RESUMEN

OBJECTIVE To explore the mechanism of Compound lizard powder reducing cisplatin resistance in gastric cancer by regulating glycolytic activity based on phosphoinositide 3-kinase(PI3K)/protein kinase B(Akt) signaling pathway. METHODS Human gastric cancer MKN45 and MKN45/DDP (cisplatin-resistant) cells were cultured in vitro and intervened with different mass concentrations of cisplatin (0.1, 0.2, 0.4, 0.8, 1.6, 3.2 μg/mL) to detect the survival rate, half inhibitory concentration (IC50) and drug resistance index. MKN45/DDP cells were inoculated subcutaneously in the right anterior axilla of nude mice to prepare a transplanted tumor model of gastric cancer. After successful modeling, they were randomly divided into model group, cisplatin group (0.002 g/kg), Compound lizard powder group (2.8 g/kg) and combination group (the same dose as each single drug group), with 8 nude mice in each group. Each administration group was given relevant solution, twice a week (cisplatin, i.p.) or twice a day (Compound lizard powder, i. g.), for 4 consecutive weeks. During the experiment, the body weight of nude mice was monitored, and tumor volume and inhibitory rate of tumor were calculated. The levels of inflammatory factors (tumor necrosis factor- α, interleukin-6) in tumor tissue, the mRNA and protein expressions of multidrug resistance-associated protein 1 (MRP1), P-glycoprotein (P-gp), glucose transporter-1 (GLUT1) and lactate dehydrogenase A (LDHA), as well as the protein expressions of PI3K, phosphorylated PI3K (p-PI3K), Akt, phosphorylated Akt (p-Akt), hexokinase-2 (HK2) and pyruvate kinase M2 (PKM2) were all detected. RESULTS With the intervention of different concentrations of cisplatin, the survival rate of MKN45/DDP-resistant cells was significantly higher than that of MKN45 parent cells (P<0.05). IC50 value of MKN45/DDP and MKN45 cells were(1.052 0±0.221 9) and (0.372 1±0.238 0)μg/mL, and the drug resistant index was 2.827. Compared with the model group, cisplatin group, Compound lizard powder group and combination group all had certain inhibitory effects on the tumor growth in nude mice; the inhibitory rates of tumor increased significantly (P<0.05); the levels of inflammatory factors, the mRNA and protein expressions of MRP1, P-gp, GLUT1 and LDHA (except for cisplatin group), the phosphorylation levels of PI3K and Akt protein (except for cisplatin group) as well as the protein expressions of HK2 and PKM2 were decreased significantly, while the combination group was significantly better than the cisplatin group (P<0.05). CONCLUSIONS Compound lizard powder may inhibit tumor growth in transplanted tumor model nude mice with gastric cancer-resistant cells by reducing the secretion of tumor-related inflammatory factors, inhibiting the expression of glycolysis, drug resistance-related proteins and genes, inhibiting the activation of the PI3K/Akt signaling pathway, thus having a certain effect of enhancing cisplatin efficacy and reversing drug resistance.

RESUMEN

Here,a styrene-based polymer monolithic column poly(VBS-co-TAT-co-AHM)with reversed-phase/hydrophilic interaction liquid chromatography(RPLC/HILIC)bifunctional separation mode was success-fully prepared for capillary electrochromatography by the in situ polymerization of sodium p-styrene sulfonate(VBS)with cross-linkers 3-(acryloyloxy)-2-hydroxypropyl methacrylate(AHM)and 1,3,5-triacryloylhexahydro-1,3,5-triazine(TAT).The preparation conditions of the monolith were optimized.The morphology and formation of the poly(VBS-co-TAT-co-AHM)monolith were confirmed by scanning electron microscopy(SEM)and Fourier transform infrared spectroscopy(FT-IR).The separation perfor-mances of the monolith were evaluated systematically.It should be noted that the incorporation of VBS functional monomer can provide π-π interactions,hydrophilic interactions,and ion-exchange in-teractions.Hence,the prepared poly(VBS-co-TAT-co-AHM)monolith can achieve efficient separation of thiourea compounds,benzene series,phenol compounds,aniline compounds and sulfonamides in RPLC or HILIC separation mode.The largest theoretical plate number for N,N'-dimethylthiourea reached 1.7×105 plates/m.In addition,the poly(VBS-co-TAT-co-AHM)monolithic column showed excellent reproducibility and stability.This novel monolithic column has great application value and potential in capillary electrochromatography(CEC).

