RESUMEN

This study aimed to investigate the mechanisms by which dark septate endophytes (DSE) regulate salt tolerance and the accumulation of bioactive constituents in licorice. First, the salt stress tolerance and resynthesis with the plant effect of isolated DSE from wild licorice were tested. Second, the performance of licorice inoculated with DSE, which had the best salt-tolerant and growth-promoting effects, was examined under salt stress. All isolated DSE showed salt tolerance and promoted plant growth, withCurvularia lunata D43 being the most effective. Under salt stress, C. lunata D43 could promote growth, increase antioxidant enzyme activities, enhance glycyrrhizic acid accumulation, improve key enzyme activities in the glycyrrhizic acid synthesis pathway, and induce the expression of the key enzyme gene and salt tolerance gene of licorice. The structural equation model demonstrated that DSE alleviate the negative effects of salt stress through direct and indirect pathways. Variations in key enzyme activities, gene expression, and bioactive constituent concentration can be attributed to the effects of DSE. These results contribute to revealing the value of DSE for cultivating medicinal plants in saline soils.

RESUMEN

Juices and beverages are produced by industry for long-distance distribution and shelf-stability, providing valuable nutrients. However, their nutritional value is often underestimated due to insufficient analytical methods. We have employed non-targeted analysis through a standardized analytical protocol, taking advantage of Data Independent Acquisition (DIA) technique and a novel Chromatographic Retention Behavior (CRB) data deconvolution algorithm. After analyzing 9 fruits and their products, correlations between fruits and their juices are accurately digitalized by similarities of their LC-MS fingerprints. We also specify non-targeted molecules primarily associate with nutrient loss in these analyzed juice products, including nitrogenous nutrients, flavonoids, glycosides, and vitamins. Moreover, we unveiled previously unreported fruit-characteristic metabolites, of which reconstituted-from-concentrate (RFC) juices contain over 40% of the content found in their fresh counterparts. Conclusively, our method establishes a quantitative benchmark for rational selection of RFC juices to substitute natural fruits.

RESUMEN

Sediment microorganisms are the main drivers of the material circulation and organic matter degradation processes in rural black and odorous water bodies(RBOWB), and the community structure of sediment microorganisms follows the changes in the external environment. Here, the pollutant indicators, including nitrogen, phosphorus, and heavy metals in the overlying water and sediment of 29 RBOWB in Dongming County of Heze City were measured, respectively. Combined with Illumina sequencing results, the composition and diversity characteristics of sediment bacterial communities in RBOWB and their correlation with environmental factors were further analyzed. The experimental results showed a wide distribution of pollutants in both of the overlying water and sediment in the RBOWB of this region. Compared with agricultural non-point source pollution, the concentrations of nitrogen and phosphorus pollutants in the overlying water with domestic sewage as the main source of pollution were 3.1 and 1.5 times higher than those of agricultural non-point source pollution, respectively. In addition, the contents of heavy metals in the sediments of RBOWB were generally lower than the soil element background value in Heze City. The dominant bacteria phyla in the sediments of the RBOWB were Proteobacteria, Actinobacteria, Chloroflexi, Firmicutes, and Acidobacteria, and the total abundance of these five dominant phyla accounted for 70.3%-83.6% of all sequences. The dominant classes were γ-Proteobacteria, α-Proteobacteria, Anaerolineae, and Actinobacteria. The dominant genera were Thiobacillus and Pseudarthrobacter. Moreover, Spearman correlation analysis showed that the environmental factors of DO, COD, TN, TP, and organic matter exerted significant effects(P<0.05) on sediment bacterial genera in RBOWB, and sediment bacterial community richness was significantly influenced by TN(P<0.05). The above results provided the microbiological knowledge for treating RBOWB.

