RESUMEN

This study aims to report a spontaneous and experimental intoxication in cattle by Melanthera latifolia (Asteraceae) for the first time, and to describe its epidemiological, clinical, and pathological findings. An outbreak of acute toxic hepatopathy in cattle occurred from December of 2021 to January of 2022 in a beef cattle farm from Rio Grande do Sul state, Southern Brazil, and resulted in 94 deaths from a herd of 430 animals. At necropsy, lesions consisted of enhanced lobular pattern of the liver, transmural gallbladder edema, and hemorrhages on the surface of multiple organs. The main microscopic lesion was marked hepatocellular necrosis in the centrilobular region associated with hemorrhage and infiltration of neutrophils. During the outbreak, multiple specimens of M. latifolia were noted in the paddocks where affected cattle were grazing and many showed signs of consumption. No other known acute hepatotoxic plants were found. Two 12-month-old steers were experimentally intoxicated with a single oral dose of the fresh plant (flowers, leaves, and less fibrous stalks). One bovine ingested a dose of 10 g/kg and was euthanized after 48 h, and the other ingested 15 g/kg and was euthanized 17 h later. Clinical signs, macroscopic, and microscopic lesions in both animals were similar to those observed in the spontaneous cases. Thus, this study demonstrates that M. latifolia was the cause of the outbreak of acute toxic hepatopathy with significant mortality in cattle. M. latifolia nor any plant of the genus Melantherahad ever been reported as toxic.

RESUMEN

Poisoning by Trema micrantha commonly causes hepatocellular necrosis in cattle, sheep, and goats and edema and cerebral hemorrhage in horses. This plant can cause toxic pneumopathy in sheep, and there is only one report of the natural form and one of the experimental form in the State of Rio Grande do Sul. This study aimed to report an outbreak of the respiratory form of natural poisoning by T. micrantha in sheep. Six sheep developed clinical respiratory signs after consumption of the plant and four of them died and two recovered after treatment with dexamethasone. The sheep presented tachypnea, noisy breathing, edema of the face, eyelids, and vulva, and subcutaneous emphysema on the face and neck. Necropsy (Sheep 2, 3, and 4) showed uncollapsed, heavy, diffuse red lungs with evident costal impressions and a moderate amount of serosanguineous fluid flowed at section. The liver had a moderate diffuse evident lobular pattern. The histopathology of the lungs of the three necropsied sheep showed congestion and edema with the formation of hyaline membranes within accentuated diffuse alveoli, in addition to thickening of the alveolar septa due to mild to moderate diffuse type II pneumocyte hyperplasia and also mild to moderate diffuse infiltrate of macrophages, lymphocytes, plasma cells, and neutrophils in the lumen of alveoli, bronchi, and bronchioles. Sheep 3 also showed type II pneumocytes with enlarged and hyperchromatic nuclei, sometimes binucleated with evident nucleoli, and, in some regions, the pneumocytes were desquamated to the alveolar lumen forming small syncytia and mild multifocal hyperplasia in the bronchial epithelium. The anti-cytokeratin IHC evaluation showed marked diffuse intracytoplasmic staining in hyperplastic type II pneumocytes in the bronchiolar epithelium of the three evaluated sheep. The liver of the three sheep had mild multifocal centrilobular necrosis. It seems to be the second report of spontaneous poisoning by T. micrantha in sheep developing lung lesions described in Brazil and the first in the State of Santa Catarina.

