RESUMO

Introduction: Plant growth-promoting bacteria (PGPB) have been primarily studied for atmospheric nitrogen (N) fixation but they also have the capacity to improve nutrition and yield of crop plants. Methods: Therefore, the objective of this research was to investigate the effects of inoculation with PGPB in association with different N rates on N uptake, grain yield, and oil concentration of dwarf castor beans in succession to legumes and grasses in Ilha Solteira, Brazil. The treatments consisted of N rates (0 to 180 kg ha-1 of N) and inoculation with three plant growth-promoting bacteria (Azospirillum brasiliense, Bacillus subtilis, and Pseudomonas fluorescens, applied by leaf) and a control with no-inoculation. Results: The grain and oil yields of castor beans were increased by 20 and 40% at a rate of 103 kg ha-1 of N in succession to grasses as compared to without N application. In addition, the grain yield of castor bean after legumes was increased by 28, 64, and 40% with estimated rates of 97, 113, and 92 kg ha-1 of N in combination with inoculations of A. brasilense, B. subtilis, and P. fluorescens as compared to without N application, respectively. Shoot, grain, and total N uptake were improved with foliar inoculation of A. brasilense, B. subtilis, and P. fluorescens at the N rates of 45, 90, and 135 kg ha-1, respectively. Discussion and conclusions: Topdressing of N at the rate of 103 kg ha-1 and foliar inoculation in succession to grasses and 180 kg ha-1 of N without the effect of foliar inoculation in succession to legumes are recommended for higher grain and oil yield of castor beans. Foliar inoculations with A. brasilense, B. subtilis, and P. fluorescens increased grain yield under reduced use of N fertilizer by 44, 37, and 49% in dwarf castor cultivation in succession to legumes, potentially contributing to sustainable agriculture.

RESUMO

Phosphate fertilization in highly weathered soils has been a major challenge for sugarcane production. The objective of this work was to evaluate the foliar levels of phosphorus (P) and nitrogen (N) and the technological quality and productivity of second ratoon cane as a function of inoculation with plant-growth-promoting bacteria (PGPBs) together with the residual effect of phosphate fertilization. The experiment was carried out at the research and extension farm of Ilha Solteira, state of São Paulo, Brazil. The experiment was designed in a randomized block with three replications in a 5 × 8 factorial scheme. The treatments consisted of five residual doses of phosphorus (0, 45, 90, 135 and 180 kg ha-1 of P2O5, 46% P) applied at planting from the source of triple superphosphate and eight inoculations from three species of PGPB (Azospirillum brasilense, Bacillus subtilis and Pseudomonas fluorescens), applied in single or co-inoculation at the base of stems of sugarcane variety RB92579. Inoculation with PGPBs influenced leaf N concentration, while inoculations with Pseudomonas fluorescens and combinations of bacteria together with the highest doses exerted a positive effect on leaf P concentration. Co-inoculation with A. brasilense + Pseudomonas fluorescens associated with a residual dose of 135 kg ha-1 of P2O5 increased stem productivity by 42%. Thus, it was concluded that inoculations with Pseudomonas fluorescens and their combinations are beneficial for the sugarcane crop, reducing phosphate fertilization and increasing productivity.

RESUMO

Water deficit is the most limiting abiotic stress to plants because it affects several physiological and biochemical processes. Brassinosteroids, including 24-epibrassinolide (EBR), are steroids that regulate growth and positively act on gas exchange. This research aims to determine whether EBR can attenuate the negative effects of water deficit, revealing possible contributions of this steroid on photosynthetic machinery of young Eucalyptus urophylla plants under water deficit. The experiment had a completely randomized factorial design with two water conditions (control and water deficit) and three levels of EBR (0, 50, and 100 nM EBR). Water deficit caused a decrease in the levels of total chlorophyll and carotenoids, but these photosynthetic pigments increased by 135 and 226%, respectively, in plants sprayed with EBR when compared to the water deficit + 0 nM EBR treatment. Regarding the antioxidant system, 100 nM EBR induced significant increments in superoxide dismutase (42%), catalase (52%), ascorbate peroxidase (147%), and peroxidase (204%). Steroid application in E. urophylla plants exposed to water deficit increased the effective quantum yield of the photosystem II (PSII) photochemistry and electron transport rate. However, interestingly, it decreased the nonphotochemical quenching and relative energy excess at the PSII level, indicating improvements related to PSII efficiency. This research revealed that application of 100 nM EBR attenuated the negative effects caused by water deficit, being explained by the positive repercussions on antioxidant enzyme activities, chloroplastic pigments, PSII efficiency, electron flux, and net photosynthetic rate.

