RESUMEN

The bird of paradise plant is a clumping tropical species native to South Africa. It is a dramatic plant with distinctive iridescent orange and midnight blue flowers that resemble an exotic bird peeking out from the broad leaves in autumn, winter and spring. An experiment was conducted during the two seasons of 2021 and 2022 at a private farm in Damanhour, Beheira Governorate, Egypt (31"°" 04 "°"N, 30"°" 47' °E) to study the effect different concentrations of nano-potassium and chitosan and their combinations on the bird of Paradise (Sterlitiza reginae). The experiment was conducted in a randomized complete block in a split-plot design with five replicates; nano-potassium was used at 0, 100, 150, and 200 mg/l and assigned to the main plots, whereas the sub-plots involved 0, 0.25, 0.50 and 0.75 g/l of chitosan. An increase in plant height and leaf length was recorded when the plants were treated with 200 mg/l nano-potassium and 0.75 g/l chitosan. Spraying plants with concentrations of 150 mg/l nano-potassium and 0.75 g/l chitosan is associated with the superiority of S. reginae plants in other traits, such as leaves wide, number of leaves/plant, days to flowering, number of inflorescence/plant, number of florets/inflorescence, stalk length and diameter, inflorescence weight, longevity of inflorescence, and flowering period, compared to the other treatments. We conclude that adding nano-potassium and/or chitosan to the bird of paradise plant leads to an improvement in terms of vegetative and yield characteristics under newly reclaimed lands.

BACKGROUND: TB remains a significant global health threat, claiming 1.3 million lives annually. The COVID-19 pandemic disrupted progress in the global TB response. Most patients with TB initially seek care from private providers, whereas only a small proportion are engaged by national programmes. The Global Fund is the major international source of funding for TB responses and supports the scale-up of innovative private-public mix (PPM) models in TB. METHODS: We collected programmatic and financial data on TB from 11 priority countries implementing PPM activities. Country examples and trends in the budget of Global Fund grants were analysed. RESULTS: These countries account for 60% of the global TB burden and Global Fund TB portfolio. PPM contributed 29% of national TB notifications in 2022 (range: 8% to 49%). During 2021-2023, US$1.4 billion was allocated for TB and US$155 million (11%) for PPM, while PPM contributed to 35% of national TB notification targets. PPM budgets increased over time from US$43 million (2002 to 2014) to US$129 million (2024 to 2026). CONCLUSION: The Global Fund's investments facilitated the expansion of innovative PPM models, improved access, and enhanced TB responses. Our indicative analysis underscores the need for evidence-based planning, collaboration, and increased domestic investment to accelerate the end of TB.


CONTEXTE: La TB reste une menace importante pour la santé mondiale, faisant 1,3 million de morts chaque année. La pandémie de COVID-19 a perturbé les progrès de la riposte mondiale à la TB. La plupart des patients atteints de TB recherchent d'abord des soins auprès de prestataires privés, tandis que seule une petite proportion est engagée par des programmes nationaux. Le Fonds mondial est la principale source internationale de financement de la lutte contre la TB et soutient l'extension de modèles innovants de partenariats public-privé (PPM, pour l'anglais « public-private mix ¼) dans le domaine de la TB. MÉTHODES: Nous avons recueilli des données programmatiques et financières sur la TB dans 11 pays prioritaires mettant en œuvre des activités de PPM. Des exemples de pays et les tendances du budget des subventions du Fonds mondial ont été analysés. RÉSULTATS: Ces pays représentent 60% de la charge mondiale de morbidité de la TB et du portefeuille du Fonds mondial de lutte contre la TB. La PPM a contribué à 29% des notifications nationales de TB en 2022 (fourchette : 8­49%). Au cours de la période 2021­2023, 1,4 milliard de dollars US ont été alloués à la TB et 155 millions de dollars US (11%) à la PPM, tandis que la PPM a contribué à 35% des cibles nationales de notification de la TB. Les budgets PPM ont augmenté au fil du temps, passant de 43 millions de dollars US (2002 à 2014) à 129 millions de dollars américains (2024 à 2026). CONCLUSION: Les investissements du Fonds mondial ont facilité l'expansion de modèles PPM innovants, l'amélioration de l'accès et le renforcement des ripostes à la TB. Notre analyse indicative souligne la nécessité d'une planification fondée sur des données probantes, d'une collaboration et d'une augmentation des investissements nationaux pour accélérer l'éradication de la TB.

