Background: Self-reported adherence scales are widely used in research and practice because they are low in cost and easy to apply. A free version in Brazilian-Portuguese of the Simplified Medication Adherence Questionnaire (SMAQ) can be a useful alternative for determining the adherent behavior of hypertensive patients. Purpose: To translate and evaluate the psychometric properties of the Brazilian-Portuguese version of the SMAQ therapeutic adherence scale for patients with arterial hypertension. Patients and methods: A multicenter, cross-sectional study was conducted in five outpatient units in Maceió-AL and Aracaju-SE between January and July 2019. A total of 117 patients aged over 18 years using antihypertensive drugs were recruited. The cross-cultural adaptation followed international methodological recommendations. Internal consistency (Cronbach's alpha) was tested as a reliability parameter. Criterion and construct validity were verified by concurrent validation, exploratory factor analysis (EFA), and validation by known groups. Results: The participants had a mean age of 56.6 years (SD = 10.7 years); most were female (72.6%). The mean number of antihypertensives prescribed per patient was 1.87 (SD = 0.87). There were 79.5% (n = 86) of patients considered non-adherent. Internal consistency was satisfactory (Cronbach's alpha = 0.63). A satisfactory correlation coefficient was verified with the Morisky-Green-Levine test as an external criterion (r = 0.56, p < 0.001). The scale's sensitivity measured through known group validity was 75.3%, specificity 29.5%, positive predictive value 63.9%, and negative predictive value 41.9%. We identified two factors of the instrument's construct from EFA: specific medication-taking behaviors and barriers to adherence. The initial KMO measure of sampling adequacy was 0.691, and Bartlett's test of sphericity was significant (χ2 = 118.342, p < 0.001). Conclusion: The Brazilian-Portuguese version of the SMAQ scale proved valid and reliable for determining adherence to the pharmacotherapy in hypertensive patients. It showed more ability to detect non-adherent patients but with low specificity, possibly influenced by high social desirability.

DNA methylation with 5-methylcytosine (5mC) has been reported in the genome of several eukaryotes, with marked differences between vertebrates and invertebrates. DNA methylation is poorly understood as its role in evolution in insects. Drosophila gouveai (cluster Drosophila buzzatii) presents larvae that develop obligatorily in necrotic tissues of cacti in nature, with the distribution of populations in South America, and plasticity of phenotypes in insect-plant interaction. We characterize organisms at developmental stages and analyze variations at multiple methylation-sensitive loci in pupae, and adult flies using methylation sensitive amplification polymorphism. We obtained 326 loci with CCGG targets in the genome of D. gouveai. Genomic regions with molecular lengths from 100 to 700 pb were most informative about methylation states. Multiple loci show differences in methylation-sensitive sites (MSL) concerning developmental stages, such as in pupae (MSL = 40), female reproductive tissue (MSL = 76), and male reproductive tissues (MSL = 58). Our results are the first evidence of genome-wide methylation in D. gouveai organisms.

Canine monocytic ehrlichiosis (CME) has been observed to impact renal function. Currently, the recognition of acute kidney injury is through the nonspecific biomarker serum creatinine (sCr). Novel markers of renal injury such as urinary clusterin (uClust) and urinary cystatin B (uCysB) may increase our understanding of the relationship between ehrlichiosis and renal cellular injury. The aim of this study was to evaluate novel renal injury biomarkers in dogs with acute CME. Twenty healthy dogs were enrolled in the control group (CG), and 16 dogs naturally infected with Ehrlichia canis were included in the Ehrlichia Group (EG). All dogs were followed for 45 days. EG dogs were treated with doxycycline twice daily for the first 30 days. Urine and serum were collected at: 0, 0.5, 1, 15, 30, and 45 days after start of treatment. Urine concentrations of uClust and uCysB were determined using a research ELISA immunoassay. A linear mixed model was used to estimate population mean of renal injury markers with patient as the random effect, and day and treatment as fixed effects. EG was observed to have higher uClust values compared to CG (estimated population mean EG: 213 ng/dL vs. CG: 84 ng/dL, P < 0.001). EG was observed to have higher uCysB values compared to CG (estimated population mean EG: 248 ng/dL vs. CG: 38 ng/dL, P < 0.001). Increases in uCysB and uClust suggest the presence of renal injury and a possible mechanism for the observed predisposition to chronic kidney disease in dogs with ehrlichiosis.

