RESUMEN

Water kefir is a sparkling, slightly acidic fermented beverage made from sugar, water, and water kefir grains, which are a mixture of yeast and bacteria. These grains produce a variety of fermentation compounds such as lactic acid, acetaldehyde, acetoin, ethanol and carbon dioxide. In this study, a high-throughput sequencing technique was used to characterize the bacterial composition of the original water kefir from which potential probiotics were obtained. We studied the bacterial diversity of both water kefir grains and beverages. DNA was extracted from three replicate samples of both grains and beverages using the Powerlyzer Microbial Kit. The hypervariable V1-V2 region of the bacterial 16S ribosomal RNA gene was amplified to prepare six DNA libraries. Between 1.4M and 2.4M base-pairs were sequenced for the library. In total, 28721971 raw reads were obtained from all the samples. Estimated species richness was higher in kefir beverage samples compared to grain samples. Moreover, a higher level of microbial alpha diversity was observed in the beverage samples. Particularly, the predominant bacteria in beverages were Anaerocolumna and Ralstonia, while in grains Liquorilactobacillus dominated, with lower levels of Leuconostoc and Oenococcus. Although the bacterial diversity in kefir grains was low because only three genera were the most represented, all of them are LAB bacteria with the potential to serve as probiotics in the artificial feeding of bees.

La rotura espontánea de bazo es una complicación infrecuente pero muy grave de la mononucleosis infecciosa. Dada la urgencia con la que suele presentarse tal situación, la esplenectomía suele ser la opción quirúrgica más utilizada. Presentamos el caso de un paciente en el que con el diagnóstico de rotura esplénica espontánea se somete a laparoscopia diagnóstica en la que se consigue la preservación del bazo con buena evolución clínica. A la luz de este paciente, consideramos que, en caso de estabilidad clínica y hematomas subcapsulares de bazo, la laparoscopia con preservación esplénica es una opción viable y con buenos resultados. Spontaneous rupture of the spleen is a rare but very serious complication of infectious mononucleosis. Given the urgency with which such a situation usually presents, splenectomy is usually the most used surgical option. We present the case of a patient diagnosed with spontaneous splenic rupture who underwent diagnostic laparoscopy in which spleen preservation was achieved with good clinical evolution. In the light of this patient, we consider that, in the case of clinical stability and subcapsular haematomas of the spleen, laparoscopy with splenic preservation is a viable option with good results. Bacteria and yeast colonies with different morphologies were isolated fromwater kefir and their antimicrobial activity was evaluated against Paenibacillus larvae andAscosphaera apis. A chemical characterization of kefir was also carried out. Our results con-firmed that bacteria and yeasts were more numerous in kefir grains compared with those in thebeverage. The counts of microorganisms declined, although an important microbial community was still present in kefir after the long storage period. Eleven strains which inhibited bee pathogens were isolated from kefir. Genotypic results demonstrated that these isolates included Lentilactobacillus hilgardii, Lentilactobacillus buchneri and Saccharomyces cerevisiae. Thus, water kefir may be an innovative source of potential probiotic strains for bee nutrition in order to control honeybee diseases.


Resumen Evaluamos la composición microbiana del kéfir de agua durante un ano para determinar si la cantidad y el tipo de microorganismos cambiaban con el tiempo. Se aislaron colonias de bacterias y de levaduras con diferentes morfologías, y su actividad antimicrobiana se evaluó frente a Paenibacillus larvaey Ascosphaeraapis. También se realizó una caracterización química del kéfir. Nuestros resultados confirmaron que las bacterias y las levaduras eran más numerosas en los gránulos de kéfir en comparación con la parte líquida. Los recuentos de microorganismos disminuyeron, aunque una cantidad igualmente importante se encontró en el kéfir después de un año. Se aislaron del kéfir once cepas que inhibieron los mencionados patógenos de abejas. Los resultados genotípicos demostraron que estos aislamientos eran Lentilactobacillus hilgardii, Lentilactobacillus buchneri y Saccharomyces cerevisiae. Por lo tanto, el kéfir de agua podría ser una fuente innovadora de potenciales cepas probióticas para contribuir a la nutrición y sanidad de las abejas.

