RESUMEN

The present study aimed to detect Bartonella DNA in cats belonging to shelters, and to evaluate risk factors, clinical signs, and hematological abnormalities associated with infection. Complete blood counts and screening for the presence of Bartonella DNA were performed on cats' ethylenediamine tetraacetic acid anticoagulant-blood samples. Eighty-three cats (39.9%) were positive for Bartonella species. Bartonella DNA was also detected in fleas and in the blood of cats infested by positive flea. Cats that had not been sterilized, had outdoor access, had histories of fights, and had concurrent flea infestation were more likely to be infected by Bartonella species (P < 0.05). Age and sex were not associated with infection. Fifty-one (38.6%) symptomatic cats were positive to Bartonella species (P > 0.05). Clinical conditions most commonly observed were signs of respiratory abnormality and Sporothrix species coinfection (P > 0.05). Regarding hematological changes, eosinophilia was associated with infection (P < 0.05). A high frequency of Bartonella species infection was found in shelter cats and highlights the importance of adequate flea-control programs to prevent infection in cats and consequently in adopters and other animals.

Chlorogenic acids (CGAs) are a group of phenolic compounds found in worldwide consumed beverages such as coffee and green tea. They are synthesized from an esterification reaction between cinnamic acids, including caffeic (CFA), ferulic and p-coumaric acids with quinic acid (QA), forming several mono- and di-esterified isomers. The most prevalent and studied compounds are 3-O-caffeoylquinic acid (3-CQA), 4-O-caffeoylquinic acid (4-CQA) and 5-O-caffeoylquinic acid (5-CQA), widely described as having antioxidant and cell protection effects. CGAs can also modulate glutamate release from microglia by a mechanism involving a decrease of reactive oxygen species (ROS). Increased energy metabolism is highly associated with enhancement of ROS production and cellular damage. Glutamate can also be used as an energy source by glutamate dehydrogenase (GDH) enzyme, providing α-ketoglutarate to the tricarboxylic acid (TCA) cycle for ATP synthesis. High GDH activity is associated with some disorders, such as schizophrenia and hyperinsulinemia/hyperammonemia syndrome. In line with this, our objective was to investigate the effect of CGAs on GDH activity. We show that CGAs and CFA inhibits GDH activity in dose-dependent manner, reaching complete inhibition at high concentration with IC50 of 52 µM for 3-CQA and 158.2 µM for CFA. Using live imaging confocal microscopy and microplate reader, we observed that 3-CQA and CFA can be transported into neuronal cells by an Na+-dependent mechanism. Moreover, neuronal cells treated with CGAs presented lower intracellular ATP levels. Overall, these data suggest that CGAs have therapeutic potential for treatment of disorders associated with high GDH activity.

Vitamin C (in the reduced form ascorbate or in the oxidized form dehydroascorbate) is implicated in signaling events throughout the central nervous system (CNS). In the retina, a high-affinity transport system for ascorbate has been described and glutamatergic signaling has been reported to control ascorbate release. Here, we investigated the modulatory role played by vitamin C upon glutamate uptake and N-methyl-d-aspartate (NMDA) receptor activation in cultured retinal cells or in intact retinal tissue using biochemical and imaging techniques. We show that both forms of vitamin C, ascorbate or dehydroascorbate, promote an accumulation of extracellular glutamate by a mechanism involving the inhibition of glutamate uptake. This inhibition correlates with the finding that ascorbate promotes a decrease in cell surface levels of the neuronal glutamate transporter excitatory amino acid transporter 3 in retinal neuronal cultures. Interestingly, vitamin C is prone to increase the activity of NMDA receptors but also promotes a decrease in glutamate-stimulated [3 H] MK801 binding and decreases cell membrane content of NMDA receptor glutamate ionotropic receptor subunit 1 (GluN1) subunits. Both compounds were also able to increase cAMP response element-binding protein phosphorylation in neuronal nuclei in a glutamate receptor and calcium/calmodulin kinase-dependent manner. Moreover, the effect of ascorbate is not blocked by sulfinpyrazone and then does not depend on its uptake by retinal cells. Overall, these data indicate a novel molecular and functional target for vitamin C impacting on glutamate signaling in retinal neurons.