RESUMEN

Objective:To establish reverse triiodothyronine (rT 3) biological reference interval suitable for laboratory by indirect method. Methods:From April to September 2019, 797 cases (332 males, 465 females, age: 12-95 years) underwent thyroid function, thyroid related antibody and rT 3 tests from hospitalized population in Dongzhimen Hospital Affiliated to Beijing University of Chinese Medicine were retrospectively analyzed. The reference individuals with normal thyroid hormone, antibody and without thyroid nodule or goiter were screened as inclusion criteria, and the factors such as acute and chronic diseases or drugs that might affect the values of rT 3 were excluded. Independent sample t test, one-way analysis of variance and least significant difference t test were used to analyze data. The rT 3 reference interval was established by non-parametric sequencing method, and 2.5% and 97.5% percentile values of data distribution were selected as the upper and the lower reference limits. In order to verify the rT 3 reference interval, 20 healthy individuals and 20 inpatients who met the inclusion and exclusion criteria were selected to test rT 3 with a simple random sampling method. Results:A total of 159 reference individuals (66 males, 93 females, age: 23-87 years) were enrolled. The rT 3 values of 23-29( n=4), 30-39( n=18), 40-49( n=29), 50-59( n=43), 60-69( n=40), 70-79( n=19) and over 80( n=6) years old groups were (0.62±0.16), (0.63±0.12), (0.64±0.11), (0.61±0.11), (0.65±0.14), (0.65±0.11) and (0.79±0.10) μg/L, respectively. There was a statistically significant difference in the rT 3 test results among different age groups ( F=2.17, P=0.049). There were statistically significant differences of rT 3 between the individuals over 80 years old and other age groups (all P<0.05), while there were no statistically significant differences among the other groups (all P>0.05). The rT 3 of males and females under 80 years old were (0.62±0.11) and (0.64±0.12) μg/L, respectively, with no significant difference between them ( t=-0.81, P=0.420). The newly established rT 3 reference interval suitable for people above 20 years old and below 80 years old was 0.47-0.92 μg/L, and the lower limit was significantly higher than that of the reference interval in the reagent specification (0.20-0.95 μg/L). The rT 3 range of 20 healthy individuals was 0.57-0.82 μg/L and that of 20 inpatients was 0.48-0.77 μg/L, which were all within the new reference interval. Conclusion:The rT 3 biological reference interval established here has clinical application value, but its applicable range of age still needs to be further improved.

Objective@#To explore the relationship and gender difference between maternal adverse childhood experiences (ACEs) with emotional and behavioral problems (EBPs) in preschool children, and to provide a reference to promote emotional and behavioral problems among preschool children.@*Methods@#Using the method of stratified cluster sampling, 9 647 children from kindergartens in three cities of Anhui Province were selected in June 2021. Maternal adverse childhood experiences were investigated with the WHO Adverse Childhood Experiences International Questionnaire(ACEs-IQ), preschool children s emotional and behavioral problems were investigated with Strengths and Difficulties Questionnaire.@*Results@#The detection rate of abnormal emotional behavior was 17.4%, emotional symptoms 16.3%, conduct problems 18.3%, hyperactivity 22.7%, peer problems 34.2%, prosocial behaviors 18.5%. Multivariate Logistic regression analysis showed that maternal adverse childhood experiences were positively associated with the risk of emotional and behavioral problems ( OR =1.51-2.97, P <0.01). Maternal cumulative adverse childhood experiences were also positively associated with the risk of emotional and behavioral problems( OR =3.13-9.61, P <0.01). The association of maternal emotional abuse, physical abuse and community violence with peer problems were stronger in boys than that of girls ( ROR =1.25, 1.26, 1.41, P <0.05).@*Conclusion@#The findings suggest maternal adverse childhood experiences were associated with emotional behavior problems among preschool children. Focusing on the maternal adverse childhood experiences is crucial for the prevention and control of childhood emotional and behavioral problems.

