Ceratocystis fimbriata Ellis & Halsted recently was recorded causing seed and seedling blight on Carapa guianensis Aubl. (andiroba), a tree species native to the Amazon Rainforest and prized for its valuable timber and medicinal seed oil. C. fimbriata more commonly causes wilt type diseases in woody hosts, especially on non-native host trees. However, on andiroba the disease occurs on seedlings and seeds, affecting the species regeneration. We studied 73 isolates of C. fimbriata on andiroba from three regions of the Amazon Basin to see if they represented natural or introduced populations. Analysis of ITS rDNA sequences and phylogenetic analysis of mating type genes revealed new haplotypes of C. fimbriata from the Latin American Clade that were closely related to other Brazilian populations of the fungus. In mating experiments, andiroba isolates were inter-fertile with tester strains of C. fimbriata from Brazil and elsewhere, confirming that they belong to a single biological species. Using microsatellite markers, 14 genotypes and populations with intermediate levels of genetic variability were found, suggesting that the fungus is indigenous to the Amazon Basin. Inoculation tests indicated that the andiroba isolates are host-specialized on andiroba, supporting the proposition of the special form C. fimbriata f. sp. carapa.

Despite the fact that E. chlorotica (Linnaeus, 1766) is common and widely distributed in South America, the reproductive aspects of the species are poorly documented. Here we present data on 18 active nests found from August to February, between 2007 and 2012. Nests were globular with a lateral entrance, and measured 97.9 ± 14.4 mm in outside height, 110.6 ± 11.6 mm in outside diameter, and were 4.88 ± 2.09 m above ground. They were often supported from bellow and were composed mainly of tiny dry leafs and leaflets, fine petioles, and plumed seeds, all compacted with spider web silk. Eggs were laid on consecutive days or with one day interval, and clutch size varied from 1-3 eggs (2.1 ± 0.6, n = 9 nests). Only females incubated the eggs, but both sexes were involved in nest construction and nestling attendance at similar rates. Incubation and nestling periods were 14 and 21 days, respectively, and overall nest survival probability was 5%. A vespiary used for nesting was not occupied by wasps and nest material was deposited only to form the incubatory chamber. Although nesting near wasps or bees is a widespread strategy among birds in general, nesting inside the nests of social insects is a poorly documented behavior.

The Mango Sudden Decline (MSD), also referred to as Mango Wilt, is an important disease of mango in Brazil, Oman and Pakistan. This fungus is mainly disseminated by the mango bark beetle, Hypocryphalus mangiferae (Stebbing), by infected plant material, and the infested soils where it is able to survive for long periods. The best way to avoid losses due to MSD is to prevent its establishment in mango production areas. Our objectives in this study were to: (1) predict the global potential distribution of MSD, (2) identify the mango growing areas that are under potential risk of MSD establishment, and (3) identify climatic factors associated with MSD distribution. Occurrence records were collected from Brazil, Oman and Pakistan where the disease is currently known to occur in mango. We used the correlative maximum entropy based model (MaxEnt) algorithm to assess the global potential distribution of MSD. The MaxEnt model predicted suitable areas in countries where the disease does not already occur in mango, but where mango is grown. Among these areas are the largest mango producers in the world including India, China, Thailand, Indonesia, and Mexico. The mean annual temperature, precipitation of coldest quarter, precipitation seasonality, and precipitation of driest month variables contributed most to the potential distribution of MSD disease. The mango bark beetle vector is known to occur beyond the locations where MSD currently exists and where the model predicted suitable areas, thus showing a high likelihood for disease establishment in areas predicted by our model. Our study is the first to map the potential risk of MSD establishment on a global scale. This information can be used in designing strategies to prevent introduction and establishment of MSD disease, and in preparation of efficient pest risk assessments and monitoring programs.

Ceratocystis wilt is among the most important diseases on mango (Mangifera indica) in Brazil, Oman, and Pakistan. The causal agent was originally identified in Brazil as Ceratocystis fimbriata, which is considered by some as a complex of many cryptic species, and four new species on mango trees were distinguished from C. fimbriata based on variation in internal transcribed spacer sequences. In the present study, phylogenetic analyses using DNA sequences of mating type genes, TEF-1α, and ß-tubulin failed to identify lineages corresponding to the four new species names. Further, mating experiments found that the mango isolates representing the new species were interfertile with each other and a tester strain from sweet potato (Ipomoea batatas), on which the name C. fimbriata is based, and there was little morphological variation among the mango isolates. Microsatellite markers found substantial differentiation among mango isolates at the regional and population levels, but certain microsatellite genotypes were commonly found in multiple populations, suggesting that these genotypes had been disseminated in infected nursery stock. The most common microsatellite genotypes corresponded to the four recently named species (C. manginecans, C. acaciivora, C. mangicola, and C. mangivora), which are considered synonyms of C. fimbriata. This study points to the potential problems of naming new species based on introduced genotypes of a pathogen, the value of an understanding of natural variation within and among populations, and the importance of phenotype in delimiting species.