Niedenzuella (Tetrapterys) multiglandulosa, a vine plant found in Brazil, has been correlated to outbreaks of poisoning in cattle and buffaloes, generating economic losses related to the death due to heart failure, miscarriage, abortion, stillbirth, and neonatal mortality. The aim of this study was to examine the embryotoxic potential of the aqueous plant extract on in vitro bovine embryos. In vitro study was performed in five replicates of bovine embryo culture assigned in two groups: control, in vitro embryo culture medium without the aqueous plant extract; treated group, with addition of 2.7mg/mL of aqueous plant extract (10%) to the embryo culture on the sixth day of culture. Cleavage rate was evaluated at day 2 of the cell culture. Viability, hatchability and underdevelopment of blastocysts on the seventh, eighth, and ninth days (D7, D8, and D9, respectively) of culture were assessed under stereoscopic microscope. On day 7, blastocysts were submitted to TUNEL assay to determine apoptotic index. In vitro exposure of bovine embryos to of N. multiglandulosa resulted in reduced embryo development and survival, evaluated by dark cytoplasm indicating poor morphology and poor quality with marked reduction of hatchability. We observed a significant reduction of blastocyst production/number of cleaved embryos (60.6% vs 41.5%); reduction of blastocysts production/total number of matured bovine oocytes (35.1% vs 21.3%); and embryonic hatching rates (38.0% vs 10.0%). However, no effects were observed on the apoptotic rate. In conclusion, aqueous extract of N. multiglandulosa leaves reduces bovine embryo viability in vitro, suggesting possible detrimental effects on embryo development.(AU)

Enterolobium contortisiliquum (Vell.) Morong (Fabaceae) is a plant widely distributed in several regions of Brazil, occurring in the phytogeographic domains of Atlantic Forest, Cerrado, and Caatinga. Cases of serious poisoning in cattle, goats, and sheep in the country caused by the ingestion of beans of this species have been reported by several studies. The present work aimed to carry out a systematic review of cases of poisoning by E. contortisiliquum in ruminants and list the main chemical compounds isolated from this plant. For this, searches were performed in the Google Academic, PubMed®, ScienceDirect®, and SciELO databases. A total of 26 articles published in the last 20 years (2001-2021) were included. Studies on cases of natural and experimental poisoning indicate that this species mainly causes photosensitization, abortions, digestive problems, and acute ruminal lactic acidosis in animals that ingest the pods of the plant. The main chemical compounds that occur in the species belong to the triterpene saponins, monoterpene, phenylpropene, and triterpene classes. It is likely that triterpene saponins isolated from E. contortisiliquum are associated with reported cases of photosensitization in cattle. New studies must be conducted to assess the mechanisms of action of chemical compounds isolated from this species in in vivo systems.

RESUMEN

Mimosa tenuiflora (Willd.) Poir. (Fabaceae) is a plant native to Brazil and occurs in the phytogeographic domains of Caatinga and Cerrado. Relevant studies have investigated the chemical components of this plant and others have already demonstrated its teratogenic potential. It has been proven that this plant causes congenital malformations in farm animals and, consequently, financial losses to farmers in the Brazilian semiarid region. The present work aimed to carry out a bibliographic survey on the teratogenic effects of M. tenuiflora in ruminants and to group the chemical compounds occurring in this species. For this, databases were consulted and twenty-four articles published in the last 30 years (1990-2020) were included. According to the scientific documents analyzed, M. tenuiflora has embryotoxic, fetotoxic and abortive potential in farm animals, especially sheep and goats. The main classes of chemical compounds present in this species are alkaloids, saponins, flavonoids, and terpenoids. It is likely that some of these substances, mainly the indole alkaloid N,N-dimethyltryptamine, are related to the teratogenic effects reported in ruminants in the Brazilian semiarid region.