BACKGROUND: During a field survey of urban flora in Alexandria city in 2019-2022, an interesting species belonging to the Solanaceae was collected from a newly archaeological excavation site and identified as Nicotiana glutinosa L. Many visits were made to the herbaria of Egypt to confirm the species records, but no single record was found. Reviewing the available literature revealed that this tropical American taxon was never recorded in the flora of Egypt. AIMS: The present study was focused on N. glutinosa growth structure and plant macro- and micromorphology. METHODS: Ten sampling sites were covered for N. glutinosa size structure. Plant samples were examined for stem anatomy, leaf, seed, and pollen morphology. RESULTS: The species size structure reveals that the individual size index ranges from 1.33 to 150 cm, while its density ranges from 4 to 273 individuals /100 m-2. N. glutinosa has successfully established itself in one of the archaeological sites in Egypt, showing a "healthy" population with a high degree of size inequality, characterized by a relative majority of the juvenile individuals. Voucher specimens were deposited in the Herbarium of Alexandria University (ALEX) Faculty of Science, another specimen is processed to make herbarium specimens at the Herbarium of the Botanic Garden (Heneidy et al. collection, deposition number. 5502). CONCLUSIONS: From our observations, N. glutinosa seems to have invasive potential, as it shows characteristics shared by most invasive species that are thought to help in their successful establishment in new habitats. This article emphasizes the importance of monitoring and regularly reporting the threats of alien invasive species to avoid any possible negative impacts on indigenous biodiversity in the future.

In the current study, we designed and synthesized a series of new quinoline derivatives 10a-p as antiproliferative agents targeting cancer through inhibition of VEGFR-2. Preliminary molecular docking to assess the interactions of the designed derivatives with the binding site of VEGFR-2 (PDB code: 4ASD) displayed binding poses and interactions comparable to sorafenib. The synthesized compounds exhibited VEGFR-2 inhibitory activity with IC50 ranging from 36 nM to 2.23 µM compared to sorafenib (IC50 = 45 nM), where derivative 10i was the most potent. Additionally, the synthesized derivatives were evaluated in vitro for their cytotoxic activity against HepG2 cancer cell line. Seven compounds 10a, 10c, 10d, 10e, 10i, 10n and 10o (IC50 = 4.60, 4.14, 1.07, 0.88, 1.60, 2.88 and 2.76 µM respectively) displayed better antiproliferative activity than sorafenib (IC50 = 8.38 µM). Compound 10i was tested against Transformed Human Liver Epithelial-2 normal cell line (THLE-2) to evaluate its selective cytotoxicity. Furthermore, 10i, as a potent representative of the series, was assayed for its apoptotic activity and cell cycle kinetics' influence on HepG2, its effects on the gene expression of VEGFR-2, and protein expression of the apoptotic markers Caspase-7 and Bax. Compound 10i proved to have a potential role in apoptosis by causing significant increase in the early and late apoptotic quartiles, a remarkable activity in elevating the relative protein expression of Bax and Caspase-7 and a significant reduction of VEGFR-2 gene expression. Collectively, the obtained results indicate that compound 10i has a promising potential as a lead compound for the development of new anticancer agents.

