Agaricus bisporus (J.E. Lange) Imbach is one the most popular species of edible mushrooms in the world because of its taste and nutritional properties. In the research, repeatability of accumulation of bioelements and biomass yield in experimentally chosen in vitro culture medium, was confirmed. The in vitro cultures were conducted on the modified Oddoux medium enriched with bioelements (Mg, Zn, Cu, Fe). The aim of the study was to create an effective method of sampling, which enabled non-invasive monitoring of metals concentrations changes in the medium, during increase of biomass in in vitro cultures. The first, indirect method of sampling was applied. The non-invasive probe (a dipper) for in vitro culture was used; hence, the highest biomass increase and metals accumulation were gained. The method also guaranteed culture sterility. The second method, a direct one, interfered the in vitro culture conditions and growth of mycelium, and as a consequence the lower biomass increase and metals' accumulation were observed. Few cases of contaminations of mycelium in in vitro cultures were observed. The proposed method of non-invasive sampling of the medium can be used to monitor changes in the concentrations of metals in the medium and their accumulation in the mycelium in natural environment. Changes in concentrations of the selected metals over time, determined by the method of atomic absorption spectrometry, made it possible to correlate the obtained results with the specific stages of A. bisporus mycelium development and to attempt to explain the mechanism of sampling metals from the liquid substrate.

Superparamagnetic iron oxide nanoparticles (SPIONs) are promising drug delivery carriers and hyperthermia agents for the treatment of cancer. However, to ensure their safety in vivo, SPIONs must be modified in order to prevent unwanted iron release. Thus, SPIONs were coated with silica layers of different morphologies: non-porous (@SiO2), mesoporous (@mSiO2) or with a combination of non-porous and mesoporous layers (@SiO2@mSiO2) deposited via a sol-gel method. The presence of SiO2 drastically changed the surface properties of the nanoparticles. The zeta potential changed from 19.6 ± 0.8 mV for SPIONs to -26.1 ± 0.1 mV for SPION@mSiO2. The Brunauer-Emmett-Teller (BET) surface area increased from 7.54 ± 0.02 m2/g for SPIONs to 101.3 ± 2.8 m2/g for SPION@mSiO2. All types of coatings significantly decreased iron release (at least 10 fold as compared to unmodified SPIONs). SPIONs and SPION@mSiO2 were tested in vitro in contact with human lung epithelial cells (A549 and BEAS-2B). Both nanoparticle types were cytocompatible, although some delay in proliferation was observed for BEAS-2B cells as compared to A549 cells, which was correlated with increased cell velocity and nanoparticles uptake.

OBJECTIVES: The aim of this study was to compare the zinc and copper concentration in the group of patients with bipolar disorder (BD) and major depressive disorder (MDD). METHODS: 110 patients with the diagnosis of BD and 114 with MDD were qualified to the study. To assess the levels of microelements, the flame atomic absorption spectrometry (FAAS) was used in the case of zinc and the electrothermal atomic absorption spectrometry (ETAAS) was used in the case of copper. RESULTS: There were no differences between concentration of zinc and copper in remission and depressive phase between patients with BD and MDD. Additionally, there were also no statistically significant differences in comparisons including type I and II, early or late phase of BD and MDD. CONCLUSIONS: The lack of differences in zinc and copper concentrations between patients with bipolar disorder and major depressive disorder might indicate that those disorders have similar etiology.

This work presents, for the first time, DPV methodology of indirect aluminum (III) determination in tea leaves and infusions using the renewable silver amalgam electrode. A self-referencing strategy was proposed which relies on alternate voltammetric curves registration with and without conditioning of the electrode surface. The developed procedure utilizing formation of aluminum-Alizarin S complex shows linearity in the range of 2-50 µg L-1 of Al(III) concentration, with r > 0.997. In the case of the electrode conditioning LOD is 0.2 µgL-1 and repeatability expressed by CV is 1-2%. Introducing an additional operation i.e. electrode activation results in decrease of sensitivity from 0.0162 to 0.0043 µA/µgL-1. The method was verified using CRM (tea leaves). Due to the wide variety of tea origin, types and producers, four most popular sorts (black, green, red and white) were chosen, made by one manufacturer. It was demonstrated that the proposed methodology may be applied in Al(III) determination in tea leaves and infusions. Al(III) extraction effectiveness was also examined. It was verified, that lemon juice addition during brewing increases aluminum concentration in infusions. Using Principal Components Analysis it was demonstrated that automatic distinction of infusions brewed with and without lemon juice, based on unprocessed voltammetric signal, may be successfully realized.

