RESUMEN

The genetic diversity and population structure of Arabidopsis thaliana populations from Norway were studied and compared to a worldwide sample of A. thaliana to investigate the demographic history and elucidate possible colonization routes of populations at the northernmost species limit. We genotyped 282 individuals from 31 local populations using 149 single nucleotide polymorphism markers. A high level of population subdivision (F(ST) = 0.85 ± 0.007) was found indicating that A. thaliana is highly structured at the regional level. Significant relationships between genetic and geographical distances were found, suggesting an isolation by distance mode of evolution. Genetic diversity was much lower, and the level of linkage disequilibrium was higher in populations from the north (65-68°N) compared to populations from the south (59-62°N); this is consistent with a northward expansion pattern. A neighbour-joining tree showed that populations from northern Norway form a separate cluster, while the remaining populations are distributed over a few minor clusters. Minimal gene flow seems to have occurred between populations in different regions, especially between the geographically distant northern and southern populations. Our data suggest that northern populations represent a homogenous group that may have been established from a few founders during northward expansions, while populations in the central part of Norway constitute an admixed group established by founders of different origins, most probably as a result of human-mediated gene flow. Moreover, Norwegian populations appeared to be homogenous and isolated compared to a worldwide sample of A. thaliana, but they are still grouped with Swedish populations, which may indicate common colonization histories.

RESUMEN

The "BF14/16xHF2/7" mapping population of meadow fescue (Festuca pratensis Huds.) was characterised for number of panicles produced by non-vernalised plants in the field, vernalisation requirement (number of weeks at 6 degrees C and 8 h photoperiod), as well as days to heading, number of panicles and proportion of shoots heading after a 12 weeks vernalisation treatment. Quantitative trait loci (QTLs) were identified and compared to QTLs and genes related to the induction of flowering in cereals and grasses. A region on chromosome 1F affected days to heading and the proportion of shoots heading. Chromosome 4F appeared to have several genes with a strong effect on vernalisation requirement. The strongest effects were located in the proximal end of 4F and may correspond to the earliness per se (eps) QTL eps6L.2 in barley and a heading time QTL in perennial ryegrass. A part of the meadow fescue orthologue of VRN1 was sequenced and mapped to another region of 4F that also had a strong effect on vernalisation requirement. The proximal end of chromosome 5F had QTLs for days to heading and proportion of heading shoots. Syntenic regions in wheat and barley contain eps-loci. A QTL for number of panicles in the field and a QTL for proportion of heading shoots were present on chromosome 6. A region on 7F affected the variation in number of panicles among plants without a vernalisation requirement, and is syntenic to regions in perennial ryegrass, barley and rice containing orthologues of Arabidopsis thaliana CO.

Genetic diversity and relatedness were studied in 30 Norwegian local populations of meadow fescue (Festuca pratensis Huds.) using amplified fragment length polymorphism (AFLP) markers. The populations were also compared with 13 Nordic meadow fescue cultivars in order to analyse the distribution of variation in local populations and cultivars and to elucidate relationships between local populations and cultivars. Analysis of molecular variance (AMOVA: ) analysis showed that most of the variation was present within populations and that little variation was found between local populations and cultivars. Separate AMOVA: analyses of local populations and cultivars revealed a higher level of variation within registered cultivars than within local populations. A cluster analysis based on corrected average pairwise differences between populations showed that the populations could be divided into three clusters, of which one also contained the cultivars. These results were supported by principal coordinates analysis. The results indicate that the Norwegian meadow fescue has a narrow genetic basis and that the local populations in Norway can be divided into three groups following the most probable routes of introduction of the species into Norway. The inland populations are closely related to the cultivars and have most probably been established as a result of migration from sown meadows. The western and southern populations probably originate from human activity-for example, trade-to the coastal western and northern parts of the country and to the central parts of southern Norway.

A genetic linkage map has been constructed for meadow fescue ( Festuca pratensis Huds.) (2n=2 x=14) using a full-sib family of a cross between a genotype from a Norwegian population (HF2) and a genotype from a Yugoslavian cultivar (B14). The two-way pseudo-testcross procedure has been used to develop separate maps for each parent, as well as a combined map. A total number of 550 loci have been mapped using homologous and heterologous RFLPs, AFLPs, isozymes and SSRs. The combined map consists of 466 markers, has a total length of 658.8 cM with an average marker density of 1.4 cM/marker. A high degree of orthology and colinearity was observed between meadow fescue and the Triticeae genome(s) for all linkage groups, and the individual linkage groups were designated 1F-7F in accordance with the orthologous Triticeae chromosomes. As expected, the meadow fescue linkage groups were highly orthologous and co-linear with Lolium, and with oat, maize and sorghum, generally in the same manner as the Triticeae chromosomes. It was shown that the evolutionary 4AL/5AL translocation, which characterises some of the Triticeae species, is not present in the meadow fescue genome. A putative insertion of a segment orthologous to Triticeae 2 at the top of 6F, similar to the rearrangement found in the wheat B and the rye R genome, was also observed. In addition, chromosome 4F is completely orthologous to rice chromosome 3 in contrast to the Triticeae where this rice chromosome is distributed over homoeologous group 4 and 5 chromosomes. The meadow fescue genome thus has a more ancestral configuration than any of the Triticeae genomes. The extended meadow fescue map reported here provides the opportunity for beneficial cross-species transfer of genetic knowledge, particularly from the complete genome sequence of rice.