This work aimed to evaluate the whole-organism and cellular level responses to different combinations of water temperature and salinity of the notothenioid Patagonotothen cornucola at the end of the yolk-sac larval stage. Egg masses of the species were collected in the wild and then maintained at natural water conditions (4 °C and 30 PSU). Newly hatched larvae were placed in aquaria with different combinations of water temperature (4 °C, 12 °C, and 16 °C) and salinity (15 and 30 PSU) during four days before yolk sac absorption. Larvae exposed to 12 °C grew more in length than those exposed to 16 °C, but yolk volume was more reduced in larvae exposed to 16 °C than those exposed to 4 °C and 30 PSU than of 15 PSU. In addition, a higher proportion of larvae exposed to 12 °C and 15 PSU completely absorbed their yolk. Whereas the more tolerant larvae to high temperatures were those exposed to 16 °C and 30 PSU, lipid peroxidation and protein oxidation were highest at natural and at 12 °C and 30 PSU conditions, respectively. The nutritional status (as standardized DNA/RNA index-sRD -) was low in all cases, even at natural conditions (average sRD ~ 1). Our study suggests that, in the context of climate change, the mortality rate of yolk-sac larvae of P. cornucola would not increase due to temperature or salinity stress. However, indirect effects (such as habitat degradation or changes in food availability) would be critical after complete absorption of the yolk.

No estamos solos. Bacterias, hongos, parásitos y virus habitan cualquier parte de nuestro organismo y pueden cumplir funciones biológicas importantes, bien sea benéficas o dañinas. Conocerlos es fundamental para un manejo adecuado. Con las herramientas que utiliza el Grupo de Investigaciones Microbiológicas se pueden identificar la presencia del microorganismo y los marcadores moleculares de interés para la salud, como los relacionados con la resistencia a antibióticos, lo que aporta una información muy útil para el manejo de los pacientes.


We are not alone. Bacteria, fungi, parasites and viruses inhabit any part of our body and can perform important biological functions, either beneficial or harmful. Knowing them is essential for proper management. With the tools used by the Microbiological Research Group, the presence of the microorganism and molecular markers of interest for health, such as those related to antibiotic resistance, can be identified, which provides very useful information for patient management.

A phylogenomic study conducted with different bioinformatic tools such as TYGS, REALPHY and AAI comparisons revealed a high rate of misidentified Streptomyces albus genomes in GenBank. Only 9 of the 18 annotated genomes available in the public database were correctly identified as S. albus species. The pangenome of the nine in silico confirmed S. albus genomes was almost closed. Lignocellulosic agroresidues were a common niche among strains of the S. albus clade while carbohydrate active enzymes (CAZymes) were highly conserved. Relevant enzymes for cellulose degradation such as beta glucosidases belonging to the GH1 family, a GH6 cellulase and a monooxygenase AA10-CBM2 were encoded by all S. albus genomes. Among them, one GH1 glycosidase would be regulated by CebR. However, this regulatory mechanism was not confirmed for other genes related to cellulose degradation. Based on AntiSMASH predictions, the core secondary metabolome of S. albus encompassed a total of 23 biosynthetic gene clusters (BGCs), where 4 were related to common metabolites within Streptomyces genus. Species specific BGCs included those related to pseudouridimycin and xantholipin. Additionally, four BGCs encoded putative derivatives of ibomycin, the lasso peptide SSV-2086, the lanthipeptide SapB and the terpene isorenieratene. Known metabolites could not be assigned to ten BGCs and three clusters did not match with any previously described BGC. The core genome of S. albus retrieved from nine closely related genomes revealed a high potential for the discovery of novel bioactive metabolites and underexplored regulatory genomic elements related to lignocellulose deconstruction.

The selection of fast-growing and high-yield-producing strains is required to satisfy the market demand on fungal food supplements. To that aim, three strains deposited in our collection as G. lucidum and G. oregonense were screened for polysaccharide production and biomass yield. Ganoderma strains deposited as G. lucidum were identified as G. sessile and G. lingzhi by nuc rDNA internal transcribed spacer ITS1-5.8S-ITS2 (ITS) and translation elongation factor 1-α (TEF1-α) phylogenies. The identity of G. oregonense was confirmed by molecular phylogeny and biogeography. Additionally, mycelial antagonism confirmed species differentiation, and strains were further distinguished by morphology and protein profiles. Biomass and polysaccharide yields of G. sessile were clearly different from those of G. lingzhi and G. oregonense in both liquid culture and solid-state fermentation. The maximum polysaccharide yield (4.52 ± 0.83 g L-1) for G. sessile was obtained from submerged cultures at day 9. G. sessile also achieved the highest linear growth in lignocellulosic solid substrates. Consequently, basidiomata were successfully obtained by solid-state fermentation in polypropylene bags, whereas G. lingzhi and G. oregonense mushrooms were not produced in artificial solid substrates. G. sessile, a species frequently collected in America, showed to be a promising polysaccharide producer for the manufacture of dietary supplements.

