RESUMEN

The problem of improving the efficiency of implantological treatment of patients with osteoporosis (OP) today remains relevant due to the high frequency of postoperative complications. Among the main factors affecting the success of dental implant treatment, the nature of the implant surface is essential. Patients with adentia and OP especially relevant in use of dental implants with optimized surface, air conditioned component influencing bone remodelling. OBJECTIVE: Evaluation of the effectiveness of dental implants with a conditioned surface based on sodium hydroxide in men with osteoporosis. MATERIAL AND METHODS: The study included 60 men 54-65 years of age in need of dental implants of which 40 were diagnosed with OP. Patients with OP were divided into groups: I - 20 men without antiresorptive therapy; II - 20 men taking ibandronate 150 mg monthly, calcium and vitamin D (1000 mg/800 IU daily). The control group consisted of 20 men 54-65 years without OP. Microarchitectonics of the mandible was studied using cone-beam computed tomography and histomorphometry. Implants with bioactive surface based on sodium hydroxide (NaOH) were used. RESULTS: The results showed that the success of osseointegration is due to the qualitative parameters of the bone tissue of the perceived bed and the initial level of remodeling processes and does not depend on the conditioning of the implant surface NaOH. Comprehensive antiosteoporotic therapy with bisphosphonates and calcium supplementation with vitamin D leads to restoration of micro-alveolar part.

The base of osteoporosis is an imbalance of bone remodeling processes, with a predominance of the resorption phase that in the maxillofacial region is manifested by a partial and complete absence of teeth with a decrease in the volume of bone tissue of the jaws and a violation of its microarchitectonics. Atrophy of the alveolar bone reduces the degree of fixation of the removable prosthesis, can develop various diseases of the underlying mucosa. In this regard, the installation of dental implants as a support for removable orthopedic structures is relevant, however, in patients with osteoporosis, the risk of complications in the early and late postoperative period remains high to 44%. Objective: to improve the efficiency of dental implantation in patients with osteoporosis. 80 men over 50 years old with complete absence of teeth on the lower jaw with alveolar atrophy and osteoporosis were under observation. Carried out condition monitoring of the micro alveolar bone in patients with osteoporosis without a comprehensive antiosteoporotic therapy and in patients receiving ibandronate for 1 tablet (150 mg), 1 time a month with calcium 1 000 mg and vitamin D (cholecalciferol) 800 IU daily. It was found that taking antiresorptive therapy for 3 months improves the quality of the alveolar bone, increases the effectiveness of dental implantation in patients with complete absence of teeth and atrophy of the alveolar part.

Micromorphology of oyster mushrooms Pleurotus ostreatus (Jacq.) Quel. and P. pulmonarius (Fr.) Quel. was studied in pure and binary culture with yeasts (Cryptococcus laurentii 1629, Rhodotorula minuta 2790, Sporidiobolus salmonicolor (see text for symbol) 31A-11, Candida krusie 3452, Pichia holstii 3438). The cultures were cultivated on malt-agar and water agar. Various mycelial structures were described: strands, rings, thin searching mycelium, clamps, crystals, head-like offshoots, mycelial fragments, chlamydospores, and coralloid hyphae. Vegetative mycelia interact in different ways (forming anastomoses, strands, system of thin anucleate hyphae) within the same culture. Head-like offshoots of mycelial cells, previously regarded as spores of asexual reproduction, appeared to lack nuclei and to be filled with polyphosphates. Coralloid hyphae, which induce yeast cell lysis after direct contact, were detected only in binary culture with yeasts under condition of nitrogen deficit. The same way of feeding is typical for carnivorous mushrooms.

RESUMEN

A comparative description of the cell wall ultrastructure was made for vegetative mycelia in Agaricus bisporus, other species of Agaricus, and Pleurotus ostreatus. Apart from well known ultrastructural features of cell walls of the examined strains, inclusions of different kinds were observed. The cell wall thickness varied in different Agaricus strains. Maximum differences between the examined strains were 1.4 times. The cell wall thickness in P. ostreatus strains was by 3 to 4 times less than that in Agaricus strains. Using electron micrographs, a brief description of mycelia and A. bisporus basidia ultrastructure was composed.

