mRNA of genes (tgfß, il6, il10, il1ß, and vegf165) involved in cell proliferation, inflammatory, anti-inflammatory, and angiogenic processes were detected and quantified in cultures of mesenchymal stromal cells of different passages derived from human extraembryonic tissues (amniotic sac, umbilical cord, chorionic villi and trophoblast of the placenta). The concentrations of IL-10, IL-6, IL-1ß, and TGFß were measured.

The expression of mRNA of cytokines and immunoregulatory molecules characterizing the interaction of mesenchymal stromal cells from chorionic villi of postpartum placenta and allogenic mononuclear blood cells was studied during 3-day co-culturing of these cells. The expression of foxp3, il2ra, and il10 mRNA in floating mononuclear cells increased from day 1 to 3 in co-culture, which can refl ect the process of induction of regulatory T cells in the lymphocyte population under the action of mesenchymal stromal cells.

We describe a method of isolation of human mesenchymal stromal cells from the umbilical cord (Wharton's jelly) and human placenta: amnion, placental villi, and trophoblast. Morphology, immunophenotypic characteristics, and differentiation potencies of isolated cells were studied. The capacity of mesenchymal stromal cells from extraembryonic tissues to osteogenic, adipogenic, and chondrogenic differentiation was demonstrated and the dynamics of this process was described. The isolated cells met the criteria for multipotent mesenchymal stem cells.

Intravenous transfusion of nucleated cells from human umbilical cord blood (10(6)/ml saline) to female rats 3 h after massive blood loss produced a pronounced corrective effect (day 5): alleviates posthemorrhagic changes and normalizes hemoconcentration parameters, metabolic disorders, and behavioral reactions.

The number of monocytes, macrophages (CD14+), and large granular lymphocytes (CD56+) in the endometrium increased in chronic endometritis. These changes are an unfavorable prognositic signs for the onset of pregnancy. Combination therapy improved the phenotypic composition of endometrial cells.

We estimated the percentage of CD4+ lymphocytes synthesizing IFN-gamma and IL-4 in the peripheral blood and measured the concentrations of IL-1, IL-6, and TNF-alpha in lochia of patients with postpartum endometritis. T lymphocytes with intracellular IFN-gamma prevailed in women with physiological course of the postpartum period. The number of cells containing IL-4 increased in patients with postpartum endometritis. On days 5-7 after childbirth the concentrations of proinflammatory cytokines IL-1beta and TNF-alpha in lochia of these patients were higher than in healthy postpartum women (by 1.4 and 2.7 times, respectively). The concentrations of cytokines IL-1beta, TNF-alpha, and IL-6 increased most significantly in patients with postpartum endometritis and active viral infection (by 3.4, 4.2, and 1.6 times, respectively).

The content of some cytokines in the peritoneal fluid and the level of their in vitro production by peritoneal macrophages and lymphocytes were evaluated in women with external endometriosis. The ratio of peritoneal lymphocytes and macrophages in the peritoneal fluid was changed in endometriosis because of increased percentage of lymphoid cells and decreased content of macrophages, in the presence of high absolute counts of both types of cells. The cytokine status of women with endometriosis was characterized by higher levels of IL-1beta, TNF-alpha, and epidermal growth factors both in the peritoneal fluid and supernatants of 24-h cultures of peritoneal macrophages; the content of IFN-alpha remained unchanged. The concentrations of IFN-gamma in the peritoneal fluid did not change in endometriosis, but increased in the supernatants of peritoneal lymphocyte cultures.

The concentrations of cytokines (tumor necrosis factor-alpha and interleukin-10) in cervical scrapes and the percentage of CD4(+)lymphocytes producing interferon-gamma and interleukin-4 in the peripheral blood were measured in women of reproductive age with typical form of genital herpes. The results suggest that genital herpes is a chronic inflammatory process characterized by high levels of proinflammatory cytokines with predominance of T-helper type 1 over T-helper type 2 cytokines.