The use of beneficial microorganisms is an important strategy to improve rice production in a sustainable way. The study was carried out to determine the effect of single and combined beneficial microorganism on the development of upland rice. The experiment was performed in greenhouse and arranged in a completely randomized design with 29 treatments and 4 replications. Treatments consisted of rice seeds cultivar BRS A501 CL treated with single and combined multifunctional microorganisms (1 (Serratia marcescens), 2 (Bacillus toyonensis), 3 (Phanerochaete australis), 4 (Trichoderma koningiopsis), 5 (Azospirillum brasilense), 6 (Azospirillum sp.), 7 (Bacillus sp.), 8 to 28 (combination of all these microorganisms in pairs) and 29 (control)). Inoculation of upland rice with sole and combined microorganism on upland rice increased the roots and shoots development, yield components and grain yield of upland rice. The combinations of Bacillus sp. (BRM 63573) and A. brasilense (AbV5), Azospirillum sp. (BRM 63574) + B. toyonensis (BRM 32110) and Phanerochaete australis (BRM 62389) + Serratia marcenscens (BRM 32114) led to improved roots and shoots development; increased number of panicles and grains per pot, 1000 grains weight and grain yield of rice plants. Besides, the combinations allow helped in increased accumulation of nutrients in roots, shoots and grains of rice plants.

BACKGROUND: Brazil lacks data from syphilis in its border areas. We aimed to describe the spatial and temporal distribution of acquired syphilis (AS), in pregnancy (SP) and congenital syphilis (CS) in Brazilian municipalities in the arches border contexts. METHODS: An ecological, cross-sectional study was conducted from 2010 to 2020. The study was based on the cases of syphilis available in the Notifiable Diseases Information System (SINAN), and on the Primary Health Care Information System. The detection rates of AS and SP, and the incidence of CS were estimated, and the time series was analyzed. Data between the border arches were compared. RESULTS: In 2020, data showed 7,603 cases of AS (detection rate 64.8/100,000 inhabitants), 3,960 cases of SP (detection rate of 21.6/1,000 live births) and 836 cases of CS (incidence of 4.6/1,000 live births) in the border region. Between 2010 and 2020, the mean annual increase of detection rate of SP was 53.4% in Brazil, 48.0% in the border region, 59.6% in the North Arch, 28.8% in the Central and 67.2% in the South. Annual variation on the incidence of CS for the same period was 31.0% in Brazil 38.4% at the border, in the North and South Arcs 18.3% and 65.7% respectively. The Central Arch showed an increase only between 2010 and 2018 (62.7%). A total of 427 (72.6%) municipalities has primary health care coverage ≥ 95% of the population. In 2019, 538 (91.8%) municipalities reported using rapid tests for syphilis, which decreased to 492 (84%) in 2020. In 2019, 441 (75.3%) municipalities reported administering penicillin, and 422 (72%) in 2020. CONCLUSION: Our data show syphilis reman problem at the Brazilian border, rates in pregnant are high. It was observed a reduction in the detection rates, SP and the incidence of CS between 2018 and 2020. Syphilis should be included on the agenda of all management levels, aiming at expanding access and quality care.

Recent reviews of data on worldwide insect decline include almost no information on Brazil. We gathered evidence from literature searches and a survey sent to researchers, to which 96 replied and 56 provided information and publications. We present 75 instances of trends recorded over an average span of 11 years for aquatic and 22 years for terrestrial insects. These include time-replicated samples and expert opinion based on long-term local collections. Most terrestrial data are for butterflies, bees and scarab beetles. Aquatic studies include several insect orders, usually sorted to genus or family. Terrestrial insects showed significantly more cases of declines than increases, both in abundance (17 : 3) and in diversity (11 : 1). In aquatic cases, no tendency was detected in abundance (2 : 2) or diversity (3 : 4), not counting cases with no trend. Differences in these results among habitats may be due to the shorter span and less change in environmental conditions in the aquatic surveys, which included sites already degraded before sampling. We offer guidelines for future long-term assessments, including resampling of legacy collection sites.