RESUMEN

Objective:To develop the minimal clinically important difference (MCID) of Quality of Life Instruments for Cancer Patients-Leukemia (QLICP-LE) (V2.0).Methods:The quality of life of 101 patients with leukemia in First Affiliated Hospital of Kunming Medical University and First People′s Hospital of Yunnan Province from October 2011 to May 2012 were measured. The QLICP-LE (V2.0) was used for data collection, and the MCID for the overall score and scores of various domains of QLICP-LE (V2.0) were established by using the distribution-based approach including indexes of effect size, standard error of measurement (SEM), reliable change index, standardized response mean and responsiveness statistic, and the recommended values of MCID were determined through the consensus method.Results:The MCID formulated by the above five indexes were as follows: the total scale 1.4-9.3, physical functional domain 1.6-15.6, psychological functional domain 2.9-15.6, social functional domain 2.2-18.0, common symptoms and side-effects domain 1.7-17.1, common module 1.8-10.0, and the specific module 1.1-12.1. Through the expert consensus method, it was recommended to use the MCID results calculated by 1.96SEM: the total scale was 4, physical domain was 8, psychological domain was 8, social domain was 9, common symptoms and side-effects domain was 9, common module was 4, and the specific module was 6.Conclusion:Each index of distribution-based approach has its own advantages and disadvantages, which can be selected based on actual conditions. There is clinical significance when the score change of QLICP-LE (V2.0) of leukemia patients after treatment exceeds its MCID.

RESUMEN

Objective To investigate the efficacy and safety of MatrixRIB fixation system in the treatment of severe rib fractures.Methods A total of 131 patients with severe rib fractures undergoing open reduction and internal fixation using MatrixRIB fixation system in our department were enrolled.Data including demographics,injury characteristics,surgical procedure details and postoperative recovery were collected.The operation experiences were summarized,and complications,postoperative and follow-up data were analyzed.Results Totally 637 (637/891)rib fractures were fixed.No internal fixation disorder was found,but 1 case had displacement after internal fixation.The majority of patients were extubated immediately after surgery.The mean time of pressure-support ventilation after surgery was 47.6(7 143)hours.The chest instability and pain were improved in all patients after surgery.At 3 to 6 months after treatment,70 patients(70/102)returned to work,and 87 patients(87/89)recovered previous normal life.Conclusions MatrixRIB fixation system is easy to operate in the treatment of severe rib fractures and has good clinical outcomes with few complications.Patients' pain is significantly relieved with satisfactory rehabilitation.

Objective To investigate the effects of endoscopic transthoracic sympathectomy on the treatment of hyperhidrosis palmaris(sweaty hands)and quality of life.Methods The clinical data of 62 patients with sweaty hands admitted from June 2010 to May 2016 were retrospectively analyzed.The 62 patients were randomly divided into two groups:thoracoscopic sympathectomy was performed at T2-3 in the control group,while at T3-4 in the experimental group.The self-reports of quality of life(QOL)before and after the operation were collected as follows:the severity of sweaty hands assessed by hyperhidrosis disease severity scale (HDSS:the higher score the more severe hyperhidrosis),the improvement of QOL assessed by sweaty hands assessment scale(the higher score the more improvement)and surgical efficacy.Results The improvements of HDSS and satisfaction score were higher in the experimental group than in the control group(P<0.05).The differences between pre-and post-operation in the five dimensions of social interaction,gestation,daily behavior,work and social psychology were better in the experimental group than in the control group(all P< 0.05).Conclusions Thoracoscopic sympathectomy at T2-4 and T3-4 is an effective treatment for sweaty hands.But surgery at T3-4 levels,which is associated with a larger improvement in quality of life and fewer complications,is recommended as a priority option for a surgical treatment.

