BACKGROUND: Polymeric nanoparticles can be used for wound closure and therapeutic compound delivery, among other biomedical applications. Although there are several nanoparticle obtention methods, it is crucial to know the adequate parameters to achieve better results. Therefore, the objective of this study was to optimize the parameters for the synthesis, purification, and freeze-drying of chitosan nanoparticles. We evaluated the conditions of agitation speed, anion addition time, solution pH, and chitosan and sodium tripolyphosphate concentration. RESULTS: Chitosan nanoparticles presented an average particle size of 172.8 ± 3.937 nm, PDI of 0.166 ± 0.008, and zeta potential of 25.00 ± 0.79 mV, at the concentration of 0.1% sodium tripolyphosphate and chitosan (pH 5.5), with a dripping time of 2 min at 500 rpm. The most representative factor during nanoparticle fabrication was the pH of the chitosan solution, generating significant changes in particle size and polydispersity index. The observed behavior is attributed to the possible excess of sodium tripolyphosphate during synthesis. We added the surfactants poloxamer 188 and polysorbate 80 to evaluate the stability improvement during purification (centrifugation or dialysis). These surfactants decreased coalescence between nanoparticles, especially during purification. The centrifugation increased the zeta potential to 40.8-56.2 mV values, while the dialyzed samples led to smaller particle sizes (152-184 nm). Finally, freeze-drying of the chitosan nanoparticles proceeded using two cryoprotectants, trehalose and sucrose. Both adequately protected the system during the process, and the sugar concentration depended on the purification process. CONCLUSIONS: In Conclusion, we must consider each surfactant's benefits in formulations for selecting the most suitable. Also, it is necessary to do more studies with the molecule to load. At the same time, the use of sucrose and trehalose generates adequate protection against the freeze-drying process, even at a 5% w/v concentration. However, adjusting the percentage concentration by weight must be made to work with the CS-TPP NPs purified by dialysis.

Garlic (Allium sativum) is an important cash food crop, and the biotechnology industry has considerable interest in the plant because of its medicinal importance. These medicinal properties are attributed to organosulphur compounds as the accumulation of these compounds varies according to genotype, locality, light quality, and cultivation practices. In this study, we compared a newly developed garlic variety NARC-G1 by National Agricultural Research Centre (NARC), Islamabad, Pakistan with three different garlic cultivars and highlighted the distinctive attributes like phenotypic characteristics, the content of allicin, elemental profile, and gene polymorphism. Phenotypic analysis showed NARC-G1 has significantly higher bulb weight (66.36g ± 18.58), single clove weight (5.87g ± 1.041), and clove width (17.41mm ± 0.95) which directly correlates to the size of the garlic. The analytical analysis showed the highest allicin content (4.82 ± 0.001) in NARC-G1. Genotyping of the alliinase in all four cultivars showed indels in the gene resulting in distinguishable changes in organosulphur compounds' profile. NARC-G1 is unique from other garlic cultivars and could be the best choice for mass production with proper cultivation and irrigation management. Moreover, for Pakistan NARC-G1 could be a potential contender to earn the industrial benefits with inland cultivation instead of importing garlic alleviating the economic burden.


O alho (Allium sativum) é uma importante cultura alimentar de rendimento e a indústria biotecnológica tem um interesse considerável na planta devido à sua importância medicinal. As propriedades medicinais são atribuídas aos compostos organossulfurados, enquanto o acúmulo de sulfóxidos de cisteína (CSOs) varia de acordo com genótipo, localidade, qualidade da luz e práticas de cultivo. Neste estudo, comparamos uma variedade de alho recém-desenvolvida NARC-G1 pelo Centro Nacional de Pesquisa Agropecuária (NARC), Islamabade, Paquistão, com três diferentes cultivares de alho, e destacamos os atributos distintivos, como características fenotípicas, conteúdo de alicina, perfil elementar e gene polimorfismo. A análise fenotípica mostrou que o NARC-G1 tem peso de bulbo significativamente maior (66.36g ± 18.58), peso de um único dente (5.87g ± 1.041) e largura do dente (17.41mm ± 0.95), o que se correlaciona diretamente com o tamanho do alho. A análise analítica indicou maior teor de alicina (4.82 ± 0.001) no NARC-G1. A genotipagem do gene da aliinase em todas as quatro cultivares mostrou indels no gene, resultando em mudanças distinguíveis no perfil das CSOs. O NARC-G1 é único em relação a outras cultivares de alho e pode ser a melhor escolha para produção em massa com cultivo adequado e manejo de irrigação. Além disso, para o Paquistão, o NARC-G1 pode ser um potencial candidato para obter os benefícios industriais com o cultivo no interior, em vez de importar alho, aliviando o ônus econômico.

