We studied the effects of stimulation of mononuclear phagocyte system on the state of CD117(+) hepatocytes of growth zone and CD117(+) hemopoietic stem cells after liver damage. Increased number of CD117(+) hepatocytes and enhanced expression of given antigen was revealed. CD117(+) hemopoietic stem cells respond to the impact by migration to the regenerating organ.

It is reputed that the ideal therapeutic approaches to treatment of patients with acute coronary syndrome (ACS) and myocardium infarction (MI) should be aimed at the inflammation reaction triggers. This study investigated the effectiveness of the impact of L- 17 compound of the group of 5- phenyl substituted-6H-1,3,4-thiadiazine-2-amines upon the course of experimental MI as compared to the impact of a preparation, officially registered in Russia as an immunomodulator, Tamerit, belonging to phthalhydrazid derivative substance. Acute MI in rats was induced by left coronary artery coagulation. Histological study of the myocardium sections and biochemical analysis has been carried out at the 1st and 7th days of the experimental MI. The conducted investigations have shown that under the action of immunocorrectors the inflammation reaction character changes, exudative/destructive inflammation is replaced by a proliferative-cellular one. Animals' blood biochemical analysis at the background of L-17 and Tamerit introduction has shown a decrease of aminotransferases and lactatedehydrogenases activity in blood as compared to the reference group of animals' indicators, which is evidently caused by epicardial injury of myocardium and lesser amount of the alternative cardiomyocytes. At the same time, no noticeable difference in biochemical characteristics in groups, having been treated to immunomodulators of different chemical composition was identified, which is the sign of the essential similarity of their impact. Thus, immunocorrectors of different chemical groups (Tamerit and compound L17) diminish the volume of initial myocardial infarction and accelerate the granulation processes in course of MI, and represent a new category of treatment agents.

The content of CD117(+)cells in the kidneys and CD45(low)CD117(+)cells in the bone marrow and blood of mice were studied after partial nephrectomy and under conditions of macrophage stimulation with 3-aminophthalhydrazide. The counts of tubular CD117(+)epitheliocytes sharply increased and the content of CD45(low)CD117(+)cells in the bone marrow decreased after renal damage. Injection of 3-aminophthalhydrazide stimulated the expression of CD117 by renal epitheliocytes and led to reduction of CD45(low)CD117(+)cell counts in the bone marrow and blood. Macrophages stimulated proliferative processes in the kidney and differentiation of stem cells in the bone marrow due to synergic effects of their cytokines and stem cell factor.

RESUMO

The effects of 3-aminophthalhydrazide and carrageenan on reparative regeneration and expression of CD117 by liver cells after partial hepatectomy were studied in mice. 3-Aminophthalhydrazide stimulated regeneration of the liver and increased the count of CD117(+) hepatocytes. By contrast, carrageenan inhibited liver reparation and CD117 expression.

AIM: To investigate the effects of Xanthigen (brown marine algae fucoxanthin + pomegranate seed oil (PSO)) on body weight, body fat, liver lipids, and blood biochemistry; and Xanthigen and its individual components on resting energy expenditure (REE) in obese, non-diabetic female volunteers with non-alcoholic fatty liver disease (NAFLD) and normal liver fat (NLF) content. METHODS: Sixteen-week, double-blind, randomized, placebo-controlled study. Food record data, body composition, REE (only 41 volunteers with NAFLD) and blood sample analysis were assessed weekly for 16 weeks in 151 non-diabetic, obese premenopausal women with liver fat content above 11% (NAFLD) n = 113, and below 6.5% (NLF) n = 38. RESULTS: Xanthigen-600/2.4 mg (300 mg PSO + 300 mg brown seaweed extract containing 2.4 mg fucoxanthin) resulted in statistically significant reduction of body weight (5.5 +/- 1.4 kg NAFLD group and 4.9 +/- 1.2 kg NLF group, p < 0.05), waist circumference (NAFLD group only), body (3.5 +/- 1.9 kg NAFLD group, p < 0.001; 3.6 +/- 0.7 kg NLF group, p < 0.05) and liver fat content, liver enzymes (NAFLD group only), serum triglycerides and C-reactive protein. Weight loss and reduction in body and liver fat content occurred earlier in patients with NLF than in patients with NAFLD. Fucoxanthin (> 2.4 mg) and Xanthigen-400/1.6 mg (200 mg PSO + 200 mg brown seaweed extract containing 1.6 mg fucoxanthin) significantly increased REE in NAFLD subjects compared to placebo. CONCLUSIONS: Xanthigen promoted weight loss, reduced body and liver fat content, and improved liver function tests in obese non-diabetic women. Xanthigen and Fucoxanthin also increased REE. This product may be considered a promising food supplement in the management of obesity.

