mRNA expression levels of NLRC5 signal pathway-related genes in secondary chicken embryo fibroblasts 7 days after infection with ALV-J. The results indicated that, compared with the control groups, the expression levels of TLR7, MHC-I, and IL-18 increased significantly in the infected groups at 7 days post-infection (d.p.i.). The expression levels of NLRC5 and IL-6 were conspicuously downregulated at 7 d.p.i., but the expression levels of NF-κB, STAT1, and STAT3 were not significantly altered. These results suggest that NLRC5 and some genes involved in the NLRC5 pathway play a key role in antiviral immunity, typically the response to ALV-J infection. Moreover, MHC-I expression levels vary between different cell types.

The P-element-induced wimpy testis (Piwi) gene is involved in germline stem cell self-renewal, meiosis, RNA silencing, and transcriptional regulation. Piwi genes are relatively well conserved in many species, but their function in poultry species is unclear. In this study, Piwi genes were sequenced using a target-sequence capture assay in quail and 28 breeds of chicken. Single nucleotide polymorphisms (SNPs) and evolutionary aspects of these chicken breeds were then analyzed. We found that SNP sites existed mainly in the introns of a few chicken breeds, and we selected an SNP on intron 4 for further verification by Sanger sequencing, the results of which were similar to those obtained by the target-capture sequencing assay. The evolutionary analysis revealed that there were more mutations in the Chahua and Leghorn breeds than in the other breeds, and that the phylogenetic tree was divided into four main categories that suggested that Piwi is evolutionarily conserved, and mutations in the introns might be associated with gametogenesis. The screened SNPs can be used as candidate markers for Piwi, and our results provide basic information for the further study of Piwi function in poultry.

OBJECTIVE: The aim of this study was to improve the understanding of FBA in children and to decrease the rate of misdiagnosis, missed diagnosis and morbidity. PATIENTS AND METHODS: We analyzed the clinical features and the three-dimensional reconstructed CT images of 590 children with foreign body aspiration (FBA) in the Xuzhou area of the Jiangsu province. RESULTS: CT imaging revealed common complications of FBA including emphysema (n = 379), pneumonia (n = 174), and atelectasis (n = 26). The remaining 120 patients had no visible complications on the three-dimensional reconstructed CT images. Serious complications including pneumothorax, pneumomediastinum, subcutaneous emphysema, pneumatorrhachis could also be observed. The types of foreign bodies were diverse: the most common were peanuts and sunflower seeds. The diagnostic accuracy of the three-dimensional CT imaging was high, with a sensitivity and specificity of 99.83% and 99.89%, respectively. CONCLUSIONS: 3D CT imaging is an accurate, non-invasive technique to evaluate children with suspected FBA that can help decrease the rate of misdiagnosis and eliminate a delay in treatment for this potentially life-threatening condition.

The aim of this study was to provide the experimental basis for effective prevention and treatment of obliterative bronchiolitis (OB) by studying the changes on the microRNA (miRNA) expression profile after an orthotopic tracheal transplantation (OTT) simulating lung transplantation (LT). The OTT was performed on inbred rats to establish an OB animal model simulating LT, which was confirmed successful through pathological examination after 4 weeks. A miRNA microarray was used to screen for the most significantly differentially expressed miRNA in the OB tissues of donor transplanted trachea and real-time quantitative PCR was then used to validate the reliability of the microarray results. The microarray detection obtained 29 OB-related miRNAs, composed of 15 and 14 significantly up- and down-regulated miRNAs, respectively, among which miR-146a, miR-155, and miR-451, whose function is involved in the immune and inflammatory reactions, were subjected to relative quantitation research. The LT-simulated OTT-induced OB showed significantly differential expressions of multiple miRNAs, among which miR-146a and miR-155 were highly expressed, while miR-451 was lowly expressed, suggesting that these miRNAs may play an important regulatory role in the OB pathological process after LT.

This study aimed at comparing the growth and mineralization of the femur, tibia, and metatarsus of male and female broiler chicks. On the day of hatch, 100 male and 100 female Ross 308 broiler chicks were transferred stainless cages with 10 birds per cage. On d 7, 14, 21, 28, 35, and 42, five males and five females were sacrificed and their femur, tibia, and metatarsus were collected. Results showed that the tibia was the heaviest and the longest and contained the highest content of ash and calcium (Ca) among the three leg bones. The femur had the greatest diameter. The weight, length, diameter, and ash weight of the femur, tibia, and metatarsus linearly increased with age. The ash, Ca, and phosphorus (P) content in the femur and the tibia quadratically increased with age; by contrast, these parameters in the metatarsus linearly increased with age. The bones grew faster in 1 to 21 d of age. The weight, diameter, and ash weight of the three bones of males were higher than those of females. The Ca to P ratio of the three bones (femur, tibia, and metatarsus) was approximately 2.0:1. These data indicate that there are differences in bone growth and mineralization among the femur, tibia, and metatarsus of male or female broiler chicks.

This study evaluated the effects of dietary non-phytate phosphorus (NPP) and 1a-hydroxycholecalciferol (1a-OH-D3) on the growth performance, bone mineralization, and carcass traits of 1- to 21-day-old broiler chickens. On the day of hatch, 600 male Ross 308 chicks were weighed and randomly assigned to 12 treatments, with five cages of 10 birds each. A 6 × 2 factorial arrangement was applied, consisting of 0.20%, 0.25%, 0.30%, 0.35%, 0.40%, or 0.45% NPP and 0 or 5 g/kg of 1a-OH-D3. The basal diet contained 0.52% calcium (Ca) and was not supplemented with vitamin D3. Dietary NPP levels significantly affected growth performance and tibia mineralization (except width) of broilers; by contrast, meat yield and organ relative weight were not influenced by NPP. The inclusion of 1a-OH-D3 improved growth performance, tibia mineralization, and carcass and breast yield, whereas it decreased the relative weights of the liver, heart, and kidney. A significant interaction between NPP and 1a-OH-D3 was observed for body weight gain (BWG), feed efficiency (FE), mortality, serum Ca and P levels, tibia breaking-strength, ash weight, and Ca content, as well as breast yield and heart relative weight. These results suggest that broilers fed with 5 g of 1a-OH-D3 per kg of diet obtain optimal growth performance and tibia mineralization when dietary NPP level was 0.30% and the analyzed Ca to NPP ratio was 1.97.

The genomic expression profile of the super-hybrid rice Liangyoupeijiu female parent Pei'ai 64S in different tissues at different developmental stages under low temperature, drought, and high temperature stresses were detected using an Affymetrix GeneChip Rice Genome Array to screen upregulated and downregulated genes. In this study, we screened the drought-resistant gene OsRCI2-5, after which a constitutive OsRCI2-5 construct was created and transferred into Nipponbare. After polyethylene glycol-6000 and drought treatment, we found that the OsRCI2-5 gene improved the drought resistance of Nipponbare. Gene expression profiling showed that the OsRCI2-5 gene was expressed in the rice leaves, stems, and flower organs. Subcellular localization revealed that the gene was located in the membranes, and hence, we can deduce that a membrane signal peptide was responsible for signal transduction.
