RESUMEN

Alternative therapies associating natural products and nanobiotechnology show new perspectives on controlled drug release. In this context, nanoemulsions (NEs) present promising results for their structural design and properties. Hesperetin (HT), a flavonoid mainly found in citrus fruits, presents highlighted bone benefits. In this context, we developed a hesperetin-loaded nanoemulsion (HT-NE) by sonication method and characterized it by dynamic light scattering, analyzing its encapsulation efficiency, and cumulative release. The biocompatibility in human osteoblasts Saos-2-like was evaluated by the cytotoxicity assay and IC50. Then, the effects of the HT-NE on osteogenesis were evaluated by the cellular proliferation, calcium nodule formation, bone regulators gene expression, collagen quantification, and alkaline phosphatase activity. The results showed that the formulation presented ideal values of droplet size, polydispersity index, and zeta potential, and the encapsulation efficiency was 74.07 ± 5.33%, showing a gradual and controlled release. Finally, HT-NE was shown to be biocompatible and increased cellular proliferation, and calcium nodule formation, regulated the expression of Runx2, ALPL, and TGF-ß genes, and increased the collagen formation and alkaline phosphatase activity. Therefore, the formulation of this NE encapsulated the HT appropriately, allowing the increasing of its effects on mechanisms to improve or accelerate the osteogenesis process.

This study assessed the mechanical properties of the Only One File Blue and the Reciproc Blue instruments. A total of 80 new 25 mm reciprocating NiTi instruments (25/.08v) were evaluated for their mechanical performance (n = 40 per group). Cyclic fatigue resistance, torsional fatigue, flexural resistance and buckling resistance tests were conducted. Statistical analysis was employed with a significance level set at 5%. The results indicated no statistically significant differences in resistance to cyclic fatigue neither in the fragment length between the instruments (p > 0.05). However, the Reciproc Blue instrument exhibited greater torque to fracture and a larger rotation angle than the Only One File Blue instrument (p < 0.05). The Only One File Blue instrument demonstrated higher flexibility but lower resistance to buckling compared to the Reciproc Blue instrument (p < 0.05). In conclusion, the tested instruments exhibit differences in mechanical properties, with the Reciproc Blue instrument generally presenting greater advantages than the Only One File Blue.

AIM: To determine the cytotoxicity mechanism of 2.5% calcium hypochlorite [Ca(OCl)2 ] in L929 fibroblasts and the effect of this solution on human osteoblast-like cells (Saos-2) mineralization, compared to that of 2.5% sodium hypochlorite (NaOCl). METHODOLOGY: L929 fibroblasts were exposed to Ca(OCl)2 and NaOCl at different dilutions for 10 min. Cell metabolism was assessed by methyl-thiazole-tetrazolium (MTT); lysosome integrity, by neutral red (NR) assay; type of cell death, by flow cytometry (apoptosis/necrosis); cytoskeleton, by actin and α-tubulin fluorescence and cell ultrastructure, by transmission electron microscopy (TEM). The alkaline phosphatase (ALP) activity and mineralized nodule formation were determined in Saos-2 by thymolphthalein release and alizarin red staining (ARS), respectively. The data were analysed by two-way anova and Bonferroni's post-test (α = .05). RESULTS: Ca(OCl)2 promoted higher cell viability and a lower percentage of apoptosis and necrosis than NaOCl (p < .05). Ca(OCl)2 and NaOCl decreased cell metabolism and lysosome integrity, induced the breakdown of microtubules and actin filaments, promoted alterations of rough endoplasmic reticulum and disruption of mitochondrial cristae. Additionally, Ca(OCl)2 did not induce ALP activity and had no effect on mineralized nodules formation. CONCLUSIONS: Although Ca(OCl)2 and NaOCl promoted the same cytotoxicity mechanism, Ca(OCl)2 was less cytotoxic than NaOCl. As for ALP activity, no differences were observed between NaOCl and Ca(OCl)2 . The production of mineralized nodules induced by Ca(OCl)2 was lower than those induced by NaOCl, but was not different from those induced by the control group.