RESUMEN

BACKGROUND: This study is to evaluate and compare the improvement of upper airway morphology and hyoid bone position in children with Class II mandibular retrusion treated with Invisalign mandibular advancement (MA) and Twin-Block (TB) appliances, utilizing cone beam computed tomography (CBCT). METHODS: 32 children aged between 8 and 11.5 years old were included in this study, with an average age of 10.2 years old. These children were divided into two groups, MA and TB, with 16 children in each group. Changes in upper airway morphology and hyoid bone position before and after treatment were analyzed using CBCT. RESULTS: (1) Changes in upper airway before and after treatment: the oropharynx volume (Or-V), the oropharynx minimum cross-sectional area (Or-mCSA), the hypopharynx volume (Hy-V), and the hypopharynx minimum cross-sectional area (Hy-mCSA) in both the MA and TB groups increased after treatment, and the differences were statistically significant (P < 0.05) compared to pre-treatment status. (2) Changes in hyoid bone position before and after treatment: The distances between H point and third cervical vertebra (H-C3), H point and pogonion (H-RGN), H point and mandibular plane (H-MP), H point and Frankfort horizontal plane (H-FH), H and S point (H-S), and H point and palatal plane (H-PP) in both the MA and TB groups increased after treatment, and the differences were statistically significant (P < 0.05). CONCLUSION: Both MA and TB appliances effectively improved the structural narrowness of the upper airway and reduced respiratory resistance, thus improving breath quality. However, MA showed more effectiveness in improving the narrowest part of the hypopharynx compared to TB. Both appliances also promoted anterior downward movement of the hyoid bone, which opens the upper airway of the oropharynx and hypopharynx and helps the upper airway morphology return to normal range.

RESUMEN

Based on the air quality data and conventional meteorological data of the Nanjing Region from January 2015 to December 2016, to analyze the characteristics of O3 concentration changes in the Nanjing Region, a light gradient boosting machine (LightGBM) model was established to predict O3 concentration. The model was compared with three machine learning methods that are commonly used in air quality prediction, including support vector machine, recurrent neural network, and random forest methods, to verify its effectiveness and feasibility. Finally, the performance of the prediction model was analyzed under different meteorological conditions. The results showed that the variation in O3 concentration in Nanjing had significant seasonal differences and was affected by a combination of its pre-concentration, meteorological factors, and other air pollutant concentrations. The LightGBM model predicted the ground-level O3 concentration in the Nanjing area more precisely to a large extent (R2=0.92), and the model outperformed other models in prediction accuracy and computational efficiency. In particular, the model showed a significantly higher prediction accuracy and stability than that of other models under a high-temperature condition that was more likely prone to ozone pollution. The LightGBM model was characterized by its high prediction accuracy, good stability, satisfactory generalization ability, and short operation time, which broaden its application prospect in O3 concentration prediction.

RESUMEN

To investigate the mechanism of this spiro conjugation magnetic behavior, we designed and synthesized three diradicals─22'SBF-NN, 44'SBF-NN, and 27SBF-NN. They are bridged by spirobifluorene and nitronyl nitroxide (NN) diradicals as the spin centers. Notably, by SQUID and electron paramagnetic resonance (EPR) zero-field splitting data analyses, the 22'SBF-NN and 27SBF-NN diradicals exhibit intramolecular, distinctly antiferromagnetic (AF) coupling, with 2J(22'SBF-NN)/kB = -5.86 K and 2J(27SBF-NN)/kB = -24.6 K, respectively. The AF of 22'SBF-NN is opposite to that predicted by the spin density alternation rule based on Hund's rule. Diradical intramolecular conjugation coupling bridged by spiro-carbon conjugation is discussed, in which the 22'SBF-NN is smaller than that of 27SBF-NN, corresponding to the room-temperature EPR characterization. This spiro conjugation is weaker than the traditional planar conjugation and generally leads to a weaker spin-spin coupling in the helical biradical molecule. The EPR spectrum of the 44'SBF-NN diradical shows a deformed nine-line curve, indicating intramolecular exchange coupling. The density functional theory calculation gives a very weak coupling constant of 2Jcalc/kB = 0.06 K, with ferromagnetic (FM) interaction as the proof, which is consistent with the spin-polarized prediction. Further analysis of magnetic susceptibility χm and VT-EPR data shows that there is indeed an extremely weak FM interaction in the 44' position diradical. We find the bridge, which is a 44' substituted SBF structure, blocks the conjugation and contains a larger twist in steric hindrance, which also hampers sufficient spin density delocalization, resulting in a much weaker spin coupling interaction. Combined with the analysis of molecular orbital calculation results, the anomalous intramolecular AF coupling mechanism of 22'SBF-NN is further explained.

