Coffee Breeding programs have traditionally relied on observing plant characteristics over years, a slow and costly process. Genomic selection (GS) offers a DNA-based alternative for faster selection of superior cultivars. Stacking Ensemble Learning (SEL) combines multiple models for potentially even more accurate selection. This study explores SEL potential in coffee breeding, aiming to improve prediction accuracy for important traits [yield (YL), total number of the fruits (NF), leaf miner infestation (LM), and cercosporiosis incidence (Cer)] in Coffea Arabica. We analyzed data from 195 individuals genotyped for 21,211 single-nucleotide polymorphism (SNP) markers. To comprehensively assess model performance, we employed a cross-validation (CV) scheme. Genomic Best Linear Unbiased Prediction (GBLUP), multivariate adaptive regression splines (MARS), Quantile Random Forest (QRF), and Random Forest (RF) served as base learners. For the meta-learner within the SEL framework, various options were explored, including Ridge Regression, RF, GBLUP, and Single Average. The SEL method was able to predict the predictive ability (PA) of important traits in Coffea Arabica. SEL presented higher PA compared with those obtained for all base learner methods. The gains in PA in relation to GBLUP were 87.44% (the ratio between the PA obtained from best Stacking model and the GBLUP), 37.83%, 199.82%, and 14.59% for YL, NF, LM and Cer, respectively. Overall, SEL presents a promising approach for GS. By combining predictions from multiple models, SEL can potentially enhance the PA of GS for complex traits.

The agouti (Dasyprocta prymnolopha) is a medium-sized, wild rodent that is highly rustic and docile. Its size and ease of management make it a viable candidate for an alternative animal model to traditional murine subjects. However, data on the epidermal strata of agoutis are lacking, with significant uncertainties persisting regarding their skin's characterization. This study aimed to describe and quantify the epidermal strata of skin biopsies from male and female agoutis raised in captivity, to further validate the species as a model for dermatological research. Ultrastructural evaluations through atomic force microscopy (AFM) and stereological analyses were conducted, revealing significant differences between the layers of the skin; notably, the dermis exhibited a greater total volume than the epidermis. The findings suggest that the epidermal strata are well-defined, with the volume likely correlating to the size and cellular density of the keratinocytes. Corneodesmosomes and tonofilaments were identified across all epidermal layers, indicating the probable maintenance of anchoring protein activity, even post-cornification of these cells. These results suggest that the agouti may serve as a promising model for dermatological studies, owing to the homogeneity of its cutaneous tissue across different body regions and the distinct volume and morphology of its epithelial stratification, which could enhance the applicability of systematic investigative methods in the future.

OBJECTIVE: To investigate the effects of combining a Pilates program with photobiomodulation therapy (PBMT) in patients with chronic nonspecific low back pain (CNLBP). METHODS: Thirty-eight adults with CNLBP were randomly assigned to two groups: Pilates exercise + active PBMT (PIL + PBMT) or Pilates exercise + sham PBMT (PIL + SHAM). Both groups performed an 8-week mat Pilates program and received PBMT on their lumbar muscles 10 min before and after each session. The following variables were assessed before and after intervention: peak pain intensity, postural balance (i.e., center of the pressure [A-COP], velocity anteroposterior [Vel AP], and velocity mediolateral [Vel ML]), perceived disability (i.e., Oswestry Disability Index [ODI] and Roland Morris Disability Questionnaire [RMDQ]), and pain-related fear of movement (i.e., Tampa Scale of Kinesiophobia [TSK], Fear Avoidance Beliefs Questionnaire [FABQ], and Pain Catastrophizing Scale [PCS]). RESULTS: Postural balance variables showed no statistically significant differences (p > 0.05) across time or between groups. The groups showed similar (p < 0.05) reductions in peak pain intensity, ODI, RMDQ, and PCS scores, but no statistically significant difference (p > 0.05) in TSK and FABQ scores. CONCLUSION: The mat Pilates program reduced peak pain intensity, perceived disability, and pain catastrophizing in adults with CNLBP, but PBMT had no additional effect on these variables. Mat Pilates alone or combined with PBMT was not able to improve postural balance.

