RESUMEN

Newcastle disease remains a major concern to the poultry industry; however, it can be managed with effective vaccination programs. The main objective of this study was to evaluate the effect of 2 1× doses of live LaSota strain Newcastle disease virus (NDV) vaccine administered oculo-nasally on d one and 21 on development of humoral and cell-mediated immune response in broilers, and to compare different immunization schedules. Two experiments were conducted. In Experiment I (n = 320), Ross 308 birds were randomly assigned to an unvaccinated control group or vaccinated treatment. [Both treatments consisted of 4 pens per treatment and 40 birds per pen]. At d one, live NDV LaSota strain vaccine was used as a primary immunization to evaluate its impact on adaptive immunity. No substantial NDV-specific humoral immune response was established. Body weights were significantly higher (P < 0.05) in the vaccinated birds on d 4 and 7. Spleen index of the vaccinated birds was significantly (P < 0.05) lower at d 28 and 35. Flow cytometry showed reduced levels of peripheral and splenic B and T lymphocytes. Interferon gamma secreted by splenocytes and in circulation was measured; the results showed a reduced expression post-secondary immunization. In Experiment II (n = 180), the role of maternal antibodies and primary vaccination at d one was evaluated using 3 vaccination protocols. Protocol 1 used live B1 strain as primary immunization, whereas protocol 2 used live LaSota strain. Protocol 3 used live LaSota strain after maternal antibodies had decayed. Protocol 3 resulted in the highest NDV-titer level during the trial. Protocol 2 had the lowest NDV titer. Feed conversion was significantly higher (P < 0.05) in protocol 3 compared to 1 and 2. Overall, the results indicate that the use of live LaSota strain NDV vaccine as primary immunization at d one has a detrimental effect on the development of adaptive immunity in broilers; however, its use after the level of maternal antibodies decays results in a robust antigen-specific humoral immune response.

RESUMEN

The production of odiferous metabolites, such as 2-methlyisoborneol (MIB), is a major concern for water utilities worldwide. Although MIB has no known biological function, the presence of the earthy/musty taste and odor attributed to this compound result in the reporting of numerous complaints by consumers, which undermines water utility performance and the safe and adequate provision of potable waters. Cyanobacteria are the major producers of MIB in natural waters, by mechanisms that have heretofore remained largely unstudied. To investigate the fundamental biological mechanism of MIB biosynthesis in cyanobacteria, the genome of a MIB-producing Pseudanabaena limnetica was sequenced using Next Generation Sequencing, and the recombinant proteins derived from the putative MIB biosynthetic genes were biochemically characterized. We demonstrate that the biosynthesis of MIB in cyanobacteria is a result of 2 key reactions: 1) a S-adenosylmethionine-dependent methylation of the monoterpene precursor geranyl diphosphate (GPP) to 2-methyl-GPP catalyzed by geranyl diphosphate 2-methyltransferase (GPPMT) and 2) further cyclization of 2-methyl-GPP to MIB catalyzed by MIB synthase (MIBS) as part of a MIB operon. Based on a comparison of the component MIB biosynthetic genes in actinomycetes and cyanobacterial organisms, we hypothesize that there have been multiple rearrangements of the genes in this operon.

BACKGROUND: This study investigates the short-term and long-term effects of thymosin alpha 1 (Talpha1) on reimplantation of avulsed teeth. METHODS AND MATERIAL: Seventy-three patients with avulsed teeth were double-blind randomly assigned to a control group and group that used Talpha1. The teeth were reimplanted after treatment with Talpha1 or saline. Patients were monitored for both short-term and long-term parameters. RESULTS: The thymosin group demonstrated a lower interferon, tumor necrosis factor alpha and interleukin-6 levels (P < .05) and higher white blood cell levels than the control group (P < .05). The thymosin group demonstrated greater periodontal healing (P < .05), less ankylosis (P < .05), less tooth movement, and greater lifetime of the replanted teeth. CONCLUSION: The use of Talpha1 has both short-term and long-term beneficial effects during reimplantation of avulsed teeth.