The functional metabolic activity of neutrophilic leukocytes underwent clinical laboratory assessment in tropical malaria. A significant inhibited activity of myeloperoxidase and decreased levels of cationic protein, glycogen, and lipids were revealed while the activities of alkaline and acid phosphatase were increased. The degree of changes in the enzymatic activity of leukocytes and the level of intraleukocytic components depended on both the stage of the disease and the degree of the pathological process and the presence of mixed infections. The maximum changes in the content of all components of the neutrophilic microbicidal system were noted in patients with tropic malaria concurrent with typhoid fever. The values of the peripheral neutrophilic microbicidal system may be used as additional criteria for evaluating the severity and prognosis of tropical malaria.

The purpose of this randomized, double-blind, placebo controlled intervention clinical trial was to investigate the effect of orally administered dietary supplement Radical Fruits (www.gardenoflife.com) consisting of highly concentrated edible fruits and berries on the concentration of plasma cholesterol, urinary 8-epi-prostaglandin F2alpha (8-epi-PGF2alpha) and 11-dehydrothromboxane B2 (11-dehydro-TXB2) in none-obese, non-smoking, non-diabetic hypercholesteremic male volunteers. Forty four (n=44) none-obese, non-smoking, non-diabetic male volunteers with an average age of 40+/-12 years, average body weight of 77,4+/-5,0 kg, average body mass index (BMI) of 22,2+/-2,7 kg/m2 and average total plasma cholesterol level 280+/-22 mg/dL were recruited to take part in a 4 week double-blind, randomized, placebo-controlled clinical trial. After evaluating 12 different antioxidant supplements for their phytochemical compositions and bioavailability, a dietary supplement Radical Fruits was chosen for this clinical trial. Radical Fruits contains standardized extracts and concentrates of prune, pomegranate, apple, grape, raspberry, blueberry, white cherry and strawberry. Subjects were randomly assigned to the treatment group and the placebo group using Simple Randomization Procedure. Subjects in the treatment group (n=22) were directed to take 900 mg of Radical Fruits supplement three times a day before meals. Subjects in the control group (n=22) were directed to take placebo according to the same schedule. Food record analysis, body composition, blood and urine samples were assessed on admission and then once a week. The duration of the clinical trial was 4 weeks. Administration of Radical Fruits(TM) for 4 weeks resulted in statistically significant reduction of total plasma cholesterol from 280+/-23 to 250+/-11 mg/dL, (p<0,001). Total plasma cholesterol changes in the placebo group were not statistically significant. The average plasma LDL was reduced from 195+/-23 to 169+/-21 mg/dL (p<0,001) in the Radical Fruits(TM) group, while in the placebo group there were no statistically significant changes. Plasma HDL increased by 3,2=/-0,6% in the Radical Fruits treated group (p<0,001). No significant changes in the HDL levels were observed in the placebo group. Urinary 8-epi-PGF2alpha level decreased from 450+/-170 to 330+/-159 pg/mg creatinine (p<0,001); urinary 11-dehydro-TXB2 level decreased from 1,200+/-420 to 790+/-320 pg/mg creatinine (p<0,001) with no changes in the placebo group. Administration of pharmacologically active antioxidant supplement Radical Fruit(TM) in hypercholesteremic men significantly increased plasma HDL and reduced total cholesterol and LDL, and urinary oxidative and inflammatory isoprostanes and thromboxane.

The effects of oral treatment with Aralox phytopreparation containing Aralia mandshurica (Araliaceae) and Engelhardtia chrysolepis (Juglandaceae) extracts on some parameters of lipid metabolism was studied in women with nondiabetic obesity receiving low-caloric diet. Our randomized placebo-controlled study comprising 32 volunteers showed that aralox treatment led to a decrease in total body weight and fat weight, reduced perilipin content in adipocytes and plasma triglyceride content, and stimulated activity of hormone-sensitive lipase.

