RESUMEN

The aim of study was to compare the content of phenolic acids and flavonoids in two cultivars of Lavandula angustifolia: 'Blue River' and 'Ellagance Purple', including flowers and leafy stalks. Total phenolics and total flavonoids contents were determined by UV-Vis spectroscopy. The contents of total phenolics in leafy stalks (3.71-4.06 mg g-1 d.m.) were higher than in flowers (1.13-1.14 mg g-1 d.m.). Similarly, higher total contents of flavonoids were determined in leafy stalks (3.41-3.51 mg g-1 d.m.), as compared with flowers (0.86-0.91 mg g-1 d.m.). Phenolic acids and flavonoids were identified and quantified using HPLC and UPLC methods. Three phenolic acids were determined: rosmarinic, ferulic and caffeic acid. Lavender extracts contained also flavonoids from group of apigenin, luteolin and quercetin. Higher amounts of luteolin diglucuronide and luteolin glucuronide were found in leafy stalks in comparison to flowers. Obtained results indicate that leafy stalks of lavender can be also valuable source of antioxidant compounds.

AIMS: Ileocaecal lymph nodes (ICLNs) of pigs, the key immune inductive site for bacterial systemic invasion, were examined in this study with emphasis on viable and cultivable bacteria. METHODS AND RESULTS: Asymptomatic and pathologically altered ICLNs of slaughter pigs (n = 16) with hyperplasia, purulence or granulomatous formations were aerobically and anaerobically cultivated. In total, 209 isolates were collected and the near full-length 16S rRNA gene from each isolate was sequenced. Taxonomic classification revealed that 68% of the isolates belonged to Proteobacteria, 27% to Firmicutes and 5% to Actinobacteria. Purulent and granulomatous ICLNs generally tended to contain more Proteobacteria than asymptomatic and enlarged ICLNs (P = 0·061). The isolates could be assigned to 25 species belonging to 17 genera including Escherichia, Carnobacterium, Lactobacillus, Staphylococcus and Acinetobacter. Furthermore, pathogens such as Streptococcus suis and Salmonella enterica were detected. The most abundant isolate (57%) was most similar (>99%) to Escherichia coli. Random amplified polymorphic DNA (RAPD) profiling revealed a high genetic diversity among E. coli isolates and 24% of these isolates were positive for at least one gene associated with enterohemorrhagic disease (eae, fliC, stx1 or hlyA). Compared with a recently published DNA-based high-throughput sequencing data set including the same ICLNs, 4% of species detected were cultivable. CONCLUSIONS: The presence of viable, commensal and pathogenic bacterial phylotypes could be proven in ICLNs with Proteobacteria being dominant. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study that broadly characterizes viable bacteria from ICLNs of pigs. The presence of bacteria in lymph nodes of farm animals has practical relevance for host colonization and possible chronic infection. It is also of great interest for basic research investigating translocation of bacteria from the gastrointestinal tract to ICLNs.

Development of ovarian follicles is controlled at the molecular level by several gene products whose precise expression leads to regression or ovulation of follicles. MicroRNAs (miRNAs) are small noncoding RNAs that regulate gene expression through sequence-specific base pairing with target messenger RNAs (mRNAs) causing translation repression or mRNA degradation. The aim of this study was to identify miRNAs expressed in theca and/or granulosa layers and their putative target genes/pathways that are involved in bovine ovarian follicle development. By using miRCURY microarray (Exiqon) we identified 14 and 49 differentially expressed miRNAs (P < 0.01) between dominant and subordinate follicles in theca and granulosa cells, respectively. The expression levels of four selected miRNAs were confirmed by qRT-PCR. To identify target prediction and pathways of differentially expressed miRNAs we used Union of Genes option in DIANA miRPath v.2.0 software. The predicted targets for these miRNAs were enriched for pathways involving oocyte meiosis, Wnt, TGF-beta, ErbB, insulin, P13K-Akt, and MAPK signaling pathways. This study identified differentially expressed miRNAs in the theca and granulosa cells of dominant and subordinate follicles and implicates them in having important roles in regulating known molecular pathways that determine the fate of ovarian follicle development.

The literature concerning the issue of canine sex pheromones includes reports presenting completely conflicting opinions about the chemical composition of the canine urine in the context of semiochemical communication. At present, the predominant report cited by many different authors is the article published in Science in 1979 by Goodwin at al., presenting methyl p-hydroxybenzoate (methyl paraben) as the main canine sex pheromone. While it has been proved that pure methyl paraben lacks semiochemical activity as do commercially available products containing this substance (Eau D'Estrus, Synbiotics, USA), in view of the conflicting published reports the aim of this study was to revaluate using modern techniques the presence of methyl p-hydroxybenzoate in canine urine during different phases of the ovarian cycle. Ten female dogs of different breeds were used. Urine samples from bitches collected during various stages of the ovarian cycle were examined with using the SPME and GC/MS methods. Methyl paraben was not detected in any of the samples. In conclusion, because of the lack of methyl-p-hydroxybenzoate in the samples examined, the present study confirmed negative opinions on the possibility of this substance playing a crucial role in semiochemical communication during reproduction in dogs (Canis familiaris).

The aim of this study was to evaluate the influence of the antibiotic treatment, including the mode of drugs administration, on bitches' attractiveness to the stud dogs during mating. Moreover, we tried to estimate the possibility of aversive effect of the drug vehicle on the male behavior. In experiment I, four bitches in oestrus without antibiotic treatment (group A), four bitches treated with intravaginal antibiotic (group B) and four bitches treated with intramuscular antibiotic (group C) were presented to four stud dogs. In experiment II, bitches in oestrus (n = 5) were presented to the males (n = 2) before and after the application to the females' vulva the antibiotic carrier--Miglyol 840 (Sasol, Germany). In both experiments the presence of the typical sexual behavior of the males (sniffing, licking the vulva and anal region, mating attempts) was evaluated. In experiment III the reaction of the males to the samples containing oestrual discharge from the bitches untreated and treated with antibiotics was evaluated. In the last part of study the aversion reaction to the samples containing antibiotic and the antibiotic carrier was evaluated. The results of experiments showed that females treated with the antibiotics were less attractive to males than untreated females, regardless of the method of administration. We did not observe adverse effect of the antibiotic carrier but samples from the bitches treated with antibiotics were significantly less attractive to the males. We concluded that the reason for reduced attractiveness of the bitches in oestrus after antibiotic treatment was the changes in semiochemical signal emitted by treated females as a consequence of elimination of the vaginal bacterial flora, which seems to be involved in creation of the typical, recognizable by the stud dogs, oestrual signal but also by the possible covering effect of used drugs.

The aim of the study was comparing the chemical composition of the essential oils from five varieties of lavender (Lavandula angustifolia L.), and its biological activity against two pathogenic bacteria, Staphylococcus aureus and Pseudomonas aeruginosa. In the comparison we included the following varieties: 'Munstead', 'Munstead Strain', 'Lavender Lady', 'Ellegance Purple' and 'Blue River'. Selected varieties of lavender (L. angustifolia) are characterised by exactly the same main compounds with only variations in the percentage of content. The primary components of the essential oils were: linalool (23.9-15.8%), linalyl anthranilate (12.3-1.6%), 1-terpinen-4-ol (9.7-5.5%), p-menth-1-en-8-ol (7.9-4.0%) and linalool oxide (4.7-1.1%). From the essential oils that were tested, the 'Blue River' and 'Munstead' varieties have the greatest antibacterial activity against S. aureus and P. aeruginosa.