Pollen dispersal and gene flow in the grass meadow fescue (Festuca pratensis Huds.) were studied using two populations which were homozygous for different allozymes at the Gpi-2 locus. The populations were established in a concentric donor-acceptor field experiment. Gene flow was found mainly to be affected by the distance between the donor and acceptor plants. Analysing 21 132 acceptor plant progenies, gene flow was shown to decrease rapidly with distance to the donor field up to 75 m, and beyond this distance much more slowly. The ability of donor pollen to fertilize acceptor plants depended very much on the density of the acceptor plants. Pairs of acceptor plants produced more compatible pollen locally, and captured significantly less donor pollen than single-plants. Despite the higher seed production of acceptor plants planted in pairs, the absolute number of heterozygous seeds carrying the donor allele was always lower than for single-plants. Wind direction had only a slight effect upon the type of pollen captured. Because of pollen production within the two plant populations being continuous and overlapping, the time when anthesis occurred had little effect on gene flow between the populations. Vigorous and tall acceptor plants with many panicles, high seed yield and high 1000-seed weight were able to capture more donor pollen than shorter plants. The results may be used to assess the risk of gene flow and to develop strategies for monitoring the spread of transgenes from genetically modified grasses.

The suitability of regression analysis for studying the phenotypic stability of grain yield was investigated using a collection of 220 Nordic barley lines. Linear regression explained 26-52% of the genotype x environment (GE) interactions in different groupings of the material. The regression coefficient, b i , measures the yield response of the i-th genotype to improved environmental conditions. Deviations from regression, S di (2) , have been used to estimate Tai's stability parameter, λ i , which is a measure of the phenotypic yield stability in the agronomic sense. Repeatability of b i , λ i , and grain yield was studied by means of correlations between estimates obtained in each experimental year. Yield had the highest repeatability, with correlations between years ranging from 0.57 to 0.85. In this study, regression coefficients and λ i -values were not repeatable, i.e. genotypes reacted differentially to the yearly climatic variations. Six-rowed (6r) barleys had higher responsiveness, but lower mean yields, than two-rowed (2r) barleys. This is partly due to the history of selection of 6r-barleys, which mainly originate from regions with low potential yield levels, i.e. Finland and Norway. In general, responsiveness and stability were not correlated with yield. The highest-yielding lines had b i ≈1. The response pattern of the different types of barleys used in this study show that responsiveness can be changed by recombination.

In a study of androgenesis in 90 Norwegian genotypes of perennial ryegrass (Lolium perenne L.), heritabilities ranged from h b (2) =0.46 to 0.80. Very high or completely positive genotypic correlations were found between most characters of androgenetic response (e.g. embryo-like structures per 100 anthers, plants per 100 embryo-like structures, albino plants per 100 anthers, green plants per 100 anthers). Three genotypes, 2 Norwegian (7-5 and 9-5) and 1 Danish (245), which had significantly different androgenetic responses were selected to study the genetic control of the processes. Genotypes 7-5 and 9-5 were highly embryogenie, 7-5 and 245 were relatively high producers of green plants, while 9-5 was unable to produce green plants. The six possible reciprocal crosses between these three genotypes were made, and 10 or 11 F1 plants from each cross were used for anther culture experiments. The cross 7-5 x 245 showed average superiority over both parents for total plant regeneration and green plant production, results not previously reported. The phenotypic correlations estimated among progenies from the crosses ranged from r=-0.99(***) to 0.81(***). These considerable changes, relative to the results of the screening experiment, are most likely the result of changed allele frequencies caused by the strong selection of parents in these crosses, and a relatively simple genetical control. This is also inferred from the large transgressive segregation observed.

Genotypes of Lolium perenne L. with different androgenetic responses were used to test effects of induction medium composition. The media tested were potato II (pII), 190-2, and modified Linsmaier and Skoog media, LS-1, LS-2, and LS-3. The effect of different gelling agents, activated charcoal in a 'double layer' design, and casein hydrolysate were also studied. From 36,696 anthers, 25,906 embryo-like structures, 1,959 albino and 173 green plants were generated. Significant differences were found between media, genotypes and medium-genotype interactions studied. All three media commonly used, pII, 190-2, and LS-3, were equivalent in production of green plants. Cold pretreatment of the anthers (4°C) significantly increased the number of embryo-like structures, the number and proportion of albino plants produced, but not the production of green plants.
