RESUMEN

Atomic resolution X-ray holography can be realized by using the atoms of the sample as inside sources or inside detectors. However, until now there were only very few experiments in which the atoms played the role of inside sources. The reason is twofold: (i) technically, inside-detector experiments are much easier and faster; (ii) by using atoms as inside detectors one can measure holograms at many energies on the same sample, which helps the reconstruction. This paper shows that, using new technical developments, inside-source holograms can be taken much faster than inside-detector holograms and, by applying a sophisticated evaluation method, high-quality reconstruction from a single-energy hologram can also be obtained.

RESUMEN

Kossel lines are formed when radiation from point x-ray sources inside a single crystal are diffracted by the crystal itself. In principle, Kossel line patterns contain full information on the crystalline structure: phase and magnitude of the structure factors. The phase is coded into the profile of the lines. Although this was known for a long time, experimental realization has not been presented. In this work we demonstrate experimentally that phases can be directly determined from the profile of the Kossel lines. These measurements are interesting not only theoretically, but they would facilitate structure solution of samples within extreme conditions, such as high pressure, high and low temperatures, high magnetic fields and extremely short times. The parallel measurement of many diffraction lines on a stationary sample will allow a more efficient use of the new generation of x-ray sources the X-ray free electron lasers (XFELs).

RESUMEN

Kossel line patterns contain information on the crystalline structure, such as the magnitude and the phase of Bragg reflections. For technical reasons, most of these patterns are obtained using electron beam excitation, which leads to surface sensitivity that limits the spatial extent of the structural information. To obtain the atomic structure in bulk volumes, X-rays should be used as the excitation radiation. However, there are technical problems, such as the need for high resolution, low noise, large dynamic range, photon counting, two-dimensional pixel detectors and the small spot size of the exciting beam, which have prevented the widespread use of Kossel pattern analysis. Here, an experimental setup is described, which can be used for the measurement of Kossel patterns in a reasonable time and with high resolution to recover structural information.

Single molecule imaging experiments at future X-ray free electron laser sources will provide large number of random 3D oriented diffraction patterns with low counting statistics. Grouping of this vast amount of data into classes of similar orientations and averaging must be performed before their orientation and structure reconstruction can take place. Classification algorithms performing all-pair pattern comparisons scale badly with the number of patterns in terms of their computing requirements, which presents a problem in case of improving resolution and decreasing signal to noise ratios. We describe an algorithm performing significantly less pattern comparisons and render classification possible in such cases. The invariance of patterns against rotation of the object about the primary beam axis is also exploited to decrease the number of classes and improve the quality of class averages. This work is the first, which demonstrates that it is possible to classify a dataset with realistic target parameters: 10 keV photon energy, 10(12) photons/pulse, 100 x 100 nm2 focusing, 538 kDa protein, 2.4 A resolution, 10(6) patterns, approximately 3 x 10(4) classes, <1 degree misorientation within classes. The effects of molecular symmetry and its consequences on classification are also analyzed.

RESUMEN

The very intense and short pulses of future X-ray free electron lasers may allow the atomic resolution imaging of small, non-periodic objects. Preliminary estimates show that images obtained from single pulses do not contain statistically enough photons to allow successful reconstruction. Therefore multiple exposures of randomly oriented identical replicas have to be taken and the individual images have to be classified according to the object's orientation. The classification has been analytically treated by Huldt et al. [Huldt, G., Szoke, A., Hajdu, J., 2003. J. Struct. Biol. 144, 219.]. In this paper we extend the analytical results with numerical model calculations. This allows us to simulate realistic situations, which we will face in real experiments. We find significant deviations from the analytical expectations, even in the ideal case of spherical particles with random atomic distributions. We introduce a new norm for the individual scattering patterns and describe a criterion to select images belonging to similar orientation, which makes the classification more reliable in practice. We also discuss the effects of particle shape and size, partial orientational ordering, the measurement's resolution and the charge error caused by the Coulomb explosion.

RESUMEN

Inverse x-ray fluorescence holography was used to explore the local atomic order of a nearly perfect quasicrystal with composition Al70. 4Pd21Mn8.6. We have demonstrated the possibility of direct 3D imaging of the atomic decoration in a quasicrystal. We have obtained the average 3D environment of selected coordination shells around the Mn atoms. These results open the way to obtaining further and more complete information about the various coordination shells in complex materials by measuring multiple energy x-ray holograms at different sites.

RESUMEN

Twelve teenaged boys referred by a juvenile court clinic because of delinquent behavior and academic failure were examined by an experienced team of pediatricians, neurologists, psychologists, social workers, nurses, and occupational, physical, recreational, speech and hearing therapists. Two boys came from troubled families with long histories of legal problems, had behavior problems antedating their school difficulties and had no evidence of perceptual handicaps. The remaining ten were the family blacksheep whose academic problems began early and whose behavior decompensated later. They all had perceptual handicaps and both groups of boys had intense psychological reactions to their school difficulties. The learning disabled require a flexible team approach in school together with individual, group and family therapy in order to achieve success.