RESUMEN

This study aimed to assess the possible occurrence of reproductive changes in male goats associated with ingestion of Cenostigma pyramidale hay. Sixteen animals divided into two experimental groups, G1 and G2 (control group) were used. Animals in G1 received 2% of forage, based on live weight (LW), composed of 100% of C. pyramidale, and animals in G2 received 2% of Panicum maximum "Massai' grass hay, based on LW. Both groups received 1% of concentrated feed supplementation based on LW, along with mineralized salt and water ad libitum. The goats were subjected to weighing, testicular biometry, and semen and blood collection every 30 days. After 120 days, the animals were castrated and their testes were collected. Testicular measurements were performed and fragments were collected for histological processing to determine the gonadosomatic index (GSI), diameter of the seminiferous tubules, height of the germinal epithelium (HGE), volumetric proportion and volume of the testicular parenchyma components, total length of the seminiferous tubules, length of the seminiferous tubules per gram of testis, and leydigosomatic and tubulosomatic indexes. The data were evaluated for normality using the Student's t-test. Data with normal distribution were assessed using analysis of variance (ANOVA) and the non-parametric data were evaluated using the Kruskal-Wallis test, both at 5% probability. Statistically significant differences (p<0.05) were observed for GSI (G1=0.48 ±0.08 and G2=0.34 ±0.09) and HGE (G1=52.95 ±2.99 and G2=.47.63 ±2.67) between treatments. Consumption of C. pyramidale hay increased LW and, consequently, testicular weight, contributing to high GSI. In conclusion, ingestion of C. pyramidale has no toxic effect on the testicular, seminal and histological parameters of goat testis. Due to its nutritional characteristics, consumption of this plant improves animal body development. Because C. pyramidale is adapted to semi-arid regions, it can be an alternative source of feed for goats during periods of shortage.(AU)

INTRODUCTION: Crotalaria spectabilis is an important species used as a pre-plant cover for soybean crops to control the proliferation of endoparasitic nematodes. Species from the Crotalaria genus are known for presenting pyrrolizidine alkaloids (PAs) in their composition, however, C. spectabilis is still considered chemically under-explored. OBJECTIVE: The goal of this manuscript is the development and validation of a method for PAs and flavonoids identification and quantification of C. spectabilis seeds and leaves, a toxic plant used for nematode proliferation control in soil, especially in soybean crops. MATERIALS AND METHODS: Seeds and leaves extracts were analysed by high-performance liquid chromatography coupled to tandem mass spectrometry (HPLC-MS/MS) for the identification of the compounds. RESULTS: PAs and phenolic compounds could be identified in both samples based on the MS/MS fragmentation pattern. Molecular formulas of the annotated compounds were confirmed by ultra-high-performace liquid chromatography-quadrupole time-of-flight (UHPLC-QToF), and monocrotaline could also be confirmed by standard comparison. The quantification of monocrotaline was performed by HPLC-MS/MS, resulting in 123 times higher monocrotaline content in seeds than in the leaves, which could explain its efficiency in combating nematode proliferation in soil. CONCLUSION: This was the first report of phenolic compounds in C. spectabilis. The current study highlights the importance of C. spectabilis for nematode control due to the presence of toxic PAs, and the employment of analytical techniques for identification and quantification of compounds present in the extracts.

RESUMEN

The aim of this study was to describe the clinical, pathological and epidemiological aspects of an outbreak of tremorgenic syndrome in suckling lambs associated to Ipomoea asarifolia poisoning occurred in the metropolitan region of Recife, northeastern Brazil. Eight suckling Dorper lambs presented clinical signs after their mothers being kept in pasture invaded by I. asarifolia. Sixty percent of adult sheep, including lactating ewe, only exhibited engorged episcleral vessels. The suckling lambs exhibited neurological signs, such as head and neck tremors, dysmetria, wide-based stance, uncoordinated gait, unusual standing and imbalance followed by falls. These signs became more evident when the animals were forced to move. No gross lesions were found in a necropsied lamb that had died spontaneously. The histological findings were axonal spheroids in the cerebellum granular layer and numerous necrotic neurons in the cerebral cortex and cerebellum molecular layer. This study demonstrates that Ipomoea asarifolia causes tremorgenic syndrome. The toxin is eliminated through milk and suckling lambs exhibit a greater susceptibility compared to adult animals.(AU)