Abstract Resin composites containing surface pre-reacted glass (S-PRG) have been introduced to reduce demineralization and improve remineralization of the tooth structure. However, water diffusion within the material is necessary for its action, which can impair its overall physicomechanical properties over time, including color stability. This study aimed to evaluate the color stability and related degree of conversion (DC) of four resin composites. Discs (6 x 4 mm, n = 5/group) of microhybrid (MH), nanofilled (NF), nanohybrid (NH), and S-PRG-based nanohybrid (S-PRG-NH) composites with two opacities (A2/A2E and A2O/A2D) were prepared. Color (CIELab and CIEDE2000) was evaluated with a spectrophotometer after aging in grape juice (2 x 10 min/10mL/7days). The DC was analyzed by using Fourier transform infrared spectroscopy before and after light-curing. Data were statistically analyzed by using two-way analysis of variance and post-hoc least significant difference tests (p<0.05). In the color stability analysis, the interaction between filler type and opacity was significant (CIELab, p = 0.0015; CIEDE2000, p = 0.0026). NH presented the highest color stability, which did not differ from that of MH. The greatest color alteration was observed for S-PRG-NH. S-PRG fillers also influenced DC (p < 0.05). The nanohybrid resin composite presented favorable overall performance, which is likely related to its more stable organic content. Notwithstanding the benefits of using S-PRG-based nanohybrid resins, mostly in aesthetic procedures, professionals should consider the susceptibility of such resins to color alteration, probably due to the water-based bioactive mechanism of action.

Purpose: To evaluate the inductive capacity of F18 bioglass putty on the induced membrane technique in a segmental bone defect of the rabbit's radius. Methods: Ten female Norfolk at 24 months of age were used. The animals were randomly separated based on postoperative time points: five rabbits at 21 and four at 42 days. A 1-cm segmental bone defect was created in both radii. The bone defects were filled with an F18 bioglass putty. Results: Immediate postoperative radiographic examination revealed the biomaterial occupying the segmental bone defect as a well-defined radiopaque structure with a density close to bone tissue. At 21 and 42 days after surgery, a reduction in radiopacity and volume of the biomaterial was observed, with particle dispersion in the bone defect region. Histologically, the induced membrane was verified in all animals, predominantly composed of fibrocollagenous tissue. In addition, chondroid and osteoid matrices undergoing regeneration, a densely vascularized tissue, and a foreign body type reaction composed of macrophages and multinucleated giant cells were seen. Conclusions: the F18 bioglass putty caused a foreign body-type inflammatory response with the development of an induced membrane without expansion capacity to perform the second stage of the Masquelet technique.

Objective: This study aimed to evaluate stem cell from human deciduous teeth (SHED) viability after exposure to different bioceramic materials. Material and Methods: Discs were constructed to obtain the material extracts according to the following groups: G1 - Bio-C Repair, G2 - MTA Repair HP, G3 - TheraCal LC, and G4 ­ Biodentine. Positive and negative control group were respectively maintained with αMEM + 10% FBS and αMEM + 1% FBS. SHED obtained through primary culture were in contact with material extracts for 24, 48, and 72h. MTT assay evaluated cell viability. Groups were plated in triplicate and the cell viability assay were repeated three times. Data were analyzed by two-way ANOVA followed by Tukey test (p<0.05). Results: The treatment and period comparisons showed statistically significant differences (p<0.000). G2 (MTA Repair HP) had greater cell viability values than the other experimental groups and negative control. MTA Repair HP and the control groups exhibited a similar behavior with cell viability values decreasing from 24h to 48h and increasing from 48h to 72h. Bio-C Repair, Biodentine, and Theracal LC did not show statistically significant differences among periods. Conclusions: SHED increased viability values after contact with MTA Repair HP in comparison with other bioceramic materials.(AU)


Objetivo: O objetivo desse estudo foi avaliar a viabilidade de células-tronco de dentes decíduos humanos (SHED) após o contato com diferentes materiais biocerâmicos. Material e Métodos: Foram confeccionados discos para obtenção dos extratos dos materiais de acordo com os seguintes grupos: G1 - Bio-C Repair, G2 - MTA Repair HP, G3 - TheraCal LC e G4 - Biodentine. Grupo de controle positivo e negativo foram mantidos respectivamente com αMEM + 10% FBS e αMEM + 1% FBS. SHED obtidas por cultura primária entraram em contato com os extratos de materiais por 24, 48 e 72h. O ensaio MTT avaliou a viabilidade celular. Os grupos foram semeados em triplicata e o ensaio de viabilidade celular foi repetido três vezes. Os dados foram analisados por ANOVA a dois critérios seguido pelo teste de Tukey (p<0,05). Resultados: As comparações de tratamentos e períodos mostraram diferenças estatisticamente significativas (p<0,000). O G2 (MTA Repair HP) apresentou maiores valores de viabilidade celular que os demais grupos experimentais e controle negativo. O MTA Repair HP e os grupos controle exibiram um comportamento semelhante com os valores de viabilidade celular diminuindo de 24h para 48h e aumentando de 48h para 72h. Bio-C Repair, Biodentine e Theracal LC não apresentaram diferenças estatisticamente significativas entre os períodos. Conclusões: SHED aumentou os valores de viabilidade após o contato com o MTA Repair HP em comparação com outros materiais biocerâmicos (AU)

