RESUMEN

Objetivo: No presente trabalho são avaliados parâmetros hematológicos e bioquímicos de ratas expostas ao extrato aquoso de Piper methysticum no período de implantação do blastocisto. Métodos: Quarenta ratas Wistar prenhes foram organizadas aleatoriamente nos grupos controle e tratado, que receberam respectivamente 1 ml de água destilada e 10 mg de extrato aquoso de kava / kg de peso corporal em 1 ml de água destilada. O tratamento foi feito por via intragástrica, duas vezes ao dia, nos dias 5, 6 e 7 pós - inseminação (1o dia=espermatozóide no esfregaço) e eutanasiadas, no 15o dia de prenhez, por exsanguinação total (punção cardíaca) sob anestesia. Resultados: houve leucopenia e aumento signifi cativo na dosagem de Transaminase Glutâmico Oxalacética (TGO) entre os animais do grupo tratado. Contudo, não foram observadas alterações histopatológicas no fígado. Conclusão: O extrato aquoso de kava-kava, administrado a ratas prenhes, parece não causar toxicidade hepática, embora tenha evidenciado leucopenia e aumento de TGO.


Aims: To evaluate biochemistry and hematological parameters in pregnant rats treated with aqueous extract of Piper methysticum. Methods: Forty pregnant rats were randomly distributed into two groups: Control that received 1ml of distilled water and treated with 10mg of aqueous extract of kava/kg/body weight, by gavage, twice a day, from day-6 to 7 post insemination (day 1 = spermatozoa visible in the vaginal smear). Euthanize was performed by exsanguinations via cardiac punction under anesthesia (ketamine plus xylazine) at 14 post insemination day. Results: Treated animal presents leucopenia and higher concentration of TGO. No histopathologic alteration was observed in the liver. Conclusion: Th e Piper methysticum aqueous extract seems do not cause hepatotoxicity, but cause leucopenia and increase the plasmatic level of TGO.

RESUMEN

St John's wort (Hypericum perforatum) is a medicinal plant used in the treatment of depression and other psychiatric disorders. In the present paper, the toxicity of H. perforatum administered to female rats during the period of organogenesis (day 9-15 of pregnancy) was evaluated. Thirty inseminated Wistar rats were randomly distributed into control and treated groups, which received, by gavage, 0.5 mL of saline and 36 mg/kg body weight of Jarsin dried extract diluted into 0.5 mL of saline, respectively. Maternal toxicity was evaluated through: water and food intake, body weight gain, piloerection, locomotor activity, diarrhea and death occurrence. Animals were killed on day 21 of pregnancy, when fetuses and placentas were removed and weighed. The indices of implantation and resorption were calculated. Clinical signs of maternal toxicity were not observed and none of the variables analysed showed statistically significant differences. In the dose administered in the experimental model used, H. perforatum does not seem to be toxic to the mother nor to interfere with the progress of gestation during organogenesis.

BACKGROUND: Saint John's wort (Hypericum perforatum) is a medicinal plant used in the treatment of depression and other psychiatric disorders. OBJECTIVE: In the present paper, the toxicity of H. perforatum administered to female rats during organogenesis (9th to 15th day of pregnancy) was evaluated. METHODS: Thirty inseminated Wistar rats were randomly distributed into Control and Treated groups, which received by gavage, respectively, 0.5 ml of saline and 36 mg/Kg body weight of Jarsin dried extract diluted into 0.5 ml of saline. Maternal toxicity was evaluated by means of: water and food intake, body weight, piloerection, walking activity, diarrhea and death. Animals were killed on the 21st day of pregnancy, when kidneys, liver and ovaries were weighed. Implantation and reabsorption indices were calculated, as well as the average number of fetuses per mother. RESULTS: Clinical signs of maternal toxicity were not observed and none of the variables analyzed showed statistically significant differences. CONCLUSION: At the dose administered in the experimental model used, H. perforatum does not seem to be toxic to the mother.

OBJETIVO: No presente trabalho foi avaliada a toxicidade do H. perforatum administrado a ratas no período de organogênese (9º ao 15º dia de gestacão). MÉTODOS: Trinta ratas Wistar inseminadas foram distribuídas aleatoriamente nos grupos controle e tratado, que receberam, respectivamente, 0,5 mL de solucão fisiológica e 36 mg/kg de extrato seco de Jarsin diluídos em 0,5 mL de solucão fisiológica por gavagem. A toxicidade materna foi avaliada por: consumo de água e racão, peso corporal, piloerecão, deambulacão, diarréia e ocorrência de mortes. As ratas foram sacrificadas no 21º dia de gestacão, quando foram removidos e pesados: rins, fígado e ovários. Foram calculados os índices de implantacão e de reabsorcão e foi verificado o número médio de fetos por rata. RESULTADOS: Não foram observados sinais clínicos de toxicidade materna e nenhuma das variáveis analisadas apresentou diferencas estatisticamente significativas entre os grupos experimentais. CONCLUSAO: Na dose administrada e no modelo experimental utilizado, o Hypericum perforatum não apresenta manifestacões tóxicas para ratas prenhas no período de organogênese.

The Jarsin extract (Hypericum perforatum L., 1753), is used as an alternative of the synthetic antidepressants. The are few paper concerning the reproductive toxicology of Jarsin extract and none related to the early embryonic development. The present work was aimed at verifying the effect of Jarsin extract, on the blastocyst development in Wistar rats. Female rats from the Centro de Biologia da Reprodução vivarium were mated with fertile males. After the insemination, the femalerats were randomly distributed in two groups: Control (0.5 ml ofdistilled water) and treated (18mg of H. perforatum/0.5 ml of distilled water). Both groups received the treatment by oral gavage, twice a day from the first to the fourth day of gestation. The animals were terminated on the fifth day of gestation. The oviduct and uterine cornua were flushedwith Talpe-Hepes medium and the embryos were collected, counting and their morphologic development performed. The dams were evaluated to clinical signs indicative of maternal intoxication (deaths, altered locomotion in the cage, piloerection, diarhoea, vaginal bleeding, reducedfood consumption and body weight gain or loss). Maternal ovary weight was measured and the number of corpora lutea was counted. The data were analyzed by the Students t test (a = 0,05).