Obesity is a complex chronic disease characterized by excess body fat (adipose) that is harmful to health and has been a major global health problem. It may be associated with several diseases, such as nonalcoholic fatty liver disease (NAFLD). Polyunsaturated fatty acids (PUFA) are lipid mediators that have anti-inflammatory characteristics and can be found in animals and plants, with capybara oil (CO) being a promising source. So, we intend to evaluate the hepatic pathophysiological alterations in C57Bl/6 mice with NAFLD, caused by obesity, and the possible beneficial effects of OC in the treatment of this disease. Eighteen 3-month-old male C57Bl/6 mice received a control or high-fat diet for 18 weeks. From the 15th to the 18th week, the animals received treatment-through orogastric gavage-with placebo or free capybara oil (5 g/kg). Parameters inherent to body mass, glucose tolerance, evaluation of liver enzymes, percentage of hepatic steatosis, oxidative stress, the process of cell death with the apoptotic biomarkers (Bax, Bcl2, and Cytochrome C), and the ultrastructure of hepatocytes were analyzed. Even though the treatment with CO was not able to disassemble the effects on the physiological parameters, it proved to be beneficial in reversing the morphological and ultrastructural damage present in the hepatocytes. Thus, demonstrating that CO has beneficial effects in reducing steatosis and the apoptotic pathway, it is a promising treatment for NAFLD.

RESUMO Este artigo apresenta perfil dos egressos, efeitos da formação e trajetória profissional dos concluintes do Curso de Mestrado Profissional em Saúde Pública, do Instituto Aggeu Magalhães, parte de pesquisa avaliativa em desenvolvimento. Dos 99 egressos convidados que concluíram seus cursos entre 2013-2020, 54 responderam a um questionário aplicado por meio do software Lime Survey. O estudo descritivo e transversal utilizou frequência absoluta e relativa e cruzamento de variáveis na análise, cujo instrumento constou dos temas: identificação do egresso e do programa/curso; atividade profissional antes e ao término do curso e expectativas; condição empregatícia atual e efeitos da formação; avaliação da trajetória formativa. Entre os respondentes, a maioria é do sexo feminino, com idade entre 31-40 anos, enfermeiros de formação, autodeclarados pretos ou pardos, não ser pessoa com deficiência, predominantemente residentes em Pernambuco e não ingressaram mediante cota. Como efeito na vida profissional, encontrou-se que o curso aumentou o prestígio/reconhecimento dos colegas, qualificou para atividades que já exerciam e para atividades diferentes, e trouxe ganhos de remuneração, proporcionando mudanças na vida pessoal e profissional. Os resultados estão em conformidade com achados científicos desse mesmo contexto, e a ferramenta desenvolvida pela Fiocruz permitiu conhecer a relação entre formação e trabalho.


ABSTRACT This article presents the profile of graduates, the effects of training, and the professional trajectory of the Professional Master's Course in Public Health graduates at the Aggeu Magalhaes Institute. Of the 99 invited graduates who completed the program between 2013-2020, 54 responded to a questionnaire applied through the Lime Survey software. The descriptive and cross-sectional study used absolute and relative frequency cross-tabulation variables in the analysis, with the instrument covering the following themes: identification of the graduate and program/course; professional activity before and at the end of the program and expectations; current employment status and formation effects; evaluation of the formative trajectory. Among the respondents, most are female, aged between 31-40 years, trained as nurses, self-declared as black or brown-skinned, not having a disability, predominantly residing in Pernambuco, and did not enter through a quota system. As an effect on their professional life, it was found that the program increased prestige/recognition among colleagues, qualified them for activities they were already performing and also different activities, and brought salary increases leading to changes in personal and professional lives. The results are consistent with scientific findings in the same context, and the tool developed by FIOCRUZ made it possible to understand the relationship between education and work.