Hydrogels are three-dimensional structures with specific features that render them useful for biomedical applications, such as tissue engineering scaffolds, drug delivery systems, and wound dressings. In recent years, there has been a significant increase in the search for improved mechanical properties of hydrogels derived from natural products to extend their applications in various fields, and there are different methods to obtain strengthened hydrogels. Cationic guar gum has physicochemical properties that allow it to interact with other polymers and generate hydrogels. This study aimed to develop an ultra-stretchable and self-healing hydrogel, evaluating the influence of adding PolyOX [poly(ethylene oxide)] on the mechanical properties and the interaction with cationic guar gum for potential tissue engineering applications. We found that variations in PolyOX concentrations and pH changes influenced the mechanical properties of cationic guar gum hydrogels. After optimization experiments, we obtained a novel hydrogel, which was semi-crystalline, highly stretchable, and with an extensibility area of approximately 400 cm2, representing a 33-fold increase compared to the hydrogel before being extended. Moreover, the hydrogel presented a recovery of 96.8% after the self-healing process and a viscosity of 153,347 ± 4,662 cP. Therefore, this novel hydrogel exhibited optimal mechanical and chemical properties and could be suitable for a broad range of applications in different fields, such as tissue engineering, drug delivery, or food storage.

Increased depressive severity has been linked to neurocognitive impairment. Aerobic exercise (AE) is an efficient technique for improving cognitive function. However, studies indicating the importance of AE to neurophysiological and neuropsychological functions in the depressed elderly using event-related potentials (ERPs) are scarce. Objectives: This study aimed to identify the potential benefits of AE on neurophysiological and neuropsychological functions. Methods: A total of 30 depressed older adults (AE group: n=15; control group (CG): n=14) were recruited based on the inclusion and exclusion criteria. The AE group was subjected to an 8-week-period AE program (3 times/week for 30 min per session) at moderate intensity, determined using heart rate maximum (HRmax). The training intensity was set at 50% HRmax and increased by 5% in subsequent weeks. Pre- and post-training measures for neurophysiological function were tested using ERP-P300 (amplitude-µV and latency-ms) and also for neuropsychological functions using the trail making test (TMT), mini mental status examination (MMSE), and everyday cognition questionnaire (ECog). Results: In the experimental group, statistically significant improvements were observed when analyzed for all 3 (group-by-time interaction effect, main effect of time, and main effect of group), in both neurophysiological functions (*p<0.001) and neuropsychological functions (*p<0.001), except for ECog scores, where the results were insignificant for the main effect of a group. Correlation analysis demonstrated no association between neurophysiological and neuropsychological functions (*p>0.05). Conclusion: Findings showed that 8 weeks of AE training may be a promising approach to improve cognitive functions in depressed older adults. However, considering relatively small number of patients, the question arises for effectiveness in other populations.