During the manufacture of ceramic restorations there is an important step of finishing and polishing and the effects of different types of these procedures on the surface characteristics of ceramics are not known for sure. AIM: To evaluate the effects of various surface treatments and immersion in coloring substances on the roughness, microhardness, and color stability of CAD-CAM monolithic ceramics. MATERIALS AND METHOD: The ceramics used were lithium disilicate reinforced with zirconium dioxide (Suprinity), lithium disilicate (E.max) or leucite (Empress). They were subjected to two surface treatments: glazing (group G) (n=20) or mechanical polishing (group P) (n=20). Then they were divided into two subgroups (n=10) to be treated with the staining substance (coffee or water). Roughness, microhardness and color were measured before and after treatment. Data were subjected to analysis of variance and multiple comparisons were performed with Tukey tests at 5% significance level. RESULTS: Roughness was lower in all tested ceramics after polishing than after glazing. Microhardness was the same for polished and glazed E.max, higher in glazed than polished Empress, and higher in polished than glazed Suprinity. Analysis of the effects of glazing and polishing on the individual ceramics showed that the ΔE2000 and ΔWID data of the E.max ceramic subjected to polishing showed greater change. Mechanical polishing is a good option for surface treatment of monolithic ceramics. CONCLUSION: Glazing was inferior and less satisfactory than polishing. Glazing generates changes that can lead to color instability.


Durante a confecção de restaurações cerâmicas existe uma importante etapa dos procedimentos de acabamento e polimento. Os efeitos de diferentes tipos desses procedimentos nas características superficiais das cerâmicas não são conhecidos com certeza. Objetivo: Avaliar os efeitos de vários tratamentos de superfície e imersão em substâncias corantes na rugosidade, microdureza e estabilidade de cor de cerâmicas monolíticas CAD-CAM. Materiais e Métodos: As cerâmicas utilizadas foram dissilicato de lítio reforçado com dióxido de zircônio (Suprinity), dissilicato de lítio (E.max) ou leucita (Empress). Foram submetidos a dois tratamentos de superfície: glazeamento (grupo G) (n=20) ou polimento mecânico (grupo P) (n=20). Em seguida, foram divididos em dois subgrupos (n=10) para serem tratados com a substância corante (café ou água). Rugosidade, microdureza e cor foram medidas antes e após o tratamento. Os dados foram submetidos à análise de variância e as comparações múltiplas foram realizadas com testes de Tukey ao nível de 5% de significância. Resultados: A rugosidade foi menor em todas as cerâmicas testadas após o polimento do que após o glazeamento. A microdureza foi a mesma para o E.max polido e vidrado, maior no Empress vidrado do que no polido, e maior no Suprinity polido do que no vidrado. A análise dos efeitos do esmaltação e polimento nas cerâmicas individuais mostrou que os dados ΔE2000 e ΔWID da cerâmica E.max submetida ao polimento apresentaram maior alteração. O polimento mecânico é uma boa opção para o tratamento superficial de cerâmicas monolíticas. Conclusão: A aplicação do glazing foi inferior e menos satisfatório que o polimento, gerando alterações que podem levar à instabilidade da cor.

BACKGROUND: Polymethylmethacrylate (PMMA) is the most used material for the manufacturing of eye prostheses. OBJECTIVES: To investigate the cytotoxicity of different cleaning agents for ocular prostheses on human conjunctival cells. MATERIAL AND METHODS: Six groups of specimens were created (saline, soap, 4% chlorhexidine, hydrogen peroxide, 1% triclosan, and citronella oil). Three specimens were made for each disinfectant at each disinfection period (1, 7, 15, 30, 60, and 90 days), totaling 108 specimens. Thus, the specimens were disinfected, with different disinfectants, for different periods of time. After each disinfection process, the specimens were washed with sterile distilled water. A human conjunctival cell line was grown on the acrylic resin specimens and then cytotoxicity tests (MTT and Neutral Red (NR)) were performed. A negative control (untreated cell cultures) and positive control (Tween 20) were created. Two-way analysis of variance (ANOVA) and Bonferroni test were performed (p < 0.05). RESULTS: For the MTT and NR tests, when there was a significant difference between the disinfectant and negative control, the disinfectant generated a significant reduction in cell proliferation most of the time. CONCLUSIONS: All reductions in cell proliferation caused by the disinfectants were clinically acceptable. All disinfectants tested in this study were found to be non-cytotoxic to human conjunctival cells.