Photobiomodulation therapy (PBMT) has been advocated as a potential intervention to improve muscle performance and recovery in the health and sports context. However, the short- and long-term effects of PBMT on endurance running performance remain under-researched and controversial. The purpose of this study was to investigate the acute dose-response effect of PBMT with light-emitting diodes (LEDs) on endurance performance and rating of perceived exertion (RPE; 6-20 Borg) during a 5-km running trial in recreational runners. In a crossover design, eighteen young adult runners (28.7 ± 7.8 years) were randomized to receive 1 of 4 PBMT conditions (placebo, 300, 900, and 1260 Joules [J]) 60 min before the 5-km running trial on four occasions, separated by a 2-wk washout period. The treatments were applied to the quadriceps, hamstrings, and gastrocnemius muscles of both legs using a device containing 200 LEDs (100 red and 100 infrared). The following variables were assessed: endurance performance (i.e. total time, mean velocity, and velocity in the split distances at the initial 200 m and every 400 m lap) and RPE in the split distances at the initial 200 m and every 400 m lap. Data normality and homogeneity were tested using Shapiro-Wilk's and Levene's tests, respectively. Differences between treatment conditions were assessed using the analysis of variance tests (one- or two-way ANOVA, depending on the comparisons), complemented by the Bonferroni post hoc test. There were significant time effects for the running velocity and RPE in the split distances (p < 0.0001), with no significant treatment-by-time interaction (running velocity, p = 0.59; RPE, p = 0.95). The mean velocity (p = 0.997), total time (p = 0.998), and total mean of the RPE (p = 0.91) were similar between treatment conditions. In conclusion, acute PBMT with LEDs at doses of 300, 900, and 1260 J is not recommended for improving endurance performance and RPE in the 5-km running trial in recreational runners.

O cenário advindo como consequência da pandemia de covid-19 acarretou diversas mudanças na vida dos profissionais de saúde. A percepção desse grupo torna-se indispensável para a compreensão do contexto, visto que esses trabalhadores são figuras essenciais no enfrentamento da doença. O objetivo deste estudo foi analisar a percepção de gestores e profissionais que atuaram durante a primeira onda da pandemia de covid-19 no município de Sobral (CE). Trata-se de uma pesquisa exploratório-descritiva, com abordagem qualitativa, feita por meio da aplicação de um instrumento de coleta de dados. O estudo foi realizado com gestores e trabalhadores que atuaram de março a dezembro de 2020 no município. A coleta foi feita no formato híbrido, com auxílio da plataforma Google Meet, fundamentada na metodologia snowball sampling e no método de saturação. Após a coleta, os discursos foram transcritos ­ utilizando a ferramenta Microsoft Word ­ e analisados ­ com suporte do software NVivo11. Foram contactados 68 profissionais. Identificou-se predominância final de enfermeiros, sexo feminino e atuantes na atenção terciária. Após análise e categorização, identificaram-se 12 unidades de registro. Foram dispostos os principais desafios e dificuldades acarretados para os processos de trabalho e suas subcategorias: a sobrecarga de trabalho e o desgaste físico; a reorganização dos serviços em resposta à nova realidade; o desconhecimento sobre a doença e seu impacto psicológico no fazer profissional; e o fator social e a dificuldade de seguimento da assistência na atenção primária. Por fim, conclui-se que os desafios identificados se inter-relacionam, repercutindo uns nos outros, e constata-se a importância de conhecer esses entraves para ações mais bem direcionadas e estratégicas.


The scenario that has arisen in response to COVID-19 has led to a variety of changes in the lives of healthcare providers. The perception of this group is essential to understanding this context since they are key figures in coping with the disease. This study aimed to analyze the perception of managers and professionals who worked during the first wave of the COVID-19 pandemic in the municipality of Sobral/CE. This is an exploratory-descriptive study with a qualitative approach that applied a collection instrument with managers and workers who worked from March to December 2020 in the municipality. Data were collected using the Snowball Smapling and the saturation method in a hybrid format using the Google Meet platform. After the collection, responses were transcribed using Microsoft Word and analyzed with the aid of NVivo11. In total, 68 professionals were contacted, with a final predominance of female nurses working in tertiary care. After analysis and categorization, 12 recording units were identified. The main challenges and difficulties for work processes and their subcategories include work overload and physical exhaustion; reorganization of services in response to the new reality; lack of knowledge about the disease and its psychological impact on professional practice; and the social factor and the difficulty of following up care in primary care. Finally, we conclude that the identified challenges are interrelated and have repercussions on each other, and that it is important to understand these obstacles to take more targeted and strategic actions.