Toxoplasmosis and neosporosis have been recognized as economically important diseases with considerable impact on the livestock industry. Little is known concerning the occurrence of Toxoplasma gondii and Neospora caninum in sheep from Tocantins state, Brazil. Here, we investigated antibodies against these parasites and associated factors in 182 sheep from Araguaína, Santa Terezinha do Tocantins, Arguianópolis and Palmeiras do Tocantins districts, Tocantins. Sheep sera were assayed for T. gondii and N. caninum IgG antibodies by indirect fluorescence antibody test (IFAT), using cut-off point at a dilution of 1:40 and 1:25 respectively. The prevalence of seropositive animal for T. gondii was 13.74% and 13.74% for N. caninum. None of the characteristics studied including reproductive problems, presence of cats, presence of dogs and veterinary care (p>0.05) was associated with occurrence of T. gondii or N. caninum infection. Only breed was identified as associated factor for the occurrence of toxoplasmosis in sheep (p<0.05). The present study is the first report on serum occurrence of T. gondii and N. caninum in sheep from the state of Tocantins, Brazil.

Pouco se sabe sobre a ocorrência de Toxoplasma gondii e Neospora caninum em ovelhas do estado do Tocantins, Brasil. Foram investigados a ocorrência de anticorpos contra estes parasitos e fatores associados em 182 ovelhas das cidades de Araguaína, Santa Terezinha do Tocantins, Arguianópolis e Palmeiras do Tocantins, Tocantins. Os soro das ovelhas foram testados para anticorpos IgG anti-T. gondii e anti-N. caninum pela Reação de Imunofluorescência Indireta (RIFI), usando os pontos de corte na diluição de 1:40 e 1:25, respectivamente. A prevalência de animais soropositivos para T. gondii foi de 13.74% e para N. caninum, 13.74%. Nenhuma das características estudadas incluindo problemas reprodutivos, presença de gatos, presença de cães e cuidados veterinários (p>0.05) foram associadas com a ocorrência infecção por T. gondii ou N. caninum. Somente raça foi identificada como fator associado à ocorrência de toxoplasmose em ovelhas (p<0.05). O presente trabalho é o primeiro relato da ocorrência sérica de T. gondii e N. caninum em ovelhas do estado do Tocantins, Brasil.

Introdução: a transmissão perinatal ou vertical do HIV pode ocorrer tanto intra-útero, como durante o trabalho de parto e no período pós-parto através do aleitamento materno. Os profissionais de saúde são coadjuvantes desta experiência e desempenham importante papel no esclarecimento da importância da realização do teste anti-HIV. Objetivo: identificar as emoções e sentimentos que as gestantes possuem ao realizar o teste anti-HIV. Métodos: de abordagem qualitativa através de entrevista semi-estruturada e consulta aos prontuários, onde foram entrevistadas 13 gestantes acompanhadas em consultas de enfermagem, na realização e entrega do teste anti-HIV em um serviço ambulatorial em DST/aids da Faculdade de Enfermagem de Passos/MG. Resultados: baseado neste estudo foi possível concluir que há falhas no acompanhamento das gestantes quanto à indicação do teste anti-HIV, devido à falta de vínculo gestante-profissional da saúde nas consultas pré-natalistas. As mesmas não são orientadas quanto à importância da realização do teste e o quanto pode beneficiar sua saúde e a do feto. Observamos também que as mulheres estão mais vulneráveis a adquirir qualquer doença transmissível, pelo fato de confiar cegamente em seus parceiros, o que as leva a pensar que basta amar para estarem previnidas. Conclusão: por se depararem com o desconhecido na realização do teste anti-HIV, foi comum estarem presentes no comportamento e falas das gestantes, diferentes estados emocionais o que sugere que é um momento propício para o profissional da saúde realizar uma relação de ajuda, diminuindo os medos e tensões das clientes proporcionando assim um report durante o acompanhamento.