RESUMEN

Long noncoding RNAs (lncRNAs) play a critical role in the regulation of atherosclerosis. Here, we investigated the role of the lncRNA growth arrest-specific 5 (lncR-GAS5) in atherogenesis. We found that the enforced expression of lncR-GAS5 contributed to the development of atherosclerosis, which presented as increased plaque size and reduced collagen content. Moreover, impaired autophagy was observed, as shown by a decreased LC3II/LC3I protein ratio and an elevated P62 level in lncR-GAS5-overexpressing human aortic endothelial cells. By contrast, lncR-GAS5 knockdown promoted autophagy. Moreover, serine/arginine-rich splicing factor 10 (SRSF10) knockdown increased the LC3II/LC3I ratio and decreased the P62 level, thus enhancing the formation of autophagic vacuoles, autolysosomes, and autophagosomes. Mechanistically, lncR-GAS5 regulated the downstream splicing factor SRSF10 to impair autophagy in the endothelium, which was reversed by the knockdown of SRSF10. Further results revealed that overexpression of the lncR-GAS5-targeted gene miR-193-5p promoted autophagy and autophagic vacuole accumulation by repressing its direct target gene, SRSF10. Notably, miR-193-5p overexpression decreased plaque size and increased collagen content. Altogether, these findings demonstrate that lncR-GAS5 partially contributes to atherogenesis and plaque instability by impairing endothelial autophagy. In conclusion, lncR-GAS5 overexpression arrested endothelial autophagy through the miR-193-5p/SRSF10 signaling pathway. Thus, miR-193-5p/SRSF10 may serve as a novel treatment target for atherosclerosis.

RESUMEN

AMA GC5000BTX was used to monitor the mixing ratio of benzene, toluene, ethylbenzene, m,p-xylene, o-xylene, and styrene (BTESX) in the atmosphere of the northern suburb of Nanjing from January 2014 to December 2016. The temporal variation characteristics of BTESX and the influence of meteorological elements on it were analyzed, and the characteristic ratio method (T/B) was used to qualitatively analyze the source of BTESX. Finally, the human exposure analysis and evaluation method of EPA was used to evaluate the health risk of BTESX. The results showed that during the observation period, the average mixing ratio of BTESX was (7.28±6.63)×10-9, and the mixing ratio of benzene was the highest at (2.45±3.91)×10-9. The mixing ratio of other species from large to small was toluene>ethylbenzene>m,p-xylene>o-xylene>styrene, which were (2.41±2.61)×10-9, (1.37±1.28)×10-9, (0.51±0.48)×10-9, (0.3±0.36)×10-9, and (0.22±0.42)×10-9, respectively. Due to the existence of stable aromatic sources, the monthly and seasonal variation in BTESX mixing ratio was not as obvious as that of other species (NOx, CO, SO2, PM2.5, etc.). The weekend effect of BTESX and other pollutants was not significant. The mixing ratio of BTESX was largely affected by the short distance transportation of chemical enterprises and traffic trunk roads in the northeast, resulting in a large mixing ratio of BTESX in the northeast. The mixing ratio of BTESX was jointly affected by relative humidity and temperature, and its high value area was mainly located in the range of 30%-70% relative humidity. In this range of relative humidity, the high value range of BTESX volume fraction increased with the elevation of temperature. The HI (hazard index) of BTESX in different seasons was within the safety range recognized by EPA, whereas the R (carcinogenic risk of benzene) value was higher than the safety threshold specified by EPA. At the same time, the HI and R values were higher in summer, to which great attention should be paid.

RESUMEN

Dynamic functional connectivity (FC) analysis can capture time-varying properties of connectivity; however, studies focusing on dynamic FC in patients with end-stage renal disease (ESRD) are very limited. This is the first study to explore the dynamic aspects of whole-brain FC and topological properties in ESRD patients. Resting-state functional magnetic resonance imaging data were acquired from 100 ESRD patients [50 hemodialysis (HD) patients and 50 non-dialysis patients] and 64 healthy controls (HCs). Independent component analysis, a sliding-window approach and graph-theory methods were used to study the dynamic FC properties. The intrinsic brain FC were clustered into four configuration states. Compared with HCs, both patient groups spent longer time in State 3, in which decreased FC between subnetworks of the default mode network (DMN) and between the dorsal DMN and language network was observed in these patients, and a further reduction in FC between the DMN subnetworks was found in HD patients compared to non-dialysis patients. The number of transitions and the variability of global and local efficiency progressively decreased from that in HCs to that of non-dialysis patients to that of HD patients. The completion time of Trail Making Test A and Trail Making Test B positively correlated with the mean dwell time of State 3 and negatively correlated with the number of transitions in ESRD patients. Our findings suggest impaired functional flexibility of network connections and state-specific FC disruptions in patients with ESRD, which may underlie their cognitive deficits. HD may have an adverse effect on time-varying FC.