RESUMO

BACKGROUND: Despite manifold benefits of nanoparticles (NPs), less information on the risks of NPs to human health and environment has been studied. Cobalt oxide nanoparticles (Co3O4-NPs) have been reported to cause toxicity in several organisms. In this study, we have investigated the role of Co3O4-NPs in inducing phytotoxicity, cellular DNA damage and apoptosis in eggplant (Solanum melongena L. cv. Violetta lunga 2). To the best of our knowledge, this is the first report on Co3O4-NPs showing phytotoxicity in eggplant. RESULTS: The data revealed that eggplant seeds treated with Co3O4-NPs for 2 h at a concentration of 1.0 mg/ml retarded root length by 81.5 % upon 7 days incubation in a moist chamber. Ultrastructural analysis by transmission electron microscopy (TEM) demonstrated the uptake and translocation of Co3O4-NPs into the cytoplasm. Intracellular presence of Co3O4-NPs triggered subcellular changes such as degeneration of mitochondrial cristae, abundance of peroxisomes and excessive vacuolization. Flow cytometric analysis of Co3O4-NPs (1.0 mg/ml) treated root protoplasts revealed 157, 282 and 178 % increase in reactive oxygen species (ROS), membrane potential (ΔΨm) and nitric oxide (NO), respectively. Besides, the esterase activity in treated protoplasts was also found compromised. About 2.4-fold greater level of DNA damage, as compared to untreated control was observed in Comet assay, and 73.2 % of Co3O4-NPs treated cells appeared apoptotic in flow cytometry based cell cycle analysis. CONCLUSION: This study demonstrate the phytotoxic potential of Co3O4-NPs in terms of reduction in seed germination, root growth, greater level of DNA and mitochondrial damage, oxidative stress and cell death in eggplant. The data generated from this study will provide a strong background to draw attention on Co3O4-NPs environmental hazards to vegetable crops.

RESUMO

OBJECTIVE: To develop and validate the Pediatric Vestibular Symptom Questionnaire (PVSQ) and quantify subjective vestibular symptom (ie, dizziness, unsteadiness) severity in children. STUDY DESIGN: One hundred sixty-eight healthy children (female, n = 91) and 56 children with postconcussion dizziness or a vestibular disorder (female, n = 32), between ages 6 and 17 years, were included. The PVSQ contains questions regarding vestibular symptom frequency during the previous month. The Strengths and Difficulties Questionnaire (SDQ), a brief behavioral screening instrument, was also completed. RESULTS: The PVSQ showed high internal consistency (10 items; Cronbach α = 0.88). A significant between-group difference was noted with higher (ie, worse) PVSQ scores for children with vestibular symptoms (P < .001); no significant differences were noted between patient groups. The optimal cut-off score for discriminating between individuals with and without abnormal levels of vestibular symptoms was 0.68 out of 3 (sensitivity 95%, specificity 85%). Emotional and hyperactivity SDQ subscale scores were significantly worse for patients compared with healthy participants (P ≤ .01). A significant relationship was noted between mean PVSQ and SDQ (parent-rated version) hyperactivity and total scores for patients (P ≤ .01) and the SDQ (self-rated) emotional, hyperactivity, and total score (P ≤ .01) in healthy controls. However, mean SDQ subscale and total scores were within normal ranges for both groups. CONCLUSIONS: Self-reported vestibular symptoms, measured by the PVSQ, discriminated between children presenting with vestibular symptoms and healthy controls and should be used to identify and quantify vestibular symptoms that require additional assessment and management.
