RESUMEN

We present scanning near-field extinction spectra of single molecules embedded in a solid matrix. By varying the tip-molecule separation, we modify the line shape of the spectra, demonstrating the coherent nature of the interaction between the incident laser light and the excited state of the molecule. We compare the measured data with the outcome of numerical calculations and find a very good agreement.

RESUMEN

We report on fluorescence fluctuations of nanoparticles diffusing through a laser focus. Subject to an intensity threshold the fluorescence signal is transformed into time traces of on and off periods. The distribution functions of the experimental on and off times follow power laws t -alpha over several orders of magnitude with exponents alpha approximately 1.5-2. At long times the distribution functions cross over to exponential decays. For the interpretation of the experimental data a diffusion-reaction equation is proposed which covers both, the diffusion controlled recurrence and the photon statistics as the relevant processes.

RESUMEN

The fluorescence of individual pairs of perylenemonoimide chromophores coupled via a short rigid linker is investigated. Photon antibunching is reported, indicating collective effects in the fluorescence, which are further substantiated by the observation of collective triplet off times and triplet lifetime shortening. The experimental findings are analyzed in terms of singlet-singlet and singlet-triplet annihilation based on Förster type energy transfer. The results reported here demonstrate that the statistical properties of the emission light of isolated single quantum systems can serve as a hallmark of intermolecular interactions.

RESUMEN

F0F1 ATP synthases are the smallest rotary motors in nature and work as ATP factories in bacteria, plants and animals. Here we report on the first observation of intersubunit rotation in fully coupled single F0F1 molecules during ATP synthesis or hydrolysis. We investigate the Na+-translocating ATP synthase of Propionigenium modestum specifically labeled by a single fluorophore at one c subunit using polarization-resolved confocal microscopy. Rotation during ATP synthesis was observed with the immobilized enzyme reconstituted into proteoliposomes after applying a diffusion potential, but not with a Na+ concentration gradient alone. During ATP hydrolysis, stepwise rotation of the labeled c subunit was found in the presence of 2 mM NaCl, but not without the addition of Na+ ions. Moreover, upon the incubation with the F0-specific inhibitor dicyclohexylcarbodiimide the rotation was severely inhibited.