Purpose: To evaluate the in vivo efficiency of commercial polymeric membranes for guided bone regeneration. Methods: Rat calvarial critical size defects was treated with LuminaCoat (LC), Surgitime PTFE (SP), GenDerm (GD), Pratix (PR), Techgraft (TG) or control (C-) and histomorphometric analysis determined the percentage of new bone, connective tissue and biomaterial at 1 or 3 months. Statistical analysis used ANOVA with Tukey's post-test for means at same experimental time and the paired Student's t test between the two periods, considering p < 0.05. Results: New bone at 1 month was higher for SP, TG and C-, at 3 months there were no differences, and between 1 and 3 months PR had greater increase growthing. Connective tissue at 1 month was higher for C-, at 3 months for PR, TG and C-, and between 1 and 3 months C- had sharp decline. Biomaterial at 1 month was higher for LC, in 3 months for SP and TG, and between 1 and 3 months, LC, GD and TG had more decreasing mean. Conclusion: SP had greater osteopromotive capacity and limitation of connective ingrowth, but did not exhibit degradation. PR and TG had favorable osteopromotion, LC less connective tissue and GD more accelerated biodegradation.

Abstract The aim of this study was to quantify the force exerted by tandem archwires in a specific system of passive self-ligating bracket. Forty-eight thermo-activated nickel-titanium orthodontic archwires were separated into four groups (n = 12): G1 - two .014" + .014" round archwires; G2 - two .014" + .016" round archwires; G3 - .014" x .025" rectangular archwire; and. G4 - .016" x .022" rectangular archwire. Brackets were fixed onto teeth 1.5 to 2.5 using a device that represented the upper teeth, maintaining an interbracket distance of 6.0 mm. The deflection tests were performed using the structure representative of tooth 1.1 as support on the Instron testing machine at a speed of 2.0 mm/min. The archwires were evaluated at deflections of 0.5 mm, 1.0 mm, and 1.5 mm. The data were analyzed by a generalized linear model, considering values at different deflections as repeated measurements in the same experimental unit (α = 0.5%). At 0.5 mm, higher forces were observed in G2 and G3, which did not differ significantly (p > 0.05). The lowest force was observed in G4 (p < 0.05). At 1.0 mm and 1.5 mm, the highest force was observed in G3, followed by G4 and G2 (p < 0.05). The lowest force was observed in G1 (p < 0.05). In general, tandem archwires (same or different calibers) in a specific passive self-ligating bracket exerted lower force when compared with rectangular archwires.

Resumo Introdução Após o advento dos termoplásticos de baixa temperatura, sua prevalência na confecção de órteses para membros superiores é identificada por vários autores. A compreensão de suas propriedades, por parte do terapeuta ocupacional e de outros profissionais que atuam na área, é importante no processo de seleção do termoplástico, que segue uma lógica para combinar as características do material à função desejável para cada órtese. Objetivos Sistematizar as características dos termoplásticos de baixa temperatura e, a partir disso, elaborar um instrumento para realizar testes práticos empíricos com os materiais a fim de estabelecer critérios para o seu manuseio e avaliação. Método Pesquisa de caráter exploratório, constituindo a criação de procedimentos de teste e, consequentemente, a elaboração de um instrumento de avaliação de abordagem qualitativa e que valoriza a experiência prática do profissional, que manipula o material e avalia cada requisito. Resultados O instrumento criado contempla 14 caraterísticas do material, acompanhadas de uma definição, um procedimento com recomendações para o teste prático e um campo destinado ao preenchimento das alternativas de resposta. Conclusão O instrumento considera características importantes a serem verificadas durante a avaliação dos materiais e poderá direcionar o olhar e os registros do profissional, auxiliando o nas decisões clínicas. Isso será importante para melhorar a qualidade das órteses e de outros dispositivos de Tecnologia Assistiva confeccionados com esse grupo de materiais. Além disso, a sistematização da avaliação prática dos materiais termoplásticos poderá auxiliar no desenvolvimento de estudos da área da saúde e de pesquisas envolvendo materiais para órteses.


Abstract Introduction After the advent of low-temperature thermoplastics, their prevalence in the manufacture of orthoses for upper limbs has been identified by several authors. The understanding of their properties by occupational therapists and other professionals working in this field is important in the process of selecting the thermoplastic, which follows a logic to match the material's characteristics to the desired function for each orthosis. Objectives Systematize the characteristics of low-temperature thermoplastics and, from that, develop an instrument for carrying out practical empirical tests with the materials, to establish criteria for their handling and evaluation. Method Exploratory study consisting of the creation of testing procedures and, consequently, the development of a qualitative assessment instrument that values ​​the practical experience of the professional who handles the material and evaluates each requirement. Results The created instrument includes 14 material characteristics, accompanied by a definition, a procedure with recommendations for the practical testing, and a field for filling in response alternatives. Conclusions The instrument considers important characteristics to be verified during the evaluation of materials and can direct the professional's observations and records, aiding in clinical decision-making. This will be important to improve the quality of orthoses and other assistive technology devices made with these thermoplastics. In addition, the systematization of the practical evaluation of thermoplastic materials can assist in the development of health studies and research involving materials for orthoses.

