RESUMEN

Climate change has prompted the search for new methods for improving agricultural practices for legume crops. The aim of the study was to test an innovative method of complementary photostimulation of seeds and plants aimed to improve the quantitative and qualitative features of soybean (Glycine hispida L. (Merr.)) yield. Complementary photostimulation of plants was shown to positively affect the yield and chemical composition of soybeans, significantly increasing the content of protein and fat in seeds of the Merlin cultivar. Significant positive effects compared to the control were obtained following irradiation of seeds and plants for 3 s (the shorter of the analyzed exposure times). The results clearly indicate the need to improve the proposed new HUGO (High Utility for Optimal Growth) technology to optimize soybean yield.

The management of hard-to-heal wounds is a significant clinical challenge. Acellular dermal matrices (ADMs) have been successfully introduced to enhance the healing process. Here, we aimed to develop protocol for the preparation of novel ADMs from abdominoplasty skin. We used three different decellularization protocols for skin processing, namely, 1M NaCl and sodium dodecyl sulfate (SDS, in ADM1); 2M NaCl and sodium dodecyl sulfate (SDS, in ADM1); and a combination of recombinant trypsin and Triton X-100 (in hADM 3). We assessed the effectiveness of decellularization and ADM's structure by using histochemical and immunochemical staining. In addition, we evaluated the therapeutic potential of novel ADMs in a murine model of wound healing. Furthermore, targeted transcriptomic profiling of genes associated with wound healing was performed. First, we found that all three proposed methods of decellularization effectively removed cellular components from abdominoplasty skin. We showed, however, significant differences in the presence of class I human leukocyte antigen (HLA class I ABC), Talin 1/2, and chondroitin sulfate proteoglycan (NG2). In addition, we found that protocols, when utilized differentially, influenced the preservation of types I, III, IV, and VII collagens. Finally, we showed that abdominoplasty skin-derived ADMs might serve as an effective and safe option for deep wound treatment. More importantly, our novel dressing (ADM1) improves the kinetics of wound closure and scar maturation in the proliferative and remodeling phases of wound healing. In conclusion, we developed a protocol for abdominoplasty skin decellularization suitable for the preparation of biological dressings. We showed that different decellularization methods affect the purity, structure, and therapeutic properties of ADMs.

Agricultural soil samples were collected from the areas where edible plants had been cultivated in southern Poland. The PHE content decreased in proportion to the median value specified in brackets (mg/kg d.m.) as follows: Zn (192) > Pb (47.1) > Cr (19.6) > Cu (18.8) > Ni (9.91) > As (5.73) > Co (4.63) > Sb (0.85) > Tl (0.04) > Cd (0.03) > Hg (0.001) > Se (< LOQ). No PHE concentrations exceeded the permissible levels defined in the Polish law. The PHE solubility (extracted with CaCl2) in the total concentration ranged in the following order: Fe (3.3%) > Cd (2.50%) > Ni (0.75%) > Zn (0.48%) > Cu (0.19%) > Pb (0.10%) > Cr (0.03%). The soil contamination indices revealed moderate contamination with Zn, ranging from uncontaminated to moderately contaminated with Pb, and, practically, no contamination with other PHEs was identified. The ecological risk indices revealed that soils ranged from uncontaminated to slightly contaminated with Zn, Pb, As, Cu, and Ni. The PCA indicated natural sources of origin of Co, Cu, Hg, Sb, Zn, Cr, and Pb, as well as anthropogenic sources of origin of Cd, Ni, As, and Tl. The human health risk assessment (HHRA) for adults and children decreased in the following order of exposure pathways: ingestion > dermal contact > inhalation of soil particles. The total carcinogenic risk values for both adults and children were at the acceptable level under residential (1.62E-05 and 6.39E-05) and recreational scenario (5.41E-06 and 2.46E-05), respectively, as well as for adults in agricultural scenario (1.45E-05). The total non-carcinogenic risk values for both adults and children under residential scenario (1.63E-01 and 4.55E-01, respectively), under recreational scenario (2.88E-01 and 6.69E-01, respectively) and for adults (1.03E-01) under agricultural scenario indicated that adverse health effects were not likely to be observed. Investigated soils were fully suitable for edible plant cultivation.

Implant treatment is safe and predictable with sufficient amount and quality of bone tissue. In case of severely reduced bone tissue after a tooth was lost, augmentation of such tissue is necessary before implant embedment. Retrospective evaluation covered 380 alveolar ridge reconstructions. The study material consisted of human grafts prepared by the Department of Transplantology and Central Tissue Bank, Medical University of Warsaw. Presentation of laboratory procedures in the context of physical parameters of frozen, radiation sterilised, allogeneic corticocancellous material was presented. The preparation process makes it possible to obtain two types of bone material: granules and blocks. Women underwent 164 procedures with the use of bone granules and 61 augmentations with bone blocks. In case of men 122 packages of granules were used as well as 33 bone blocks. Based on the results an evaluation of usability of available allogeneic grafts was performed with reference to planned alveolar ridge augmentation procedures, which they were used for. 1. The opportunity to prepare allogeneic material of different textures allowed selection to meet augmentation requirements while providing biological safety. 2. Allogeneic granules should be used in multi-wall defects, such as a double, closed sinus lift and post-extraction socket augmentation. 3. Owing to their superior mechanical parameters, bone blocks were successfully used in extending the width and height of the alveolar ridge and in open sinus lifts with one-wall or two-wall defects and adequate location of the lamellar bone in a graft prevented substantial graft resorption.