O aumento da gravidade de quadros de depressão tem sido associado a comprometimento neurocognitivo. O exercício aeróbico (EA) é uma técnica eficiente para melhorar a função cognitiva. No entanto, estudos sugerindo a importância de EA para funções neurofisiológicas e neuropsicológicas usando potenciais relacionados a eventos (PRE) em idosos deprimidos são escassos. Objetivo: Identificar os potenciais benefícios do EA nas funções neurofisiológicas e neuropsicológicas. Métodos: Vinte e nove idosos deprimidos (grupo EA: n=15; grupo controle: GC: n=14) foram recrutados segundo critérios de inclusão e exclusão. O grupo EA foi submetido a um programa de EA de oito semanas (3 vezes/semana em sessões de 30 minutos cada) em intensidade moderada, determinada pela frequência cardíaca máxima (FCmáx). A intensidade do treinamento foi fixada em 50% da FCmax e aumentada em 5% nas semanas subsequentes. As medidas pré- e pós-treinamento para a função neurofisiológica foram testadas pelo ERP-P300 (amplitude-µV e latência-ms) e também para as funções neuropsicológicas usando o Trail Making Test (TMT), o Mini Mental Status Examination (MMSE) e o Everyday Cognition Questionnaire (ECog). Resultados: No grupo experimental, melhorias estatisticamente significativas foram observadas quando analisadas para todos os 3 (efeito de interação grupo por tempo, efeito principal do tempo e efeito principal do grupo), em ambas as funções neurofisiológicas (*p<0,001) e funções neuropsicológicas (*p<0,001), exceto para escores ECog, onde os resultados foram insignificantes para o efeito principal de um grupo. Being the first or second cause of death worldwide, cancer represents the most significant clinical, social, and financial burden of any human illness. Despite recent progresses in cancer diagnosis and management, traditional cancer chemotherapies have shown several adverse side effects and loss of potency due to increased resistance. As a result, one of the current approaches is on with the search of bioactive anticancer compounds from natural sources. Neopeltolide is a marine-derived macrolide isolated from deep-water sponges collected off Jamaica's north coast. Its mechanism of action is still under research but represents a potentially promising novel drug for cancer therapy. In this review, we first illustrate the general structural characterization of neopeltolide, the semi-synthetic derivatives, and current medical applications. In addition, we reviewed its anticancer properties, primarily based on in vitro studies, and the possible clinical trials. Finally, we summarize the recent progress in the mechanism of antitumor action of neopeltolide. According to the information presented, we identified two principal challenges in the research, i) the effective dose which acts neopeltolide as an anticancer compound, and ii) to unequivocally establish the mechanism of action by which the compound exerts its antiproliferative effect.

Introduction: Extraction of the impacted mandibular third molar is a common procedure in dentistry. Many complications may arise after this operation, the most common being pain, trismus, and swelling. Systemic medications have been used in an attempt to manage these problems, but because of their side effects, the need for non-medication treatment arises to treat these complications without side effects, such as cryotherapy, ice packs, low-level laser therapy, and ozone. Ozone is one of the most effective antimicrobials used in the dentistry field, and it also has a positive effect on soft tissue healing, activates cellular metabolism, and can react with blood components; for these reasons ozone is used to manage trismus, swelling, and pain after removal of the mandibular third molar. Aim: The purpose of the study was to assess the effects of topical ozone gel on complications from the extraction of the impacted mandibular third molar. Materials and Methods: Thirty patients were enrolled in the current study and were randomly divided into two equal groups. Preoperatively clinical examination included measurement of facial swelling measurements and maximum mouth opening. The position and configuration of the impacted lower third molar, the surrounding bone, the mandibular canal, and the neighboring tooth were all assessed using a panoramic X-ray. On days 2 and 7, after surgery, the facial swelling dimensions and maximum mouth opening were again assessed. Statistics were used to analyze results. Results: Findings indicate statistical significance for pain, but not for swelling or mouth opening. Conclusions: After lower third molar surgery, topical ozone gel helps reduce postoperative pain.