The Sudety Mts. form a chain of mountains in the South of Poland and during the last 200 years were subjected to strong industrial and agricultural pressure. The records of these human-induced changes are stored in natural archives like lake sediments. For the comprehensive study, three sediment cores taken from Maly Staw Lake (Sudety Mts.) were analyzed for the concentration of K, Na, Mn, Fe, Cu, Mg, Zn, Cd, Cr, Ni, Pb and radioactivity of 137Cs and 210Pb. As a result of the studies, the bathymetry map was developed and the sources of solid material supplied to the lake were identified. The geochronology studies of the cores were performed using 210Pb method, to evaluate model of time changes in the sediment. Radioactivity of 210Pbuns (determined indirectly by 210Po) ranged from 1051 ± 64 to 12 ± 8 Bq kg-1. The 137Cs radioactivity was determined directly by gamma spectrometry and varied from 525 ± 37 Bq kg-1 for top layers to 9.80 ± 5.40 Bq kg-1 for the bottom of the core. Two characteristic peaks of 137Cs radioactivity related to the global fallouts after nuclear weapons testing and the Chernobyl accident were observed and used to confirm 210Pb dating method. Chemometrics analysis of the chosen metal's concentrations combined with sample dating showed distinct imprint of human activity on the studied area.

OBJECTIVES: Some scientific reports indicate the changes in the concentration of serum copper in patients with bipolar disorder (BD), however the data are inconclusive. The aim of this study was to assess the concentration of copper in the blood serum of patients in various phases of BD compared to healthy volunteers, taking into consideration the specific clinical features, and the stage of illness. METHODS: The study enrolled 133 patients with a diagnosis of BD (type I, II and NOS), including 61 people in depressive episode, 23 in mania or hypomania and 49 in remission. The control group consisted of 50 people. Atomic absorption spectrometry was used to measure the concentration of copper. RESULTS: There were no statistically significant differences in the serum copper concentration between patients in various phases of BD (mania/hypomania, depression, remission), sub-types (Type I, Type II + NOS) or stages and healthy volunteers. However, serum copper concentrations in patients in stage 1 was significantly higher than in advanced stages (2+3+4), (ß = 0.22; p = 0.02). Serum copper concentration was also the higher, the later the age of onset was (ß = 0.33; p < 0.001), and the lower, the greater the number of illness episodes (ß = - 0.23; p = 0.02) (multiple regression model, adj R2 = 0.19, p = 0.0001). CONCLUSIONS: The dependencies demonstrated above may reflect pathophysiological processes that occur in the course of BD (e.g., inflammatory response and oxidative stress) with a different intensity depending on its stage.

Despite many clinical trials assessing the role of zinc in major depressive disorder (MDD), the conclusions still remain ambiguous. The aim of the present clinical study was to determine and comparison the zinc concentration in the blood of MDD patients (active stage or remission) and healthy volunteers (controls), as well as to discuss its potential clinical usefulness as a biomarker of the disease. In this study 69 patients with current depressive episode, 45 patients in remission and 50 controls were enrolled. The zinc concentration was measured by electrothermal atomic absorption spectrometry (ET AAS). The obtained results revealed, that the zinc concentration in depressed phase were statistically lower than in the healthy volunteers [0.89 vs. 1.06 mg/L, respectively], while the zinc level in patients achieve remission was not significantly different from the controls [1.07 vs. 1.06 mg/L, respectively]. Additionally, among the patients achieve remission a significant differences in zinc concentration between group with and without presence of drug-resistance in the previous episode of depression were observed. Also, patients in remission demonstrated correlation between zinc level and the average number of depressive episodes in the last year. Serum zinc concentration was not dependent on atypical features of depression, presence of psychotic symptoms or melancholic syndrome, age, age of onset or duration of disease, number of episodes in the life time, duration of the episode/remission and severity of depression measured by the Hamilton Rating Scale for Depression (HDRS), and the Montgomery-Asberg Depression Rating Scale (MADRS). Concluding, our findings confirm the correlation between zinc deficit present in the depressive episode, and are consistent with the majority of previous studies. These results may also indicate that serum zinc concentration might be considered as a potential biological marker of MDD.

