This study investigated the occurrence of plastic particles in the digestive tracts of fish from headwater streams in a human-thinly populated region of the subtropical Sinos River basin in southern Brazil. In total, 258 individuals from 17 species were collected using electric fishing. Thirty-eight percent (38%) of the specimens contained plastic particles. All of them were fibers, with a maximum count of 43 per individual. Plastic fibers were the fourth most abundant food category. Results showed that the uptake of these plastic particles was proportional to the number of ingested food items. Fiber counts in the guts correlated with the uptake of Trichoptera, which are invertebrates using plastic particles to construct their protective cases. No significant difference in plastic uptake was detected between benthic and water column fish. No evidence of bioaccumulation of plastic particles was found in the intestines. The distance from urban areas was not related to the number of ingested plastic particles, concluding that plastics are ubiquitous and available to biota, even in remote locations. The most probable source of these particles is residences close to the streams which discharge the sewage of washing machines without any treatment.

BACKGROUND: The prevalence of persistent respiratory symptoms tends to be low in patients with a longer recovery time after COVID-19. However, some patients may present persistent pulmonary abnormalities.OBJECTIVE: To evaluate the prevalence of tomographic abnormalities 90 days after symptom onset in patients with COVID-19 and compare two chest high-resolution computed tomography (HRCT) analysis techniques.METHODS: A multicentre study of patients hospitalised with COVID-19 having oxygen saturation <93% on room air at hospital admission were evaluated using pulmonary function and HRCT scans 90 days after symptom onset. The images were evaluated by two thoracic radiologists, and were assessed using software that automatically quantified the extent of pulmonary abnormalities.RESULTS: Of the 91 patients included, 81% had at least one pulmonary lobe with abnormalities 90 days after discharge (84% were identified using the automated algorithm). Ground-glass opacities (76%) and parenchymal bands (65%) were the predominant abnormalities. Both chest HRCT technical assessments presented high sensitivity (95.9%) and positive predictive value (92%), with a statistically significant correlation at baseline (R = 0.80) and after 90 days (R = 0.36).CONCLUSION: The prevalence of pulmonary abnormalities on chest HRCT 90 days after symptom onset due to COVID-19 was high; both technical assessments can be used to analyse the images.

The aim of this case study was to evaluate the clinical and productive data of Nellore cows during pregnancy and lactation, supplemented or not with P. Ninety-five pregnant heifers were divided into two groups (G40 and G0). Until the date of calving animals from G40 received a mineral mixture composed of 224g of Na and 40g of P/kg, and G0 received only NaCl. After calving heifers were divided into four treatments: from G40, 28 first-calf cows continued to receive the mineral mixture containing 224g of Na and 40g of P/kg (group 40/40) and 12 started to receive only NaCl (group 40/0); from G0, 26 continued to receive only NaCl (group 0/0) and 15 started to receive a mixture containing 224g of Na and 40g of P/kg (group 0/40). In the second experimental phase, 205 multiparous cows were divided into two groups: G1 (40/40) consisting of 40 cows, receiving a mineral mixture containing 244g of Na and 40g/P per kg, during pregnancy and lactation, and group 2 (0/0) formed by 165 cows, which were supplemented only with NaCl. In both experiments, whether in pregnant or lactating breeders, there was not any sign of P deficiency or differences in calves LW at 120 and 210 days.


Este estudo avaliou os aspectos clínicos e produtivos de matrizes Nelore, suplementadas ou não com P. Noventa, e cinco nulíparas prenhes foram distribuídas em dois grupos (G40 e G0). Os animais do G40 receberam uma mistura mineral com 224g de Na e 40g de P/kg, até a data do parto, e o G0 recebeu apenas NaCl. Após o parto, as primíparas foram divididas em quatro grupos: do G40, 28 continuaram a receber mistura contendo 224g de Na e 40g de P/kg (grupo 40/40), e 12 passaram a receber apenas NaCl (grupo 40/0); do G0 26 continuaram recebendo apenas NaCl (grupo 0/0), e 15 passaram a ser suplementadas com a mistura contendo 224g de Na e 40g de P/kg (grupo 0/40). Na segunda fase experimental, 205 vacas foram divididas em dois grupos: G1 (40/40), composto por 40 vacas, suplementadas com a mistura mineral contendo 244g de Na e 40g de P/kg, durante a gestação e a lactação, e o G2 (0/0) foi formado por 165 vacas, as quais foram suplementadas apenas com NaCl, durante a gestação e a lactação. Em ambos os experimentos, seja nas matrizes em gestação ou naquelas em lactação, não houve qualquer sinal da deficiência de P, e isso se refletiu na ausência de diferenças nos pesos dos bezerros aos 120 e 210 dias.