Blood content of MDA in rats increased 1 and 2 weeks after mandibular bone fracture at stages of cellular fibrous and chondroid callus and decreased 4 weeks after fracture at the stage of primary bone callus. Treatment with Se (intragastrically and electrophoretically) reduced this increase by activating selenium-containing glutathione peroxidase. In order to clear out the relationship between Se and carbohydrate metabolism in different ages, the distribution of Se between the blood and mandibular bone, diaphysis and metaepiphyseal zone of the femoral bone was studied using the bone/blood relative radioactivity coefficient after intraperitoneal injection of [(75)Se]selenate. In control 1-month-old rats the radioactivity had 2 peaks corresponding to 6 and 48 h. The first peak was presumably caused by Se adsorption on hydroxyapatite, the second by chemosorption on hydroxyapatite and protein binding. Only one peak of relative radioactivity (after 12-48 h) was observed in 3-month-old control rats, and it could be increased by sucrose diet. The relative radioactivity was higher in rats receiving sucrose ration for 2 months starting from the age of 1 month in comparison with the control.

The study is devoted to the registration of local H+ gradients on the inner membrane of mitochondria under conditions of H+ pump functioning were recorded. By using a covalently linked pH probe (fluorescein isothiocyanate), a local increase in the activity of hydrogen ions on the outer face of the inner mitochondrial membrane in the presence of the respiration substrate at increased permeability of the membrane for K+ was registered. It was also found that the buffer capacity of medium affects the respiration rate of completely uncoupled mitochondria; a change in respiration rate strictly correlates with changes in local H+ gradients on the mitochondrial membrane. It was concluded that local gradients of H+ activity can control the rate of functioning of H+ pumps. It was shown that, under certain conditions, the system of H+ pumps incorporated into succinate oxidase of mitochondria functions as a nonliner system.

The paper presents the results of examining the central part of the rat mandible for a month after its fracture. The rate of free radical oxidation was estimated by the levels of diene conjugates (DC), malonic dialdehyde (MDA) and by the activity of selenium-containing glutathione peroxidase (GPO). The findings suggest that there are profound injury-induced changes in free radical oxidation and antioxidative defense. The levels of DC and MDA showed the maximum upward change as a double-peaked curve and longer restoration to the level seen in control animals. These rats had the minimum enhanced GPO activity. In the animals receiving intragastric selenium methionine or selenium electrophoresis applied to the site of mandible fracture, alterations in DC and MDA were less pronounced and GPO showed a higher activity.

The effect of swelling of cultured primary astrocytes from rat brain in hypotonic medium on K+ influx has been studied. A decrease in osmolality from 310 to 180 mOsm increased the activity of sodium pump (ouabain-inhibited 86Rb+ influx) and Na+,K+,2Cl- cotransport (ouabain-insensitive bumetanide-inhibited 86Rb+ influx) by 70 and 35%, respectively. It is suggested that activation of these transport systems makes it possible to retain a high potassium concentration in the cells under regulatory volume decrease.

Effect of dalargin, an opioid peptide (a synthetic analogue of Leu-enkephalin), on proliferation and intensity of DNA synthesis of C6 glioma cells was studied. Specific conditions of cultivation were selected, with a low control value of proliferation, which permitted to assess growth-stimulating effect of the peptide. Growth curves were plotted to assess peptide activity, which demonstrated that reaction was a many-phase process: a significant increase in cell number under peptide effect was observed only at the beginning of the logarithm phase and at the beginning of the prestationary phase of the growth curve. Cell number increased on average by 25-27% in the presence of dalargin as compared to control. Reaction of glioma DNA synthesis to dalargin also demonstrates complexity of the process: the peptide changes DNA synthesis, but as a rule, this process has a three-phase character and is not directly associated with the duration of cultivation in the presence of dalargin. Effect of naloxone, an opiate receptor blocker, was analysed to assess the receptor mechanism. It was found that reaction for naloxone and for combined effect of naloxone and dalargin was not the same.

Effect of opioid peptide Leu-enkephalin (Leu-enk) and synthetic analogue of Leu-enk on fetal rat spinal cord cells in culture was studied. Activity of peptides was measured in dissociated culture on aggregation assay. The aggregated place was calculated on days 3 and 6 of culturing, because this parameter indicates the cell adhesion and neuronal and glial survival. The aggregation place was shown to increase due to peptide action: with Leu-enk and dalargin by 1.6 and 1.7, resp., compared to the control; both calculations being made on days 3 and 6 of culturing. The results of the analysis of opioid peptide activity during culturing of dissociated cells of fetal rat spinal cord demonstrate that the peptides changed the adhesion of cells and increased their survival. The results of the present and our earlier experiments suggest that the opioid peptides may be growth factors for CNS cells and may play a role in development and regeneration of the nervous tissue.

