RESUMEN

Short heat waves (SHW), defined as periods of several consecutive days with high temperatures above the developmental optimum, will become more frequent due to climate change. The impact of SHW on yield and yield-related parameters has received considerable interest, but their effects on grain quality remain poorly understood. We employed a simulation approach to investigate the impact of SHW on durum wheat quality over a 7 day period, starting 1 week after anthesis. During the SHW treatment, carried out using portable polyethylene tents, the temperature in the treated plots increased by 10-15 °C during daily hours. The SHW treatment reduced the number of grains per spike, thousand kernel weight, and total carotenoid content in grains in stressed plants in comparison to control plants. However, no differences in the protein content or percentage of vitreous grains were observed. The behavior of individual carotenoids in response to SHW appears to differ, suggesting a differential change in the balance between ß,ε- and ß,ß-branches of the carotenoid biosynthetic pathway as a consequence of SHW-induced stress. The present study highlights the importance of developing efficient breeding strategies for reduced sensitivities to heat stress. Such strategies should not only prioritize yield but also encompass grain quality.

Surface enhanced Raman spectroscopy (SERS) is gaining importance as sensing tool. However, wide application of the SERS technique suffers mainly from limitations in terms of uniformity of the plasmonics structures and sensitivity for low concentrations of target analytes. In this work, we present SERS specimens based on periodic arrays of 3D-structures coated with silver, fabricated by silicon top-down micro and nanofabrication (10 mm × 10 mm footprint). Each 3D-structure is essentially an octahedron on top of a pyramid. The width of the top part-the octahedron-was varied from 0.7 µm to 5 µm. The smallest structures reached an analytical enhancement factor (AEF) of 3.9 × 107 with a relative standard deviation (RSD) below 20%. According to finite-difference time-domain (FDTD) simulations, the origin of this signal amplification lies in the strong localization of electromagnetic fields at the edges and surfaces of the octahedrons. Finally, the sensitivity of these SERS specimens was evaluated under close-to-reality conditions using a portable Raman spectrophotometer and monitoring of the three vibrational bands of 4-nitrobenzenethiol (4-NBT). Thus, this contribution deals with fabrication, characterization and simulation of multiscale 3D-structures with SERS activity.

We have investigated the lithium capacity of the 2H phase of niobium sulfide (NbS2) using density functional theory calculations and experiments. Theoretically, this material is found to allow the intercalation of a double layer of Li in between each NbS2 layer when in equilibrium with metal Li. The resulting specific capacity (340.8 mAh/g for the pristine material, 681.6 mAh/g for oxidized material) can reach more than double the specific capacity of graphite anodes. The presence of various defects leads to an even higher capacity with a partially reversible conversion of the material, indicating that the performance of the anodes is robust with respect to the presence of defects. Experiments in battery prototypes with NbS2-based anodes find a first specific capacity of about 1,130 mAh/g, exceeding the theoretical predictions.

Introduction: Understanding immune cell dynamics in kidney transplantation may provide insight into the mechanisms of rejection and improve patient management. B cells have gained interest with a special relevance of the "regulatory" subsets and their graft outcome prognostic value. In this study, we aimed to prove that the direct immunophenotyping and target gene expression analysis of kidney transplant patients' fresh whole blood will help to identify graft rejection risk and assist in the monitoring of kidney transplanted patients. Methods: We employed flow cytometry and qPCR techniques to characterize B and T cell subsets within fresh whole blood samples, with particular emphasis on transitional B cells (TrB) identified as CD19+CD24hiCD38hi. TrB are a relevant population in the context of kidney transplantation and are closely associated with regulatory B cells (Bregs) in humans. Patients were monitored, tracking pertinent clinical parameters and kidney-related events, including alterations in graft function and episodes of biopsy proven rejection. Results: Higher percentages of TrB cells at 3 months after transplantation were positively associated with better graft outcomes and lower biopsy-proven acute rejection risk. Furthermore, a novel panel of B cell regulatory associated genes was validated at 3 months post-transplantation by qPCR analysis of peripheral blood mononuclear cell (PBMC) mRNA, showing high predictive power of graft events and prognostic value. Discussion: These findings suggest that monitoring TrB may provide interesting patient management information, improve transplant outcomes, and allow for personalized drug regimens to minimize clinical complications.

