RESUMO

THE AIM OF THE STUDY: The study by the method of tissue polymerase chain reaction of the species composition of the microbiota of lesions of the oral mucosa in patients with bullous lesions. MATERIAL AND METHODS: Biopsy specimens of the oral mucosa of 51 patients were studied by the polymerase chain reaction method, of which 14 patients with pemphigus vulgaris, 17 patients with pemphigoid bullosa, and 20 patients with the bullous form of ruber lichen planus. 4 types of microorganisms have been identified - Fusobacterium, Streptococcus pneumoniae, Candida albicans, Ureaplasma spp. and viruses - Human Papillomavirus 16, Epstein-Barr virus and Citomegalovirus. RESULTS: In the study of the microbiota of bullous lesions, associations of microorganisms and viruses were established in a significant number of cases. Associations of Str.pneumoniae and C. albicans were quite common in patients with pemphigus vulgaris in 26.3%, pemphigoid bullosa in 20.0%, and in patients with the bullous form of ruber lichen planus in 14.3% of cases. In patients with pemphigus vulgaris, the association of Str.pneumoniae, C. albicans and EBV was noted in 31.6% of cases. In patients with the bullous form of ruber lichen planus in a high percentage of cases (28.6%), the associations of Str. pneumoniae, EBV and CMV. CONCLUSION: Identification at earlier stages of management of patients with bullous lesions Str. pneumoniae, Candida albicans, and Fusobacterium associated with herpes viruses should be regarded as one of the triggering mechanisms of an autoimmune conflict, which subsequently causes a specific clinical picture of these diseases.

The emergence of new concepts and mechanisms for remineralization is of great interest to study. The study objective was to evaluate the clinical efficiency of use of modern remineralizing formulas: "Enamel-sealing liquid" ("Tiefenfluorid", Humanchemie, Germany), "FluoroLux" (TechnoDent, Russia), "InnoDent" (PLC, "InnoDent", Kazakhstan) and ClinproTM XT Varnish (3M ESPE, Germany) in treatment of focal demineralization. According to the number of studied drugs in the framework of a clinical study, four groups of patients with a diagnosis of caries of enamel were formed (n=36): Group 1 (n=9 people, 30 teeth); Group 2 (n=9 people, 29 teeth); Group 3 (n=9 people, 28 teeth); Group 4 (n=9 people, 31 teeth). Evaluation of the clinical efficacy of enamel caries treatment was carried out using 1) vital staining with a 2% aqueous solution of methylene blue followed by evaluation using a control gradation 10-point grayscale; 2) laser-fluorescence method using the apparatus "DiagnoDent Pen" (KaVo). Statistical processing of the obtained results was performed using the Wilcoxon and Money-Whitney test. The confidence level was considered at p≤0.05. According to the data of laser fluorescence of enamel and vital staining, the median index with high statistical significance decreased in all groups after 3 months compared with the data before treatment. Thus, all the studied drugs showed remineralizing activity in the elimination of enamel demineralization foci. This study is the basis for the search and development of new remineralizing compounds for the prevention and treatment of focal enamel demineralization.

As a result of studying different populations of crustaceans of the genus Eurytemora Giesbrecht from the inland water bodies of the basins of the White Sea, the Pechora Sea, the Lena River delta, and Pacific coast of the United States, the species Eurytemora gracilicauda Akatova 1949 has been found along the entire northern border of the Russian Eurasia, while North American E. americana Williams 1906 has not been registered within these limits. Clear morphological and molecular genetic differences between E. gracilicauda and E. americana are presented.

RESUMO

The article is devoted to the study of immunohistochemical parameters in patients with bullous lesions of the oral mucosa. The biopsy samples of the oral mucosa were studied in 57 patients, including patients with pemphigoid bullosa (38 people) and pemphigus vulgaris (19 people). The results of immunohistochemical studies indicate the cellular mechanisms of damage to the epithelium, in which IL-1, IL-6 and HPV16 are involved.