Este estudo investigou a ocorrência de partículas plásticas no trato digestivo de peixes de riachos de cabeceira em uma região de baixa densidade humana da bacia subtropical do rio dos Sinos no sul do Brasil. No total, 258 indivíduos de 17 espécies foram coletados por meio da pesca elétrica. Trinta e oito por cento (38%) dos espécimes continham partículas de plástico. Todos eram fibras, com contagem máxima de 43 por indivíduo. As fibras plásticas foram a quarta categoria de alimentos mais abundante. Os resultados mostraram que a absorção dessas partículas plásticas foi proporcional ao número de itens alimentares ingeridos. A contagem de fibras nos intestinos se correlacionou com a absorção de Trichoptera, que são invertebrados que usam partículas de plástico para construir suas capas protetoras. Nenhuma diferença significativa na absorção de plástico foi detectada entre peixes bentônicos e de coluna d'água. Nenhuma evidência de bioacumulação de partículas plásticas foi encontrada nos intestinos. A distância das áreas urbanas não foi relacionada ao número de partículas plásticas ingeridas, concluindo que os plásticos são ubíquos e disponíveis para a biota, mesmo em locais remotos. A fonte mais provável dessas partículas são as residências próximas aos córregos que descarregam os esgotos das máquinas de lavar sem nenhum tratamento.

Manganese (Mn) is an essential metal for mammals. It can modulate the action of endogenous substances, as neurotransmitters, but in excess also can trigger known neurotoxic effects. Many studies have been conducted assessing Mn neurotoxicity. However, Mn bioaccumulation in different brain tissues and behavior effects involving gender-specific studies are conflicted in the literature. Therefore, the aim of this work was to compare Mn effects, after 30days of intraperitoneal treatment, in male and female rats, submitted to forced swim and open field tests. After that, were evaluated Mn and Fe tissue levels in CNS, liver, and kidneys. Wistar rats were divided into saline, Mn 1mg/kg, Mn 5mg/kg, and imipramine (as forced swim control). Then, animals were euthanized by anesthesia overdose followed by decapitation and the collected tissue were striatum, hippocampus, brainstem, cortex, cerebellum, hepatic tissue, and renal tissue. Mn and Fe were determined by ICP-MS. There was a dose-dependent effect on accumulation of Mn in the cerebellum and brainstem to the dosage of 5mg/kg. In hippocampus there were bioaccumulation differences between gender and dose, and an increase of Fe in the groups exposed to Mn. Excess metals in the brain dissected has a strong influence on memory and learning processes and suggests pro-depressive effects, possibly triggered by the reduction of monoamines due to excessive metal bioaccumulation. It was concluded that, under this experimental design, Mn exposure cause metal deposition on dissected CNS, liver and kidney. There an effect at lower doses that was gender-dependent and males had more pronounced behavioral damage compared to females, although with increasing dose, females had an indication of motor damage.

This study aimed to evaluate serum and urine concentrations of several trace metals of a non-directly exposed population in southern Brazil and establish reference values. Serum and urine samples were obtained from 240 volunteers (175 males and 65 females, age ranging from 18 to 74 years old). Levels of arsenic, chromium, cobalt, copper, lead, nickel, manganese and zinc were determined by means of dynamic reaction cell inductively coupled plasma mass spectrometry (DRC-ICP-MS). Comparison between genders resulted in no significant difference for all metals but serum copper, as concentrations are higher in females than males. For most metals assessed, a negative correlation between serum concentrations and age was found, but no significant correlation was found between urine concentrations and age.

Polymorphisms present in the TNF promoter region has shown to influence the gene transcription. Leprosy displays different clinical manifestations according to the immune responses of the individual infected with Mycobacterium leprae. In this study, we evaluated the single nucleotide polymorphisms (SNPs) -238 G/A (rs361525), -308 G/A (rs1800629), -857 C/T (rs1799724), -863 A/C (rs1800630) and -1031 T/C (rs1799964) in the promoter region of the TNF to see whether these SNPs influence host-susceptibility to leprosy and the different clinical manifestation. Nucleotide sequencing was performed with DNA samples from 108 leprosy patients and 253 control subjects. An association between -1031 C/C genotype and protection from leprosy was observed when leprosy patients were compared to controls (OR 0.11; 95% CI=0.01-0.82; p=0.012). The -857 C/T genotype may be associated with susceptibility to leprosy (OR=1.81; 95% CI=1.09-3.00; p=0.028). Similar genotype and allele frequencies for the SNPs -308 G/A and -238 G/A were observed between leprosy patients and control subjects. Altogether, TNF polymorphisms in the promoter region may be predictive of leprosy outcome.