Functional regression of the mammary gland is partly reflected by proteolysis of milk protein and tissue protein. The involvement of the plasminogen activation system in degradation of milk protein and mammary tissue damage has been demonstrated under inflammatory conditions. In this study, mammary secretion from 23 dairy goats primarily grouped as lactation (milking twice daily) or involution (milking once daily or less) was used to determine the ratio of gravity-precipitated casein to total milk protein (casein ratio) as an index of caseinolysis, and activities of components of plasminogen activation system as well as their expressions on somatic cells. Based on the casein ratio, lactation goats were subcategorized as very active (71.8 +/- 1.0%) or less active (29.9 +/- 1.0%) in mammary function; involution goats were subcategorized as gradual (21.7 +/- 1.0%) or acute (5.9 +/- 0.2%) involution. This result suggests that caseinolysis occurred during regular lactation as well as during involution. On the other hand, activities of components of the plasminogen activation system in mammary secretion were increased along with the decreasing casein ratio, in contrast to the similar activities of their counterparts in circulation throughout various mammary statuses. Correlation analysis between casein ratio and activities of plasminogen activation system of goat milk indicated a significant negative relationship for plasmin (r = -0.64), plasminogen (r = -0.69), and urokinase-type plasminogen activator (uPA; r = -0.78) during involution but not during lactation. As for the cellular components of plasminogen activation system, there was an increase in immunoreactivity on somatic cells toward both monoclonal antibodies of human uPA and human uPA receptor under involution conditions suggesting their upregulation relative to lactation condition. Collectively, these results suggest that plasminogen activation system within the mammary gland differentially contribute to milk caseinolysis along the various stages of goat lactation. Meanwhile, a somatic cell-mediated local elevation of plasmin activity may be committed to extensive caseinolysis during involution.

The safety and efficacy of patient-controlled analgesia used for postoperative pain relief were evaluated. Cumulative 24-hour requirements were analyzed for possible correlation with patient characteristics. All patients who used a patient-controlled analgesia device for postoperative pain relief were reviewed from June to October 1991. The device Baxter's basal/bolus infusor with patient control module, was used to deliver fentanyl in 379 patients. The fentanyl requirement, verbal analog pain score, first passage of flatus, side effects, sedative score, and degree of satisfaction were examined. The fentanyl requirement during the first 24 hours after operation was analyzed with regard to age, body weight, and sex. The daily fentanyl consumption in the first three postoperative days was 928 +/- 352 micrograms (n = 338), 553 +/- 259 micrograms (n = 220), and 490 +/- 222 micrograms (n = 71), respectively. The requirement for fentanyl during the first 24 hours after surgery was significantly higher than for the next two days (p-value < 0.001). Fentanyl consumption correlated well with body weight, and inversely with age. No difference was found between fentanyl consumption and sex (p-value = 0.4687). The mean time to the first passage of flatus in patients with abdominal surgery was 54.6 +/- 26.4 hours. The incidence of nausea, vomiting, and dizziness was similar, about 20% of patients. Itching was noted in 7% of patients. Oversedation (class 4) was found in three patients during the first operative day, the sedative score for other patients were around class 1-3. No patient exhibited signs of respiratory depression or withdrawal syndrome.(ABSTRACT TRUNCATED AT 250 WORDS)

The use of a disposable non-electronic patient-controlled analgesia (PCA) device (Baxter) was evaluated in this study. Every patient under trial was instructed thoroughly how to use the PCA device before operation. The PCA device delivers a 25 mg IV injection of pethidine upon patient demand with a delay of 15 min between allowable administrations. The degree of patient satisfaction, the drug consumption, and the duration of using analgesia were compared with those in IM pethidine group. The results showed that, in the PCA group, the duration of using analgesic was longer; the drug consumption was greater (especially during the first 20 h after operation); and the degree of patient satisfaction was higher. The series comprised a total of 54 patients, 14 of whom complained of mild dizziness or nausea which did not require specific treatment. In sum, giving analgesic by the Baxter PCA device was a safe and effective alternative for IM analgesic in postoperative pain control.