Introducción: La extracción del tercer molar mandibular retenido es un procedimiento común en odontología. Pueden surgir muchas complicaciones después de esta operación, siendo las más comunes dolor, trismo y edema. Se han utilizado medicamentos sistémicos en un intento de controlar estos problemas, pero debido a sus efectos secundarios, surge la necesidad de tratamientos sin medicamentos para tratar estas complicaciones sin efectos secundarios, como crioterapia, bolsas de hielo, terapia con láser de baja intensidad y ozono. El ozono es uno de los antimicrobianos más eficaces utilizados en el campo de la odontología, además tiene un efecto positivo en la cicatrización de los tejidos blandos, activa el metabolismo celular y puede reaccionar con los componentes sanguíneos; Por estas razones, el ozono se utiliza para controlar el trismo, la hinchazón y el dolor después de la extracción del tercer molar mandibular. Objetivo: El propósito del estudio fue evaluar los efectos del gel de ozono tópico sobre las complicaciones de la extracción del tercer molar mandibular impactado. Materiales y Métodos: Se inscribieron treinta pacientes en el estudio actual y se dividieron aleatoriamente en dos grupos iguales. El examen clínico preoperatorio incluyó la medición de la hinchazón facial y la apertura máxima de la boca. La posición y configuración del tercer molar inferior impactado, el hueso circundante, el canal mandibular y el diente vecino se evaluaron mediante una radiografía panorámica. Los días 2 y 7, después de la cirugía, se evaluaron nuevamente las dimensiones de la hinchazón facial y la apertura máxima de la boca. Se utilizaron estadísticas para analizar los resultados. Resultados: Los hallazgos indican significación estadística para el dolor, pero no para la hinchazón o la apertura de la boca.Conclusión: Después de la cirugía del tercer molar inferior, el gel de ozono tópico ayuda a reducir el dolor postoperatorio.

Performance management (PM) reforms have been introduced in health systems worldwide to improve accountability, transparency and learning. However, gaps in evidence exist regarding the ways in which PM contributes to organizational-level outcomes. Between 2015 and 2017, the government of El Salvador and the Salud Mesoamerica Initiative (SMI) introduced team-based PM interventions in the country's primary health care (PHC) system including target setting, performance measurement, provision of feedback and in-kind incentives. The programme's evaluation showed widespread improvements in performance for community outreach and service timeliness, quality and utilization. The current study characterizes how the implementation of team-based PM interventions by SMI implementers contributed to PHC system performance improvements. We used a descriptive, single-case study design informed by a programme theory (PT). Data sources included qualitative in-depth interviews and SMI programme documents. We interviewed the members of four PHC teams (n = 13), Ministry of Health (MOH) decision makers (n = 8) and SMI officials (n = 6). Coded data were summarized, and thematic analysis was employed to identify broader categories and patterns. The outcomes chain in the PT was refined based on empirical findings that revealed the convergence of two processes: (1) increased social interactions and relationships among implementers that enhanced communication and created opportunities for social learning and (2) cyclical performance monitoring that generated novel flows of information. These processes contributed to emergent outcomes including the uptake of performance information, altruistic behaviours in service delivery and organizational learning. Through time, the cyclical nature of PM appears to have led to the spread of these behaviours beyond the teams studied here, thus contributing to system-wide effects. Findings illustrate the social nature of implementation processes and describe plausible pathways through which lower-order implementation programme effects can contribute to higher-order changes in system performance.

INTRODUCTION: Antimicrobial resistance developed through the inadequate use of antibiotics; is an overriding task for global public health. OBJECTIVE: To explore awareness, knowledge, and practices, and compare the elements associated with antibiotic misuse in different University students and uneducated people of Khyber Pakhtunkhwa Province, Pakistan. METHODS: Cross-sectional study was conducted from July to December 2020 using a validated questionnaire. Data were collected from eleven different university students and uneducated people of Khyber Pakhtunkhwa, Pakistan. RESULTS: 3,600 questionnaires were completed, consisting of 56.9% Male and 43.0% Female. 1,999 (55.5%) of the antibiotic users reported through the survey used non-prescription antibiotics within a one-month study period. Out of the participants, 230 (6.3%) were uneducated or their education level was below matric rest were university students. 1999 (55.5%) reported buying Antibiotics with Medical Prescription. Most self-medicated participants (56.9%) stop taking antibiotics when they feel better. More than 90% of the respondents answered that doctors and pharmacist staff do not guide them well that how to use antibiotics. 2,171 (60.03%) respondents mistakenly believed that antibiotics improve restoration from coughs and colds. Only 720 (20%) respondents knew that antibiotics also disturb normal flora and 547 participants (15.9%) agree that unnecessary use of antibiotics causes bacterial resistance. CONCLUSION: Finding from this study may have important implications for public health policy in Khyber Pakhtunkhwa, Pakistan given the growing global resistance to antibiotics and the reported health issues related to their improper use.