Mushroom Lentinula edodes has been widely studied therapeutically. However, there is no data regarding its daily intake level safety. Since L. edodes has many active compounds known to bind to metals, we evaluated macro and micronutrients in liver and kidney of healthy rats after subchronic exposure to L. edodes. Rats were divided into four groups, receiving water and L. edodes at 100, 400 and 800 mg/kg/day. The treatment lasted 30 days. Essential elements (Zn, Cu, Mg, Fe, Mn, Se, Co, Mo, and Li) were analyzed in an inductively coupled plasma mass spectrometer. Our results demonstrated a significant decrease in Cu, Fe, Mn and Co levels in liver of rats receiving L. edodes at the highest doses. In kidney, Mn, Mo and Li concentrations significantly dropped in the groups exposed to the highest doses. In this way, an important point is revealed concerning the food safety from L. edodes, once its chronic and high consumption could contribute to macro and micronutrients deficiency. Additionally, we speculate that the daily use of L. edodes could be unsuccessful for patients in mineral therapy besides being able to be unsafe for individuals with some propensity to mineral deficiency.

Caffeic acid phenethyl ester (CAPE) is a botanical compound abundant in honeybees' propolis. It has anti-inflammatory, antiviral, antioxidant, immunomodulatory and antitumor properties. Its beneficial effects against neurodegenerative diseases, including Parkinson's disease, have also been suggested and some mechanisms have been proposed. Mitochondrial damage and oxidative stress are critical events in neurodegeneration. Release of cytochrome c from mitochondria to cytosol and the downstream activation of caspase-3 have been suggested as targets of the protective mechanism of CAPE. Most of the studies addressing the protective effect of CAPE have been performed in cell culture. This is the first study to demonstrate the protective effect of CAPE against the dopaminergic neuronal loss induced by 6-hydroxydopamine (6-OHDA) in rats. It also demonstrates, for the first time, the inhibitory effect of CAPE on mitochondrial permeability transition (MPT), a mediator of neuronal death that triggers cytochrome c release and caspase-3 activation. Scavenging of reactive oxygen species (ROS) and metal chelation was demonstrated in the brain-affected areas of the rats treated with 6-OHDA and CAPE. Additionally, we demonstrated that CAPE does not affect brain mitochondrial function. Based on these findings and on its ability to cross the blood-brain barrier, CAPE is a promising compound to treat Parkinson's and other neurodegenerative diseases.

There is scientific evidence to suggest that the medicinal mushroom Agaricus blazei Murrill (AbM) has immunomodulatory effects on cytokine synthesis, both in vitro and in vivo. This study was the first randomized, double-blind, placebo-controlled trial designed to investigate these purported actions in elderly women. The objective of this study was to ascertain the effects of AbM intake on serum levels of interleukin-6 (IL-6), interferon-gamma (IFN-γ) and tumour necrosis factor-alpha (TNF-α) in community-living seniors. The sample consisted of 57 elderly females who were carriers or homozygous for the majority allele of functional polymorphisms for the chosen cytokines. Subjects were randomly allocated to receive placebo (n = 29) or AbM dry extract (n = 28), 900 mg/day for 60 days. Body mass index, abdominal girth, body composition, blood pressure and cytokine (IL-6, IFN-γ, and TNF-α) levels were measured, and food intake was assessed as a possible confounder. Analysis of these parameters showed the sample was characterized by overweight and excess adiposity. After the study period, no changes from baseline were detectable for any parameter in either group. In this study, AbM extract had no modulating effect on IL-6, IFN-γ or TNF-α levels in elderly females.

Twenty-seven rice samples from Brazil, four parboiled brown, seventeen white and six parboiled white were analysed by ICP-MS for trace element determination. Concentrations of arsenic varied from 58.8 to 216.9 ng g(-1), for cadmium from 6.0 to 20.2 ng g(-1), for antimony from 0.12 to 1.28 ng g(-1), and for uranium from 0.025 to 1.28 ng g(-1). The estimated daily intake through rice consumption was 9.5 µg for As, 2.4 µg for Cd, 0.029 µg for Sb, 0.013 µg for U, 3.1 µg for Co, 0.2 µg for Cu, 85.6 mg for Mg, 1.9 mg for Mn, 333 mg for P, 3.0 µg for Se, 1.6 mg for Zn, 0.9 mg for Rb, and 0.3 µg for V. Found values represent a considerable percentage of the dietary reference intakes and provisional tolerable daily intake for essential and toxic elements, respectively.

