RESUMEN

AIM To study the chemical constituents from the fruits of Evodia rutaecarpa (Juss.) Benth.and their biological activities.METHODS The ethyl acetate fraction of 80% ethanol extract from E.rutaecarpa was isolated and purified by silica column and recrystallization,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The antitumor and antifungal activities were determined by MTT and mycelium growth method,respectively.RESULTS Ten compounds were isolated and identified as rutaecarpine (1),evodiamine (2),1-methyl-2-undecyl-4 (1H)-quinolone (3),1-methyl-2-tridecyl-4 (1H)-quinolone (4),wuchuyuamide Ⅰ (5),wuchuyuamide Ⅲ (6),wuchuyuamide Ⅳ (7),β-sitosterol (8),β-daucosterol (9),dehydroevodiamine (10).Compounds 1,2,10 had certain inhibitory effects on MDA-MB-231,LoVo,A2780 and HeLa with the IC50values of 0.65-29.45 μmol/L.Compound 1 had a certain inhibitory effect on Botrytis cinerea,Glomerella cingulata and Rhizoctonia solani with the inhibitory rates of 18.12%-49.57%.CONCLUSION Compounds 3-7 are isolated from this plant for the first time,compounds 1,2,10 have strong biological activities.

RESUMEN

OBJECTIVES: Bioactive calcium phosphate cement (CPC) has been used widely to repair bone defects because of its excellent biocompatibility and bioactivity. However, the poor handling properties, low initial mechanical strength, and long setting time of CPC limit its application in vital pulp therapy (VPT). The aim of this study was to synthesize biphasic calcium phosphate/sulfate cements and evaluate the feasibility of applying these cements in VPT. METHODS: The physical, chemical, and mechanical properties of CPC were improved by mixing the cement with various amounts of α-calcium sulfate hemihydrate (CSH). The hydration products and crystalline phases of the materials were characterized using scanning electron microscopy and X-ray diffraction analysis. In addition, the physical properties, such as the setting time, compressive strength, viscosity, and pH were determined. Water-soluble tetrazolium salt-1 and lactase dehydrogenase were used to evaluate cell viability and cytotoxicity. RESULTS: The developed CPC (CPC/CSH cement), which contains 50wt% CSH cement, exhibited no obvious temperature increase or pH change during setting when it was used as a paste. The initial setting time of the CPC/CSH biphasic cement was substantially shorter than that of CPC, and the initial mechanical strength was 23.7±5.6MPa. The CPC/CSH cement exhibited higher viscosity than CPC and, thus, featured acceptable handling properties. X-ray diffraction analysis revealed that the relative peak intensity for hydroxyapatite increased, and the intensity for calcium sulfate dehydrate decreased as the amount of CPC was increased. The cell viability and cytotoxicity test results indicated that the CPC/CSH cement did not harm dental pulp cells. SIGNIFICANCE: The developed CPC/CSH biphasic cement exhibits substantial potential for application in VPT.

<p><b>OBJECTIVE</b>To investigate the cholesterol 7alpha-hydroxylase gene -204A/C polymorphism and its relationship with serum lipids and apolipoproteins (apo) levels in patients with endogenous hypertriglyceridemia (HTG) in Chinese population in Chengdu area.</p><p><b>METHODS</b>The genotype and allele frequencies of cholesterol 7alpha-hydroxylase gene -204A/C polymorphism were analyzed by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP). Serum lipids were measured by enzymatic kits and apolipoproteins AI, AII, B100, CII, CIII and E were measured by the RID kits in 132 HTG patients and 212 control subjects.</p><p><b>RESULTS</b>Allele frequencies of A and C were 0.602 and 0.398 in HTG group and 0.601 and 0.399 in control group, respectively. There was no significant difference of allele and genotypes frequencies between HTG and control groups (P> 0.05). In HTG group, carriers with the genotypes CC and AC were associated with significantly higher concentrations of triglycerides and apoCIII compared with those with genotype AA (P< 0.05). In the control group, carriers with the genotypes CC and AC were associated with significantly lower serum high density lipoprotein cholesterol (HDL-C) level compared with those with genotype AA (P< 0.05). In the male control group, carriers with the genotypes CC and AC had elevated levels of serum triglycerides than those with genotype AA (P< 0.05).</p><p><b>CONCLUSION</b>These results suggest that -204A/C polymorphism in the CYP7A1 gene does not relate with HTG but may has an effect on serum triglyceride and apoCIII levels in patients with endogenous HTG, the serum HDL-C level in control subjects and the serum TG level in male control subjects.</p>