BACKGROUND: Cartilaginous disorders comprise a wide range of diseases that affect normal joint movement, ear and nose shape; and they have great social and economic impact. Mesenchymal stem cells (MSCs) provide a promising regeneration alternative for treatment of degenerative cartilaginous disorders. This study aimed to compare therapeutic potential of different types of laser activated MSCs to promote auricular cartilage regeneration. Twelve adult rabbit allocated equally in four groups, all animals received a surgical mid auricular cartilage defect in one ear; Group I (Positive control) injected sub-perichondrially with phosphate-buffered saline (PBS), Group II (ADMSC-transplanted group) injected adipose-derived MSCs (ADMSCs), Group III (BMMSCs-transplanted group) received bone marrow-derived MSCs (BMMSCs), and Group IV (EMSC-transplanted group) received ear MSCs (EMSCs) in the defected ear. The auricular defect was analyzed morphologically, histopathologically and immunohistochemically after 4 weeks. In addition, a quantitative real-time polymerase chain reaction was used to examine expression of the collagen type II (Col II) and aggrecan as cartilage growth factors. RESULTS: The auricles of all treatments appeared completely healed with smooth surfaces and similar tissue color. Histopathologically, defective areas of control positive group, ADMSCs and EMSCs treated groups experienced a small area of immature cartilage. While BMMSCs treated group exhibited typical features of new cartilage formation with mature chondrocytes inside their lacunae and dense extracellular matrix (ECM). In addition, BMMSC treated group showed a positive reaction to Masson's trichrome and orcein stains. In contrary, control positive, ADMSC and EMSC groups revealed faint staining with Masson's trichrome and Orcein. Immunohistochemically, there was an intense positive S100 expression in BMMSCs (with a significant increase of area percentage + 21.89 (P < 0.05), a moderate reaction in EMSCs (with an area percentage + 17.97, and a mild reaction in the control group and ADMSCs (area percentages + 8.02 and + 11.37, respectively). The expression of relative col II and aggrecan was substantially highest in BMMSCs (± 0.91 and ± 0.89, respectively). While, Control positive, ADMSCs and EMSCs groups recorded (± 0.41: ± 0.21, ± 0.6: ± 0.44, ± 0.61: ± 0.63) respectively. CONCLUSION: BMMSCs showed the highest chondrogenic potential compared to ADMSCs and EMSCs and should be considered the first choice in treatment of cartilaginous degenerative disorders.

Garlic (Allium sativum) is an important cash food crop, and the biotechnology industry has considerable interest in the plant because of its medicinal importance. These medicinal properties are attributed to organosulphur compounds as the accumulation of these compounds varies according to genotype, locality, light quality, and cultivation practices. In this study, we compared a newly developed garlic variety NARC-G1 by National Agricultural Research Centre (NARC), Islamabad, Pakistan with three different garlic cultivars and highlighted the distinctive attributes like phenotypic characteristics, the content of allicin, elemental profile, and gene polymorphism. Phenotypic analysis showed NARC-G1 has significantly higher bulb weight (66.36g ± 18.58), single clove weight (5.87g ± 1.041), and clove width (17.41mm ± 0.95) which directly correlates to the size of the garlic. The analytical analysis showed the highest allicin content (4.82 ± 0.001) in NARC-G1. Genotyping of the alliinase in all four cultivars showed indels in the gene resulting in distinguishable changes in organosulphur compounds' profile. NARC-G1 is unique from other garlic cultivars and could be the best choice for mass production with proper cultivation and irrigation management. Moreover, for Pakistan NARC-G1 could be a potential contender to earn the industrial benefits with inland cultivation instead of importing garlic alleviating the economic burden.