Neurochordins are a family of high Mr P-epitope-bearing neural tissue glycoproteins. Comparison of SDS-agarose electrophoretic patterns of extracts from human, rat, mouse and chicken brain after immunostaining of blots with an anti-P-epitope MAb At5 demonstrated that in each case neurochordins from the A, B and C group were present. A similar result was obtained when polyclonal serum against total human neurochordin was used. The data favours the idea of high evolutional conservatism of neurochordins of higher vertebrate species. Expression of the P-epitope on glial cells in culture was studied. Strong immunostaining of oligodendrocytes with MAb At5 and the absence of this staining in astrocytes and Schwann cells make it possible to use At5 for identification of cell types in tissue culture experiments as well as for differential diagnostics of brain tumours.

The immunofluorescence technique was used to trace auto-antibodies to neural structures in the blood serum of 180 patients with various cardiomyopathies and 20 healthy probands (controls). Incubation of cryostat slices of heart, kidney, spinal cord and medulla oblongata of Wistar-rats or of cell cultures of embryonal spinal cord with the blood serum of patients with cardiomyopathies resulted in immunofluorescence of nerve fibres and neuronal perikaryon. The controls were negative. The question remained unanswered for the nature of the involved antigen. Neurohistochemical findings suggested auto-antibodies in sera of patients to be orientated to cholinergic endings. The authors suggest the immunomorphological approach as practicable for revealing auto-antibodies in neurohistological studies.

RESUMEN

The effect of Balis-2 drug on the growth and differentiation of nervous tissue was studied on organotypic culture of the sympathetic ganglia. It was established that this agent is able to stimulate fiber outgrowth from explant and increase mean value of the maximal magnitude of the zone of the growth by concentration 0.001% and 0.0001%. It was found that Balis-2 drug is able to increase the intensity of reaction by 1.8-2 times. It is suggested that Balis-2 drug can be used as a new neurotropic agent.

The effect of the baliz-2 drug (0.01%, 0.001%, 0.0001%) on the growth and differentiation of nervous tissue was studied on organotypic cultures of the sympathetic ganglia of newborn Wistar, Wag, August rats. The growth zone of explants in living cultures and histochemical stained preparations were analyzed for catecholamine-containing neurites. It has been established that control value of growth intensity for August ganglia is 2.1 2.0 times lower than for Wistar and Wag. The baliz-2 drug at concentration 0.001% and 0.0001% is able to stimulate fibre outgrowth from the explant. Mean values of the reaction are different: they are maximal for August rats (2.3-2.6 times higher than the control), while for Wistar and Wag rats they are 1.8-2.0 times higher than the control. These results provide evidence that the baliz-2 drug is a neurotrophic agent for sympathetic ganglia in culture. The results are discussed in terms of the role of the sympathetic-adrenal system in the nervous tissue regeneration.

The proliferative response of phytohaemagglutinin (PHA) - stimulated lymphocytes of healthy donors and hypertensive subjects was analysed according to 3H-thymidine incorporation and cell cycle phase position on a flow cytofluorimeter. 3H-thymidine uptake and the percentage of cells in (S + G2) phase were significantly lower in hypertensive patients. After short-term propranolol administration the lymphocyte blastogenic response in essential hypertension increased by 30--100%. The data suggest that the change in mitogenic response of lymphocytes in essential hypertension results from changes in the distribution of lymphocyte subclasses. The connection between the sympathoadrenal and immune system is discussed.

Studies with organotypic culture of sympathetic ganglia and spinal cord revealed that substance P at concentrations of 10(-5) to 10(-12) M and 10(-5) to 10(-14) M exerts a marked growth-stimulating effect on sympathetic ganglia and spinal cord, respectively. In the presence of substance P, the intensity of sympathetic ganglion growth exceeds control values 3.0-4.8 times. The growth zone size of spinal cord explants increases under these conditions 2.0-5.2-fold. A feasible physiological significance of regulatory peptides in the growth and regeneration of nervous tissue as well as the role of noci-antinociceptive systems in histogenesis and regeneration are discussed.