OBJECTIVE Based on the data available in modern literature, to conduct a study on the epidemiology, etiology and pathogenesis of bullous lesions of the oral mucosa (pemphigus vulgaris, pemphigoid bullosa, lichen ruber planus). MATERIAL AND METHODS: The article is based on the study of materials of foreign and domestic research databases Pubmed and Elibrary. Used literature data over the past 15 years. RESULTS: According to domestic and foreign studies, among dermatological diseases, the most frequently detected diseases are: lichen ruber planus, pemphigoid bullosa and pemphigus vulgaris. The initial signs of these diseases are often neglected, both in patients and doctors, which leads to irreversible changes and severe course of the pathological process. In this connection, the number of patients with these lesions of the oral mucosa increases every year in all countries of the world. Bullous lesions of the oral mucosa are inflammatory and destructive diseases characterized mainly by recurrent course, variety of clinical manifestations, resistance to various therapeutic drugs. The specific features of the course of these diseases are explained both by the anatomical and physiological features of the oral mucosa, and by the etiologic and pathogenetic mechanisms of development. Currently, autoimmune processes play a leading role in the genesis of diseases, developing in response to changes in the antigenic structure of epidermal and epithelial cells under the influence of various damaging agents. CONCLUSION: Thanks to the study, it was found that currently there are no data confirming the presence of microbial contamination of the tissues of the oral mucosa in the pathology under study. Not enough information on the factors of the immune response, in particular, on proinflammatory cytokines in the tissues of the oral mucosa.

The content of regulatory proteins involved in adaptation to hypoxia and ischemia was studied in brain rat homogenate under conditions of normoxia and after bilateral ligation of the common carotid arteries. Preconditioning with amtizole in combination with moderate hypoxia increased the levels of HIF-1α, erythropoietin, vascular endothelial growth factor under conditions of normoxia. During experimental ischemia, combined preconditioning led to stabilization of the content of these regulatory proteins at the level of intact control and to a decrease in glycogen synthase-3ß kinase activity. This pattern of changes in regulatory proteins was noted during the early and late periods of preconditioning.

Characteristics of the effect of attenuated rabies virus strain «Moscow 3253¼ on morphological parameters of transplantable line Vero cells were studied by atomic force microscopy (AFM). Methods based on phase contrast microscopy and immunofluorescence were used to confirm the specificity of interaction and to identify the infectious activity of the rabies virus. Images of intact Vero cells and Vero cells infected with rabies virus were obtained at different periods of cultivation. The character of changes in the cell dimensions (length, width, height) and the cell membrane roughness depending on the rabies virus cultivation time was determined. During the observation period both increases and decreases in the size of the cells were recorded. The size of the infected cells exceeded that of the intact. An increase in the membrane roughness in cells exposed to rabies occurred during the entire period of observation, since the first hours of the interaction of the virus with the cell, while the intact Vero cells exhibited only minor changes in the membrane surface roughness, which were not dependent on the age of the culture. The dependence of the increase in the cell membrane roughness on the infecting dose of the rabies virus was determined. The obtained results open up the prospect of developing a methodological approach to the quantitative in vitro evaluation of the rabies virus using AFM. Changes in the cell membrane roughness appear to be the most indicative parameter for such evaluation.

Based on personal clinical experience the authors provide clinical guidelines for periodontal anesthesia. The by-step protocol for intraligamentary and intraseptal anesthesia is presented. The basic mistakes in technique and possible complications are described. Recommendations for a local anesthetic choice and dosage for different groups of teeth are given.

The accumulation of nickel ions in the roots and shoots of vetch seedlings (Vicia sativa L.) at increasing concentrations of nickel chloride in the medium was studied. It was shown that the accumulation of nickel in the shoots was increased when the concentration of nickel chloride in the medium was more than 50 µM. The bioconcentration factor and sustainability index for vetch seedlings were calculated under the experimental conditions. The obtained results were similar to parameters for other plants, grown on a nutrient medium or soil substrate. First, the obtained results allowed estimate the limits of nickel chloride concentrations for four of five zones, which correspond to the theoretical concept of dose-response curves in the studies on the influence of physiologically essential heavy metals on plants (Prasad 2010). Some parameters of oxidative stress caused by the presence of nickel chloride in the medium were shown. It seems that at low nickel concentrations in the medium in vetch seedlings the increase of several biochemical parameters (catalase activity and proline) caused by the high amylase activity in seeds.

RESUMO

The aim of the study was to prove the rationale for antiviral therapy combined with surgical procedures for treatment of severe oral leukoplakia. Complex clinical and laboratory evaluation and treatment was performed in 56 patients divided in 2 groups. Control group was presented by 13 patients receiving dental treatment, local and systemic keratoplastic formulations. Main group involved 43 patients in which conventional treatment protocol was completed by antiviral therapy and surgical procedures. Leukoplakia diagnosis was based on clinical findings, histological and immunohistochemical studies as well as optic coherent tomography data. The obtained results evidently prove the necessity for including antiviral therapy and surgical procedures in treatment scheme of severe oral leukoplakia.