El escenario que ha surgido en respuesta a la pandemia de la COVID-19 ha provocado diversos cambios en la vida de los profesionales de la salud. La percepción de este grupo es indispensable para comprender el contexto, ya que estos trabajadores son figuras esenciales en el enfrentamiento a la enfermedad. El objetivo de este estudio fue analizar la percepción de gestores y profesionales que actuaron durante la primera ola de la pandemia de la COVID-19 en el municipio de Sobral (Ceará, Brasil). Se trata de un estudio exploratorio-descriptivo, con enfoque cualitativo, realizado mediante la aplicación de una herramienta de recolección de datos. En esta investigación participaron gestores y trabajadores que actuaron de marzo a diciembre de 2020 en el municipio. Se realizó la recolección en formato híbrido, con ayuda de la plataforma Google Meet, basada en el método bola de nieve y en el método de saturación. Después, se transcribieron los discursos, utilizando la herramienta Microsoft Word, y se analizaron con el apoyo del software NVivo11. Se contactaron 68 profesionales. Se identificó predominio de enfermeros, mujeres y trabajadores de atención terciaria. Después del análisis y de la categorización, se identificaron 12 unidades de registro. Se presentaron los principales retos y dificultades que generan los procesos de trabajo y sus subcategorías: la sobrecarga de trabajo y el agotamiento físico; la reorganización de los servicios en respuesta a la nueva realidad; el desconocimiento de la enfermedad y su impacto psicológico en la práctica profesional; y el factor social y la dificultad de seguimiento de los cuidados en atención primaria. Finalmente, se concluye que los retos identificados están interrelacionados, impactándose entre sí, y se señala la importancia de conocer estos obstáculos para emprender acciones más específicas y estratégicas.

Objective: To conduct a meta-analysis to investigate the effects of Photobiomodulation (PBM) therapy on running performance. Introduction: PBM has recently been advocated as a valuable non-pharmacological ergogenic strategy, however, the efficacy of PBM on running performance remains unproven. Methods: A computerized literature search was conducted until June 2023. The databases searched were PubMed/Medline, Embase, Scopus, SPORTDiscus, and Web of Science. Inclusion/exclusion criteria were determined through the PICO process. The running variables analyzed were time-trial or time-to-exhaustion. Results were combined with the standardized mean differences (SMD) and the 95% confidence intervals. Results: Twelve studies fulfilled the inclusion criteria. No significant effects in favor of PBM were found (SMD = 0.13; p = 0.11). There was no effect considering the presence (SMD = 0.16; p = 0.38) and absence (SMD = 0.11; p = 0.25) of training, and there was no dose-response effect (p = 0.82). Conclusion: Our findings indicate that PBM alone or combined with a training program does not improve running performance in terms of time-trial and time-to-exhaustion testing. More studies involving PBM plus training and doses higher than 1000 J are needed to determine if PBM is effective in improving running performance.

(1) Background: Species of the genus Cymbopogon and its essential oil are known for their antioxidant and hypoglycemic effects. This study aimed to investigate the impact of the essential oil of Cymbopogon flexuosus (EOCF), and its major component, citral, on glycemic, lipid, antioxidant parameters, and oxidative stress in a type 1 diabetes (DM1) rat model. (2) Methods: Initially, EOCF was analyzed by Gas chromatography-mass spectrometry (GC-MS) and the antioxidant activity of EOCF and citral was evaluated. Next, male Wistar rats (3 months old, 200-250 g) induced with DM1 using Streptozotocin (STZ) were divided into four groups: negative control supplemented with an 80% Tween solution, two groups of animals supplemented with EOCF (32 mg/kg and 64 mg/kg) and with citral (32 mg/kg), and treated for 14 days. Measurements of blood glucose levels and body weight were taken; after euthanasia, biochemical markers, including lipid profile, uric acid, alanine aminotransferase (ALT), and aspartate aminotransferase (AST), were evaluated. (3) Results: The predominant compounds in EOCF were α-citral (53.21%) and neral (19.42%), constituting 72.63% citral. EOCF showed good antioxidant activity, significantly greater than citral. EOCF supplementation demonstrated a mitigating effect on glycemic, lipid, and hepatic abnormalities induced by DM1. (4) Conclusions: EOCF emerges as a promising therapeutic option for the management of DM1.

