Little information is currently available concerning the mechanisms controlling palm seed germination. We compared the anatomical and physiological aspects of seeds of two neotropical palm species showing different levels of dormancy. The seeds of Attalea vitrivir and Butia capitata were evaluated for the endogenous contents of hormones (ABA, GAs, CKs, BRs, IAA, JA, SA and the ethylene precursor ACC) in their cotyledonary petiole and operculum (structures involved in germination control), the force necessary to displace the operculum, endo-ß-mannanase activities, and embryo cell elongation. The analyses were carried out on with intact dry and imbibed seeds as well as with seeds with the operculum mechanically removed, 2, 5 and 10 days after sowing. The germinabilities of the intact seeds of A. vitrivir and B. capitata were 68% and 3%, respectively; the removal of the operculum increased germination to more than 90% in both species. Reductions of ABA and increases in GAs contents coincided with cell elongation, although there is no evidence that hormonal balance and endo-ß-mannanase activity are involved in operculum weakening. The ratio between the embryo length and the force required for operculum displacement (EL/OF) was found to be 1.9 times greater in A. vitrivir than in B. capitata, which means that very small elongations in each cell would be sufficient to promote germination, resulting in a lower level of dormancy in the former species. EL/OF and cell growth control are therefore important for defining dormancy level in palm seeds.

Zingiberaceae containing over 1,000 species that are divided into four subfamilies and six tribes. In recent decades, there has been an increase in the number of studies about vessel elements in families of monocotyledon. However, there are still few studies of Zingiberaceae tribes. This study aims to establish systematic significance of studying vessel elements in two subfamilies and three tribes of Zingiberaceae. The vegetative organs of 33 species processed were analysed by light and scanning electron microscopy and Principal Component Analysis was used to elucidate genera boundaries. Characteristics of vessel elements, such as the type of perforation plate, the number of bars and type of parietal thickening, are proved to be important for establishing the relationship among taxa. Scalariform perforation plate and the scalariform parietal thickening are frequent in Zingiberaceae and may be a plesiomorphic condition for this taxon. In the Principal Component Analysis, the most significant characters of the vessel elements were: simple perforation plates and partially pitted parietal thickening, found only in Alpinieae tribe, and 40 or more bars composing the plate in Elettariopsis curtisii, Renealmia chrysotricha, Zingiber spectabile, Z. officinale, Curcuma and Globba species. Vessel elements characters of 18 species of Alpinieae, Zingibereae and Globbeae were first described in this work.

Pera is a neotropical genus that currently belongs to the family Peraceae. This circumscription resulted from an inclusion of the Rafflesiaceae between the old tribe Pereae and all other Euphorbiaceae, and wherein Pereae was elevated to family rank making Euphorbiaceae monophyletic again. These changes are necessary although Rafflesiaceae are holoparasitic with extremely reduced vegetative bodies and large flowers while Peraceae and Euphorbiaceae have well developed vegetative parts and reduced flowers. As the embryology of Peraceae was poorly known, and embryological processes are conservative, we studied the embryology of Pera glabrata, searching for similarities between Peraceae, Rafflesiaceae, and Euphorbiaceae that could support this grouping. Usual methods of light microscopy and scanning electron microscopy were utilised. The results show endothecium with reversed-T-shaped cells, prismatic crystals in the tapetum, and disintegrated aerenchymatous septum in the mature fruit as unique features for Peraceae and possibly apomorphies for the family. In addition to the unisexual flowers, porogamous fertilization is present and one ovule per carpel which may support the Peraceae-Rafflesiaceae-Euphorbiaceae clade. The comparative approach also suggests possible (syn-)apomorphies for linoids and phyllanthoids, only linoids, Rafflesiaceae, Euphorbiaceae, and Ixonanthaceae. The presence of a placental obturator found previously unknown in Peraceae emerged as a possible synapomorphy for the euphorbioids (including Ixonanthaceae, Linaceae, Phyllanthaceae, Picrodendraceae, Peraceae, Rafflesiaceae, and Euphorbiaceae), which appeared in a common ancestor of the group and has been lost in Rafflesiaceae.