Aim: This study assessed the color and translucency stability of a polymer infiltrated ceramic network (PICN) and compared it with a resin composite (RC) and a feldspathic ceramic (FEL). Methods: Disc-shaped samples of a PICN (Vita Enamic), a feldspathic ceramic (Vitablocks Mark II), and a resin composite (Brava block) were prepared from CAD/CAM blocks. PICN and RC surfaces were finished with a sequence of polishing discs and diamond paste. FEL samples received a glaze layer. The samples were subjected to 30-min immersions in red wine twice a day for 30 days. CIEL*a*b* coordinates were assessed with a spectrophotometer at baseline and after 15 and 30 days of immersion. Color alteration (ΔE00) and translucency parameter (TP00) were calculated with CIEDE2000. Average roughness was measured before the staining procedures. Color difference and translucency data were analyzed with repeated-measures ANOVA and Tukey's tests. Roughness was analyzed with the Kruskal-Wallis test. Results: Roughness was similar among the experimental groups. All materials had their color alteration significantly increased from 15 to 30 days of staining. PICN reached an intermediate ΔE00 between FEL and RC at 15 days. PICN revealed a color alteration as high as the composite after 30 days. No statistical difference was observed regarding translucency. Conclusion: PICN was not as color stable as the feldspathic ceramic at the end of the study. Its color alteration was comparable to the resin composite when exposed to red wine. However, the translucency of the tested materials was stable throughout the 30-day staining

Objetivo: Determinar el efecto de la intensidad de dos unidades de fotopolimerización sobre la biocompatibilidad, resistencia flexural y módulo elástico de una resina compuesta. Metodología: Se crearon dos grupos de resina compuesta Filtek Z250XT cada uno fotopolimerizado con intensidades diferentes (800 mW/cm2 por 20s). La viabilidad celular fue analizada mediante ensayo de MTT a las 24 y 48 horas siguiendo la normativa ISO 10993-5. La resistencia flexural y módulo elástico fueron analizadas siguiendo la normativa ISO 4049. Resultados: En el grupo fotopolimerizado con una intensidad <400 mW/cm2, la citotoxicidad fue estadísticamente mayor tanto a las 24 como a las 48 horas y la resistencia flexural y módulo elástico fueron estadísticamente menores Conclusión: Una intensidad de polimerización <400 mW/cm2, aumenta los niveles de citotoxicidad y disminuye las propiedades mecánicas de las resinas compuestas. Se destaca la importancia del control periódico de las unidades de fotopolimerización.

Metodologia: Foram fabricados dois grupos de resina composta Filtek Z250XT, cada um deles foi fotopolimerizado com intensidades diferentes ( 800 mW/cm2 por 20s). A viabilidade celular foi analisada por ensaio de MTT em 24 e 48 horas seguindo a norma ISO 10993-5. A resistência à flexão e o módulo de elasticidade foram analisados ​​seguindo a norma ISO 4049. Resultados: No grupo fotopolimerizado com intensidade <400mW/cm2, a citotoxicidade foi estatisticamente maior nas 24 e 48 horas e a resistência à flexão e o módulo de elasticidade foram estatisticamente menores. Conclusão: Uma intensidade de polimerização <400 mW/cm2 aumenta os níveis de citotoxicidade e diminui as propriedades mecânicas das resinas compostas. Destaca-se a importância do controle periódico das unidades de fotopolimerização.


Objective: To determine the effect of the intensity of two light curing units on the biocompatibility, flexural strength and elastic modulus of a composite resin. Methodology: Two groups of Filtek Z250XT (3M ESPE) composite resin were created, each one photopolymerized using different intensities ( 800 mW/cm2 for 20s). Cell viability was analyzed by MTT assay at 24 and 48 hours following the ISO 10993-5 standard. The flexural strength and elastic modulus were analyzed following the ISO 4049 standard. Results: In the group photopolymerized with an intensity <400 mW/cm2, cytotoxicity was statistically higher both at 24 and 48 hours and flexural strength and elastic modulus were statistically lower. Conclusion: A polymerization intensity <400 mW/cm2 increases the levels of cytotoxicity and decreases the mechanical properties of composite resins. The importance of the periodic control of the light curing units is emphasized.