Leptospira is a genus of bacteria that includes free-living saprophytic species found in water or soil, and pathogenic species, which are the etiologic agents of leptospirosis. Besides all the efforts, there are only a few proteins described as virulence factors in the pathogenic strain L. interrogans. This work aims to perform L. biflexa serovar Patoc1 strain Paris global proteome and to compare with the proteome database of pathogenic L. interrogans serovar Copenhageni strain Fiocruz L1-130. We identified a total of 2327 expressed proteins of L. biflexa by mass spectrometry. Using the Get Homologues software with the global proteome of L. biflexa and L. interrogans, we found orthologous proteins classified into conserved, low conserved, and specific proteins. Comparative bioinformatic analyses were performed to understand the biological functions of the proteins, subcellular localization, the presence of signal peptide, structural domains, and motifs using public softwares. These results lead to the selection of 182 low conserved within the saprophyte, and 176 specific proteins of L. interrogans. It is anticipated that these findings will indicate further studies to uncover virulence factors in the pathogenic strain. This work presents for the first time the global proteome of saprophytic strain L. biflexa serovar Patoc, strain Patoc1. SIGNIFICANCE: The comparative analysis established an array of specific proteins in pathogenic strain that will narrow down the identification of immune protective proteins that will help fight leptospirosis.

Equine farming generates a significant amount of waste, prompting the need for effective management. Composting enhanced by filamentous fungi holds promise for this purpose. This study focused on inoculating Aspergillus fumigatus isolates in composting horse bedding made with wood shavings (Pinus elliottii). The experiment lasted 90 days, with two treatment groups, control and inoculated, analyzing temperature, pH, electrical conductivity, total organic carbon and nitrogen content, and cellulose, hemicellulose, and lignin contents. Both treatments entered the thermophilic phase by the fourth day, reaching temperatures above 55°C and mesophilic maturation at 35 days (41 ± 0.2°C). The inoculated treatment exhibited higher electrical conductivity after 30 days and a more pronounced reduction in the total carbon content (42.85% vs. 38.29%) compared to the control. While there was no significant nitrogen difference, the inoculated treatment had a sharper reduction in carbon/nitrogen ratio, and cellulose and hemicellulose contents. Both treatments showed low coliform counts, no Salmonella sp., and reduced Strongyloides sp. larvae. Inoculating A. fumigatus in saturated horse bedding made from wood shavings improved compost quality, providing a possibility for sustainable equine farming waste treatment.

In the context of multi-environment trials (MET), genomic prediction is proposed as a tool that allows the prediction of the phenotype of single cross hybrids that were not tested in field trials. This approach saves time and costs compared to traditional breeding methods. Thus, this study aimed to evaluate the genomic prediction of single cross maize hybrids not tested in MET, grain yield and female flowering time. We also aimed to propose an application of machine learning methodologies in MET in the prediction of hybrids and compare their performance with Genomic best linear unbiased prediction (GBLUP) with non-additive effects. Our results highlight that both methodologies are efficient and can be used in maize breeding programs to accurately predict the performance of hybrids in specific environments. The best methodology is case-dependent, specifically, to explore the potential of GBLUP, it is important to perform accurate modeling of the variance components to optimize the prediction of new hybrids. On the other hand, machine learning methodologies can capture non-additive effects without making any assumptions at the outset of the model. Overall, predicting the performance of new hybrids that were not evaluated in any field trials was more challenging than predicting hybrids in sparse test designs.