OBJECTIVES: To estimate the risk of recurrence of adverse events following immunization (AEFIs) upon revaccination and to determine among patients with suspected vaccine allergy whether allergy skin test positivity was associated with AEFI recurrence. STUDY DESIGN: This prospective observational study included patients assessed in the Canadian Special Immunization Clinic Network from 2013 to 2019 with AEFIs who required revaccination with the vaccine temporally associated with their AEFI. Participants underwent standardized assessment and data collection. Special Immunization Clinic physicians used guidelines to inform their recommendations. Participants were followed up after revaccination to capture AEFI recurrences. Data were transferred to a central database for descriptive analysis. RESULTS: Overall, 588 participants were assessed for 627 AEFIs; 570 (91%) AEFIs occurred in children <18 years of age. AEFIs included immediate hypersensitivity (130/627; 21%), large local reactions (110/627; 18%), nonurticarial rash (51/627; 8%), seizures (26/627; 4%), and thrombocytopenia (11/627; 2%). Revaccination was recommended to 513 of 588 (87%) participants. Among participants recommended and due for revaccination during the study period, 63% (299/477) were revaccinated. AEFI recurrence was 10% (31/299) overall, 31% (15/49) for large local reactions, and 7% (5/66) for immediate hypersensitivity. No recurrence was serious. Among 92 participants with suspected vaccine allergy who underwent skin testing and were revaccinated, the negative predictive value of skin testing for AEFI recurrence was 96% (95% CI 92.5%-99.5%). CONCLUSIONS: Most individuals with AEFIs were safely revaccinated. Among those with suspected vaccine allergy, skin testing may help determine the safety of revaccination.

Ultraviolet radiation (UV) is causally linked to cutaneous melanoma, yet the underlying epigenetic mechanisms, known as molecular sensors of exposure, have not been characterized in clinical biospecimens. Here, we integrate clinical, epigenome (DNA methylome), genome and transcriptome profiling of 112 cutaneous melanoma from two multi-ethnic cohorts. We identify UV-related alterations in regulatory regions and immunological pathways, with multi-OMICs cancer driver potential affecting patient survival. TAPBP, the top gene, is critically involved in immune function and encompasses several UV-altered methylation sites that were validated by targeted sequencing, providing cost-effective opportunities for clinical application. The DNA methylome also reveals non UV-related aberrations underlying pathological differences between the cutaneous and 17 acral melanomas. Unsupervised epigenomic mapping demonstrated that non UV-mutant cutaneous melanoma more closely resembles acral rather than UV-exposed cutaneous melanoma, with the latter showing better patient prognosis than the other two forms. These gene-environment interactions reveal translationally impactful mechanisms in melanomagenesis.

Objective: To evaluate the interaction of the dental anxiety precedents at the first dental examination and the AB secretor status of blood groups (A, B, AB, and O) on dental caries experience and severity in a group of school children. Material and Methods: This is a cross-sectional study included 81 healthy children aged 6-8 years from primary schools in Baghdad; with negative previous dental visit. Dental anxiety represented by physiological measures was measured by pulse oximeter. Dental caries experience and severity was recorded. Unstimulated salivary samples were taken from the children to determine the salivary secretor status for AB blood type. Results: The anxiety was decreased as age increases; boys were more anxious than girls; and more anxious children were found in non-secretor group; Dental caries was higher in non-secretor anxious children all with non-significant difference. Conclusion: There is little evidence that the AB secretor status and anxiety to have influence on dental caries experience and severity, as there are many factors may interact to propagate the caries experience and severity. Objetivo: Avaliar a interação dos precedentes da ansiedade odontológica no primeiro atendimento e o status secretor do grupo sanguíneo ABO (A, B, AB, and O) na experiência e severidade de carie dental em um grupo de crianças em fase escolar. Material e Métodos: Este estudo cruzado incluiu 81 crianças saudáveis, com 6-8 anos de idade de escola primária em Baghdad; sem atendimento odontológico prévio. A ansiedade odontológica representada por medidas fisiológicas foi avaliada pelo oxímetro de pulso. A experiência à cárie bem como a severidade foram registradas. Amostras de saliva não estimulada foram coletadas das crianças para se determinar o status secretor salivar para tipo sanguíneo AB. Resultados: A ansiedade diminuiu conforme a idade aumentava; meninos foram mais ansiosos do que as meninas; e crianças mais ansiosas foram encontradas no grupo não secretor. A cárie dental foi maior no grupo não secretor de crianças ansiosas sem diferença significante. Conclusão: Há uma pequena evidência de que o status secretor AB e ansiedade tem influência na experiência e na severidade da cárie dental, assim como há vários fatores que podem interagir para propagar a experiência de cárie e sua severidade.(AU)

