RESUMO

Aspergillus niger ß-glucosidase was modified by covalent coupling to periodate activated polysaccharides (glycosylation). The conjugated enzyme to activated starch showed the highest specific activity (128.5 U/mg protein). Compared to the native enzyme, the conjugated form exhibited: a higher optimal reaction temperature, a lower Ea (activation energy), a higher K m (Michaelis constant) and Vmax (maximal reaction rate), and improved thermal stability. The calculated t 1/2 (half-life) values of heat in-activation at 60 °C and 70 °C were 245.7 and 54.5 min respectively, whereas at these temperatures the native enzyme was less stable (t 1/2 of 200.0 and 49.5 min respectively). The conjugated enzyme retained 32.3 and 29.7%, respectively from its initial activity in presence of 5 mM Sodium Dodecyl Sulphate (SDS) and p -Chloro Mercuri Benzoate ( p -CMB), while the native enzyme showed a remarkable loss of activity (retained activity 1.61 and 13.7%, respectively). The present work has established the potential of glycosylation to enhance the catalytic properties of ß-glucosidase enzyme, making this enzyme potentially feasible for biotechnological applications.

BACKGROUND: Maxillary sinus inverted papilloma entails medial maxillectomy and is associated with high incidence of recurrence. OBJECTIVE: To study the impact of prior surgery on recurrence rate after transnasal endoscopic medial maxillectomy. METHODOLOGY: Eighteen patients with primary and 33 with recurrent maxillary sinus inverted papilloma underwent transnasal endoscopic medial maxillectomy. Caldwell-Luc operation was the primary surgery in 12 patients, transnasal endoscopic resection in 20, and midfacial degloving technique in one. The follow-up period ranged between 2 to 19.5 years with an average of 8.8 years. RESULTS: Recurrence was detected in 8/51 maxillary sinus inverted papilloma patients (15.7 %), 1/18 of primary cases (5.5 %), 7/33 of recurrent cases (21.2 %); 3/20 of the transnasal endoscopic resection group (15%) and 4/12 of the Caldwell-Luc group (33.3%). Redo transnasal endoscopic medial maxillectomy was followed by a single recurrence in the Caldwell-Luc group (25%), and no recurrence in the other groups. CONCLUSION: Recurrence is more common in recurrent maxillary sinus inverted papilloma than primary lesions. Recurrent maxillary sinus inverted papilloma after Caldwell-Luc operation has higher incidence of recurrence than after transnasal endoscopic resection.

BACKGROUND: The frequency of ear, nose, and throat (ENT) involvement in pemphigus vulgaris (PV) is not clear; thereby, the importance of setting routine ENT examination for patients with PV could not be deduced. OBJECTIVE: Determine the prevalence of ENT involvement in patients with PV in Egypt; to modify the routine protocol and achieve a step towards better management. PATIENTS AND METHODS: Thirty-four patients with PV were included. Patients were asked about ENT symptomatology and evaluated for ENT manifestations. RESULTS: Twenty-five patients complained from ENT symptoms (74%). The pharyngeal/laryngeal-related symptoms were the most common. Eighty-two percent of patients had positive endoscopic findings. The most common were pharyngeal/laryngeal (76.5%). In total, the positive endoscopic findings superseded the positive symptomatic findings. More severe involvement was documented in non-smoking patients (P < 0.05). CONCLUSION: Full ENT examination as a routine for all patients with PV could be of great value, as it would lead to more accurate diagnosis, therefore improved management.

The Aspergillus niger NRC1ami pectinase was evaluated according to its hydrolysis efficiency of dry untreated orange peels (UOP), HCl-treated orange peels and NaOH-treated orange peels (HOP and NOP). Pectinase was entrapped in polyvinyl alcohol (PVA) sponge and the optimum pH and temperature of the free and immobilized enzymes were shifted from 4, 40 °C to 6, 50 °C respectively. The study of pH stability of free and immobilized pectinase showed that the immobilization process protected the enzyme strongly from severe alkaline pHs. The immobilization process improved the enzyme thermal stability to great instant. The unique feature of the immobilization process is its ability to solve the orange juice haze problem completely. Immobilized enzyme was reused 12 times in orange juice clarification with 9% activity loss from the original activity. Maximum reaction rate (V(max)) and Michaelis-Menten constant (K(m)) of the partially purified form were significantly changed after immobilization.

Bacillus subtilis NRC 33a was able to produce both inducible and constitutive extracellular levansucrase, respectively, using sucrose and glucose as carbon source. The optimal production of the levansucrase was at 30 degrees C. The effect of different nitrogen sources showed that baker's yeast with 2% concentration gave the highest levansucrase activity. Addition of 0.15 g/L MgSO(4) was the most favorable for levansucrase production. The enzymic synthesis of levan was studied using 60% acetone fraction. The results indicated that high enzyme concentrations produced increasing amounts of levan, and hence conversion of fructose to levan reached 84% using 1,000 microg/ml enzyme protein. Sucrose concentration was the most effective factor controlling the molecular weight of the synthesized levan. The conversion of fructose to levan was maximal at 30 degrees C. The time of reaction clearly affected the conversion of fructose to levan, which reached its maximum productivity at 18 hours (92%). Identification of levan indicated that fructose was the building unit of levan.