Cabomba Aubl. is a genus that presents a range of features that have made it to be considered a potential genetic model for studies of early angiosperm evolution. Therefore, any study that expands our knowledge of this genus is potentially useful for the understanding of the evolution of early angiosperms. This paper reports the study of the anatomy and the ultrastructure of the stigma and the style of Cabomba caroliniana Gray during the 2 days of anthesis using bright-field microscope, fluorescence microscope and transmission electron microscope. The stigma is dry and has pluricellular papillae. The style is hollow with a central canal coated by an epithelium. The papillae have fewer organelles than those typical of glandular cells, and they are covered by a cuticle that is broken when pollen germinates. The ultrastructure of epithelial cells indicates that the cells lining the canal are secretory. The canal is filled with a fibrillar and granular substance. The pollen tubes grow inside the canal through this substance. The results are discussed in the context of what is known for other species of angiosperms.

During the diversification of angiosperms, seeds have evolved structural, chemical, molecular and physiologically developing changes that specially affect the nucellus and endosperm. All through seed evolution, programmed cell death (PCD) has played a fundamental role. However, examples of PCD during seed development are limited. The present review examines PCD in integuments, nucellus, suspensor and endosperm in those representative examples of seeds studied to date.

Drought tolerance is a key factor for the establishment and survival of tree species in tropical ecosystems. Specific mechanisms of drought resistance can be grouped into four functional ecotypes based on differences in leaf fall behavior: deciduous, brevi-deciduous, stem succulent and evergreen. To identify the key factors influencing phenology and cambial activity and thus drought tolerance, we tested the stomatal conductance, leaf water potential and stable carbon isotopes in the leaves and wood of 12 species from a tropical dry forest in Costa Rica. With wood anatomical techniques, we further studied seasonal cambial activity and a suite of wood traits related to water transport for each of the functional ecotypes. Using a principal component analysis, we identified two groups of variables that can be related to (i) hydraulic conductivity and (ii) control of transpiration and water loss. Hydraulic conductivity is controlled by vessel size as the limiting variable, water potential as the driving force and wood density as the stabilizing factor of the anatomical structure of an effective water transport system. Stomatal control plays a major role in terms of water loss or saving and is the dominant factor for differences in phenological behavior. Stem succulent species in particular developed a rarely identified but highly effective strategy against drought stress, which makes it a successful pioneer species in tropical dry forests.

The phylogenetic position of a new species of Anhellia (Myriangiales) was investigated by analysis of nucleotide sequences of ribosomal large subunit (LSU) and ITS regions. The new sequence was aligned with 28 sequences obtained from GenBank, including four species of Davidiellaceae (Capnodiales) used as outgroup. This study is the first attempt to resolve the placement of the genus Anhellia within Myriangiales. The genus Anhellia was strongly supported in Myriangiaceae by phylogenetic analyses. In addition, A. nectandrae sp. nov., collected on Nectandra rigida from a fragment of Atlantic forest in Brazil, is described, illustrated and a table with morphological features to all known Anhellia species is provided.

The sporogenesis and development of gametophytes in Tetracentron sinense Oliv. were studied with light microscopy. The anther has four microsporangia; its primary anther wall consists of an epidermis, an endothecium, one or two middle layers and one glandular tapetum. Simultaneous cytokinesis follows meiosis, forming a tetrahedral tetrad. Mature pollen grains are two-celled at the time of anther dehiscence. Its ovule is anatropous, bitegmic and crassinucellate; the development of the female gametophyte is of the monosporic 8-nucleate Polygonum type. Significantly, some striking features were first found in T. sinense: (1) anther dehiscence occurs soon after the endothecium fibrously thickens and the intersporangial septum degenerates; (2) tapetum degeneration is retarded, persisting up to the stage of two-celled pollen grain; (3) a few cellular events such as the vacuolization and the contraction and deformation of the pollen mother cell (PMC) and microspore are not normal at the PMC, dyad and tetrad stages. The abnormalities during male reproduction might be one of important factors resulting in the poor natural regeneration of T. sinense.

In flowering plants, the formation of gametes depends on the differentiation of cellular precursors that divide meiotically before giving rise to a multicellular gametophyte. The establishment of this gametophytic phase presents an opportunity for natural selection to act on the haploid plant genome by means of epigenetic mechanisms that ensure a tight regulation of plant reproductive development. Despite this early acting selective pressure, there are numerous examples of naturally occurring developmental alternatives that suggest a flexible regulatory control of cell specification and subsequent gamete formation in flowering plants. In this review, we discuss recent findings indicating that epigenetic mechanisms related to the activity of small RNA pathways prevailing during ovule formation play an essential role in cell specification and genome integrity. We also compare these findings to small RNA pathways acting during gametogenesis in animals and discuss their implications for the understanding of the mechanisms that control the establishment of the female gametophytic lineage during both sexual reproduction and apomixis.