Copper may be involved in the pathophysiology of depression. Clinical data on this issue are very limited and not conclusive. The purpose of the study was to determine the copper concentration in the serum of patients with major depressive disorder and to discuss its potential clinical usefulness as a biomarker of the disease. A case-control clinical study included 69 patients with current depressive episode, 45 patients in remission and 50 healthy volunteers. Cu concentration was measured by electrothermal atomic absorption spectrometry (ETAAS). The mean serum copper level in depressed patients was slightly lower (by 11 %; not statistically significant) than in the control group. Furthermore, there was no significant difference in Cu2+ concentration between depressive episode and remission, nor between remission and control group. In the remission group were observed significant correlations between copper levels and the average number of relapses over the past years or time of remission. There was no correlation between serum copper and severity of depression, as measured by HDRS and MADRS. The obtained results showed no significant differences between the copper concentration in the blood serum of patients (both with current depressive episode and in remission) and healthy volunteers, as well as the lack of correlations between the copper level in the active stage of the disease and clinical features of the population. Our study is the first conducted on such a large population of patients, so the results may be particularly important and reliable source of knowledge about the potential role of copper in depression.

Agaricus bisporus species (J.E. Lange) Imbach one of the most popular Basidiomycota species was chosen for the research because of its dietary and medicinal value. The presented herein studies included determination of essential mineral accumulation level in the mycelium of A. bisporus, cultivated on liquid cultures in the medium supplemented with addition of the chosen metals' salts. Quantitative analyses of Zn, Cu, Mg, and Fe in liquid cultures made it possible to determine the relationship between accumulation of the selected mineral in A. bisporus mycelium and the culture conditions. Monitoring of the liquid cultures and determination of the elements' concentrations in mycelium of A. bisporus were performed using the flame technique of AAS method. Concentration of Zn in the mycelium, maintained in the medium with the addition of its salt, was in a very wide range from 95.9 to 4462.0 mg/g DW. In the analyzed A. bisporus mycelium, cultured in the medium enriched with copper salt, this metal concentration changed from 89.79 to 7491.50 mg/g DW; considering Mg in liquid cultured mycelium (medium with Mg addition), its concentration has changed from 0.32 to 10.55 mg/g DW. The medium enriched with iron salts has led to bioaccumulation of Fe in mycelia of A. bisporus. Determined Fe concentration was in the range from 0.62 to 161.28 mg/g DW. The proposed method of liquid A. bisporus culturing on medium enriched with the selected macro- and microelements in proper concentrations ratio have led to obtaining maximal growth of biomass, characterized by high efficiency of the mineral accumulation. As a result, a dietary component of increased nutritive value was obtained.

Element content, loss-on-ignition, chironomid analysis and (210)Pb dating were applied on a sediment core from a subalpine Tatra lake (Popradské pleso) to reveal the response of aquatic biota to eutrophication induced by human activities in the lake catchment. The lead dating indicates that the 0-8 cm section of the core represents the past ca 200 years, ending at ~1814 AD. Comparing the key changes of the proxies with human activities that are historically well documented, four phases of the recent lake development were distinguished: (1) a pre-tourism phase, (2) a phase of increasing touristic activity and early cottage development, (3) a phase of eutrophication, and (4) a phase of post-eutrophication. Neither touristic activity, nor early cottage development around the lake (1st and 2nd phases) had considerable influence on the chironomid assemblage structure or organic content of the lake. The most significant change both in chironomid assemblage structure and loss-on-ignition occurred during the 3rd phase, when a big tourist hotel was built close by the lake and started contaminating it via direct wastewater input. However, the structure of the chironomid assemblage has not changed significantly over time and the dominating taxa remained the same during the whole period. Parallel with the nutrient signal of the paleo assemblage, a secondary signal has been identified as the ratio of rheophilic taxa on total abundance that did not correlate with the sediment's organic content, and is most likely driven by local climatic oscillations. Changes of most of metal elements concentrations reflected rather bigger scale changes of industrial activities than local scale human disturbances. Our results indicate that hydromorphological properties can moderate the impact of organic pollution on the lake biota.