In continuation of our efforts to discover new structural chemotypes with significant chemotherapeutic activities, a novel series of pyrazolo[3,4-d]pyrimidine-based compounds linked to a piperazine ring, bearing different aromatic moieties, through different linkages was designed and synthesized as FLT3 inhibitors. All of the newly synthesized compounds were evaluated for their cytotoxicity on 60-NCI cell lines. Compounds with the piperazine acetamide linkage XIIa-f & XVI exhibited a remarkable anticancer activity among all of the tested compounds, especially against non-small cell lung cancer, melanoma, leukemia and renal cancer models. Furthermore, compound XVI (NSC no - 833644) was further screened with a 5-dose assay on nine subpanels and exhibited a GI50 between 1.17 and 18.40 µM. On the other hand, molecular docking and dynamics studies were performed to predict the binding mode of the newly synthesized compounds in the FLT3 binding domain. Finally, through a predictive kinetic study, several ADME descriptors were calculated.

Foreign body (FB) in the urinary bladder (UB) is uncommon and rarely reported in pediatric patients. FB migration into the UB is an extremely rare and unpredictable condition that needs a high index of suspicion with meticulous history taking and clinical reasoning, so diagnosis may be challenging. In this study, we report two cases of male pediatric patients from Sudan with FB in the UB, with a history of penetrating perineal trauma, both were presented with irritative lower urinary tract symptoms, history of penetrating perineal trauma, and unremarkable clinical examination. Both were diagnosed by abdominal Ultrasound study (USS) and confirmed by cystoscopy. One child was treated by endoscopic extraction, while the other was treated by open surgical extraction. The outcome of treatment of both the cases was satisfactory.

Objective: This study was performed to (I) evaluate the potential effect of advanced paternal age on global DNA methylation in spermatozoa, and (II) to investigate the association between the outcome of intracytoplasmic sperm injection (ICSI), semen parameters, and advanced paternal age. Material and Methods: This study comprised 230 semen samples collected from males with a mean age of 38.2±8.5 years. Medical records were used to gather clinical information related to the female partner. The participants were divided into three groups depending on age: age <30 years; age 30-40 years; and age >40 years. The DNA was extracted from purified spermatozoa. Then the sperm global DNA methylation, sperm DNA fragmentation, and chromatin decondensation were evaluated by an ELISA, TUNEL, and Chromomycin A3 staining, respectively. Results: The sample counts were n=50 (21.8%), n=90 (39.1%) and n=90 (39.1%) for the <30, 30-40 and >40 year age-groups, respectively. A significant variation was found in the age of males included in this study (p<0.001). There was a significant reduction in sperm count, total motility, and non-progressive motility in the older group compared to the younger group (p<0.001). There was also a significant elevation in chromatin decondensation, DNA fragmentation, and global DNA methylation of spermatozoa in the older age group (p<0.001). Finally, there was a significant positive correlation between the percentage of non-motile sperm, sperm chromatin decondensation, DNA fragmentation, global DNA methylation status, and paternal age (p<0.001). Conclusion: These results suggest that advanced paternal age increased the DNA fragmentation, chromatin decondensation, and global DNA methylation level in human spermatozoa, which negatively affects the ICSI outcomes in couples undergoing ICSI cycles.

We report the total synthesis and configurational assignment of pargamicin A, a highly oxidized nonribosomal peptide that potently inhibits the growth of drug-resistant bacteria. Our synthetic approach relies on late-stage piperazine ring formation and careful selection of condensation reagents to assemble the densely substituted hexapeptide backbone. This work enables the synthesis of pargamicin congeners for the development of structure-activity relationships and informs strategies for accessing other sterically congested piperazic acid-containing natural products.