Aim: This study was fulfilled to evaluate the flexural strength, micro-hardness, and release of two fluoride ions of bioactive restorative materials (Cention N and Activa Bioactive), a resin modified glass ionomer (Fuji II LC), and a resin composite (Filtek z250). Methods: Forty samples from four restorative materials (Activa Bioactive, Fuji II LC, Cention N, and Filtek Z250) were provided according to the current standards of ISO 4049/2000 guide lines. Subsequently, the samples were stored for 24 hours and 6 months in artificial saliva, and successively, flexural strength and micro-hardness of the samples were measured. For each studied groups the pH was decreased from 6.8 to 4 in storage solution. The rate of changes in fluoride ion release was measured after three different storage periods of 24 hours, 48 hours, and 6 months in distilled water, according to the previous studies' method. Two-way ANOVA, One-way ANOVA, Tukey HSD Pair wise comparisons, and independent t-tests were used to analyze data (α= 0.05). Results: The highest flexural strength and surface micro-hardness after 24 hours and also after 6 month were observed for Cention N(p<0.001).Flexural strength of all samples stored for 6 months was significantly lower than the samples stored for 24 hours(p<0.001). The accumulative amount of the released fluoride ion in RMGI, after six-month storage period in distilled water was considerably higher (p<0.001) than 24 hours and 48 hours storage. The amount of fluoride ion release with increasing acidity of the environment (from pH 6.8 to 4) in Fuji II LC glass ionomer was higher than the bioactive materials (p<0.05). Conclusion: The flexural strength of RMGI was increased after storage against the Activa Bioactive,Cention N and Z250 composite. Storage of restorative materials in artificial saliva leads to a significant reduction in micro hardness. The behavior and amount of released fluoride ions in these restorative materials, which are stored in an acidic environment, were dependent on the type of restorative material

Introduction: This study aimed to prepare a new root repair material including Portland cement, bismuth oxide, and nano-hydroxyapatite and analyze its physicochemical properties and its effects on the proliferation and differentiation of human dental pulp stem cells (hDPSCs). Material and Methods: Bismuth oxide as a radiopaque component and nano-hydroxyapatite particles were added to white Portland cement at 20% and 5% weight ratio, respectively. Characterization of the prepared cement was done using conventional methods. To examine the bioactivity of this new material, atomic absorption spectroscopy (AAS) was used for the investigation of the rate of calcium ions dissolution in simulated body fluid media. The viability of hDPSCs was assessed by an MTT assay after 1, 3 and 7 days. The odontogenic potential of this substance was evaluated by measuring alkaline phosphatase activity and alizarin red S staining. Results: Based on the bioactivity results, the cement presented high bio-activity, corroborating sufficiently with the calcium release patterns. The cell viability was significantly increased in new root repair material containing hydroxyapatite nanoparticles after 3 and 7 days (p<0.05). Conclusion: Moreover, alkaline phosphatase activity increased over 7 days in all experimental groups. The new cement containing nano-hydroxyapatite particles could be a good root repair material.


Objetivo: Este estudio tuvo como objetivo preparar un nuevo material de reparación de raíces que incluye cemento Portland, óxido de bismuto y nano-hidroxiapatita y analizar sus propiedades fisicoquímicas y sus efectos sobre la proliferación y diferenciación de células madre de pulpa dental humana. Material y Métodos: El óxido de bismuto como compo-nente radiopaco y las partículas de nano-hidroxiapatita se agregaron al cemento Portland blanco en una proporción en peso del 20 % y el 5 %, respectivamente. La caracterización del cemento preparado se realizó utilizando métodos con-vencionales. Para examinar la bioactividad de este nuevo material, se utilizó la espectroscopia de absorción atómica para investigar la velocidad de disolución de los iones de calcio en medio fluido corporal simulado. La viabilidad de las células madre de pulpa dental humana se evaluó mediante un ensayo MTT después de 1, 3 y 7 días. El potencial odontogénico de esta sustancia se evaluó midiendo la actividad de la fosfatasa alcalina y la tinción con rojo de alizarina S.Resultados: Con base en los resultados de bioactividad, el cemento presentó alta bioactividad, corroborando suficientemente con los patrones de liberación de calcio. La viabilidad celular aumentó significativamente en el nuevo material de reparación de raíces que contenía nanopartículas de hidroxiapatita después de 3 y 7 días (p<0,05). Conclusión: Además, la actividad de la fosfatasa alcalina aumentó durante 7 días en todos los grupos experimentales. El nuevo cemento que contiene partículas de nanohidroxiapatita podría ser un buen material de reparación radicular.