OBJECTIVES: Zinc may be involved in the pathophysiology and treatment of depressive disorder. However, data on this issue in bipolar disorder (BD) are limited. The aim of the study was to assess zinc concentrations in the blood serum of patients at various phases and stages of bipolar disorder. METHODS: The study included 129 patients with a diagnosis of bipolar disorder type I (n=69) or type II (n=60). Fifty-eight were in a depressive episode, 23 in a manic episode and 48 in remission. Fifty healthy volunteers made a control group. Zinc concentration was measured using flame atomic absorption spectrometry. RESULTS: Serum zinc level in patients diagnosed with BD type I in the depressive phase was significantly reduced as compared with mania, remission and healthy subjects. In the BD type II, serum zinc level in hypomania, depression or remission phase was not significantly different from the control group. In the whole group, lower level of zinc in depression compared to remission and control subjects was found during late stage of the illness but not in the early stage. Zinc concentration was not dependent on the severity of manic or depressive symptoms and subtype of depression but correlated positively with the number of manic/hypomanic relapses in the past year. LIMITATIONS: Lack of prospective model, heterogeneity of pharmacological treatment, small number of subgroups presenting specified clinical features. CONCLUSIONS: Decreased serum zinc concentration occurs in depression in BD type I and probably in depression in the late stage of BD.

Zinc can regulate neural function in the brain via the GPR39 receptor. In the present study we investigated whether inhibition of serotonin, noradrenaline and dopamine synthesis and potentialization of glutamate, via administration of p-chlorophenylalanine (pCPA), α-methyl-p-tyrosine (αMT) and N-methyl-D-aspartatic acid (NMDA), respectively, would cause changes in GPR39 levels. Western blot analysis showed GPR39 up-regulation following 3-day administration of αMT and NMDA in the frontal cortex, and GPR39 down-regulation following 10-day administration of pCPA, αMT, and NMDA in the hippocampus of CD-1 mice. There were no changes in serum zinc levels. Additionally, we investigated tryptophan, tyrosine and glutamate concentrations in the hippocampus and frontal cortex of GPR39 knockout (GPR39 KO) mice. Liquid chromatography-mass spectrometry (LC-MS) showed a significant decrease in tryptophan and tyrosine, but not in glutamate concentrations in the hippocampus of GPR39 KO mice. There were no changes in the frontal cortex between GPR39 KO and wild type. These results indicate a possible role of the GPR39 receptor in monoaminergic and glutamatergic neurotransmission, which plays an important role in the pathophysiology of depression.

AIM: The growing body of evidence suggests that magnesium levels can serve as a marker of major depressive disorder (MDD), but findings from clinical trials remain inconclusive. The aim of the presented study was to determine the magnesium concentration in serum of patients with MDD (in the active stage of the disease or in remission) and to analyze the role of magnesium levels as apotential marker of the disease. METHODS: Sixty-nine patients with current depressive episode, 45 patients in remission and 50 healthy volunteers were enrolled into the case-control study. The magnesium concentration was measured by flame atomic absorption spectrometry (FAAS). RESULTS: The mean serum magnesium concentration of patients in the depressed phase was significantly higher, compared to the control group. Moreover, magnesium levels of patients in the remission were not significantly different from the concentrations recorded in the healthy volunteers. There was also a positive correlation between the magnesium levels and the severity of depression measured by the Hamilton Rating Scale for Depression (HDRS) and the Montgomery-Asberg Depression Rating Scale (MADRS). CONCLUSIONS: The obtained results may suggest a role of magnesium as a state marker reflecting the pathophysiological changes underlying MDD and accompanying severe depressive episodes.