A naturally occurring polyphenol called trans-resveratrol has received a lot of attention due to its possible health advantages for humans. The low solubility of trans-resveratrol and its isomerization upon UV exposure strongly limit its application as a skin-whitening agent. In the present study, to increase trans-resveratrol solubility, a new nanoformula was created by combining hydrophilic surfactants and oils. Trans-Resveratrol nanoformula has been prepared, characterized, and applied as a skin-whitening agent on the dorsal skin of Guinea pigs. The optimized trans-resveratrol nanoformula with a particle size of 63.49 nm displayed a single peak and a polydispersity index [0.36 ± 0.02]. In addition, the zeta potential of the optimized formula was -30.4 mV, confirming the high stability of this nanoformula. The melanin contents in the trans-resveratrol nanoformula-treated group were substantially lower than those of the control and the blank nanoformula-treated groups after staining of the dorsal skins [black areas] of guinea pigs with Fontana Mountain dye. The pigmentation index in the control, blank nanoformula, and optimized trans-resveratrol nanoformula were 329.4 ± 36.9, 335.8 ± 71.4, and 124.8 ± 19.6 respectively. Confirming this finding, immunohistochemistry analysis of skin tissues revealed that the expressions of melanogenesis-regulating proteins such as tyrosinase and microphthalmia-associated transcription factor were down-regulated. The safety of topical application of trans-resveratrol nanoformula was validated by no changes in free radical levels and oxidative stress markers proteins in the livers and kidneys of guinea pigs at the end of the experiment. Conclusions: A novel trans-resveratrol nanoformula as well as the mechanism whereby it promotes skin whitening effects were presented. Furthermore, the study illustrated that trans-resveratrol nanoformula is safe, non-toxic, and can be applied for skin whitening, although more research on human skin is needed.

OBJECTIVES: Ranula is one of the commonest salivary gland cysts that mostly occur due to mucus extravasation from the sublingual salivary gland. Treatment of this lesion is still somewhat shrouded in controversy and varies from conservative treatment to surgical excision of the causative gland. MATERIALS AND METHODS: This was a case series in quasi-experimental design that evaluated the outcome and complications of the modified micromarsupialization technique as a newly introduced treatment at our center for simple sublingual ranula over a 2-year period. RESULTS: Twenty patients were treated by the modified micromarsupialization technique and followed up for 1 year. There was complete resolution following this technique in 17 patients (85%) with no evidence of recurrence or complications, whereas 2 patients (10%) showed partial resolution and the remaining 1 patient (5%) showed a failure and recurrence. The age of the patient, the size of the ranula, and the retention of sutures throughout the study period did not significantly affect the treatment outcome. CONCLUSIONS: The modified micromarsupialization technique was a simple and effective treatment that should be used as a primary treatment option for simple ranulas and we recommend it to be the first-choice treatment before surgical excision of the sublingual gland, especially in a resource-challenged economy like ours.

Computer vision and image processing techniques have been extensively used in various fields and a wide range of applications, as well as recently in surface treatment to determine the quality of metal processing. Accordingly, digital image evaluation and processing are carried out to perform image segmentation, identification, and classification to ensure the quality of metal surfaces. In this work, a novel method is developed to effectively determine the quality of metal surface processing using computer vision techniques in real time, according to the average size of irregularities and caverns of captured metal surface images. The presented literature review focuses on classifying images into treated and untreated areas. The high computation burden to process a given image frame makes it unsuitable for real-time system applications. In addition, the considered current methods do not provide a quantitative assessment of the properties of the treated surfaces. The markup, processed, and untreated surfaces are explored based on the entropy criterion of information showing the randomness disorder of an already treated surface. However, the absence of an explicit indication of the magnitude of the irregularities carries a dependence on the lighting conditions, not allowing to explicitly specify such characteristics in the system. Moreover, due to the requirement of the mandatory use of specific area data, regarding the size of the cavities, the work is challenging in evaluating the average frequency of these cavities. Therefore, an algorithm is developed for finding the period of determining the quality of metal surface treatment, taking into account the porous matrix, and the complexities of calculating the surface tensor. Experimentally, the results of this work make it possible to effectively evaluate the quality of the treated surface, according to the criterion of the size of the resulting irregularities, with a frame processing time of 20 ms, closely meeting the real-time requirements.