RESUMEN

The main objectives of this study were to determine the 1,2-dehydropyrrolizidine alkaloid (DHPA) content in Senecio grisebachii Baker (Compositae), to experimentally demonstrate its toxicity in calves and to describe the main clinical and pathological findings of this toxicity. S. grisebachii plants were collected in Paysandú, Uruguay. The concentration and identification of DHPA and associated N-oxides were determined using liquid chromatography-mass spectrometry. Three calves weighing 85-89 kg received doses of 15, 24 or 45 g of dry S. grisebachii per kg of body weight for 6, 10 or 20 days of treatment, respectively. Two animals received no treatment and served as controls. The animals were clinically evaluated, and blood samples were taken to study the serum levels of gamma glutamyltransferase (GGT), aspartate aminotransferase (AST) and alkaline phosphatase (FAS). After death, necropsy was performed and organ samples were taken for histopathological examination. The concentration of DHPA in S. grisebachii was found to be 0.29% (dry weight basis) as free base and 0.08% as N-oxide for a total DHPA concentration of 0.37%. Individual alkaloids identified included seneciophylline, senecionine and retrorsine. The disease was clinically characterized by depression, anorexia, emaciation, colic, dehydration and death in the three animals. Serum concentrations of GGT, AST and FAS were higher than normal beginning on day 7 after start of treatments. Necropsy findings included generalized edema, hemorrhage, ascites and a grayish liver with increased consistency. The main histological lesions were hepatic necrosis, fibrosis, hepatomegalocytosis and bile duct proliferation. The control calves showed no clinical signs of disease.

ABSTRACT Plants produce a wide variety of secondary metabolites, which are frequently related to a plant’s protective mechanism against predators and pathogens. Toxic species are those capable of producing compounds that can cause metabolic changes harmful to humans and animals. The toxicity of plant species can be associated with aspects related to the individual, the plant, the manner of exposure, and to environmental issues. Acute or chronic intoxication caused by plants is difficult to diagnose and the association between the symptoms and the consumption of and/or contact with plants is hard to establish. In the public health sector, intoxications caused by plants have a wide impact. In Brazil, 1,026 cases were registered in 2012, most of which occurred with children between the ages of 0 to 4 years, according to data reported by SINITOX. The purpose of this study was to perform a bibliographic survey of Brazilian or acclimatized exotic plant species, which have been reported as toxic even though they are used for ornamental or medicinal purposes.

The first occurrence of sodium monofluoroacetate in plants was reported in Dichapetalum cymosum. Further, this toxic substance was found in other plant species, as Palicourea marcgravii, which causes /"sudden death/". Amorimia rigida, which is well known and frequent in northeastern Brazil, it is among the poisonous plants that have identical lethal effects in cattle to that caused by the ingestion of plants containing sodium fluoroacetate. These intoxication signs are the same that ones presented by experimentally poisoned animals with this toxic. Significant economic losses can be imputed to A. rigida, therefore our aim was to identify its toxic compound and also to establish its biomarkers of effect. The study by cellulose thin layer chromatography using as mobile phase solution of ethanol /ammonium hydroxide/pyridine/water (95:3:1:1, v/v/v/v) and Nile blue as the derivatizing agent - of the aqueous extract of milled dried plant showed the presence of MFA-Na. The fractionation of the hydroalcoholic extract of A. rigida by XAD-2 column chromatography gave four fractions: 100% H2O (F1), H2O: MeOH 80/20 (F2) H2O: MeOH 40/60 (F3) and 100% MeOH (F4); only F1 fraction contained MFA-Na, according to CCDC monitoring. This fraction was subjected to preparative thin layer chromatography, in order to isolate the toxic principle which, in a study by infrared spectroscopy and by comparison with standard MFA-Na, confirmed being a mixture of mono-, di and trifluoroacetate. In the biochemical study, animals intoxicated by MFA-Na and by A. rigida extract showed serum calcium/citrate imbalances; such alterations may be considered biomarkers of MFA-Na exposure effect