To evaluate the fracture strength of extended Class I restorations with different restorative techniques using nanofilled and nanohybrid composites. Sixty extracted human third molars were prepared with extended Class I cavities and divided into six groups: groups FS-F (Filtek bulk-fill Flow + Filtek Supreme Ultra, 3 M) and GR-F (X-tra base + GrandioSO, VOCO), restored with a flowable bulk-fill composite as a base covered by a nanofilled or nanohybrid composite; groups FB (Filtek One Bulk-Fill, 3 M) and AF (Admira Fusion X-tra, VOCO), restored with a bulk-fill resin composite; and groups FS (Filtek Supreme Ultra, 3 M) and GR (GrandioSO, VOCO), restored incrementally with a nanofilled or nanohybrid composite. Sound extracted teeth (n = 10) were used as a control group (CTL). The specimens were axially loaded until failure. Data were analyzed using one-way ANOVA and Games Howell test (α = 0.05). Groups CTL and GR-F demonstrated significantly higher mean fracture strength when compared to FS, AF, and GR (p < 0.05). Group AF obtained more repairable fractures than the other groups. Restorations made with a nanofilled bulk-fill composite or with conventional resin composites associated with a flowable bulk-fill base were able to reestablish the fracture strength to that of sound teeth.

Purpose: Poultry by-products can contribute as an innovative natural source for the development of composites based on polymers and minerals aiming at bone regeneration. The objective of this study was the physicochemical and biological characterization of collagen-based hydrogels crosslinked with ultraviolet (UV)-riboflavin. Methods: Pure hydrogels of 100% collagen (G1) or hybrid hydrogels, 90% collagen:10% apatite (G2), 90% collagen:10% nanokeratin (G3), and 90% collagen:5% apatite:5% nanokeratin (G4) were characterized by scanning electron microscope, Fourier-transform infrared spectroscopy, differential scanning calorimetry, swelling degree and quali-quantitative histological analysis. Ectopic implantation in subcutaneous tissue in mice at one, three and nine weeks allowed to assess the inflammation (neutrophils, lymphocytes, macrophages, and giant cells) and repair (neovascularization, and connective tissue) to determine biocompatibility and the integrity of biomaterials to score their biodegradability. Histomorphometry on critical size defects in rat calvaria at one and three months evaluated the percentage of bone, connective tissue, and biomaterials in all groups. Results: The hydrogels presented porous microstructure, water absorption and physicochemical characteristics compatible with their polymeric and/or mineral composition. All materials exhibited biocompatibility, biodegradability, and low osteoconductivity. G2 showed greater density of new bone and biomaterial than the G1, G3 and G4. Conclusions: The collagen-apatite group formulation suggests potential for development as osteopromoting membrane.

Abstract Filling materials should be restricted to the root canal space. However, sometimes it is impossible to control the apical extrusion, in this case, the fate of the filling material and the result of the treatment will depend on its physicochemical properties and biocompatibility. Objective To evaluate the tissue response and bone repair capacity of endodontic sealers that were implanted in the calvaria of Wistar rats, forming the groups (n=16): AH Plus and Sealer Plus, compared to the clot group. Methodology On days 30 and 60, the animals were euthanized, the calvaria was removed and processed for hematoxylin-eosin, immunohistochemistry for collagen type I, Picrosirus red and microtomographic analysis. Data were subjected to ANOVA and Tuckey tests (p<0.05). Results At 30 days, all groups showed an intense inflammatory reaction (p>0.05). At 60 days, the AH Plus and Sealer Plus maintained an intense inflammatory infiltrate compared to the clot group (p<0.05). We observed immunopositive areas for type I collagen in all groups at 30 days and 60 days (p>0.05). We observed more red collagen fibers for the Sealer Plus compared to the clot group at 30 days (p<0.05). Considering the total fibers, the clot group at 30 days compared to 60 days after surgery showed an increase in the amount of matrix (p<0.05). There were no statistical differences between groups for green and yellow fibers (p>0.05). Regarding morphometric parameters, at 30 days, the newly formed bone volume and number of bone trabeculae were higher in the groups with sealers compared to the clot group (p<0.05). At 60 days, AH Plus and Sealer Plus showed greater bone neoformation compared to the clot group (p<0.05). Conclusions Despite AH Plus and Sealer Plus induced an intense inflammatory reaction, they can be considered biocompatible materials, since they allowed bone repair.