Different activities related to uranium mining and nuclear industry may have a negative impact on the environment. Bioremediation of nuclear pollutants using microorganisms is an effective, safe, and economic method. The present study compared the uranium biosorption efficiency of two immobilized algae: Nostoc sp. (cyanophyte) and Scenedesmus sp. (chlorophyte). Effects of metal concentration, contact time, pH, and biosorbent dosage were also studied. The maximum biosorption capacity (60%) by Nostoc sp. was obtained at 300 mg/l uranium solution, 60 min, pH 4.5, and 4.2 g/l algal dosage, whereas Scenedesmus sp. maximally absorbed uranium (65 %) at 150 mg/l uranium solution, 40 min, pH 4.5, and 5.6 g/l of algal dosage. The interaction of metal ions as Na2SO4, FeCl3, CuCl2, NiCl2, CoCl2, CdCl2, and AlCl3 did not support the uranium biosorption by algae. The obtained data was adapted to the linearized form of the Langmuir isotherm model. The experimental qmax values were 130 and 75 mg/g for Nostoc sp. and Scenedesmus sp., respectively. Moreover, the pseudo-second-order kinetic model was more applicable, as the calculated parameters were close to the experimental data. The biosorbents were also characterized by Fourier-transform infrared spectroscopy (ATR-FTIR), energy-dispersive X-ray spectroscopy (EDX), and scanning electron microscopy (SEM) analyses. The results suggest the applicability of algae, in their immobilized form, for recovery and biosorption of uranium from aqueous solution.

OBJECTIVES: To estimate the risk of malignancy in indeterminate thyroid nodules and to determine whether certain clinical or radiological parameters can predict the risk of malignancy. METHODS: This retrospective study enrolled all adult patients (age ≥14 years) with a cytological diagnosis of atypia/follicular lesion of undetermined significance and follicular neoplasm/suspicious for a follicular neoplasm between January 2014 and January 2020. Fifty patients with surgically treated primary thyroid nodules, documented final histological diagnosis, and ultrasound examination records were included. Thyroid nodules were evaluated radiologically using Thyroid Imaging Reporting and Data System introduced by the American College of Radiology (2017). RESULTS: Forty-two (84.0%) female and 8 (16.0%) male patients were enrolled in the study. The malignancy risks were 44.8% for Bethesda III and 28.6% for Bethesda IV. The malignancy risks for the Thyroid Imaging Reporting and Data System categories were 33.3% (TR2), 39.1% (TR3), 35.3% (TR4), and 50% (TR5). No significant associations were observed between age, gender, Bethesda category, and Thyroid Imaging Reporting and Data System and the risk of malignancy. CONCLUSION: None of the clinical or radiological characteristics evaluated in this study contributed to the cancer risk stratification of thyroid nodules with indeterminate cytology. A prospective multicenter study is needed to better understand cytologically indeterminate thyroid nodules.

The challenge continues globally triggered by the absence of an approved antiviral drug against COVID-19 virus infection necessitating global concerted efforts of scientists. Nature still provides a renewable source for drugs used to solve many health problems. The aim of this work is to provide new candidates from natural origin to overcome COVID-19 pandemic. A virtual screening of the natural compounds database (47,645 compounds) using structure-based pharmacophore model and molecular docking simulations reported eight hits from natural origin against SARS-CoV-2 main proteinase (Mpro) enzyme. The successful candidates were of terpenoidal nature including taxusabietane, Isoadenolin A & C, Xerophilusin B, Excisanin H, Macrocalin B and ponicidin, phytoconstituents isolated from family Lamiaceae and sharing a common ent-kaurane nucleus, were found to be the most successful candidates. This study suggested that the diterpene nucleus has a clear positive contribution which can represent a new opportunity in the development of SARS-CoV-2 main protease inhibitors.

Leiomyosarcoma is a rarely seen neoplasm of the ureter. Malignant tumors of smooth muscle of the ureter are extremely rare, and about 22 cases of leiomyosarcoma of ureter have been reported to date. A 57-year-old diabetic Pakistani man presented with a dull ache pain in the right flank. Past surgical history was three ureteroscopic surgeries for a ureteric stricture. Computed tomography showed a stricture with a peri-ureteral soft tissue mass of 11 mm x 5 mm at the middle third of the ureter at the level of common iliac vessels. laparoscopic excision with safety margin and right ureterovesical reimplantation is performed. Diagnosis of leiomyosarcoma of the right ureter was made, and one iliac lymph node was excised and was positive for tumor by pathologic examination. Although leiomyosarcoma is rarely seen in urinary tract, it should be considered in the differential diagnosis of ureteral stricture disease and retroperitoneal tumors.

