RESUMO

The aqueous root extract of Securidaca longepedunculata (polygalaceae) was investigated for possible antinociceptive and central nervous system (CNS) effects in mice. Three nociceptive models; acetic acid, formalin and tail-flick tests were used to study the antinociceptive activity. Rectal temperature test was employed as an adjunct to the nociceptive models. The extract at 200 and 400 mg/kg significantly and dose dependently reduced the nociception induced by the acetic acid and in the early phase of formalin test (P<0.05). The extract exerted significant (P<0.05) hypothermic effect in the 15 and 30 min of the rectal temperature test. The antinociceptive and hypothermic effects were partially reversed by naloxone (1mg/kg). The tail-flick test produced an insignificant increase in tail-flick latency at 400 mg/kg after 60 min of the test, but significantly (P<0.05) increase tail-flick latency in the 400mg/kg group of animals pre-treated with naloxone (1 mg/kg) after 120 min of the test. The extract also produced a significant (P<0.05) naloxone reversible antidepressant like effect in the forced swimming test (an animal model of depression). Collectively, these results suggest that the extract possess antinociceptive and antidepressant like effects with possible involvement of opioidergic pathways. The extract at limit dose of 2 g/kg body weight appeared to be safe in oral formulation.

RESUMO

The non-lymphoid elements of the peripheral leucocyte pool were examined in the present study to determine their response to chemotactic stimulation. Our results indicate that granulocytes are effectively mobilized during malaria infections and are not deactivated by complement-derived chemotactic factors. These findings provide further evidence for the restriction of immunosuppression to some specific T and B-cell related functions only.

RESUMO

Chemotaxis has gained some prominence in recent years and available today are various methods that are applicable to both in vitro and in vivo measurements of the phenomenon. Some of these methods are discussed. Many are adaptable for use in the clinical laboratory as an aid in the diagnosis of disease syndromes which are characterized by or due to disorders of leucocyte chemotaxis.

Living, heat or formalin killed Bacteroides fragilis and a crude preparation of their cell walls were examined by the Boyden technique for chemotactic activity upon guinea pig peritoneal exudate cells. Their relative chemotactic activity ranged from 3.0 to 5.2 compared to an average value of 6.4 for the positive control, an endotoxic culture filtrate of Escherichia coli. A culture filtrate of B. fragilis and an index of 3.7. Miocrogram quantities of cytoplasmic preparations obtained by ammonium sulphate precipitation had chemotactic indices ranging from 2.8 to 6.4, the highest value being displayed by the precipitate formed between 50 and 75% saturation with ammonium sulphate. This fraction retained leucotactic activity after exposure to strong acid and heat. The leucotactic potency of these fractions did not correlate directly with their protein content. Further precipitation of the most active fraction with 80% ethanol revealed that there was little chemotactic activity attributable to polysaccharides. Gas liquid chromatography of a chloroform-methanol extract of the cells which had a chemotactic index of 6.1 revealed the presence of more than thirty fatty acids ranging in carbon length from C8 to C25. These results suggest a role of lipids as initiators of the leucotactic response associated with infections caused by B. fragilis.