The Argentine hake, Merluccius hubbsi, is one of the most important fishing species in the Argentine Sea due to its great abundance and high-quality meat. The study of the nutritional condition is widely used to determine the physiological state of the fish larvae and to estimate their survival possibilities. The larval nutritional condition reflects the environmental conditions to which they have been exposed and represents a useful instrument to determine favourable nursery areas. It also provides tools for the comprehensive management of a population subjected to fishing exploitation. This study aimed to determine potential differences in the nutritional condition of M. hubbsi larvae from the two fishing stocks (Northern and Southern) of the species. The authors assessed the nutritional condition of larvae captured during the 2012 main reproductive peak in the nursery areas of each population. Two different methodologies were applied: a morphometrical approach, by recording five morphometric variables, and a biochemical technique, employing the RNA/DNA index (RDs ) and its derived index of growth performance. The morphometrical indexes did not show differences in the larval condition between both stocks. Nonetheless, the RDs index did detect differences in the nutritional condition of larvae from different stocks. The RDs index of larvae in pre-flexion and flexion stages showed significant differences between stocks, indicating that these stages are more susceptible to starvation. The results suggest that the biochemical indexes prove to be more sensitive than the morphometric indexes to detect slight differences in hake larvae nutritional condition. The scope and limitations of these techniques for the analysis of the nutritional condition of larvae are discussed.

The presence, biosynthesis and functional role of sterols in the green microalga Haematococcus pluvialis remain poorly understood. In this work we studied the effect of high-light (HL) stress on sterol synthesis in H. pluvialis UTEX 2505 cells. HL stress induced the synthesis of sterols in parallel with that of triacylglycerides (TAG), giving rise to the synthesis of cholesterol over that of phytosterols. Blockage of the carotenogenic 1-deoxy-D-xylulose 5-phosphate (MEP) pathway is shown to be involved in HL-induced sterol synthesis. In addition, high irradiance exposure induced MEP- and fatty acid (FA)-biosynthetic transcripts. The pharmacological inhibition of these pathways suggests a possible feedback regulation of sterol and FA homeostasis. Finally, both lipid classes proved crucial to the adequate photosynthetic performance of H. pluvialis grown under HL intensity stress. Our findings reveal new insights into H. pluvialis lipid metabolism that contribute to the development of value-added bioproducts from microalgae.

The Argentine hake Merluccius hubbsi is an important fishery resource of the Southwestern Atlantic Ocean and it is also a potential species for cultivation. In this work, the digestive system development in field-caught hake larvae was studied using histological and histochemical approaches. The digestive tract of larvae was divided into: oropharyngeal cavity (OPC), esophagus, stomach (that develops in the preflexion stage), and intestine. The annexed digestive glands consisted of the liver and the exocrine pancreas. At the beginning of the preflexion stage, teeth were developed in the OPC. There were mucous cells in the esophagus secreting different glycoconjugates from hatching. The enterocytes in the posterior intestine exhibited supranuclear vesicles associated with protein absorption. Mucous cells were observed in the posterior intestine in the preflexion stage and, in the anterior region, ending the flexion stage. Each type of glycoconjugates has a specific role. Acidic mucins lubricate and protect from mechanical damage, sialomucines protect from bacterial infections and neutral mucins regulate the acidity of mucus secretion, protect against abrasion and participate in the formation of the chyme, indicating a pregastric digestion. The liver was present since hatching with pancreatic tissue inside and increased in size acquiring the typical structure with hepatocyte cords, sinusoids, vacuoles, and hepatic duct. The hepatocytes vacuolization increased with larval development. The pancreas became extra-hepatic, with basophilic acinar cells and acidophilic zymogen granules. Throughout the ontogeny, the increased structural and functional complexity of the digestive system reflected the transition to exogenous feeding and nutritional increasing needs.