Here, we evaluated the influence of outdoor environmental conditions (synoptic weather conditions) on human thermal discomfort in the five macro-regions of Pelotas city, located in the southernmost region of Brazil. To do this, meteorological sensors (HOBO MX2301A) were installed outside the residences to measure the air temperature, dew point temperature, and relative humidity between 18 January and 20 August 2019. Two well-established simplified biometeorological indices were examined seasonally: (i) humidex for the summer months and (ii) effective temperature as a function of wind for the autumn and winter months. Our findings showed seasonal differences related to human thermal discomfort and outdoor environmental conditions. The thermal discomfort was highest in the afternoons during the summer months and at night during the winter months. The seasonal variation in human thermal discomfort was highly associated with the meteorological conditions. In summer, the presence of the South Atlantic Subtropical Anticyclone (SASA) contributed to heat stress. The SASA combined with the continent's low humidity contributed to the perceived sensation of thermal discomfort. In the winter, thermal discomfort was associated with the decrease in air humidity caused by high atmospheric pressure systems, which led to a decrease in both air temperature and air moisture content. Our findings suggest that a better understanding of the complex interplay between outdoor environmental factors and human thermal comfort is needed in order to mitigate the negative effects of thermal discomfort.

Water availability is a limiting factor for the cultivation of sour passion fruit. Soil management techniques and the use of water-retaining polymers can increase soil water retention, reducing the frequency of irrigation in the crop. In this context, the objective of the research was to evaluate the gas exchange, the chlorophyll index, and the yield of the sour passion fruit cv. BRS GA1 as a function of irrigation depths, pit volumes, and doses of water-retaining polymer. The experiment was carried out in randomized blocks, in plots subdivided in a 2 × (2 × 5) arrangement, with irrigation depths of 70 and 100% of the crop evapotranspiration (ETc) as the main plot, the subplots with the volumes of pit of 64 and 128 dm3, and doses of the water-retaining polymer of 0, 0.5, 1.0, 1.5, and 2.0 g dm-3. The interaction of irrigation depths × pit volumes × doses of water-retaining polymer influences chlorophyll indexes, gas exchange, and water productivity, with positive impacts on yield of the sour passion fruit. The water depth of 70% of ETc increased the yield of sour passion fruit, in pits of 64 dm3. The application of doses of up to 1.1 g dm-3 of the water-retaining polymer and irrigation with water of 70% of ETc is recommended, and a dose of 2.0 g dm-3 of the water-retaining polymer in a pit volume of 128 dm3, associated with an irrigation depth of 100% ETc causes stress in sour passion fruit plants due to excess water.

BACKGROUND: Blood plasma is the main source of extracellular vesicles (EVs) in clinical studies aiming to identify biomarkers and to investigate pathophysiological processes, especially regarding EV roles in inflammation and thrombosis. However, EV isolation from plasma has faced the fundamental issue of lipoprotein contamination, representing an important bias since lipoproteins are highly abundant and modulate cell signaling, metabolism, and thromboinflammation. OBJECTIVES: Here, we aimed to isolate plasma EVs after depleting lipoproteins, thereby improving sample purity and EV thromboinflammatory analysis. METHODS: Density-based gradient ultracentrifugation (G-UC) was used for lipoprotein depletion before EV isolation from plasma through size-exclusion chromatography (SEC) or serial centrifugation (SC). Recovered EVs were analyzed by size, concentration, cellular source, ultrastructure, and bottom-up proteomics. RESULTS: G-UC efficiently separated lipoproteins from the plasma, allowing subsequent EV isolation through SEC or SC. Combined analysis from EV proteomics, cholesterol quantification, and apoB-100 detection confirmed the significant reduction in lipoproteins from isolated EVs. Proteomic analysis identified similar gene ontology and cellular components in EVs, regardless of lipoprotein depletion, which was consistent with similar EV cellular sources, size, and ultrastructure by flow cytometry and transmission electron microscopy. Importantly, lipoprotein depletion increased the detection of less abundant proteins in EV proteome and enhanced thromboinflammatory responses of platelets and monocytes stimulated in vitro with EV isolates. CONCLUSION: Combination of G-UC+SEC significantly reduced EV lipoprotein contamination without interfering in EV cellular source, gene ontology, and ultrastructure, allowing the recovery of highly pure EVs with potential implications for functional assays and proteomic and lipidomic analyses.