The balance between the CO2 entry for photosynthesis and transpiration water loss is crucial for plant growth, and ABA signaling can affect this equilibrium. To test how ABA balances plant growth and environmental adaptation, we performed molecular genetics studies in the biotech crop Nicotiana benthamiana under well-watered or drought conditions. Studies on ABA signaling in crops are complicated by the multigenic nature of the PYR/PYL/RCAR ABA receptor family and its functional redundancy, which is particularly challenging in polyploid plants. We have generated a pentuple pyl mutant in the allotetraploid Nicotiana benthamiana through CRISPR/Cas9 gene editing. The pentuple mutant is impaired in 2 NbPYL1-like and 3 NbPYL8-like receptors, affecting the regulation of transpiration and several ABA-dependent transcriptional processes. RNA-seq and metabolite analysis revealed that the synthesis of galactinol, an essential precursor for the osmoprotective raffinose family of oligosaccharides, is ABA-dependent and impaired in the mutant under osmotic stress. In contrast, our results show that, under well-watered conditions, partial inactivation of ABA signaling leads to higher CO2 entry and photosynthesis in the mutant than in WT. Photosynthesis analyses revealed an increased CO2 diffusion capacity mediated by higher stomatal and mesophyll conductances, and higher substomatal CO2 concentration in the pentuple mutant. RNA-seq analyses revealed that genes associated with cell wall loosening (e.g., expansins) and porosity were strongly downregulated by ABA in WT. In summary, a partial relief of the ABA control on transpiration mediated by ABA receptors positively affects photosynthesis when water is not limited, at the expense of reduced water use efficiency.

Recent developments in the application of aperiodic fiber Bragg gratings (AFBGs) in astrophotonics, such as AFBG for astronomical near-infrared OH suppression and gas detection based on cross-correlation spectroscopy, have illuminated the problem that the optimization for AFBG with certain fabrication constraints has not been fully investigated and solved. Previous solutions will either sacrifice part of the spectral features or consume a significant amount of computation resources and time. Inspired by recently successful applications of artificial neural networks (ANNs) in photonics inverse design, we develop an AFBG optimization approach employing ANNs in conjunction with genetic algorithms (GAs) for the first time, to the best of our knowledge. The approach maintains the spectral notch depths and preserves the fourth-order super-Gaussian spectral features with improvements of interline loss by ∼100 times. We also implement, to our knowledge, the first inverse scattering neural network based on a tandem architecture for AFBG, using a first-order Gaussian notch profile. The neural network successfully converges but has a poor predictive capability for the phase part of the design. We discuss possible ways to overcome these limitations.

We present a low-cost alternative to more complex laser metrology systems that uses a single-mode fiber Fabry-Perot etalon to generate an emission spectrum of evenly spaced lines with similar intensities, ideal for calibrating spectrographs (both in terms of wavelength and image quality). The system uses the hyperfine transition lines of 87 R b near 780.24 nm as an absolute reference. By controlling the cavity dimensions by small changes in temperature, we can tune and thus stabilize the transmission spectrum. A 20 Hz PID loop controls the etalon temperature and locks it to the 87 R b transitions. Through this method, we achieve a centroid error/precision of <1m/s (2.6 fm or 1.3 MHz) for 1 s integrations and 1 cm/s (0.026 fm or 13 kHz) for 30 min integrations of the reference line. We also show that a solution can be found to mathematically describe the spectrum. With the correct calibration and environmental controls in place, we show that this setup has the potential to be competitive with the best existing methods based on expensive and cumbersome laser combs.

Mosquitoes are a complex nuisance around the world and tropical countries bear the brunt of the burden of mosquito-borne diseases. Rwanda has had success in reducing malaria and some arboviral diseases over the last few years, but still faces challenges to elimination. By building our understanding of in situ mosquito communities in Rwanda at a disturbed, human-occupied site and at a natural, preserved site, we can build our understanding of natural mosquito microbiomes toward the goal of implementing novel microbial control methods. Here, we examined the composition of collected mosquitoes and their microbiomes at two diverse sites using Cytochrome c Oxidase I sequencing and 16S V4 high-throughput sequencing. The majority (36 of 40 species) of mosquitoes captured and characterized in this study are the first-known record of their species for Rwanda but have been characterized in other nations in East Africa. We found significant differences among mosquito genera and among species, but not between mosquito sexes or catch method. Bacteria of interest for arbovirus control, Asaia, Serratia, and Wolbachia, were found in abundance at both sites and varied greatly by species.

Parkinson's disease (PD) is characterized by progressive motor as well as less recognized non-motor symptoms that arise often years before motor manifestation, including sleep and gastrointestinal disturbances. Despite the heavy burden on the patient's quality of life, these non-motor manifestations are poorly understood. To elucidate the temporal dynamics of the disease, we employed a mouse model involving injection of alpha-synuclein (αSyn) pre-formed fibrils (PFF) in the duodenum and antrum as a gut-brain model of Parkinsonism. Using anatomical mapping of αSyn-PFF propagation and behavioral and physiological characterizations, we unveil a correlation between post-injection time the temporal dynamics of αSyn propagation and non-motor/motor manifestations of the disease. We highlight the concurrent presence of αSyn aggregates in key brain regions, expressing acetylcholine or dopamine, involved in sleep duration, wakefulness, and particularly REM-associated atonia corresponding to REM behavioral disorder-like symptoms. This study presents a novel and in-depth exploration into the multifaceted nature of PD, unraveling the complex connections between α-synucleinopathies, gut-brain connectivity, and the emergence of non-motor phenotypes.