Clostridium perfringens is the etiological agent of NE, a disease that greatly affects the poultry industry. Experiments on the induction of NE are difficult to carry out, as it is a multifactorial disease, and thus different predisposing factors have been used. This study evaluated the effect of the Gumboro disease vaccine virus vaccine (IBDV-vac) associated or not with infection by Eimeria spp. in broilers, as a predisposing factor for NE. Broilers (n = 99) were divided into groups (11) challenged with IBDV-vac, Eimeria spp. CP type G (CP13, CP14 and CP03) or both. The macroscopic evaluation revealed that the highest average (3.45) of injury occurred for the CP13 + IBDV-vac group. The microscopic analysis showed that Eimeria spp. increased the population of intraepithelial lymphocytes and reduced the villus/crypt ratio in duodenum and jejunum when associated with CP13 or CP14. There was a synergistic effect between the CP strain used and the predisposing factors; nevertheless, it was not clear which was the most effective predisposing factor to potentiate the lesions, suggesting that the association of the strain with the factors should first be evaluated for each experimental protocol.

Local communities and individual species jointly contribute to the overall beta diversity in metacommunities. However, it is mostly unknown whether the local contribution (LCBD) and the species contribution (SCBD) to beta diversity can be predicted by local and regional environmental characteristics and by species traits and taxonomic relatedness, respectively. We investigated the LCBD and SCBD of stream benthic diatoms and insects along a gradient of land use intensification, ranging from streams in pristine forests to agricultural catchments in southeast subtropical Brazil. We expected that the LCBD would be negatively related to forest cover and positively related to the most unique streams in terms of environmental characteristics and land use (hereafter environmental and land use uniqueness, respectively). We also expected that species with a high SCBD would occur at sites with reduced forest cover. We found that the LCBD of diatoms and insects was negatively related to forest cover. The LCBD of insects was also positively related to environmental and land use uniqueness. As forest cover was negatively related to uniqueness in land use, biologically unique streams were those that deviated from the typical regional land cover. We also found that diatom traits, insect traits, and taxonomic relatedness partly explained SCBD. Furthermore, the SCBD of diatoms was positively correlated with forest cover, but the inverse was found for insects. We showed that deforestation creates novel and unique communities in subtropical streams and that species that contribute the most to beta diversity can occur at opposite ends of a land use gradient.

While land use intensification is a major driver of biodiversity change in streams, the nature of such changes, and at which scales they occur, have not been synthesized. To synthesize how land use change has altered multiple components of stream biodiversity across scales, we compiled data from 37 studies where comparative data were available for species' total and relative abundances from multiple locations including reference (less impacted) streams to those surrounded by different land use types (urban, forestry, and agriculture). We found that each type of land use reduced multiple components of within-stream biodiversity across scales, but that urbanization consistently had the strongest effects. However, we found that ß-diversity among streams in modified landscapes did not differ from ß-diversity observed among reference streams, suggesting little evidence for biotic homogenization. Nevertheless, assemblage composition did experience considerable species turnover between reference and modified streams. Our results emphasize that to understand how anthropogenic factors such as land use alter biodiversity, multiple components of biodiversity within and among sites must be simultaneously considered at multiple scales.