RESUMEN

Objective To assess the perioperative efficacy and complications of intra-aortic balloon pump(IABP) in patients with cardiac operation.Methods A total of 36 cases with IABP patients from January 2011 to September 2016 were studied in Luhe Hospital of Beijing Affiliated to Capital Medical University,inluding 27 patients with coronary atherosclerotic heart disease(CAD),8 patients with valvular heart diseaseand and 1 patient with left atrial myxoma.The clinical data was collected to analyze IABP efficacy and complications.Results There were 10(27.8%) patients died,the mortality rate of CAD patients was lower than that of non-CAD patients(18.5% vs.55.6%,X2=4.615,P=0.032).The related complications after IABP implantation included:6(16.7%) cases suffered from thrombocytopenia,3(8.3%) cases with lower limb ischemia,2(5.6%) cases with lower extremity arterial thromboembolism,2(5.6%) cases with femoral artery pseudoaneurysm,2(5.6%) cases with acute pancreatitis,1(2.8%) cases with intestinal necrosis by mesenteric ischemia,2(5.6%) cases with gastrointestinal bleeding,3(8.3%) cases with hemorrhage in puncture site,4(11.1%) cases of fever,1(2.8%) case with pipeline leak,and 1(2.8%) case no pressure lumen data.After treatment with IABP,in 26 cases survival patients,the hemodynamic indexes(mean arterial pressure,heart rate,central venous pressure,urine volume) were significantly improved and the amount of dopamine and epinephrine decreased(t=-22.35,64.41,53.31,-23.82,26.07,14.15 respectively,P<0.05).Conclusion IABP can significantly improve the perioperative efficacy.The active treatment on complications is important to reduce the death induced by IABP.

RESUMEN

Objective To compare the clinical safety and efficacy of conservative treatment and surgery for traumatic flail chest in elderly patients.Methods From January 2013 to January 2017,68 elderly patients with traumatic flail chest in the Department of Thoracic Surgery,Beijing Lu He Hospital were enrolled.Based on different treatment methods,patients were divided into a conservative treatment group (n =34) and a surgery group (n =34),and the clinical safety and efficacy were compared between the two different treatment groups.Results Patients in the surgery group were associated with significantly decreased mechanical ventilation time,and days stayed in ICU and hospital,compared with the conservative treatment group (all P<0.05).However,incidences of atelectasis and/or pneumonia,duration of pain and oxygen therapy,total thoracic drainage volume and extubation time were similar between the two groups (all P>0.05).Rates of delayed or nonunion (2.9% vs.17.6%) and thoracic deformity (0.0% vs.11.8%) were significantly lower in the surgery group than those in the conservative treatment group (x2=3.981 and 4.250,both P<0.05).Conclusions Surgery can safely and effectively reduce incidences of complications in elderly patients with traumatic flail chest disease and shorten the time of hospitalization.Therefore,the short-and longterm effects are promising and warrant further investigation.

Objective To investigate the changes of serum levels of CysC and NT-proBNP in patients with hypertension .Meth-ods 89 patients with hypertension were selected and divided into five subgroups:Group A (n= 20) with simple hypertension;Group B(n=18) with hypertension and left ventricular hypertrophy ;Group C(n=15) with hypertension and heart failure;Group D(n=17) with hypertension and early renal damage;Group D(n=19) with hypertension and coronary heart disease .20 healthy in-dividuals were selected as control group .The serum levels of CysC and NT-proBNP were measured in all the groups .Results The serum levels of CysC in group B ,C and D were higher than control group (P<0 .05) ,and serum CysC level in group E was lower than control group(P<0 .05) .The serum level of NT-proBNP in group D was higher than control group(P<0 .05) ,and the serum NT-proBNP levels in group B ,C and E were significantly higher than control group (P<0 .01) .Conclusion The detection of CysC and NT-proBNP in patients with hypertension is helpful for understanding the progression of the disease and the occurrence of amalgamative diseases ,and it provides positive proof for clinical treatment .