A assistência nutricional em cuidados paliativos possui papel preventivo e visa assegurar as necessidades nutricionais do paciente na tentativa de auxiliar o controle dos sintomas, manter a hidratação satisfatória, ressignificar a alimentação, reduzir a ansiedade, retardar o desenvolvimento da caquexia, preservar o peso e a composição nutricional. Objetivo: Desenvolver e validar o mnemônico NUTRIFICO como abordagem de assistência nutricional e comunicação de notícias difíceis em cuidados paliativos. Método: Pesquisa de desenvolvimento e validação de conteúdo que utilizou a técnica Delphi com a aplicação do delineamento misto com abordagens qualitativas e quantitativas para a coleta e análise dos dados. Os participantes, nutricionistas com atuação em cuidados paliativos e/ou formação na área, foram recrutados por amostragem do tipo bola de neve. O percentual de concordância mínimo definido para validação foi de 80% pela escala Likert de cinco pontos. Resultados: Quarenta participantes, incluindo oito "sementes", foram convidados a participar do estudo. Destes, 16 assinaram o Termo de Consentimento Livre e Esclarecido (TCLE). A análise de conteúdo seguiu duas rodadas Delphi e foi obtida uma taxa de concordância superior a 80% entre os participantes para todos os componentes. A versão final do mnemônico NUTRIFICO resultou em dois princípios-chave, oito componentes e 12 recomendações. Conclusão: O mnemônico NUTRIFICO foi considerado válido quanto ao conteúdo e, portanto, pertinente à aplicação na abordagem nutricional e comunicação de notícias difíceis em cuidados paliativos.


Nutritional assistance in palliative care has a preventive role and seeks to ensure the patient's nutritional needs in an attempt to help to control the symptoms, maintain satisfactory hydration, reframe food, reduce anxiety, delay the development of cachexia, preserve weight and nutritional composition. Objective: To develop and validate the NUTRIFICO mnemonic as an approach to nutritional assistance and communication of dismal news in palliative care. Method: Research development and validation of content utilizing the Delphi technique with the application of a mixed design with qualitative and quantitative approaches for data collection and analysis. The participants, nutritionists with experience in palliative care and/or training in the area, were recruited by sampling using the snowball method. The minimum agreement percentage defined for validation was 80% on the five-point Likert scale. Results: Forty participants were invited to participate in the study, including eight "seeds" and 16 signed the Informed Consent Form (ICF). The content analysis followed two Delphi rounds, an agreement rate greater than 80% was obtained among participants for all the components. The final version of the NUTRIFICO mnemonic presented two key principles, eight components and twelve recommendations. Conclusion: The mnemonic NUTRIFICO was validated in terms of content and relevant for the application in the nutritional approach and communication of dismal news in palliative care

Leptospira is a genus of bacteria that includes free-living saprophytic species found in water or soil, and pathogenic species, which are the etiologic agents of leptospirosis. Besides all the efforts, there are only a few proteins described as virulence factors in the pathogenic strain L. interrogans. This work aims to perform L. biflexa serovar Patoc1 strain Paris global proteome and to compare with the proteome database of pathogenic L. interrogans serovar Copenhageni strain Fiocruz L1–130. We identified a total of 2327 expressed proteins of L. biflexa by mass spectrometry. Using the Get Homologues software with the global proteome of L. biflexa and L. interrogans, we found orthologous proteins classified into conserved, low conserved, and specific proteins. Comparative bioinformatic analyses were performed to understand the biological functions of the proteins, subcellular localization, the presence of signal peptide, structural domains, and motifs using public softwares. These results lead to the selection of 182 low conserved within the saprophyte, and 176 specific proteins of L. interrogans. It is anticipated that these findings will indicate further studies to uncover virulence factors in the pathogenic strain. This work presents for the first time the global proteome of saprophytic strain L. biflexa serovar Patoc, strain Patoc1.