In a 4-week randomized placebo-controlled clinical trial we investigated the effect of 300 mg Blueberin, a phytomedicine containing 250 mg Blueberry leaves (Vaccinium arctostaphylos L, Ericaceae) extract providing minimum 50 mg 3,4-caffeoylquinic (chlorogenic) acid, and 50 mg myricetin, on fasting plasma glucose, alanine aminotransferases (ALT), aspartate aminotransferases (AST), glutamyltransferase (GGT) enzymes levels, and serum inflammatory C-Reactive proteins (CRP) in forty-two volunteer subjects (46+/-15 year of age, BMI 25+/-3 kgs/(m2)) diagnosed with Type 2 diabetes. During the 4-week trial, the Blueberin supplement was administered three times per day, 15-30 minutes prior to a meal along with 100 ml of water. Results of this trial revealed that the supplementation of Blueberin reduced fasting plasma glucose from 143+/-5,2mg/L to 104+/-5,7 mg/L (p<0,001), whereas there was no statistically significant changes in the Placebo group from 138+/-4,8 mg/L to 126+/-5,1mg/L (p>0,05). The reduction of fasting glucose was correlated with the reduction of serum CRP and in the Blueberin group from 5,18+/-1,4 mg/l to 2,14+/-1,8 mg/L (p<0,05), whereas in the Placebo group CRP levels were not significantly reduced from 5,11+/-1,7 mg/l to 4,94+/-1,1mg/L (p>0,05). Furthermore, the Blueberin also significantly reduced the levels of plasma enzymes ALT, AST and GGT, indicating that, in addition to anti-diabetes effects, the Blueberin also possess pharmacologically relevant anti-inflammatory properties.

RESUMO

Results of examination of systemic immunity state, microbiocenosis and immunohistological changes of tissues of the esophagus and stomach in 37 adult patients and 69 children with erosive-ulcerous diseases of the gastrointestinal tract were analyzed. Based on these examinations, up-to-date immunomodulator tamerit inhibiting pathologic activity of monocytes/macrophage system with simultaneous activation of neutrophil granulocytes was used in 154 adult patients and 69 children. Positive result was achieved in all the cases when tamerit was used.

The effects of extracts of Rhodiola rosea radix on blood levels of inflammatory C-reactive protein and creatinine kinase were studied in healthy untrained volunteers before and after exhausting exercise. Rhodiola rosea extract exhibited an antiinflammatory effect and protected muscle tissue during exercise.

We compared the effects of Tamerit, Polyoxidony, and Licopid on spontaneous and lipopolysaccharide-stimulated production of interleukin-1 and tumor necrosis factor by mouse peritoneal macrophages in vitro. The test preparations were equally potent in stimulating nonactivated cells. Licopid produced a costimulatory effect on macrophages primed with endotoxin. Tamerit in different doses suppressed cytokine production by cells. Polyoxidony in low doses activated, but in high doses suppressed this process.

We studied the effects of oral treatment with extracts from Rhodiola rosea (50 mg/kg) and Rhodiola crenulata (50 mg/kg) roots on the duration of exhaustive swimming and ATP content in mitochondria of skeletal muscles in rats. Treatment with R. rosea extract significantly (by 24.6%) prolonged the duration of exhaustive swimming in comparison with control rats and rats treated with R. crenullata. R. rosea extract activated the synthesis or resynthesis of ATP in mitochondria and stimulated reparative energy processes after intense exercise. Experiments proved different pharmacological characteristics of R. rosea and R. crenulata: R. rosea is most effective for improving physical working capacity.

In 77 patients with critical limb ishaemia treated surgically (arterialization of venous blood flow, creation of arteriovenous fistulas, transplantation of great omentum on microvascular anastomosis) regional perfusion of both legs was evaluated with radionuclide angiography. High perfusion of the ishaemic limb was revealed in the majority of patients. In these patients positive results were achieved most often, particularly in creation of arteriovenous fistulas on the leg low third and foot.

Serum MDA level and concentration of superoxide anion in leukocytes were measured in patients with acute hepatitis B and C at the peak of the disease, during regression, and during early convalescence. The concentration of active oxygen forms in leukocytes was maximum at the peak of the disease and decreased before discharge from the hospital.