RESUMEN

The purpose of this pilot study was to investigate the effects of the transfusion of one erythrocyte concentrate on the number of circulating red blood cell extracellular vesicles (RBC-EVs) and their clearance time. Six, healthy volunteers donated their blood and were transfused with their RBC concentrate after 35-36 days of storage. One K2 EDTA and one serum sample were collected before donation, at four timepoints after donation and at another six timepoints after transfusion. RBC-EVs were analyzed on a Cytek Aurora flow cytometer. A highly significant increase (p < 0.001) of RBC-EVs from an average of 60.1 ± 19.8 (103 /µL) at baseline to 179.3 ± 84.7 (103 /µL) in the first 1-3 h after transfusion could be observed. Individual differences in the response to transfusion became apparent with one volunteer showing no increase and another an increased concentration at one timepoint after donation due to an influenza infection. We concluded that in an individualized passport approach, increased RBC-EVs might be considered as additional evidence when interpreting suspicious Athletes Biological Passport (ABPs) but for this additional research related to sample collection and transport processes as well as method development and harmonization would be necessary.

Two strains of the halophilic alga Dunaliella parva, a wild type (WT) and a transgenic strain (D-PSY) containing an exogenous phytoene synthase gene (PSY), were used to investigate the growth, carotenoid accumulation, and carotenoid antioxidant properties under nitrogen starvation, cobalt and biochar treatments. D-PSY had higher carotenoid content (1.8 times) compared to the WT. The applied stressors stimulated the carotenoid content of both WT and D-PSY especially. The carotenoids were assayed for the potential antioxidant activities by five different assays. Generally, the antioxidant activities of D-PSY carotenoids were superior to that of WT. The biochar and nitrogen treatments generally enhanced the antioxidant activities of the carotenoids, whereas cobalt came third in this respect. The D-PSY transgenic algal strain has both high carotenoids content and antioxidant properties which enhanced under the relatively lower concentrations of the applied stressors. The results have shown to lead to an accurate application of the transgenic alga as a source of potent antioxidant compounds. Intra-city railway areas are deemed large greenspaces and are believed to be key in enhancing the diversity and dynamics of urban flora. In the current study, the floristic composition and diversity along intra-city railway and tram tracks in Alexandria were surveyed. The floristic composition of the plant communities in relation to environmental factors, ecological indicators, and level of human impact was analyzed using multivariate analysis (two-way indicator species analysis (TWINSPAN) for classification and detrended correspondence analysis (DECORANA) for ordination. The multivariate ordination techniques (CCA) revealed differences in the environmental factors and climatic factors influencing the floristic composition of the railway and tram track habitats. Tram tracks suffered higher human impact while maintaining higher vitality and cover compared to train tracks. Species recorded were mainly therophytes, followed by phanerophytes and hemicryptophytes dominated by native species; however, invasive species' contribution was higher compared to surrounding regions. The number of invasive species was greater in railway areas compared to tram track areas (19 and 15, respectively). The occurrence of two endemic species (Sinapis allionii and Sonchus macrocarpus) with limited national distribution highlights the importance of these habitats as valuable refuge areas for rare and endangered species worthy of conservation action.

RESUMEN

Osteoid osteoma (OO) of the elbow is a very rare entity as it typically occurrs in the lower extremity of extra-articular long bones. Hereby, we report a case for a 28-year-old female patient has been diagnosed with a unique presentation of sub-periosteal osteoid osteoma in the intra-articular portion of distal humerus. She underwent arthroscopic excision for the lesion which is considered a reliable minimally invasive management modality. In addition to this, findings of a 1-year follow-up after excision is reported.
