RESUMEN

The genus Helicoverpa includes several agricultural pests globally. Helicoverpa armigera was reported in several countries in South America in 2013, and in Puerto Rico, in 2014. This territory is considered an agricultural hub, with a high-input system of seed production in the southern region of the island, and also at the edge of the continental U.S. Possible natural dispersion of populations of H. armigera from the Caribbean or other Central American regions poses a continuing risk to the U.S. This study was performed during the post-detection scenario of H. armigera in Puerto Rico, from 2018 to 2021. A year-round pheromone trapping program of adult males indicated an increase in the population from October to March and differences in the occurrence of Helicoverpa spp. between the municipalities Juan Diaz and Salinas. The proportion of H. armigera/H. zea and detection of congeneric hybrids between these species were assessed based on genital morphology and DNA analysis. Interestingly, neither H. armigera nor expected hybrids were detected in the present study. The susceptibility of H. zea populations to the insecticides Spinetoram, Emamectin benzoate, Chlorantraniliprole, and Esfenvalerate was assessed, and an overall significant effect of insecticide susceptibility was detected. Chlorantraniliprole and Emamectin benzoate had the highest efficacy. These results contribute to the Integrated Pest Management and Insect resistance management programs to Helicoverpa spp. in Puerto Rico. In addition, provide validated information to be considered in mitigation plans, in the scenario of an invasion of H. armigera in the continental U.S.

RESUMEN

Given the increasing importance of Elaphria agrotina (Guenée) (Lepidoptera: Noctuidae) in corn crops, especially in the Brazilian Savannah biome, the present research aimed to study its development and survival on Greenes and Poitout & Buess artificial diets (25±1°C, 70±10% relative humidity [RH] and 12h photophase). Poitout & Buess diet was more suitable than Greenes diet, providing higher survival percent (77.51% vs. 5.57%), lower development time (49.81 days vs. 55.24 days) and higher fecundity (167.65 vs. 84.9 eggs), respectively. All the caterpillars reared on Poitout & Buess diet passed through six instars, while almost half of the larvae reared on Greenes diet went through one less instar stage. Regarding the main reproductive parameters, higher average time of generation (T) and lower values of net rates of reproduction (Ro), and increased (rm) intrinsic and finite reason of increase (λ), were observed for larvae reared on Greene's diet. Results presented in this study indicated that Poitout & Bues's diet is more suitable for maintenance of colonies of E. agrotina as compared to Greene's diet.(AU)

The micro-hymenopterans of genus Trichogramma are eggs parasitoids used in the biological control of lepidopteran pests. The objectives of this study was to record the interaction of species of Trichogramma and their hosts on crops in the Mid-North, in the states of Maranhão and Piauí, Brazil. Lepidopteran eggs were sampled on crops and non-crops. Trichogramma atopovirilia , T. manicobai , T. galloi and T. pretiosum occur naturally in eggs of eight lepidopteran species. Natural parasitism shown by Trichogramma species reveals the importance of these parasitoids as agents for biological control in the Mid-North region.(AU)

RESUMEN

Herein described is the biochemical characterisation, including in vitro and in vivo assays, for a proteinase inhibitor purified from Clitoria fairchildiana seeds (CFPI). Purification was performed by hydrophobic interaction and gel filtration chromatography. Kinetic studies of the purified inhibitor showed a competitive-type inhibitory activity against bovine trypsin and chymotrypsin, with an inhibition stoichiometry of 1:1 for both enzymes. The inhibition constants against trypsin and chymotrypsin were 3.3 × 10(-10) and 1.5 × 10(-10)M, respectively, displaying a tight binding property. SDS-PAGE showed that CFPI has a single polypeptide chain with an apparent molecular mass of 15 kDa under non-reducing conditions. However, MALDI-TOF analysis demonstrated a molecular mass of 7.973 kDa, suggesting that CFPI is dimeric in solution. The N-terminal sequence of CFPI showed homology with members of the Bowman-Birk inhibitor family. CFPI remained stable to progressive heating for 30 min to each temperature range of 37 up to 100 °C and CD analysis exhibited no changes in spectra at 207 nm after heating at 90 °C and subsequent cooling. Moreover, CFPI was active over a wide pH range (2-10). In contrast, reduction with DTT resulted in a loss of inhibitory activity against trypsin and chymotrypsin. CFPI also exhibited significant inhibitory activity against larval midgut trypsin enzymes from Anagasta kuehniella (76%), Diatraea saccharalis (59%) and Heliothis virescens (49%). Its insecticidal properties were further analysed by bioassays and confirmed by negative impact on A. kuehniella development.