RESUMEN

Atherosclerotic carotid plaques have been shown to be closely associated with the risk of stroke. Since patients with symptomatic carotid plaques have a greater risk for stroke, stroke risk stratification based on the classification of carotid plaques into symptomatic or asymptomatic types is crucial in diagnosis, treatment planning, and medical treatment monitoring. A deep learning technique would be a good choice for implementing classification. Usually, to acquire a high-accuracy classification, a specific network architecture needs to be designed for a given classification task. In this study, we propose an object-specific four-path network (OSFP-Net) for stroke risk assessment by integrating ultrasound carotid plaques in both transverse and longitudinal sections of the bilateral carotid arteries. Each path of the OSFP-Net comprises of a feature extraction subnetwork (FE) and a feature downsampling subnetwork (FD). The FEs in the four paths use the same network structure to automatically extract features from ultrasound images of carotid plaques. The FDs use different object-specific pooling strategies for feature downsampling based on the observation that the sizes and shapes in the feature maps obtained from FEs should be different. The object-specific pooling strategies enable the network to accept arbitrarily sized carotid plaques as input and to capture a more informative context for improving the classification accuracy. Extensive experimental studies on a clinical dataset consisting of 333 subjects with 1332 carotid plaques show the superiority of our OSFP-Net against several state-of-the-art deep learning-based methods. The experimental results demonstrate better clinical agreement between the ground truth and the prediction, which indicates its great potential for use as a risk stratification and as a monitoring tool in the management of patients at risk for stroke.

RESUMEN

Saline wastewater poses a challenge during bio-treatment process due to salinity affecting the physiological activity of microorganisms and inhibiting their growth and metabolism. Thus, screening and engineering the salt-tolerant strains with stronger performances are urgent. Shewanella aquimarina XMS-1, a salt-tolerant dissimilated metal reducing bacteria (DMRB), was isolated from seawater environment. Its ability for reducing pollutants and generating electricity was enhanced by overexpression of riboflavin synthesis pathway encoding genes from S. oneidensis MR-1 under salt stress. Furthermore, upon contact with graphene oxide (GO), the engineered strain XMS-1/pYYDT-rib with enhanced flavins synthesis could reduce GO and self-assemble to form a three-dimensional (3D) biohybrid system named XMS-1/flavins/rGO. This 3D biohybrid system significantly enhanced the EET efficiency of S. aquimarina XMS-1. Our findings provide a feasible strategy for treatment of salt-containing industrial wastewater contaminated by metal and organic pollutants.

RESUMEN

Objective:To preliminarily investigate the effect of circIGF2BP3 on autophagy in photoaged dermal fibroblasts. Methods:Human dermal fibroblasts were isolated from circumcised foreskin tissues from 6 children in the Department of Urological Surgery, Third Affiliated Hospital of Sun Yat-sen University. An ultraviolet A (UVA) -induced photoaged human dermal fibroblast model (UVA radiation group) was established by repeated UVA radiation at a dose of 10 J/cm 2 for 14 consecutive days, and human dermal fibroblasts receiving no treatment served as control group. The photoaged cell model was verified by β-galactosidase staining, Western blot analysis for determining P21 protein expression, and cell counting kit-8 (CCK8) assay for evaluating cell viability. Moreover, Western blot analysis was performed to determine the protein expression of autophagy-related proteins P62, microtubule-associated protein 1 light chain 3 (LC3) -Ⅰand LC3-Ⅱ in photoaged human dermal fibroblasts, and real-time quantitative RCR (qRT-PCR) to verify the differential expression of circIGF2BP3 between photoaged and normal human dermal fibroblasts. Furthermore, circIGF2BP3 was biologically annotated. Some cultured primary human dermal fibroblasts were divided into 4 groups: empty vector group transfected with an empty vector, UVA + empty vector group transfected with an empty vector followed by repeated UVA radiation, circIGF2BP3 group transfected with a circIGF2BP3-overexpressing lentiviral vector, UVA + circIGF2BP3 group transfected with a circIGF2BP3-overexpressing lentiviral vector followed by repeated UVA radiation. Western blot analysis was performed to determine the expression of autophagy-related proteins. Statistical analysis was carried out by using t test, one-way analysis of variance and least significant difference- t test. Results:Compared with the control group, the UVA radiation group showed significantly increased proportions of β-galactosidase-positive cells (61.33% ± 5.78% vs. 6.37% ± 0.32%, t = 9.49, P < 0.01) and P21 expression (1.25 ± 0.03 vs. 1.00 ± 0.05, t = 4.26, P < 0.05), but significantly decreased cell viability (74.33% ± 3.48% vs. 100%, t = 7.38, P < 0.01). Moreover, the P62 expression and LC3-Ⅱ/Ⅰ ratio were significantly higher in the UVA radiation group than in the control group (both P < 0.05). The relative expression of circIGF2BP3 was 0.72 ± 0.04 in the photoaged human dermal fibroblasts, which was significantly lower than that in the normal human dermal fibroblasts (1.00 ± 0.03, t = 5.46, P < 0.01). The P62 expression and LC3-Ⅱ/Ⅰ ratio were significantly lower in the circIGF2BP3 group (0.60 ± 0.01, 0.71 ± 0.01, respectively) than in the empty vector group (1.00 ± 0.02, 1.00 ± 0.01, t = 16.25, 2.75, P < 0.01, < 0.05, respectively), and lower in the UVA + circIGF2BP3 group (1.05 ± 0.02, 2.04 ± 0.05, respectively) than in the UVA + empty vector group (1.31 ± 0.02, 2.72 ± 0.14, t = 10.493, 6.472, respectively, both P < 0.01) . Conclusion:circIGF2BP3 can regulate autophagy in UVA-induced photoaged dermal fibroblasts, which provides a new potential therapeutic target for the prevention and treatment of skin photoaging.