Physic nut (Jatropha curcas) is a plant cultivated for biofuel production. Pericarp is a potential livestock food source by-product. However, its use may be limited due to the presence of toxic compounds, mainly phorbol esters. Thus, this study aimed to evaluate pericarp toxicity. Twenty sheep were divided in four groups, one control group which did not receive the plant and three experimental groups which received pericarp in 15% (G15), 30% (G30) and 45% (G45) concentrations for 23 days. After 10 days of treatment, pericarp ingestion produced food intake decrease, diarrhea, dehydration and loss of body condition. All treated groups showed decrease in alkaline phosphatase activity. G30 animals presented reductions in urea and total protein concentrations, and increase in potassium and sodium levels. G45 animals showed increase in serum aspartate aminotransferase activity and in albumin, creatinin, total and indirect bilirubin levels. Anatomohistopathologic findings included ascites, hydropericardium, congestion of the gastintestinal tract and lungs, pulmonary edema and adhesions in the thoracic cavity, renal tubular cells and centrilobular cytoplasmic vacuolation and lymphohistiocytic pneumonia and lymphoplasmacytic and histiocytic enteritis. On the physiochemical analysis 0.3845mg of phorbol esters/g of pericarp were detected. It is concluded that J. curcas pericarp is toxic and is not recommended for sheep feeding.(AU)

RESUMEN

Clinical and laboratory changes were evaluated in rabbits after intoxication by Amorimia rigida, a plant that causes sudden death. Nine New Zealand male rabbits, averaging 3.54 kg, were categorized into three groups (n = 3) and received, for eight consecutive days, the equivalent of 30 g/kg dry matter of A. rigida water-soluble (SG) and water-insoluble (IG) extracts via nasoesophageal route. The control group received water. There were no alterations in creatine kinase enzyme (CK), CK myocardial fraction (CKMB) or troponine I (cTnI). None of the animals had clinical or electrocardiographic (conventional and Holter) alterations. There were progressive decreases in the left ventricular ejection fraction and systolic fractional shortening. Doppler echocardiography alterations suggested a systolic dysfunction in the SG and IG groups and diastolic dysfunction in IG group. It was concluded that the soluble and insoluble extracts of A. rigida cause deficit of cardiac function.

A study was carried out to evaluate the biochemical profile in Mascagnia rigida poisoning, a toxic plant that generates a significant economic problem to livestock, causing "sudden death". Nine New Zealand rabbits, male, 3,54kg mean body weight were divided into three groups (G) (n = 3). The animals received the equivalent of 30g/kg of dry matter in two types of extracts: water-soluble (GS) and insoluble in water (GI), and the control group (CG) (ultra-pure water) for eight consecutive days. Biochemical exams were done prior to the beginning of the experiment until the ninth day. It was concluded that the administration of extracts of Mascagnia rigida cause electrolyte imbalances that may justify some clinical signs and act significantly in the cause of death.

RESUMEN

Buchenavia tomentosa Eichler is a common plant in Brazilian cerrado. Fruits of thisplant are employed in human feeding and folk medicine. Cattle producers affirm thatconsumption of the fruits cause abortion in cows, and even death. Considering that the plantmay be consumed by pregnant women and animals, the present study was undertaken toevaluate the possible toxic effects of the ingestion of B. tomentosa fruit (10% added to the diet),from the first to the twenty-first days of gestation, on reproductive parameters and on physicaland neurobehavioral development of rats offspring. An increase in food consumption atpregnancy days 11 and 17, and weight increase at day 17 of pregnancy were observed. Besidesthat, we verified an increase in weight of male offspring on post natal day 1. Other parameterswere not affected by plant consumption. These results indicate that the consumption ofB. tomentosa at 10% during pregnancy cause slight toxicological effects. The changes verified inthe present study indicate toxic action of the fruit possibly induced by flavonoids with hormonalaction; however, further studies must be accomplished to corroborate this hypothesis.(AU)