To explore the molecular pathways underlying thiazolidinediones effects on pancreatic islets in conditions mimicking normo- and hyperglycemia, apoptosis rate and transcriptional response to Pioglitazone at both physiological and supraphysiological glucose concentrations were evaluated. Adult rat islets were cultured at physiological (5.6 mM) and supraphysiological (23 mM) glucose concentrations in presence of 10 µM Pioglitazone or vehicle. RNA expression profiling was evaluated with the PancChip 13k cDNA microarray after 24-h, and expression results for some selected genes were validated by qRT-PCR. The effects of Pioglitazone were investigated regarding apoptosis rate after 24-, 48- and 72-h. At 5.6 mM glucose, 101 genes were modulated by Pioglitazone, while 1,235 genes were affected at 23 mM glucose. Gene networks related to lipid metabolism were identified as altered by Pioglitazone at both glucose concentrations. At 23 mM glucose, cell cycle and cell death pathways were significantly regulated as well. At 5.6 mM glucose, Pioglitazone elicited a transient reduction in islets apoptosis rate while at 23 mM, Bcl2 expression was reduced and apoptosis rate was increased by Pioglitazone. Our data demonstrate that the effect of Pioglitazone on gene expression profile and apoptosis rate depends on the glucose concentration. The modulation of genes related to cell death and the increased apoptosis rate observed at supraphysiological glucose concentration raise concerns about Pioglitazone's direct effects in conditions of hyperglycemia and reinforce the necessity of additional studies designed to evaluate TZDs effects on the preservation of ß-cell function in situations where glucotoxicity might be more relevant than lipotoxicity.

The scrotal-testicular biometry was evaluated in goats raised in Piaui state, Brazil, presenting different levels of scrotal division, in rainy and dry periods of the year. For this study, eighteen male goats at mating age were accomplished and arranged into three groups (6 animals each), obeying the classification as goats with no scrotal bipartition (GI), goats showing scrotal bipartition up to 50 percent of testicular length (GII), and goats with more than 50 percent of scrotal bipartition (GIII). The biometry of the scrotal-testicular was made evaluating the scrotal length (SL), scrotal circumference (SC), testicular length (TL) and testicular volume (TV). The results were evaluated following the variance analysis (ANOVA) and the SNK test applied on the average comparisons. The analysis of the data demonstrated high values, in dry and rainy periods, of SC (24.63cm/ 26.97cm), SL (16.61cm/ 18.24cm), TL (5.32cm/ 5.93cm), TV (173.81cm³/ 203.01cm³). This supports the hypothesis of the influence of the period of the year and of the scrotal bipartition on the scrotal-testicular biometry in goat.(AU)


Objetivou-se neste trabalho avaliar a biometria escroto-testicular em caprinos com escroto simples e bipartido, criados no Estado do Piauí, Brasil, nos períodos seco e chuvoso do ano. Foram utilizados 18 caprinos machos, divididos em três grupos de seis caprinos. O grupo I (escroto sem bipartição), o grupo II (escroto bipartido até 50 por cento de comprimento testicular) e o grupo III (bipartição escrotal superior a 50 por cento do comprimento testicular). A biometria escroto-testicular consistiu do comprimento escrotal (COE), circunferência escrotal (CE), comprimento testicular (CT) e volume testicular (V). Os resultados foram submetidos à análise de variância (ANOVA), seguido do teste SNK para comparação das médias. Os dados mostraram que os animais do grupo GIII apresentaram, no período seco, os valores de CE, COE, CT e V de 24,63cm, 16,61cm, 5,32cm, e 173,81cm³, respectivamente e de 26,97cm para CE, 18,24cm para COE, 5,93cm para CT e 203,01cm³ para V, no período chuvoso. Todos esses valores foram superiores (p<0,05) aos encontrados para os animais dos demais grupos. Conclui-se que o grau de bipartição escrotal e o período do ano interferem na biometria escroto-testicular de caprinos.(AU)
