RESUMEN
The described microbiotesting technique provides the recording of pH changes, redox potential, electrical conductivity, optical density, light scattering and luminescence intensities, and other parameters of samples with viable test microorganisms incubated in a liquid nutrient medium in the presence and absence of the tested factors. The results of using this system for the analysis of pro- and antibiotic activity of various oil products, as well as weak electromagnetic fields of the megahertz range, are presented. It has been shown that the proposed technique, compared to the standard methods, yields more detailed and objective information and is less time- and labor-consuming.

A biotesting procedure that makes it possible to record the changes in the intensities of elastic light scattering, light absorption, and intrinsic photofluorescence of the protein component, as well as the determination of the concentration and structuring coefficients of the genomic component, of the samples with viable unicellular test organisms incubated in a liquid nutrient medium in the presence and absence of various external chemical factors, is described. The results of the analysis of the antibiotic activity of various metal cations with the use of this technique are presented. This technique is much more rapid, objective, and comprehensive for the assessment of the effect on the reproduction rate, metabolic activity, and genome structure of the test organisms from the samples of various products, wastes, etc. than standard visual microbiotesting methods.

The results of long-term author's studies of the optical and complex-forming properties of more than 30 synthetic low-molecular-weight fluorophores specific for DNA are described. These studies made it possible to significantly expand the already existing database of properties of such compounds, clarify the ideas about the patterns linking the mentioned properties of fluorophores with their structure, and formulate recommendations on designing new effective DNA-specific fluorophores. The results of these studies can be used, in particular, in the development of new rapid methods for diagnosing various diseases, biotesting of probiotic and antibiotic properties of various products and wastes, etc.

Spectral data and nucleic acid complex formation properties for more than 30 both newly synthesized and widely used fluorescent nucleotide-specific compounds of various classes have been analyzed. These include phenylbenzene, bisbenzimidazole, psoralen, angelicin, tetrahydrocarbazole, oxophenoxazine, and others. The main rules of a generalized model adequately explaining changes in fluorescent properties of synthetic, low molecular weight nucleotide-specific dyes depending on their chemical structure, mode of interaction with substrate, properties of assay medium, etc. are proposed. Fluorescent nucleotide-specific dyes have been originally used in newly developed methods for: express evaluation of "generalized microbial dissemination" of liquid media; evaluation of possible genotoxic effects of various foodstuffs, pharmaceutical drugs, hazardous environmental factors (including their combined effects on living organisms), etc.

Absorption and luminescence excitation and emission spectra of newly synthesized 2-(4-methylphenylimino)-3-(2 -benzothiazolyl)benz-alpha-chromene (BCBT) have been studied in the presence of various DNA concentrations. BCBT is characterized by the existence of two different fluorescent systems, exhibiting radiationless fluorescence resonance energy transfer between them. In the range of molar ratios of polynucleotide/dye concentrations from 0 to 50, BCBT preferentially intercalates into DNA due to its benz-alpha-chromene fragment, whereas the 2-benzothiazolyl fragment is responsible for fluorescence.

The experimental data on the effects of a widespread carcinogen, benzo[a]pyrene (BP), on individual reactions of rats were treated using mathematical-statistical methods. The individual reactions were analyzed in dependence of doses and modes of administration (single or chronic). The analysis revealed a statistically significant correlation between life span and urinary content of (+/-)-trans-7,8-dihydroxy-7,8-dihydrobenzo[a]pyrene (7,8-BP) in rats treated with BP. The calculated regression equations revealed that the individual sensitivity to carcinogen in case of the BP single administration to rats is mainly determined by efficiency of excretion of the BP active forms out of the organism, whereas after chronic BP administration it is determined by mechanisms of enzymatic deactivation of BP.

Absorption and fluorescence spectra for six new synthetic dyes of 4,8,4'-trimethylpsoralen and 4,4'-dimethylangelicin derivatives containing various terminal substituents at 5'-position have been investigated in different environments using a wide range of the DNA/ligand concentrations. Various spectral and binding characteristics of the DNA-ligand systems have been determined. General principles characterizing mechanisms responsible for changes in the fluorescent properties of nucleotide-specific dyes have been proposed; they take into consideration chemical structure of the dyes, properties of the environment, and degree of sorption on substrate.

Changes in absorbance and fluorescence excitation and emission spectra in the ultraviolet and visible regions of the system containing ethidium bromide (EtBr) and Hoechst-33258 (Ht) were investigated depending on various DNA quantities and the composition of the medium. It is noted that spectral properties of this system are determined by interactions of EtBr and Ht both with nucleic acid and with one another (for example, joint sorption of EtBr and Ht on DNA may involve fluorescent resonance energy transfer between the dye molecules). Thus, different modes of EtBr and Ht specificity to substrate and assay conditions suggest that combined use of these dyes provides some additional effects that may be interesting in terms of structure-functional study of nucleic acid. Some of these effects are considered in this paper.

Various models of complex formation of low-molecular ligands with DNA are considered. Using the Scatchard model, parameters of binding of fluorescent monophenylindole, monophenylbenzimidazole, and bisbenzimidazole dyes with calf thymus DNA were evaluated. By means of graphic (nonparametric) and correlation analysis, various spectral and complexation properties of these dyes in the presence of DNA are compared.

Mathematical and statistical procedures were used to identify individual response in rats to benzo(a)pyrene (BP) treatment versus dose and regimen (single or repeated administration). Primary experimental data were suitably processed to exclude extreme values. A significant correlation was established between life-span and urine level of (+/-)-trans-7,8-dihydroxy-7,8-dihydrobenzo(a)pyrene (7,8-BP) after BP administration. It was found that, with single administration regimen, individual response to the carcinogenic agent correlated primarily with efficiency of excretion of the active forms of BP while in chronic experiment--with deactivation of the agent by enzymes.