Developing a biomimetic material to wound care is an emerging need for the healing process. Poly (ε-caprolactone) (PCL) is a polymer with the necessary dressing's requirements often used in medicine. Their surface, physic-chemical and biological properties can be modified by adding bioactive compounds, such as andiroba seed oil (Carapa guianensis). This Amazonian natural plant has medicinal and pharmacological properties. For this purpose, PCL polymeric films incorporated with andiroba oil were investigated. The synthesis of hybrids materials was carried out in the solvent casting method. Thermal properties were evaluated using thermogravimetric analysis (TGA/DTGA) and differential scanning calorimetry (DSC). The solvent type on the surface and hydrophilicity of samples was studied using a scanning electron microscope (SEM). Additionally, contact angle measurements, functional groups analysis, fluid absorption capacity, and cell viability were performed. The results demonstrated the influences of andiroba oil under the morphology and thermal properties of the polymeric matrix; the hydrophilicity of the hybrid film obtained by acetic acid was reduced by 13%; the porosity decreased as the concentration of oil increased, but its higher thermal stability. The L929 cell line's proliferation was observed in all materials, and it presented nontoxic nature. It was demonstrated the ability of PCL hybrid film as a matrix for cell growth. Then, the materials were proved potential candidates for biomedical applications.

Petroleum has been extracted from oil reservoirs using different techniques. This activity is accompanied for a large amount of water and sometimes mixed with gas. This produced water has a high oil concentration and other toxic chemical compounds, thus, it must be treated to be reused or released to environment according to environmental protection regulations. Currently, ceramic membrane technology has been employed in the wastewater treatment, due to its high benefit-cost ratio. In this sense, this work aims to study the oil-water mixture separation process using a new configuration of tubular ceramic membrane module by computational fluid dynamic (ANSYS Fluent software). The proposed model is composed of mass and linear momentum conservation equations coupled to Darcy's law and SST k-ω turbulence model. Results of the volumetric fraction, pressure, and velocity distribution of the oil and water phases are presented and discussed. The results indicated that the proposed model and new device both have great potential to be used on the water/oil separation process and that the transmembrane pressure remains constant in the axial direction and decreases radially through the membranes, indicating an efficient system that favors the transport of clean water and oil retention.

DICER is a key enzyme in microRNA (miRNA) biogenesis. Here we show that aerobic exercise training up-regulates DICER in adipose tissue of mice and humans. This can be mimicked by infusion of serum from exercised mice into sedentary mice and depends on AMPK-mediated signaling in both muscle and adipocytes. Adipocyte DICER is required for whole-body metabolic adaptations to aerobic exercise training, in part, by allowing controlled substrate utilization in adipose tissue, which, in turn, supports skeletal muscle function. Exercise training increases overall miRNA expression in adipose tissue, and up-regulation of miR-203-3p limits glycolysis in adipose under conditions of metabolic stress. We propose that exercise training-induced DICER-miR-203-3p up-regulation in adipocytes is a key adaptive response that coordinates signals from working muscle to promote whole-body metabolic adaptations.

Acai seed was used herein as an Amazon biomass waste for the synthesis of activated and modified carbon in order to find a possible use for the large volume of residues generated during the processing of this fruit and to add value to this residue. Activated carbon materials were used to remove Pb2+, Fe2+, and Mg2+ metal ions from water. The efficiency of removal of these ions by the acai seed activated carbon was compared with that by commercial activated carbon. Activated carbon materials were prepared by carbonization and chemical activation using two KOH impregnation ratios, namely 1:1 (ACK1) and 5:1 (ACK5), by mass. These samples were modified by treatment with nitric acid under microwave heating (ACK1-M) and (ACK5-M), respectively. The result of the elemental analysis indicated that this biomass has carbon and sulfur contents of 43.29% and 0.10% wt, respectively. The textural parameters showed that the obtained activated carbon samples presented high surface areas between 1462 and 2774 m2 g-1. Raman analysis revealed the different degrees of graphitization of the activated carbon materials. Boehm titration identified the presence of phenolic, carboxylic, and lactonic groups in samples that were confirmed by Fourier transform infrared spectroscopy. In the metal adsorption tests, ACK5-M showed better removal efficiency, reaching 86% removal for Pb2+, 69% for Fe2+, and 8% for Mg2+in 1 h of contact time; these results were superior to those obtained for commercial carbon. The results indicated that acai seed can be used for the production of activated carbon and can also be used for metal removal.