During the early ontogeny of fish larvae, the accurate development of the visual system plays a key role, because it is involved in locating food, orientation, selection of favorable habitat, and evasion of predators. The structure of the eye of the fish is typical of vertebrates, with some modifications related to the aquatic environment. In the present work, we describe the development of the larval eye of Engraulis anchoita for the first time. Larvae were collected at the Permanent Station of Environmental Studies (EPEA) in coastal waters of the Southwestern Atlantic Ocean during research cruises in 2015 and 2016. We describe the histology of the retina layers, determine the beginning of the functionality of the eye, and discuss a possible synchronization with the development of the digestive tract. This study provides information about the biology of E. anchoita, the most abundant fish species in the southwestern Atlantic Ocean. Also, recent studies have shown responses of the retina and other tissues to the increase in environmental acidity. Therefore, results of this study are also discussed with respect to the possible effect of acidification on the larvae of this species. The continuity of the time series developed at the EPEA will allow monitoring the effect of long-term environmental and biological variables on the early ontogeny of anchovy in the context of climate change. The high commercial fishing potential of E. anchoita due to its high abundance, as well as its essential role in the trophic web of other commercially valuable fishing resources of Argentina, reinforce the need to continue deepening knowledge about this species. Research highlights: Eyes of Engraulis anchoita larvae are functional from early larval stages. At hatching, the retina is formed by only few layers from which the other layers differentiates during ontogeny. Focal distance increases with larval growth.

Understanding the genetic basis of cold tolerance is a key step towards obtaining new and improved crop varieties. Current geographical distribution of durum wheat in Argentina exposes the plants to frost damage when spikes have already emerged. Biochemical pathways involved in cold tolerance are known to be early activated at above freezing temperatures. In this study we reported the transcriptome of CBW0101 spring durum wheat by merging data from untreated control and cold (5 °C) treated plant samples at reproductive stage. A total of 128,804 unigenes were predicted. Near 62% of the unigenes were annotated in at least one database. In total 876 unigenes were differentially expressed (DEGs), 562 were up-regulated and 314 down-regulated in treated samples. DEGs are involved in many critical processes including, photosynthetic activity, lipid and carbohydrate synthesis and accumulation of amino acids and seed proteins. Twenty-eight transcription factors (TFs) belonging to 14 families resulted differentially expressed from which eight families comprised of only TFs induced by cold. We also found 31 differentially expressed Long non-coding RNAs (lncRNAs), most of them up-regulated in treated plants. Two of these lncRNAs could operate via microRNAs (miRNAs) target mimic. Our results suggest a reprogramming of expression patterns in CBW0101 that affects a number of genes that is closer to the number reported in winter genotypes. These observations could partially explain its moderate tolerance (low proportion of frost-damaged spikes) when exposed to freezing days in the field.

Fatty acid desaturases (FADs) catalyze the introduction of a double bond into acyl chains. Two FAD groups have been identified in plants: acyl-acyl carrier proteins (ACPs) and acyl-lipid or membrane-bound FAD. The former catalyze the conversion of 18:0 to 18:1 and to date have only been identified in plants. The latter are found in eukaryotes and bacteria and are responsible for multiple desaturations. In this study, we identified 82 desaturase gene and protein sequences from 10 grass species deposited in GenBank that were analyzed using bioinformatic approaches. Subcellular localization predictions of desaturase family revealed their localization in plasma membranes, chloroplasts, endoplasmic reticula, and mitochondria. The in silico mapping showed multiple chromosomal locations in most species. Furthermore, the presence of the characteristic histidine domains, the predicted motifs, and the finding of transmembrane regions strongly support the protein functionality. The identification of putative regulatory sites in the promotor and the expression profiles revealed the wide range of pathways in which fatty acid desaturases are involved. This study is an updated survey on desaturases of grasses that provides a comprehensive insight into diversity and evolution. This characterization is a necessary first step before considering these genes as candidates for new biotechnological approaches.

