RESUMEN

The possibility of long-term changes in the innate characteristics of the behavior of mice of different genotypes (C57Bl/6 and BALB/c) after active immunization with dopamine-BSA conjugates was studied. These experiments revealed significant differences in the effects of anti-dopamine antibody on behavioral responses in an open field and neurotransmitter contents of brain structures in mice of different lines. Immunization with the dopamine-protein conjugate led to increases in the functional activity of the corresponding brain receptors, to different extents in mice of different lines.

RESUMEN

A possibility of the long-term modification of inborn behavioral features of different mice genotypes (C57B1/6 and BALB/c) by active immunization with dopamine-bovine serum albumin conjugate was investigated. Significant interstrain differences were found in the effects of dopamine antibodies on the open-field behavior and the content of neurotransmitters in the brain cortex and striatum. It was shown that the active immunization of mice to dopamine produces an increase in the functional activity of brain dopamine receptors. The extent, to which this increase is pronounced, is genotype-dependent.

RESUMEN

Effects of active immunization with the serotonin-protein conjugate on open-field behavior, learning and retention of the conditioned passive avoidance response, neurotransmitter contents in brain structures, and functional activity of central serotonin receptors were studied on C57Bl/6 and BALB/c mice. Serotonin antibodies produced opposite effects on the studied parameters in genetically different mouse strains.

RESUMEN

Single i.p. doses of anti-dopamine antibodies were given to C57Bl/6 mice. This resulted in inhibition of motor activity in a large proportion of the animals in the "open field" test, which lasted five days. Hyperalgesia, detected 1.5 h and 1 day after doses of antibody, was replaced by analgesia on day 5. There was a sharp reduction in the levels of dopamine and its metabolites in the cerebral cortex at 1 and 5 days; the serotonin level was increased 1 day after doses of antibody, and was significantly decreased at 5 days. There was no effect on cells of the immune system. The possible mechanisms of the neurotropic action of these antibodies are discussed.

Dopamine antibodies (AB) were singularly injected to C57B1/6 mice intraperitoneally. The locomotor activity in the open field was suppressed for 5 days in the majority of the animals. Hyperalgesia revealed 1.5 h and 1 day after the AB injection changed for analgesia on the 5th day. A sharp reduction of the brain level of dopamine and its metabolite was revealed 1 and 5 days after the AB injection, the serotonin content was increased within 1 day and decreased within 5 days after the injection. No action of the AB on cells of the immune system was observed. The possible mechanisms of the AB neurotropic action are discussed.

RESUMEN

Mice of C57BL/6 strain were singly injected intraperitoneally with antibodies (AB) to serotonin (5-HT). The "open-field" testing in different periods after the AB injection revealed a depression of behavior within 1.5 h which changed for activation within 1 day and, again, depression within 5 days after the injection. The analysis of neurotransmitter content in the sensorimotor cortex and hypothalamus revealed increased levels of serotonin and, especially, dopamine in the cortex within 1 day. The cortex serotonin level within 5 days was also increased. The possible mechanisms are discussed of neurotropic action of AB to 5-HT.

RESUMEN

A study was made of the effect of zixorin, an inductor of liver cytochrome P-450, on the intensity of the immediate type allergic reaction--active food anaphylaxis in response to ovalbumin in guinea-pigs. Intraperitoneal injection of zixorin in a dose of 50 mg/kh for 3 days before administration of the antigen resolution dose was found to lower 2,3-fold the level of lethal outcomes due to the anaphylactic shock. It is suggested that zixorin may be used in multimodality treatment of food allergy.

Activity of cytochrome P-450-dependent monooxygenase system is significantly lower in hepatoma H-2-73 and Lewis lung carcinoma-bearing mice as compared to that in control animals. An inhibition of the cytochrome P-450 system was observed after the injection of ftorafur. Treatment of the mice with perfluorodecalin markedly increased the antineoplastic activity of ftorafur determined by a loss of the leucocytes and of the weight hepatoma H-2-73- and Lewis lung carcinoma resistant to ftorafur alone.

RESUMEN

The effect of adamanthylamides of 1,3-diphenylptrazol-4-carbonic acids on the system of the liver microsomal monoxygenases was studied. A potent induction of the liver cytochrome P-450 by 1,3-diphenylpyrazol-5-methyl-4-carbonic acid, N-methyl-N-(adamant-1-il)amide was found. As a consequence of the induction of the liver cytochrome P-450, the compound reduced by 40% the degree of the delayed type hypersensitivity in mice.

The effect of zixoryn, a hepatic inductor of cytochrome P-450, on the pharmacokinetics of antipyrine in intact and sensibilized guinea-pigs was studied. It was found that sensibilization of albumin increased the half-time (T1/2), AUC and decreased the total body clearance (Clt) of antipyrine and the renal clearance (Clr) of metabolites in urine. The administration of zixoryn in sensibilized animals decreased T1/2 and AUC and increased the clearance of antipyrine and its metabolites.

Antipyrine oxidation was studied in C57BL/6 and BALB/c inbred mice. It was found that C57BL/6 are weak oxidant but BALB/c are strong oxidants of antipyrine. Animals F1 hybrids inherited the high capacity of antipyrine oxidation.

RESUMEN

The effect of a single intraperitoneal injection of perfluorodecaline (0.5 ml) and perfluorotributylamine on the cytochrome P-450-dependent monooxygenase system of male hybrid mouse (CBA X C57Bl) liver was studied. It was shown that perfluorodecaline is a potent inducer of the cytochrome P-450 system in the liver. The sharp increase of the cytochrome P-450 content and of the enzymatic activity was observed within 5 months. The biological effect of perfluorotributylamine on the monooxygenase system of the liver was much less pronounced. A slight inhibition of the cytochrome P-450 system was observed at short times after the injection followed by its slight stimulation after 2 weeks; 30 days thereafter all the parameters under study returned to control values.

Intraperitoneal injection of hydrocortisone in single daily doses from 0.1 to 25.0 mg/kg, for 3 days was followed by reduced activity of liver microsomal monooxygenases and decreased content of cytochrome P-450 and b5. Hydrocortisone diminished the activity of the liver cytochrome P-450 system in vivo that significantly potentiated hexenal sleep. The hydrocortisone-inhibited activity of the liver chromosome P-450 system was followed by intensified food anaphylaxis in guinea-pigs.