BACKGROUND: Outpatient total joint arthroplasty is increasing in frequency as reimbursement models change. Potential benefits include same-day surgery for patients and decreased exposure to nosocomial pathogens. This study aims to determine if total elbow arthroplasty (TEA) is also trending toward an outpatient setting, and if there is any impact on complication rates as a result. METHODS: A retrospective chart review of the American College of Surgeons National Surgical Quality Improvement Program was performed. Specifically, the database was queried for all patients with CPT code 24363 from 2010-2017. The percentage of TEAs performed each year as an outpatient was trended from 2010-2017. Additionally, the complication rate between the inpatient and outpatient cohorts was compared. RESULTS: A total of 524 TEAs were analyzed. Of these, 111 procedures (21.2%) were performed as an outpatient. There was a statistically significant increase in the percentage of outpatient TEAs from 2010-2017 (P = .0016). In 2010, 2.4% of TEAs were outpatient, compared with 34.5% in 2017. The total complication rate trended toward being lower in the outpatient group, but this difference was not statistically significant (P = .08). CONCLUSIONS: There is a significant trend toward TEA being performed as an outpatient procedure, with more than one-third currently being performed in this manner. In our study, there was no difference in the complication rate between inpatient and outpatient TEAs; in fact, outpatient TEAs trended toward having a lower complication rate than inpatient TEAs. Taken together, the outpatient setting comprises an ever-increasing segment of TEA without an increase in morbidity to patients.

Ecological drift can override the effects of deterministic niche selection on small populations and drive the assembly of some ecological communities. We tested this hypothesis with a unique data set sampled identically in 200 streams in two regions (tropical Brazil and boreal Finland) that differ in macroinvertebrate community size by fivefold. Null models allowed us to estimate the magnitude to which ß-diversity deviates from the expectation under a random assembly process while taking differences in richness and relative abundance into account, i.e., ß-deviation. We found that both abundance- and incidence-based ß-diversity was negatively related to community size only in Brazil. Also, ß-diversity of small tropical communities was closer to stochastic expectations compared with ß-diversity of large communities. We suggest that ecological drift may drive variation in some small communities by changing the expected outcome of niche selection, increasing the chances of species with low abundance and narrow distribution to occur in some communities. Habitat destruction, overexploitation, pollution, and reductions in connectivity have been reducing the size of biological communities. These environmental pressures might make smaller communities more vulnerable to novel conditions and render community dynamics more unpredictable. Incorporation of community size into ecological models should provide conceptual and applied insights into a better understanding of the processes driving biodiversity.

Accurate and precise detection of anthropogenic impacts on stream ecosystems using macroinvertebrates as biological indicators depends on the use of appropriate field and laboratory methods. We assessed the responsiveness to anthropogenic disturbances of assemblage metrics and composition by comparing commonly employed alternative combinations of field sampling and individuals counting methods. Four datasets were derived by, in the field 1) conducting multihabitat sampling (MH) or 2) targeting samples in leaf packs (single-habitat sampling - SH) and, in the laboratory A) counting all individuals of the samples, or B) simulating subsampling of 300 individuals per sample. We collected our data from 39 headwater stream sites in a drainage basin located in the Brazilian Cerrado. We used a previously published quantitative integrated disturbance index (IDI), based on both local and catchment disturbance measurements, to characterize the intensity of anthropogenic alterations at each site. Family richness and % Ephemeroptera, Plecoptera and Trichoptera (% EPT) individuals obtained from each dataset were tested against the IDI through simple linear regressions, and the differences in assemblage composition between least- and most-disturbed sites was tested using Permutational Multivariate Analysis of Variance (PERMANOVA). When counting all individuals, differences in taxonomic richness and assemblage composition of macroinvertebrate assemblages between least- and most-disturbed sites were more pronounced in the MH than in the SH sampling method. Leaf packs seemed to concentrate high abundance and diversity of macroinvertebrates in highly disturbed sites, acting as 'biodiversity hotbeds' in these situations, which likely reduced the response of the assemblages to the disturbance gradient when this substrate was targeted. However, MH sampling produced weaker results than SH when subsampling was performed. The % EPT individuals responded better to the disturbance gradient when SH was employed, and its efficiency was not affected by the subsampling procedure. We conclude that no single method was the best in all situations, and the efficiency of a sampling protocol depends on the combination of field and laboratory methods being used. Although the total count of individuals with multihabitat sampling obtained the best results for most of the evaluated variables, the decision of which procedures to use depends on the amount of time and resources available, on the variables of interest, on the availability of habitat types in the sites sampled, and on the other methods being employed in the sampling protocol.