Los ceparios o colecciones de microorganismos son fuentes de recursos genéticos cuyo propósito es la preservación de la diversidad biológica, garantizando su disponibilidad para actividades de docencia, investigación y comerciales. En este trabajo se verificó la viabilidad, pureza y características biológicas de las bacterias que conforman el cepario del Instituto de Ciencias Biológicas de la Universidad de Ciencias y Artes de Chiapas, y se organizó y estructuró la información obtenida en un portal virtual, para propiciar la cooperación académica. El cepario cuenta con 33 microorganismos, la mayoría del género Streptococcus y Escherichia (45,1 y 21,2%, respectivamente). Del primer género, se confirmó la identificación de S. pyogenes (40%), exhibiendo la mayoría genes que codifican para DNAsas. Con respecto al segundo género, un 58,3% de las bacterias fueron confirmadas taxonómicamente como E. coli. De esta especie, la colección cuenta con las cepas prototipo causantes de diarrea y que han preservado sus rasgos genéticos por más de cinco años. Dicho acervo ha impulsado actividades de docencia e investigación, a nivel local e internacional. Es importante que los ceparios sean fuentes sustentables de recursos biológicos, para la adquisición y suministro de especies bacterianas, con la finalidad de fomentar la interacción con la comunidad académica.


Strain collections or bacterial culture collections are genetic resources whose purpose is the preservation of biological diversity, ensuring their availability for teaching, research and trade activities. In this work viability, purity and biological characteristics of bacteria from the bacterial collection of the Institute of Biological Sciences, University of Science and Arts of Chiapas were studied. Information was structured and organized in a virtual site, to promote academic cooperation. The strain bank includes 33 microorganisms, most of the genus Streptococcus and Escherichia (45.1 and 21.2%, respectively). For Streptococcus, the identification of S. pyogenes (40%) was confirmed, by determination of most DNAses encoding genes. For Escherichia 58.3% were taxonomically confirmed as E. coli. For this species, the collection includes typical strains that produce diarrhea and their genetic traits have been preserved for more than five years. This bacterial culture collection has stimulated teaching and research activities at local and international levels. Strain collections are important sources of biological material which can provide bacterial species, in order to encourage interaction with the academic community.

Morphometrical and histological techniques were employed to characterize Pagrus pagrus larvae nutritional condition. Larvae were reared in laboratory under controlled conditions with the main objective of testing whether these methodologies allowed finding differences between larvae from different feeding treatments. Once yolk was consumed (three days after hatching) larvae were assigned to a feeding treatment: starved during the whole experiment; delayed feeding, starved during three days; fed during the entire experiment. Algae (Nannochloropsis oculata) and rotifers (Brachionus plicatilis) were provided to larvae for feed treatments. Larvae were fixed daily; for morphometrical purposes in 5% formaldehyde solution, and in Bouin for histological sections. Results herein obtained showed that both methodologies are sensitive enough to distinguish larvae characterized by different nutritional condition states obtained from the feeding treatments. Consequently, these methodologies could be employed in wild red porgy larvae in order to asses their nutritional condition. These techniques could also be employed to check larval quality obtained with aquaculture purposes to estimate the effects of changes in rearing protocols or kind of food supply and thus, to guaranty a higher survival of early developmental stages of reared larvae.


Con el objeto de determinar si las técnicas morfométricas e histológicas permiten diferenciar larvas en estado de inanición de las sometidas a tratamientos de alimentación, ejemplares de Pagrus pagrus fueron criados en laboratorio bajo condiciones controladas. Una vez que el vitelo fue consumido (tres días luego de la eclosión) las larvas fueron sometidas a diferentes tratamientos de alimentación: completamente privadas de alimento, privadas de alimento durante tres días y luego alimentadas, y alimentadas durante todo el experimento. Algas (Nannochloropsis oculata) y rotíferos (Brachionus plicatilis) fueron proporcionados a las larvas de los tratamientos de alimentación. Diariamente se fijaron ejemplares de los tres tratamientos en formol 5% para estudios morfométricos y en Bouin para estudios histológicos. Los resultados obtenidos dan evidencias de que las metodologías presentadas son suficientemente sensibles para determinar diferencias en la condición de larvas sometidas a los tratamientos de alimentación. Por lo tanto, podrían ser empleadas para determinar la condición nutricional de las larvas de besugo recolectadas en el mar. Por otra parte, éstas metodologías podrían ser empleadas para determinar la calidad de larvas producidas en acuicultura y evaluar los efectos de modificaciones introducidas en los protocolos de crianza o en el tipo de alimento suministrado con el objeto de mejorar la supervivencia de los estadios tempranos de desarrollo.