Therefore, the aim of this study was to evaluate the serum magnesium level in patients with bipolar disorder (in different phases of the disease) in comparison to healthy volunteers. METHODS: The study included 129 patients (58 subjects in depressive episode, 23 in manic episode and 48 patients in remission) with the diagnosis of bipolar disorder type I or II. The control group consisted of 50 healthy people. Magnesium concentration was measured using flame atomic absorption spectrometry (FAAS). RESULTS: Patients with a current depressive or manic/hypomanic episode had statistically significantly elevated serum magnesium levels compared to healthy volunteers. Moreover, a positive correlation between the duration of the manic/hypomanic episode and the relapse frequency in the last year was observed. The concentration of magnesium in patients in remission was unchanged in relation to the control group. CONCLUSIONS: Presented findings suggest a role of serum magnesium level as a potential state marker, reflecting the pathophysiological changes associated with acute episodes of bipolar disorder.

Bacopa monnieri is one of the most interesting plants from the Ayurveda system. The aims of present research were, basing on in vitro shoot culture of B. monnieri, to determine content and to evaluate the influence of physiologically important metabolites on the selected bioelements accumulation in biomass. The most significant increase in biomass production was observed in the culture medium enriched with 0.5 mg/L of anthranilic acid. In this medium also, the highest accumulation of Mg was noted. The highest concentration of iron was determined in B. monnieri in vitro culture enriched with 0.25 g/L of serine. The addition of L-tryptophan, magnesium sulfate, and zinc hydroaspartate caused only a small increase in the accumulation of copper in B. monnieri. Increase in Zn accumulation was obtained in biomass from in vitro culture of B. monnieri with the addition of magnesium sulfate and zinc hydroaspartate. In the case of Na, the maximum level of this element was in biomass from medium enriched with zinc hydroaspartate. Twofold increase in K concentration was obtained in biomass from cultures on medium with addition of serine and magnesium sulfate. The concentrations of Ca in biomass of all studied media were at the similar level.

It has been demonstrated that zinc exerts its beneficial influence on skin fibroblasts. Propolis, a complex mixture of plant-derived and bees' products, was reported to stimulate cicatrization processes in skin and prevent infections. The aim of this study was to find out how zinc and propolis influence human skin fibroblasts in cell culture and to compare the effect of individual compounds to the effect of a mixture of zinc and propolis. In this study, zinc, as zinc aspartate, at a concentration of 16 µM, increased human fibroblasts proliferation in cell culture, whereas propolis at a concentration of 0.01% (w/v) revealed antiproliferative and cytotoxic action followed by mild cell necrosis. In culture, zinc was effectively transported into fibroblasts, and propolis inhibited the amount of zinc incorporated into the cells. An addition of propolis to the medium caused a decrease in the Zn(II) amount incorporated into fibroblasts. The obtained results also indicate an appreciable antioxidant property of propolis and revealed its potential as a supplement when applied at doses lower than 0.01% (w/v). In conclusion, the present study showed that zinc had a protective effect on human cultured fibroblasts' viability, although propolis revealed its antiproliferative action and caused mild necrosis.

RESUMEN

The aim of the study is to present the results of determination of radioactivity of artificial 137Cs and natural 40K and certain heavy metals in soil samples collected from the eastern part of the Main Ridge of Carpathians, including the Beskid Niski Mts and the Bieszczady Mts. The evaluation of level of radionuclides was based on the bulk density analysis of the soil. A valuable finding of the study was a good linear correlation between the level of 137Cs concentration and bulk density of the soil as well as an inverse correlation between radioactivity of natural 40K and tested soil density. This might indicate though a high competitiveness of these elements between each other. Moreover, a good correlation between the concentrations of artificial element 137Cs and Pb has been also observed in soil samples collected from the Beskid Niski Mts. In most cases, the level of artificial 137Cs was lower comparing to an average 137Cs concentration established for soils in Poland.