OBJECTIVES: Here, we assessed the ability of immature stages of the lacewing Ceraeochrysa caligata (Neuroptera: Chrysopidae) to prey upon different developmental stages of the red palm mite Raoiella indica (Acari: Tenuipalpidae), one of the most destructive invasive pests of palm trees in Neotropical regions. METHODS: Increasing densities of three stages of R. indica (eggs, immature stages, and adult females) were offered to C. caligata in coconut leaf arenas. The immature stages of C. caligata were less than 24 h old and were starved before being transferring to the arenas. The amount of prey consumed was recorded 6 h after releasing the C. caligata. RESULTS: Our results indicated that the ability of C. caligata to feed upon R. indica increased with the larval development of the predator. Higher feeding levels and shorter handling times were recorded for the first and second instars of C. caligata when preying upon the eggs and immature stages of R. indica. Furthermore, C. caligata individuals of different stages exhibited differential functional responses according to prey type (i.e., eggs, immatures, or adult females of R. indica). Ceraeochrysa caligata second instar individuals exhibited a sigmoid increase in consumption rate with increasing prey availability (i.e., a type III functional response) when preying upon immature stages of R. indica. However, when preying upon R. indica adult females, C. caligata second instar individuals exhibited a type II functional response (i.e., an increase in consumption rate with increasing prey availability, before reaching a plateau). Predator individuals of the first and third instar stages exhibited a type II functional response for all prey types. CONCLUSIONS: Collectively, our findings demonstrate that C. caligata, especially at the second instar stage, has potential as a tool for ecological management of the red palm mite.

Despite the importance of assessing beta diversity to understand the effects of human modifications on biological communities, there are almost no studies that properly addressed how beta diversity varies along anthropogenic gradients. We developed an algorithm to calculate beta diversity among a set of sites included in a moving window along any given environmental gradient. This allowed us to assess beta diversity among sites with similar conditions in terms of human modifications (e.g., land use or instream degradation). We investigated beta diversity using stream fish community data and indicators of human modification quantified at four spatial scales (whole catchment, riparian, local, and instream). Variation in beta diversity was dependent on the scale of human modifications (catchment, riparian, local, instream, and all four scales combined) and on the type of diversity considered (taxonomic or functional). We also found evidence for non-linear responses of both taxonomic and functional beta diversity to human-induced environmental alterations. Therefore, the response of beta diversity was more complex than expected, as it depended on the scale used to quantify human impact and exhibited opposite responses depending on the location along the environmental impact gradient and on whether the response was taxonomic or functional diversity. Anthropogenic modifications can introduce unexpected variability among stream communities, which means that low beta diversity may not necessarily indicate a degraded environmental condition and high beta diversity may not always indicate a reference environmental condition. This has implications for how we should consider beta diversity in environmental assessments.