OBJECTIVE@#To investigate the effect of PPP2R5C to the activity of Molt-4 cells in childhood acute T lymphocytic leukemia and its mechanism.@*METHODS@#The small interfering RNA (siRNA) technology targeting PPP2R5C gene was used to down-regulate the expression of PPP2R5C in Molt-4 cells. At the same time, a blank control group, a negative control group and a 17-DMAG group were set up. The cells in the negative control group were transfected with siRNA-NC, the cells in 17-DMAG group were treated with the HSP90 inhibitor 17-DMAG at a final concentration of 6.4 μmol/L for 48 h. Real-time fluorescent quantitative PCR (RT-qPCR) and Western blot were used to detect transfection efficiency; CCK-8 method was used to detect the proliferation activity of the cells in each group, EdU was used to detect the proliferation level of the cells in each group, flow cytometry was used to detect the cell cycle distribution ratio of the cells in each group, Annexin V-FITC/PI staining was used to detect the apoptosis of the cell, RT-qPCR and Western blot were used to detect the expression changes of heat shock protein 90 (HSP90) and glucocorticoid receptor (GR) of the cells in each group.@*RESULTS@#After Molt-4 cells were transfected with siRNA-PPP2R5C, the expression of PPP2R5C mRNA and protein in the cells were down-regulated significantly compared with those in the blank control group and the si-NC group (P<0.05); compared with cells in the blank control group and the si-NC group, the proliferation activity of the cells in the siRNA-PPP2R5C group and the 17-DMAG group significantly decreased (P<0.05), and the rate of EdU positive cells was significantly reduced (P<0.05); the proportion of the cells in G1 phase decreased while the proportion of the cells in G2 phase increased (P<0.05), the apoptosis rate of the cells also increased significantly (P<0.05); in addition, the expression of PPP2R5C mRNA and protein of the cells in siRNA-PPP2R5C group was significantly down-regulated compared with those in the blank control group and si-NC group (P<0.05). The expressions of PPP2R5C mRNA and protein in the 17-DMAG group were also significantly down-regulated compared with those in the blank control group and si-NC group (P<0.05).@*CONCLUSION@#Down-regulation of PPP2R5C gene expression can inhibit Molt-4 cell activity in childhood acute T lymphocytic leukemia, block the cells in G2 phase, and promote cell apoptosis, the mechanism may be related to the inhibition of HSP90-GR signaling pathway.

RESUMEN

A visualized, rapid, simple method was developed based on loop-mediated isothermal amplification (LAMP) to detect Vibrio splendidus in aquatic products and aquaculture waters. TakingVibrio splendidus as the research object, and the toxR gene of Vibrio splendidus as the target gene, the boiling method was determined as a fast method suitable for the genomic DNA extraction of Vibriosplendidus. The optimized and screened primers could specifically detect Vibrio splendidus, and the sensitivity of nucleic acid concentration detection could reach 10