The aim of the work was to evaluate the usefulness of the in vitro multiplication of Biscutella laevigata calamine ecotype for in situ reclamation of post-flotation wastes polluted with Pb and Cd. The experiment was conducted on three steps: (i) plant shoots' production under in vitro condition, (ii) establishment of the material in greenhouse experiment, and finally (iii) field cultivation directly on the mining-waste heap of Olkusz Ore-Bearing Region, Poland. This region is known to be one of the most chemically-degraded area in central Europe. The laboratory-set in vitro analysis enabled to obtain the high-quality plant shoots, which multiply the most effectively (with growth tolerance index 130-150%) on medium containing 5.0µM CdCl2 and 0.5mM Pb(NO3)2. These plants were used for the next two ex vitro experiments. Several biometric and physiological analysis (i.e. of photosystem II activity Fv/Fm and PI, photosynthetic pigment contents) were done to indicate plant physiological status during these experiments. The main novelty of the work was to prove that in vitro-multiplied shoots of B. laevigata - the representative of native flora from Olkusz Ore-Bearing Region - can be successfully implemented in situ for the restoration of these degraded area. Moreover, the addition of sewage sludge as a source of organic compounds significantly improved plants' growth and development what is especially important due to the lack of other legal solutions for the management of the sewage sludge in some countries.

Zinc-lead mining wastes remain largely unvegetated and prone to erosion for many years because of phytotoxic levels of residual heavy metals, low nutrient status and poor physical structure. The optimal solution for these areas is to restore plant cover using species which spontaneously appear on the spoils. These species are adapted to the conditions of tailings, and their establishment will promote further vegetation by increasing soil organic matter and development of a soil system capable of supporting the nutrient and water requirements of plants and microoorganisms. The potential of Dianthus carthusianorum and Biscutella laevigata to stabilize mine spoils was analysed in a three-year pot experiment. Post-flotation wastes accumulated after Zn and Pb recovery from ores, were collected from tailings and used as a substrate for plant growth. Seeds for seedling production were collected from plants growing spontaneously on mine tailings. Prior to the establishment of the three-year pot experiment, the substrate was amended with fertilizer NPK or municipal sewage sludge, supplemented with K2O (SS). Substrate samples were collected for chemical analyses, dehydrogenase and urease activities measurements each year at the end of the growing season. The plants were harvested three years after the amendments. Both tested plant species were equally suitable for revegetation of the tailings. The amendment including both SS and NPK resulted in an increase of Corg, Nt, available P, K, Mg contents, an increase of dehydrogenase (DHA) and urease activities and a decrease in the concentrations of the soluble forms of Zn, Pb and Cd. However, nutrient content, DHA activity and plant biomass were higher with SS than NPK addition. NPK application enhanced the substrate properties after the first growing season, while positive effects of SS use were still observed after three years. A longer-lasting positive effect of SS than NPK application was probably due to the high organic matter content in SS, which was gradually decomposing and releasing nutrients.

The objective of this study was the investigation whether the administration of the elderberry fruit lyophilizate under exposure to cadmium(Cd) and (Pb) lead may protect against some effects of their toxic action in Wistar rats. Rats were fed with diets containing Cd (Cd 0.025 mg/kg b.m.) or Pb (Pb 0.025 mg /kg b.m.) with the addition of the freeze-dried elderberry fruits (BEF) in the amount of 5 %. BEF added to the diet with Cd significantly decreased the activity of AST and ALT compared to the rats fed with the control diet with Cd (C + Cd). Activity of glutathione peroxidase was significantly higher in the blood of rats fed with BEF diet compared with animals fed with BEF + Cd, BEF + Pb, and C + Pb diets. Addition of BEF to the diets with Cd or Pb significantly decreased the uric acid concentration compared to the level of this parameter in the serum of animals fed with control diets containing Cd or Pb. The level of the Cd significantly decreased in the livers of rodents fed with BEF + Cd diet as compared to the concentration of this metal in the livers of rats fed with C + Cd diet. Elderberry fruit lyophilizate did not protect against the increased concentration of Cd or Pb in kidneys and bones of experimental rats; however, it improved the function of livers and kidneys, especially of rats intoxicated with Cd.

The main objective of this study was to determine and describe the lead transfer in the soil-plant-animal system in areas polluted with this metal at varying degrees, with the use of mathematical forecasting methods and data mining tools contained in the Statistica 9.0 software programme. The starting point for the forecasting models comprised results derived from an analysis of different features of soil and plants, collected from 139 locations in an area covering 100km(2) around a lead-zinc ore mining and processing plant ('Boleslaw'), at Bukowno in southern Poland. In addition, the lead content was determined in the tissues and organs of 110 small rodents (mainly mice) caught in the same area. The prediction models, elaborated with the use of classification algorithms, forecasted with high probability the class (range) of pollution in animal tissues and organs with lead, based on various soil and plant properties of the study area. However, prediction models which use multilayer neural networks made it possible to calculate the content of lead (predicted versus measured) in animal tissues and organs with an excellent correlation coefficient.