A esporotricose é uma micose subcutânea, causada por um fungo dimórfico do complexo Sporothrix schenckii que acomete várias espécies, especialmente a felina, causando comprometimento local, disseminado ou sistêmico. Objetivou-se relatar a ocorrência de um surto de esporotricose felina na Região Metropolitana do Recife. Foram coletados suabes de úlceras cutâneas de felinos atendidos no Hospital Veterinário da Universidade Federal Rural de Pernambuco e em Clínicas Veterinárias particulares. O diagnóstico foi realizado por meio de exame citológico e cultura fúngica. Dos 115 casos suspeitos, 59 casos foram confirmados, número nunca registrado anteriormente no estado de Pernambuco. Os casos de esporotricose felina registrados têm demonstrado aspectos clínicos e epidemiológicos semelhantes aos ocorridos em outras regiões do país, caracterizando-se como uma doença predominantemente de machos jovens em idade reprodutiva e semi-domiciliados, de difícil tratamento e de rápida disseminação.(AU)


Sporotrichosis is a subcutaneous mycosis caused by a dimorphic fungus from Sporothrix schenckii complex, which affects various species, especially feline, able to cause local, disseminated or systemic impairment. We aimed to report the occurrence of an outbreak of feline sporotrichosis in the Metropolitan Region of Recife. Swabs of skin ulcers in cats treated in the Veterinary Hospital of the Federal Rural University of Pernambuco and in particular veterinary establishments were collected. Diagnosis was made by cytological examination and fungal culture. From 115 suspicious cases, 59 cases were confirmed, unprecedented results in the state of Pernambuco. These registered cases of feline sporotrichosis have demonstrated clinical and epidemiological characteristics similar to those occurring in other regions of the country, characterized as a quick-spreading disease, of difficult treatment, which affects predominantly semi-domiciled young males in reproductive age.(AU)

Salmonella detection is a key point in food safety testing, because of the frequent association of this pathogen with food poisoning in humans. The standard bacteriological tests currently used for Salmonella-detection are time-consuming; therefore, there is a need to develop alternative methods to accelerate the detection. In order to accelerate Salmonella diagnosis, we used the immunomagnetic separation assay associated with bacteriophage P22 for the rapid detection of the following Salmonella serovars in chicken rinses of drumsticks, artificially contaminated with 5, 10, and 100 CFU/25mL of bacteria: Salmonella enterica subsp. enterica serovar Heidelberg (S. Heidelberg), Salmonella enterica subsp. enterica serovar Enteritidis (S. Enteritidis) and Salmonella enterica subsp. enterica serovar Typhimurium (S. Typhimurium). The efficiency of the technique, represented by the time required for detection of positive and negative samples, was compared with that of the standard diagnostic tests used for this pathogen, the bacteriological assay and the polymerase chain reaction (PCR)-based test. This study confirmed the ability of the bacteriophage-associated immunomagnetic separation assay to identify 99.6% of Salmonella-positive samples of the three serovars tested. In contrast, the bacteriological assay and PCR-based test detected 95.1% and 98.5% of the Salmonella-positive samples respectively.(AU)


A detecção de Salmonella é um ponto crucial para a segurança alimentar, devido a frequente associação deste patógeno com infecções alimentares em humanos. O método padrão para detecção de Salmonella é o bacteriológico, mas o tempo requerido para o processamento das amostras e o diagnóstico final é longo, por isso existe a necessidade de desenvolvimento de métodos alternativos que visem acelerar esta etapa. Para isto utilizamos a separação imunomagnética associada ao bacteriófago P22 como técnica de detecção rápida para os seguintes sorovares de Salmonella: Salmonella enterica subsp. enterica sorovar Heidelberg (S. Heidelberg), Salmonella enterica subsp. enterica sorovar Enteritidis (S. Enteritidis) e Salmonella enterica subsp. enterica sorovar Typhimurium (S. Typhimurium), os quais foram inoculados artificialmente em lavados de sobre-coxas de frango nas seguintes concentrações: 5, 10 e 100 UFC/25mL. A eficiência da técnica, representada pelo tempo requerido para detecção de amostras positivas ou negativas, foi comparado com os testes rotineiramente utilizados para detecção de Salmonella, o exame bacteriológico e a reação em cadeia da polimerase (PCR). Este estudo confirmou a capacidade do teste de separação imunomagnética associado a bacteriófago, o qual identificou 99,6% das amostras positivas para Salmonella, dos três sorovares testados. Já o bacteriológico e PCR identificaram respectivamente 95,1% e 98,5% das amostras positivas.(AU)