Studies show that genetic polymorphisms in apolipoproteins, which are in charge of lipid transport, predispose to atherogenic dyslipidemia. This study aimed to investigate the impact of apolipoprotein E, A5, and B genotypes and dietary intake on lipid profile in a sample of elderly women in Brazil. Two hundred and fifty-two women (60 years or older) living in the outskirts of the Brazilian Federal District underwent clinical and laboratory assessments to characterize glycemic and lipidemic variables, and also to exclude confounding factors (smoking, drinking, hormone replacement, cognitive impairment, physical activity). Three-day food records were used to determine usual dietary intake, whereas genotypic evaluations were in accordance to established methodologies. Genotype frequencies were consistent with the Hardy-Weinberg equilibrium. Prior to adjustment, individuals carrying the epsilon2 allele showed higher serum levels of triglycerides (P<0.05) and VLDL (P<0.005) compared to epsilon4 carriers, whereas LDL levels were considerably elevated in epsilon4 compared to epsilon2 carriers. In the presence of high intake of total fat or a low ratio of polyunsaturated to saturated fatty acid, epsilon4 carriers lost protection against hypertriglyceridemia. There was no association of the apolipoprotein A5 and B genotypes with lipidemic levels independently of the fat intake regimen. Results are suggestive of a dysbetalipoproteinemic-like phenotype in postmenopausal women, with remarkable gene-diet interaction.

O tipo de tarefa realizada determina o tipo de preensao utilizada a firca aplicada, bem como as posturas adotadas para sua execucao. Diferentes posturas e preensoes envolvidas nessas tarefas apresentam maiores ou menores riscos de lesoes musculoesqueleticas. Objetivo: descrever os tipos de preensao empregados durante atividades de manuseio de cargas de 5 e 10kg entre superficies de alturas diferentes e, quantificar os movimentos de flexo/extensao, devio radial e ulnar do punho registrados por meio da eletrogonometria. Metodo: 10 voluntarios masculinos manusearam caixa entre superficies de alturas diferentes. Os tipos de preensao utilizados foram filmados. diferentes categorias de preensao utilizadas durante essa tarefa foram identificadas. As amplitudes de movimentos e a porcentagem de tempo gasto dentro e fora das faixa de amplitudes pre-estabelecidas (15o. de flexao e extensao, 10o. de dsevio radial, e 15o. desvio ulnar)0rosp foram determinadas. Uma analise de univariancia foi utilizada para avliar diferencas significativas entre a antropometria das maos dos voluntarios, e entre os movimentos utilizados para o manuseio de cargas e alturas diferentes. Resultados e conclusoes: os resultados mostraram que as alturas das superficies para as quais os objetos eram manuseados influenciaram significativamente (p=0,000) as amplitudes articulares, contudo nao houve diferenca nos movimentos para as diferentes massas das cargas manuseadas (p=0,43). tempo acima do considerado recomendavel foi gasto em desvio radial quando os manuseios envolviam superficies altas. Recomendacoes sobre local adequado para o acondicionamento de material a ser manuseado deveriam constar nos programas de treinamento visando a prevencao de lesoes musculoesqueleticas

PURPOSE: To evaluate thrombolytic therapy with rt-PA for acute stroke within 6h of symptom onset, and assessment of neurologic outcome. METHODS: We studied 6 patients, four women, mean age 63 +/- 18 years, with severe neurologic deficit within 6h of stroke onset, and with no spontaneous improvement. The stroke was embolic in 3, and thrombotic in the others. All patients were submitted to a head CT scan followed by either a cerebral angiography in 3 patients, or a transcranial Doppler, in the other 3 for assessment of arterial obstruction, and patency after thrombolytic therapy. We used 0.9 mg/kg of rt-PA, IV, over 60 min in 5 patients, and 0.5 mg/kg by intra-arterial infusion, over 60 min, in one. At the beginning a bolus of 10% of the total dose was delivered during 1 to 2 min. Head scan was repeated 24h and 7 days after treatment to detect ischemic areas and hemorrhagic complications. RESULTS: Middle cerebral artery occlusion was observed in 5 patients and posterior cerebral artery occlusion in one. The obstruction was cleared in 4 patients with persistence of the patency after 24h. A complete neurologic recovery was found in one patient, and a partial recovery in three. In two patients there was failure of arterial recanalization with no neurologic recovery. Only one patient had hemorrhagic transformation of ischemic tissue, without neurologic worsening. Death occurred in one patient due to pulmonary infection. CONCLUSION: Arterial patency with thrombolytic therapy was effective in 4 of our 6 patients. All 4 patients also disclosed a certain degree of neurologic improvement, rt-PA can be successfully used in selected patients up to 3h of the event onset, as shown in randomized studies.

RESUMEN

Os autores relatam caso de um homem com 56 anos de idade que se queixava de dor na regiäo lombar com irradiaçäo para os membros inferiores por três meses, dois anos após a correçäo de aneurisma da aorta infra-renal. Após estudos diagnósticos - como tomografia computadorizada - e angiográficos, o paciente foi encaminhado ao centro cirúrgico com diagnóstico de falso aneurisam do terço médio da prótese. No ato cirúrgico verificou-se que o falso aneurisma teve origem devido à deiscência da anastomosoe artéria mesentérica inferior que havia sido implantada na prótese.