Abstract Green synthesis has been introduced as an alternative to chemical synthesis due to the serious consequences. Metal nanoparticles synthesized through green approach have different pharmaceutical, medical and agricultural applications. The present study followed a green and simple route for the preparation of potentially bioactive gold nanoparticles (Au NPs). Au NPs were prepared via green synthesis approach using crude basic alkaloidal portion of the tuber of Delphinium chitralense. The green synthesized Au NPs were characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), X-ray diffraction (XRD) fourier transform infrared (FTIR), and UV-Visible spectrophotometer. Morphological analysis shows that Au NPs have cubic geometry with different sizes. UV-Vis spectroscopic analysis confirmed the synthesis of Au NPs while XRD proved their pure crystalline phase. The Au NPs showed promising dose dependent inhibition of both AChE and BChE as compared to the crude as well as standard drug.


Resumo A síntese verde foi introduzida como uma alternativa à síntese química devido às graves consequências. As nanopartículas metálicas sintetizadas através da abordagem verde têm diferentes aplicações farmacêuticas, médicas e agrícolas. O presente estudo seguiu uma rota verde e simples para a preparação de nanopartículas de ouro potencialmente bioativas (Au NPs). As NPs de Au foram preparadas via abordagem de síntese verde usando a porção alcaloide básica bruta do tubérculo de Delphinium chitralense. As NPs de Au sintetizadas verdes foram caracterizadas por microscopia eletrônica de transmissão (TEM), microscopia eletrônica de varredura (MEV), difração de raios X (DRX), infravermelho com transformada de Fourier (FTIR) e espectrofotômetro UV-Visível. A análise morfológica mostra que as NPs de Au possuem geometria cúbica com tamanhos diferentes. A análise espectroscópica UV-Vis confirmou a síntese de Au NPs enquanto a XRD provou sua fase cristalina pura. O Au NPs mostrou inibição dependente da dose promissora de AChE e BChE em comparação com a droga bruta e padrão.

Abstract Diabetic mellitus is an emerging disease in Saudi Arabia. In this regard, a cross-sectional retrospective study was conducted to evaluate drug utilization pattern and the cost associated with non-insulin-dependent diabetes mellitus disease management in Saudi Arabia. Data retrieved from the electronic pharmacy records during the last one year were employed in this study. World Health Organization (WHO) Defined Daily Dose (DDD) method was employed to compute the daily price of each oral hypoglycaemic agent. The American Diabetes Association (ADA) guidelines and protocols were used to evaluate the level of adherence. A total of 17057 patients were enrolled in the study. Out of the 17057 patients enrolled in the study, 60.06 % (10246) were males and the rest females. In monotherapy, biguanides (metformin) were the most recommended and utilised drugs among 5673 patients (33.25%). The most commonly used drug combination was found to be sitagliptin+metformin (1754 units). The cost per unit dose was highest for liraglutide (A10BJ02) 258.32SR (68.79USD), and lowest for metformin (A10BA02) 0.49SR (0.13 USD). Metformin was the choice drug for the diabetes patients; biguanides (metformin) and DPP-4 (sitagliptins) were the most familiar established dose combination employed. Generic drugs should be used in order to reduce overall cost.