BACKGROUND: There is some evidence coming from preclinical and clinical studies suggesting a relationship between dietary zinc intake and depressive symptoms. The aim of the study was to determine whether zinc deficiency alters the response to antidepressants with a different mechanism of action. We examine also whether these changes are related to activity of the hypothalamic-pituitary-adrenal HPA axis. METHODS: Male CD-1 mice were assigned to groups according to diet and antidepressant administration. To evaluate animal behavior, the immobility time in the forced swim test (FST) and locomotor activity were measured. To determine serum zinc levels the flame atomic absorption spectroscopy (FAAS) was used. The serum corticosterone was determined by radioimmunoassay (RIA). RESULTS: Antidepressants administered to zinc-deprived mice induced an altered response in the FST when compared to animals fed with an adequate diet. There were no changes in locomotor activity. Animals subjected to a zinc-deficient diet showed a significant reduction in serum zinc levels, which was normalized by antidepressant treatment. An increase in serum corticosterone concentrations in mice fed with a zinc-deficient diet and treated with antidepressants was observed, so it can be concluded that reduced levels of zinc contribute hyperactivation of the HPA axis. CONCLUSION: The results of this study suggest that a diet with a reduced zinc level alters antidepressant action, which is associated with a reduction in the serum zinc level and rise in the corticosterone level. The results of this study may indicate the involvement of zinc deficiency in the pathogenesis of depression.

Cantharellus cibarius Fr. (chanterelle) and Boletus badius Pers. (bay bolete) harvested from natural sites in Poland were used to derive in vitro cultures. The optimal medium composition for cultures was developed. Concentrations of the chosen elements (Zn, Cu, Fe, Mg, Ni, and Cd) in mycelium samples were measured by means of atomic absorption spectrometry. Fe concentration in the analyzed mushroom materials was in the range 215.4-680.3 µg/g dry weight. Mean values of Mg were respectively (in micrograms per gram dry weight) 541.8 for mycelium of C. cibarius cultured in vitro and 1,004.1 for C. cibarius fruiting bodies and 928.9 for the mycelium of B. badius cultured in vitro and 906.4 for B. badius fruiting bodies. The mean concentrations of Zn were 442.7 µg/g dry weight in mycelium from in vitro cultures of B. badius and 172.1 in B. badius fruiting bodies and 131.9 in the case of C. cibarius in mycelium from in vitro cultures and 95.5 for the C. cibarius fruiting bodies. Cu exhibited a reversal tendency, i.e., the element concentrations in naturally grown mushrooms were significantly higher (43.57 µg/g dry weight for C. cibarius and 43.54 µg/g for B. badius) than in cultured in vitro mycelium (12.47 µg/g for C. cibarius and 4.17 µg/g for B. badius). Ni was found in lowest concentrations ranging from 0.33 to 1.88 µg/g dry weight. Toxic metal Cd was found in relatively high concentrations in naturally grown species (0.79 µg/g dry weight-1.02). The lowest was the concentration of Cd in C. cibarius mycelium from in vitro culture-0.06 µg/g dry weight-a bit higher than it was in the B. badius mycelium (0.21 µg/g).

The aim of the study was to determine whether using chemical and radiochemical analysis of lake sediments can highlight changes in the climate. Also it was studied whether human impact on the environment can be observed and to what extent such changes are in agreement with historical data. Samples of 16 cm thick sediment cores from the Smreczynski Staw Lake were collected and divided into 1 cm thick sub-samples. The samples were air dried and homogenized. The quantitative analysis of Fe, Mn, Zn, Cr, Cu, Ni, Cd, and Pb in the digested sediment samples was made by using atomic absorption spectrometry. Simultaneously, the radioactivity of 137Cs using gamma spectrometry and 210Pbuns using alpha spectrometry, were measured for sediment layer dating. Results showed that iron concentration was in the range 0.3-over 1 % (w/w), and zinc 0.01-0.05 % (w/w). Lesser concentrations were found for copper 18.37-43.6 ppm, manganese 37.5-50.7 ppm, lead 146.1-432 ppm, chromium 12.3-37.4 ppm, nickel 3.1-10.8 ppm and cadmium 0.9-34.6 ppm. Changes in 137Cs radioactivity was in the range of 89 ± 11 to 865 ± 62 (Bq kg-1). Sediments composition can accurately reflect (in terms of time and to what extent) air pollution and natural geo-chemical processes in the environment. However, the choice of the analysed object is crucial in this respect. The Smreczynski Staw Lake, due to its location in the mountains and hydrological situation, proved to be very useful for providing undisturbed analytical samples.
