Abstract Pleurolophocercous cercariae found in the invasive gastropod Melanoides tuberculata (Müller, 1774) collected in a stream of the Vila do Abraão, Ilha Grande, Angra dos Reis, Rio de Janeiro, Brazil were used for experimental infection that enabled the identification of the heterophyid trematode Centrocestus formosanus (Nishigori, 1924). The parasite has been found in the locality since 2007, after two years of the introduction of M. tuberculata. Recently, from a sample of 483 specimens collected in June 2013, 101 (21%) were found infected with parasite. The potential environmental impacts caused by the parasite occurrence could be underestimated in the country, and actions to monitor and control both the parasite and the mollusk are necessary.


Resumo Cercárias do tipo pleurolofocerca encontradas no gastrópode invasor Melanoides tuberculata (Müller, 1774) coletados em um riacho da Vila do Abraão em Ilha Grande, Angra dos Reis, Rio de Janeiro, Brasil foram utilizadas em estudos de infecção experimental que possibilitaram a identificação do trematódeo heterofiídeo Centrocestus formosanus (Nishigori, 1924). O parasito vem sendo encontrado na localidade desde 2007, dois anos após a introdução do molusco. Recentemente, de uma amostra de 483 espécimes coletados em junho de 2013, 101 (21%) apresentavam-se infectados pelo parasito. Os potenciais impactos ambientais ocasionados pela ocorrência deste parasito podem estar sendo subestimados no país, sendo necessárias ações visando o monitoramento e controle tanto do parasito quanto do molusco.

Metacercarial stages of digeneans were collected from decapod crustaceans inhabiting intertidal rocky zones in central Chile. The digeneans were identified through a molecular analysis based on the V4 region of the 18S ribosomal RNA gene. We analyzed 356 crustaceans belonging to 10 species, 115 intertidal fish belonging to 6 species, and 4 specimens of 1 coastal bird species. In total, 74.1% of crustaceans were parasitized with metacercariae. We found 1 species of Opecoelidae. This species showed low genetic divergence (0% and 0.1%) with adult digeneans found in intertidal fish and with the species Helicometrina labrisomi infesting a subtidal fish from northern Chile (Labrisomus philippii). Additionally, we found 2 species of Microphallidae, 1 closely related to Maritrema (1.3% genetic distance) and the other related to Microphallus (5% genetic distance). Therefore, our findings showed that the decapod crustaceans are relevant hosts in food webs from the southeastern Pacific coast. Furthermore, we found 5 species of crustaceans as second intermediate hosts for H. labrisomi and 2 species as secondary intermediate hosts for 2 Microphallidae, which contribute to elucidate parts of their life cycles through molecular markers and extended the host distribution of H. labrisomi in the southeastern Pacific coast.

The aim of the present study was to evaluate the infection levels of the eye flukes Sphincterodiplostomum musculosum Dubois, 1936 (metacercariae) in Steindachnerina insculpta (Fernádez-Yépez, 1948) from three ecosystems under the influence of the Jurumirim reservoir (Paranapanema, Taquari and Veados Rivers). A total of 174 fish specimens were sampled between April 2011 and April 2012. There were high levels of infection by S. musculosum metacercariae in the eyes of S. insculpta from all the sampled ecosystems, thus presenting broad geographical distribution. The histological analyses revealed that the metacercariae were apparently lodged behind the retina. The smaller fish from the Taquari River were less parasitized by metacercariae than the adults. This pattern can be explained by the cumulative effect of parasitism, as demonstrated by the positive correlation between the abundance of metacercariae and fish body size.

Novas ocorrências e efeitos do parasitismo por metacercárias em olhos de peixe têm sido recentemente discutidas em muitos estudos. O objetivo deste estudo foi avaliar os níveis de infecção dos vermes de olho Sphincterodiplostomum musculosum Dubois, 1936 (metacercárias) em Steindachnerina insculpta (Fernádez-Yépez, 1948) de três ecossistemas influenciados pelo reservatório de Jurumirim (Rios Paranapanema, Taquari e Veados). Um total de 174 espécimes de peixes foi amostrado de Abril/2011 a Abril/2012. Metacercárias de S. musculosum mostraram altos níveis de infecção nos olhos de S. insculpta em todos os ecossistemas amostrados apresentando ampla distribuição geográfica. As análises histológicas revelaram que, aparentemente, estas metacercárias estavam alojadas por trás da retina. Os peixes menores do rio Taquari estavam menos parasitados por metacercárias do que os adultos. Esse padrão pode ser explicado pelo efeito cumulativo de parasitismo, evidenciado pela correlação positiva entre a abundância de metacercárias e tamanho do corpo dos peixes.

Traira (Hoplias malabaricus) is a neotropical fish that is widely distributed in freshwater environments in South America. In the present study, we documented the occurrence of metacercariae of Austrodiplostomum spp. (Diplostomidae) in the eyes and cranial cavity of H. malabaricus and described parasite-induced behavioral changes in the host. The fish were collected from the upper São Francisco River, in the Serra da Canastra mountain range, Minas Gerais, transported alive to the laboratory, observed for 2 weeks, and subsequently examined for parasites. Of the 35 fish examined, 28 (80 %) had free metacercariae in the vitreous humor (mean intensity=95.4; mean abundance=76.3), and 24 (68.57 %) had free metacercariae in the cranial cavity, mainly concentrated below the floor of the brain, at the height of the ophthalmic lobe (mean intensity=12.91; mean abundance=8.85). Specimens of H. malabaricus with a high intensity of infection in the brain displayed changes in swimming behavior.

Trypsin and bile salts have been identified as important triggers for excystation of Echinostoma metacercariae. Although excystation in trematodes is a well-known phenomenon, some morphological developmental changes remain to be elucidated. In order to gain further insight into the in vitro development of metacercariae, we assayed different cultivating conditions: 0.5% trypsin and 0.5% bile salts; 1% trypsin and 1% bile salts; 1% trypsin and 0.5% bile salts; 0.5% bile salts; or 0.5% trypsin. By means of light microscopy and confocal microscopy, we characterized each encysted, activated, breached and excysted stage based on the morphological features. However, breached and excysted stages were not revealed in both bile salts and trypsin-free medium. Excretory concretions (25 ± 3.9) were visualized within excretory tubules, close to the ventral sucker and genital anlage. The oral sucker armed with spines and digestive system was similar to those of adult worms. The reproductive system is composed of a genital anlage and the cirrus sac primordium. In short, trypsin and bile salts associated were fundamental for the in vitro metacercariae excystation of Echinostoma paraensei. This article presents the first detailed information of all stages of metacercariae excystation obtained through light and confocal microscopy.

This study was aimed at evaluating the infection levels of Austrodiplostomum compactum metacercariae in 378 specimens of Plagioscion squamosissimus ('corvina' or 'pescada branca') from the Nova Avanhandava reservoir, low Tietê River, São Paulo State, Brazil. High prevalence, mean intensity of infection and abundance were observed in P. squamosissimus during most of the study, with the exception of March 2004. The relative condition factor (Kn) did not differ between parasitized and non-parasitized fish. A statistically significant positive correlation was observed between host standard length and intensity of infection. The analysis of biotic and abiotic variables showed that no abiotic variable correlated with parasitic infection levels. Moreover, P. squamosissimus status was demonstrated to be unaffected by A. compactum metacercariae infection.