The growth in thickness of monocotyledon stems can be either primary, or primary and secondary. Most of the authors consider this thickening as a result of the PTM (Primary Thickening Meristem) and the STM (Secondary Thickening Meristem) activity. There are differences in the interpretation of which meristem would be responsible for primary thickening. In Cordyline fruticosa the procambium forms two types of vascular bundles: collateral leaf traces (with proto and metaxylem and proto and metaphloem), and concentric cauline bundles (with metaxylem and metaphloem). The procambium also forms the pericycle, the outermost layer of the vascular cylinder consisting of smaller and less intensely colored cells that are divided irregularly to form new vascular bundles. The pericycle continues the procambial activity, but only produces concentric cauline bundles. It was possible to conclude that the pericycle is responsible for the primary thickening of this species. Further away from the apex, the pericyclic cells undergo periclinal divisions and produce a meristematic layer: the secondary thickening meristem. The analysis of serial sections shows that the pericycle and STM are continuous in this species, and it is clear that the STM originates in the pericycle.The endodermis is acknowledged only as the innermost layer of the cortex.
O crescimento em espessura do caule de monocotiledônea pode ser primário, ou primário e secundário. A maioria dos autores consideram o espessamento resultante do MEP (Meristema de Espessamento Primário) e do MES (Meristema de Espessamento Secundário). Há divergências de qual seria o meristema responsável pelo espessamento primário. Em Cordyline fruticosa o procâmbio forma feixes vasculares de dois tipos: traços foliares colaterais (com proto e metaxilema e proto e metafloema), e feixes caulinares concêntricos (com metaxilema e metafloema). O procâmbio também forma o periciclo, a camada mais externa do cilindro vascular, constituída por células menores e menos coradas que se dividem irregularmente, formando novos feixes vasculares. O periciclo dá continuidade à atividade procambial, originando somente feixes concêntricos. Concluiu-se ser o periciclo responsável pelo espessamento primário desta espécie. Mais distante do ápice as células pericíclicas passam a sofrer divisões periclinais originando o Meristema de Espessamento Secundário. A análise dos cortes seriados mostra que o periciclo e o MES são contínuos nesta espécie, ficando claro que o periciclo origina oMES. A endoderme é reconhecida, apenas, como a camada mais interna do córtex.

The roles of mucilage cells were investigated through morphological and cytological analysis during leaf development in young Araucaria angustifolia plants. Differentiation began in leaf primordia in the shoot apex, when the young cells underwent a greater increase in volume in comparison with other mesophyll cells. The mucilage polysaccharides were synthesized by dictyosomes, from where they were taken by large vesicles and released into a cavity formed by detachment of the tonoplast, which was separated from the cytoplasm. At the end of differentiation, the cell was completely filled with mucilage, a gel consisting of a denser reticular structure surrounding less dense regions. The nucleus and cytoplasm were degenerated in mature cells. The A. angustifolia mucilage cells presented some cytological resemblances to the mucilage cells of members of some dicotyledonous families; however, differences in the dictyosomes and the secretion route were observed. Translocation and water storage of solutes was suggested by the use of the hydroxy pyrenetrisulfonic acid tri-sodium salt apoplastic tracer. The tonoplast detachment, dechromatinization, nuclear condensation, and general degeneration of the membrane systems observed during maturity indicated a programmed cell death process, one not yet described for angiosperm mucilage cells.

A proposta deste trabalho é mostrar uma nova interpretação do meristema de espessamento primário em monocotiledôneas. Anatomia dos órgãos vegetativos das seguintes espécies foi examinada: Cephalostemon riedelianus (Rapataceae), Cyperus papyrus (Cyperaceae), Lagenocarpus rigidus, L. Junciformis (Cyperaceae), Echinodorus paniculatus (Alismataceae) and Zingiberofficinale (Zingiberaceae). A atividade meristemática da endoderme foi observada nas raizes de todas as espécies, no caule de Cyperus, Cephalostemum e Lagenocarpus rigidus, e no traço foliar de Cyperus e folha de Echinodorus. Considerando a continuidade dos tecidos através da raiz, caule e folha, as autoras concluem que no caule o periciclo permanece ativo durante a vida da planta, como um gerador de tecidos vasculares. O "Meristema de Espessamento Primário" é o periciclo em fase meristemática, juntamente com a endoderme e suas derivadas (ou apenas o periciclo). Próximo ao ápice caulinar, esses tecidos se assemelham a um único meristema, dando origem ao córtex interno e aos tecidos vasculares.

