RESUMEN

Ceruloplasmin as a modulator was used for the first time in the clinical practice in 30 patients with gastric cancer in the course of postoperative intraabdominal adjuvant chemotherapy. It was established that ceruloplasmin was well tolerated by the patients and its use during polychemotherapy is accompanied by normalization of the levels of natural ceruloplasmin and iron in the blood serum that are usually reduced in patients with gastric cancer, reduces activation of lipid peroxidation and improves the course of postoperative period. Ceruloplasmin possesses a week immunomodulating action in polychemotherapy which is expressed in the increase of the percentage content of T-lymphocytes. Use of ceruloplasmin tended to improve the functional state of the cardiovascular system.

RESUMEN

Ceruloplasmin administered 60 min before irradiation diminished cAMP and cGMP levels, which were increased by irradiation at LD50 and LD100, and normalized cAMP/cGMP ratio in the rat liver during the first 24 h following irradiation with a dose of 6.24 Gy. The content of phospholipids increased and that of cholesterol decreased under the effect of ceruloplasmin leading to normalization of the molar cholesterol/phospholipid ratio in the rat liver on the 7th day of radiation sickness (LD50, 6.24 Gy).

There were found certain changes in the total content of phospholipids as well as in the content of their separate fractions in the membranes of endoplasmic reticulum in the rat liver at early stages of carcinogenesis. In the first week under the carcinogen DENA exposure a considerable reduction of the total phospholipids content was noted in the fraction of granular endoplasmic reticulum, while in agranular reticulum during this period of embryogenesis the total content of phospholipids is not changed, but subsequently the former increases as compared with normal values. During the whole period of carcinogenesis under study changes in the phospholipid content are observed in both fractions of endoplasmic reticulum membranes, the greatest changes being noted in the fractions of lysophosphatidicholines, phosphatidilcholines, and phosphatidilinosites.

RESUMEN

The level of the ATPase activity in the skeletal muscles nuclei with experimental muscular dystrophy and the sensitivity to bivalent (Mg2+, Ca2+) and univalent (Na+, K+) cations under conditions of delipidation were studied. It is established that among ATPases of the young rabbit skeletal muscles nuclei there is ATPase sensitive to monovalent cations: the presence of Na+ or K+ produces a 45% increase in its activity in some experiments as compared to the initial level. This activation is attributed to Mg2+, Ca2+-ATPase the action of which is not realized in the presence of EGTA-chelator of calcium ions. A decrease in the total ATPase activity in the skeletal muscles nuclei resulted from the experimental muscular dystrophy development occurs due to a decrease in the Mg2+, Ca2+-ATPase activity as the ability for activation by the monovalent cations is practically lost under these conditions. Delipidation of the skeletal muscles nuclei, which results in their loss of some phospholipids and cholesterin, is accompanied by the ATPase activity decrease. At the same time the nuclei ATPase activity lose its ability to activate by monovalent cations: Na+, K+. A conclusion is made that during delipidation the decrease in the total ATPase activity is due to a decrease in the activity of its Mg2+, Ca2+-part.

In the rabbit sarcolemma both in norm and under experimental muscular dystrophy besides cholesterol there are found squalene and lanosterol - predecessors of its synthesis as well as a series of other sterols which are not yet identified. The cholesterol fraction of sarcolemma of the rabbits in norm and under muscular dystrophy contain so-called rapid sterols. Under experimental muscular dystrophy in the rabbit sarcolemma side by side with an increase in the content of cholesterol content the concentration of rapid sterols increases. It is also found that under mentioned pathology desmosterol arises in the rabbit sarcolemma.

It is shown that simultaneous treatment with sarcolemma with triton X-100 (0.1-1.0%) and KCl (0.6-1.2 M) solubilizes about 60% of acetyl cholinesterase, however its specific activity in the intact sarcolemma and in the obtained fractions is the same. A considerable part of lipids (about 90% of phospholipids and 50% of cholesterol) may be removed by treating sarcolemma with 0.1% triton X-100; it may be fixed in the membrane by hydrophoric bonds. Other phospholipids and cholesterol are bound with sarcolemma proteins more strongly and, probably, form the protein-lipid complexes. In the composition of phospholipids strongly bound with sarcolemma proteins a relative content of sphingomyelin increases and that of lysolecithin decreases.

The cholesterin content, total content and phsopholipid composition were studied in sarcolemma of normal rabbits and those with experimental muscular dystrophy. The total content of phospholipids in the sarcolemma of young normal rabbits is 4.8 mg of phosphorus per 1 mg of protein which is higher than that in the sarcolemma of adult rabbits 33%. With experimental muscular dystrophy the content of cholesterin is 2.7 as high as its content in the sarcolemma of the normal rabbits. It is established that the content of phosphatidilserine in the sarcolemma of young normal rabbits is by 61% higher than that in adult animals. The content of phosphatidilserine of lysophosphatidilcholine decreases by 40 and 39%, respectively, with dystrophy. The content of phosphatidilcholine increases by 16% as compared with sarcolemma extracted from the muscles of the young normal rabbits. Molar ratio of phospholipid/cholesterine in the sarcolemma of normal rabbits is 1.8 and decreases with the experimental muscular dystrophy to 0.75.