BACKGROUND: SARS-CoV-2 infection is associated with worse maternal and fetal outcomes. This study aims to describe the characteristics of pregnant and postpartum women with severe Covid-19 admitted to ICU. METHODS AND FINDINGS: It's a retrospective cohort study evaluating pregnant and postpartum women referenced to a specialized ICU between May 2020 and June 2022. Covid-19 was confirmed with RT-PCR or rapid antigen test on a nasopharyngeal swab. Variables were described by median and IQR when numerical, and by frequency and percentage when categorical. OR with 95% CI were calculated for the evaluation of factors related to death. P-values were calculated using Pearson's ꭓ2-test, Fisher's exact test, Wilcoxon rank sum test, and Kruskall-Wallis test, and statistical significance was established as < 0·05. Missing data were excluded. All statistical analysis were performed using R software version 4.2.2. Of the 101 admissions, 85 (84·2%) were of pregnant women. Obesity (23·0%) and systemic arterial hypertension (13·0%) were the most prevalent medical conditions. Sixty-six (65·3%) were admitted using some type of oxygen support. Forty-seven (46·5%) evolved to mechanical ventilation. There were 61 events considered obstetric complications, with 8 stillbirths/fetal losses. The overall lethality was 15·8%. Pregnancy interruption, need for non-invasive mechanical ventilation, level of oxygen support at admission, prone maneuver, hemodialysis, and healthcare-related infections were factors associated with death. Evaluating the WHO 7-category ordinary scale, there was a trend of increase in the risk of death with higher punctuation, with a statistically significant difference of women with 5 (OR = 7·27; 95% IC = 1·17-194; p = 0·031) or 6 points (OR = 12·0; 95% IC = 1·15-391; p = 0·038) when compared to the ones with 3 points, i.e., of women admitted with a high-flow non-rebreathing mask or invasive mechanical ventilation, compared with the ones admitted at room air, respectively. The main limitations of this study are the relatively small number of participants, and the use of data derived of medical records-which are susceptible to misclassification and variable amounts of missing data. CONCLUSIONS: Pregnant and postpartum women with severe Covid-19 have high lethality and a high incidence of clinical and obstetric complications. These findings support that this population should be prioritized in public health strategies that address Covid-19.

KEY MESSAGE: An approach for handling visual scores with potential errors and subjectivity in scores was evaluated in simulated and blueberry recurrent selection breeding schemes to assist breeders in their decision-making. Most genomic prediction methods are based on assumptions of normality due to their simplicity and ease of implementation. However, in plant and animal breeding, continuous traits are often visually scored as categorical traits and analyzed as a Gaussian variable, thus violating the normality assumption, which could affect the prediction of breeding values and the estimation of genetic parameters. In this study, we examined the main challenges of visual scores for genomic prediction and genetic parameter estimation using mixed models, Bayesian, and machine learning methods. We evaluated these approaches using simulated and real breeding data sets. Our contribution in this study is a five-fold demonstration: (i) collecting data using an intermediate number of categories (1-3 and 1-5) is the best strategy, even considering errors associated with visual scores; (ii) Linear Mixed Models and Bayesian Linear Regression are robust to the normality violation, but marginal gains can be achieved when using Bayesian Ordinal Regression Models (BORM) and Random Forest Classification; (iii) genetic parameters are better estimated using BORM; (iv) our conclusions using simulated data are also applicable to real data in autotetraploid blueberry; and (v) a comparison of continuous and categorical phenotypes found that investing in the evaluation of 600-1000 categorical data points with low error, when it is not feasible to collect continuous phenotypes, is a strategy for improving predictive abilities. Our findings suggest the best approaches for effectively using visual scores traits to explore genetic information in breeding programs and highlight the importance of investing in the training of evaluator teams and in high-quality phenotyping.