This paper proposes a new interpretation for primary thickening in monocotyledons. The anatomy of the vegetative organs of the following species was examined: Cephalostemon riedelianus (Rapataceae), Cyperus papyrus (Cyperaceae), Lagenocarpus rigidus, L. junciformis (Cyperaceae), Echinodorus paniculatus (Alismataceae) and Zingiber officinale (Zingiberaceae). The endodermis with meristematic activity was observed in the root of all the species, in the stem of Cyperus, Cephalostemum and Lagenocarpus rigidus, and in the leaf trace of Cyperus and leaf of Echinodorus. Considering the continuity of tissues through the root, stem and leaf, the authors conclude that in the stem the pericycle remains active throughout the life of the plant as the generator of the vascular tissue. The "Primary Thickening Meristem" is in fact the pericycle plus the endodermis and its derivatives (or only the pericycle). Close to the stem apex, the assemblage of seems to be a unique meristem, giving rise to the inner cortex and vascular tissues.

A rapid in vitro propagation system leading to the formation of shoots, calli, roots, cell suspensions and plantlets was developed for the Andean medicinal plant Fabiana imbricata (Solanaceae). Massive propagation of shoots and roots was achieved by the temporary immersion system (TIS), morphogenesis and maintenance of cell suspensions by standard in vitro culture techniques. Oleanolic acid (OA), rutin, chlorogenic acid (CA) and scopoletin content in aerial parts of wild growing Fabiana imbricata plants as well as in plantlets regenerated in vitro, callus cultures, cell suspensions and biomass, obtained by the TIS system was assessed by HPLC. On a dry weight basis, the OA content in the aerial parts of the plant ranged between 2.26 and 3.47% while in vitro plantlets, callus and root cultures presented values ranging from not detected up to 0.14%. The rutin content of the samples presented a similar trend with maxima between 0.99 and 3.35% for the aerial parts of the plants to 0.02 to 0.20% for plantlets, 0.12% for cell suspensions and 0.28% for callus. Rutin was not detected in the roots grown by the TIS principle. The CA and scopoletin content in the aerial parts of F. imbricata ranged between 0.22-1.15 and < 0.01-0.55%, respectively. In the plantlets, the concentration of CA was 0.29 to 1.48% with scopoletin in the range 0.09 to 0.64% while in the callus sample, the CA and scopoletin content were 0.46 and 0.66%, respectively. A very different result was found in roots grown by TIS, where both OA and rutin were not detected and its main secondary metabolite, scopoletin was found between a range of 0.99 and 1.41% with CA between of 0.11 and 0.42%.

The organogenetic cycle of main-branch shoots of Nothofagus dombeyi (Nothofagaceae) was studied. Twelve samples of 52-59 parent shoots were collected from a roadside population between September 1999 and October 2000. Variations over time in the number of nodes of terminal and axillary buds, and the length, diameter and number of leaves of shoots derived from these buds (sibling shoots) were analysed. The number of nodes of buds developed by parent shoots was compared with the number of nodes of buds developed, I year later, by sibling shoots. The length, diameter and number of leaves of sibling shoots increased from October 1999 to February 2000 in those shoots with a terminal bud. However, extension of most sibling shoots, including the first five most distal leaf primordia, ceased before February due to abscission of the shoot apex. Axillary buds located most distally on a shoot had more nodes than both terminal buds and more proximal axillary buds. The longest shoots included a preformed part and a neoformed part. The organogenetic event which initiated the neoformed organs continued until early autumn, giving rise to the following year's preformation. The absence of cataphylls in terminal buds could indicate a low intensity of shoot rest. The naked terminal bud of Nothofagus spp. could be interpreted as a structure less specialized than the scaled bud found in genera of Fagaceae and Betulaceae.

Candida guilliermondii cells were immobilized in Ca-alginate beads and used for xylitol production from concentrated sugarcane bagasse hydrolysate during five successive fermentation batches, each lasting 48 hours. The bioconversion efficiency of 53.2%, the productivity of 0.50 g/l x h and the final xylitol concentration of 23.8 g/l obtained in the first batch increased to 61.5%, 0.59 g/l x h and 28.4 g/l, respectively, in the other four batches (mean values), with variation coefficients of up to 2.3%.