O presente estudo teve por objetivo avaliar a resistência à flexão de um pino experimental de fibra de vidro vazado, e a resistência adesiva entre este pino experimental à dentina radicular, associando a utilização de um dispositivo iluminador em fibra óptica, sob o processo de envelhecimento, além de determinar a distribuição de tensão pela Análise de Elementos Finitos (FEA). O pino experimental e o dispositivo iluminador de fibra óptica têm por finalidade aumentar a efetividade de fotoativação dos cimentos resinosos nos terços médios e apicais dos canais radiculares. Para o teste de flexão de 3 pontos, estritamente do pino de fibra de vidro, as amostras foram divididas em 4 grupos (n=10): pino convencional (pc), pino experimental sem preenchimento (pp), com preenchimento de cimento resinoso (ppc) e com preenchimento de fibra óptica (ppf). Para o teste de resistência adesiva Pull-out, 100 raízes de dentes bovinos foram restauradas com retentores intrarradiculares e coroas de resina composta, distribuídos em 10 grupos (n=10), tendo como variáveis: o tipo de pino de fibra de vidro [convencional (C) ou perfurado (P)], o sistema de fotoativação [com ou sem o iluminador de fibra óptica (O)], o tipo de cimento resinoso [fotoativado (F) ou polimerização dual (D)] e o envelhecimento das amostras [com e sem ciclagem mecânica (C)]. O processo de envelhecimento foi realizado através da ciclagem mecânica e as amostras foram submetidas ao teste de resistência adesiva Pull-out. A análise do modo de falha foi realizada em Estereomicroscópio. A Análise em Elementos Finitos avaliou a distribuição de tensão pela análise de von Mises, deformação total e tensão máxima principal no Software Ansys 19.3. A análise estatística foi constituída pelo teste de normalidade Shapiro-Wilk, análise de variância (ANOVA) 1, 2 e 3 fatores e teste Tukey 5%. As médias e desvios padrão de resistência à flexão foram (MPa): pc = 677 ± 81,1; ppc = 419 ± 23,2; ppf = 200 ± 32,5 e pp = 177 ± 32,8. As médias e desvios padrão de resistência adesiva foram (MPa): CD = 12,2 ± 1,21; PD = 11,1 ± 1,32; PF = 10,9 ± 1,29; POD = 13,7 ± 1,16; POF = 11,9 ± 1,48; CDC = 10,1 ± 1,33; PDC = 9,25 ± 1,11; PFC = 8,37 ± 1,25; PODC = 10,8 ± 0,95 e POFC = 9,82 ± 1,02. Os resultados do modo de falha foram: adesiva entre pino e cimento (56%), mista predominantemente adesiva entre cimento e dentina (22%), adesiva entre cimento e dentina (14%) e a mista predominantemente adesiva entre pino e cimento (8%). Concluiu-se que entre os pinos experimentais, o grupo do pino perfurado preenchido com cimento resinoso obteve o maior valor de resistência à flexão, a utilização do dispositivo iluminador aumentou os resultados de resistência adesiva nos grupos dos pinos perfurados e não houve diferença na distribuição de tensões entre os grupos. The present study aimed to evaluate the flexural strength of an experimental hollow glass fiber post, and the bond strength between this experimental post and root dentin, associating the use of an optical fiber illuminating device, under the aging process, in addition to determine the stress distribution by Finite Element Analysis (FEA). The experimental post and optical fiber illuminating device are intended to increase the polymerization effectiveness of resin cements in the middle and apical regions of root canals. For the 3-point bending test, strictly of the fiber post, the samples were divided into 4 groups (n=10): conventional post (pc), experimental post without filling (pp), with resin cement filling (ppc) and with optical fiber filling (ppf). For the Pull-out test, 100 roots of bovine teeth were restored with intraradicular retainers and composite resin crowns, distributed into 10 groups (n=10), with the following variables: glass fiber post type [conventional (C) or hollow (P)], the photocuring system [with or without the optical fiber illuminating device (O)], the type of resin cement [light cured (F) or dual cured (D)] and aging of the samples [with and without mechanical cycling (C)]. The aging process was performed by mechanical cycling and the samples were submitted to the Pull-out bond strength test. Failure mode analysis was performed using a stereomicroscope. Finite Element Analysis evaluated the stress distribution by von Mises analysis, total strain and maximum principal stress in Ansys Software 19.3. Statistical analysis consisted of the Shapiro-Wilk normality test, 1-, 2- and 3-way analysis of variance (ANOVA) and Tukey test 5%. The means and standard deviations of flexural strength were (MPa): pc = 677 ± 81.1; ppc = 419 ± 23.2; ppf = 200 ± 32.5 and pp = 177 ± 32.8. The means and standard deviations of bond strength were (MPa): CD = 12.2 ± 1.21; PD = 11.1 ± 1.32; PF = 10.9 ± 1.29; POD = 13.7 ± 1.16; POF = 11.9 ± 1.48; CDC = 10.1 ± 1.33; PDC = 9.25 ± 1.11; PFC = 8.37 ± 1.25; PODC = 10.8 ± 0.95 and POFC = 9.82 ± 1.02. The failure mode results were: adhesive between post and cement (56%), mixed predominantly adhesive between cement and dentin (22%), adhesive between cement and dentin (14%) and mixed predominantly adhesive between post and cement (8 %). It was concluded that among the experimental posts, the hollow post group filled with resin cement obtained the highest flexural strength value, the use of the illuminating device increased the results of bond strength in the hollow post groups and there was no difference in the stress distribution among the groups (AU)