Changes in the proteasome chymotrypsin-like activity in mammary and thyroid carcinomas in comparison with the adjacent tissue were studied at stages T(1-4)N(0-3)M(0) and T(2-3)N(0-1)M(0), respectively. The activities changed in a wave-like manner over the course of mammary carcinoma growth in cases with and without metastases. The minimum increment of the activity in the tumor was recorded during the T(2)N(0) stage in the absence of local metastases. The increment of the activity reached the peak in N(1) tumors of the same size with metastases. The activities in the tumor and adjacent tissues virtually did not differ during the T(3-4)N(1-3) stages. The time course of proteasome activity changes in thyroid tumors of the studied stages was similar to that in mammary carcinoma. The results can be used for development of methods for evaluating the aggressiveness of mammary and thyroid tumors.

RESUMO

The aim of the study was to assess likopid effectiveness in complex treatment of oral dysbiosis. The study included 76 patients: 23 patients receiving antifungal and antimicrobial treatment, eubiotics and bacteriophages and 53 patients in which the above mentioned therapy was combined with licopid as immunocorrective agent. Laser flow cytometry was used to examine oral fluid bacteriostatic properties. The results of the study confirm licopid to be useful for complex treatment of oral dysbiosis.

The dynamics of the expression of LMP7 and LMP2 proteasome subunits in embryonic and early postnatal development of rat spleen and liver is investigated in comparison with the dynamics of chymotrypsin-like and caspase-like proteasome activities and expression of MHC (major histocompatibility complex) class I molecules. The immune subunits LMP7 and LMP2 distribution in spleen and liver cells in the development process is also studied. A mutual for both organs tendency to the increase of the expression of both LMP7 subunit and LMP2 one on P21 (the 21st postnatal day) as compared to the embryonic period is discovered. However, the total proteasome level is shown to be constant. At definite development stages, the dynamics of immune subunits expression in the spleen and liver was different. In the spleen gradual enhancement of both immune subunits level being detected on P1, P18 and P21, in the liver gradual enhancement periods on E16 (the 16th embryonic day) and E18 changed to the stage of the shrink of immune subunits level on P5. This level did not reliably change till P18 and was augmented on P21. The alterations revealed were accompanied by chymotrypsin-like activity raise and caspase-like activity drop in spleen by P21 as compared with the embryonic period, which proves the enlargement of proteasome ability to form antigenic epitopes for MHC class I molecules. In the liver, both activities increased by P21 in comparison with the embryonic period. Such dynamics of caspase-like activity can be explained not only by the change of proteolytic constitutive and immune subunits, but also by additional regulatory mechanisms. Besides, it is discovered that the increment of immune subunits expression in the early spleen development is connected with the process of successive forming the white pulp by B- and T-lymphocytes enriched by immune subunits. In the liver, the growth of immune subunits level by P21 was accompanied by their expression expansion in hepatocytes, while their plunge by P5 may be related to the loss of liver function of a primary lymphoid organ of the immune system by this stage and disappearance of B-lymphocytes enriched by immune proteasomes in it. In the spleen and liver, MHC class I molecules were revealed at the periods of the raise of proteasome immune subunits level. On E21 , the liver was enriched by neuronal NO-synthase, its level decreased after birth and enhanced to P18. This fact indicates the possibility of the induction of the immune subunits LMP7 [character: see text] LMP2 expression in hepatocytes in signal way with neuronal NO-synthase participation. The results obtained prove that T-cell immune response with spleen participation as regards rat liver cells is possible starting with P19-P21 stage. First, at this period, white pulp T-area is formed in the spleen. Second, enhanced immune proteasomes and MHC class I molecules levels in hepatocytes can procure antigenic epitopes formation from foreign proteins and their delivery to cell surface for their subsequent presentation for cytotoxic T-lymphocytes.

The possibility of examination of oral fluid bacteriocidal activity in complex diagnostics of oral mucosa disbiosis was evaluated. Thirty-seven patients were included in complex clinical and laboratory studies. The patients were divided in two groups: main group (30 patients exhibiting various grades of oral mucosa disbiosis) and control group (7 patients with no signs of oral disbiosis). The oral fluid bacteriocidal activity was examined by means of laser flow cytometry. Study results proved oral fluid bacteriocidal activity increase to correlate with the grade of oral mucosa disbiosis thus confirming the usefulness of the method in complex diagnostics of oral disbiosis.

The phase-field model of Echebarria, Folch, Karma, and Plapp [Phys. Rev. E 70, 061604 (2004)] is extended to the case of rapid solidification in which local nonequilibrium phenomena occur in the bulk phases and within the diffuse solid-liquid interface. Such an extension leads to the fully hyperbolic system of equations given by the atomic diffusion equation and the phase-field equation of motion. This model is applied to the problem of solute trapping, which is accompanied by the entrapment of solute atoms beyond chemical equilibrium by a rapidly moving interface. The model predicts the beginning of complete solute trapping and diffusionless solidification at a finite solidification velocity equal to the diffusion speed in bulk liquid.