INTRODUCTION: Emotional stimuli elicit changes in the electroencephalographic (EEG) activity of several brain structures. Prefrontal cortex is involved in the processing of emotional stimuli and executive functions. The correlation analysis of EEG provides information about the functional coupling between areas. It is reasonable to expect that emotional activation will modify prefrontal coupling during the performance of executive tasks such as Tower of Hanoi or Wisconsin Card Sorting Test (WCST). AIM: Determine whether the prefrontal EEG correlation during the performance of the Tower of Hanoi and WCST is affected by previous exposure to videos with sexual or aggressive content. MAIN OUTCOME MEASURES: Prefrontal EEG coupling was determined by the Pearson correlation. Valence and general arousal were evaluated by the Self-Assessment Manikin Scale and sexual arousal with a Sexual Arousal Scale. Computerized versions of the Towers of Hanoi and WCST provided data on prefrontal executive functions. METHODS: EEG from the left and right prefrontal zones was recorded during the performance of the Tower of Hanoi and WCST immediately after the subjects were exposed to one of the videos (neutral, aggressive, and erotic). RESULTS: There was no difference between videos in the task performance parameters. Only the erotic video produced an increased prefrontal coupling in the slow bands (delta and theta) during the performance of the Tower of Hanoi, whereas a decreased coupling in the delta, theta, and alpha bands was observed during the WCST. CONCLUSIONS: Prefrontal coupling was changed after exposure to the erotic video, and it is likely that enhanced sexual arousal was the main cause of this change. The correlation patterns obtained could be associated with particular cognitive strategies or to functional adaptations while being sexually aroused. The results of this study may contribute to an understanding of the central nervous mechanisms underlying the cognitive effects of sexual arousal.

Three rusts, Puccinia nassellae, Uromyces pencanus, and Puccinia graminella, are being studied as potential biological control agents for Nassella neesiana (Chilean needle grass) in Australia and New Zealand. An understanding of the life cycle of a pathogen is desirable before its release as a biocontrol agent is considered, to enable the assessment of the risks involved in such a release. Field observations and experiments have been carried out to elucidate the life cycles of these three pathogens. Puccinia nassellae cycles as urediniospores and produces dormant teliospores. Dormancy of teliospores has been broken through manipulation in the laboratory, but resulting basidiospores have failed to infect N. neesiana plants under the conditions tested. Uromyces pencanus cycles as urediniospores and its telia appear to have lost the capacity to produce basidiospores. Aecia have been reported for this rust in the literature. However, evidence is provided that these aecia in fact belong to the life cycle of P. graminella. Puccinia graminella produces only aecia and telia. The aeciospores have been shown to be repetitive (aecidioid urediniospores). Teliospores germinate directly without a dormant phase, but resulting basidiospores failed to infect N. neesiana plants under the conditions tested. The role of teliospores in the life cycle of all three rusts remains unknown. Although the autoecious nature of their life cycles has not been proven experimentally, neither is there any evidence that they are heteroecious. Practical and theoretical implications of these findings are discussed.

OBJECTIVE: To assess the association between recent cigarette smoking (CS) in female and male partners and assisted reproduction technology (ART) outcomes. DESIGN: Cohort prospective study. SETTING: University ART program in Chile. PATIENT(S): One hundred sixty-six couples seeking pregnancy through ART. INTERVENTION(S): Follicular fluid (FF) and serum cotinine concentrations were measured in female partners. Self-reported CS data were collected through personal interviews. MAIN OUTCOME MEASURE(S): The association between female recent smoking, assessed by FF and serum cotinine concentrations, and ART outcomes, such as number of ova retrieved and implantation rates, and the association between self-reported male recent smoking and live birth rates. RESULT(S): A significant age-adjusted association between increased FF cotinine level and decreased number of ova retrieved was found. The male partner's smoking habit significantly decreased the live birth rate from 21.1% to 7.8%. Serum cotinine concentrations paralleled those of FF. CONCLUSION(S): The hypothesis of a detrimental effect of recent female smoking over implantation rates is rejected. However, recent male smoking is associated with significantly decreased live birth rates even after adjusting for confounders. Female recent smoking was significantly associated with decreased number of retrieved ova.
