RESUMO

Workers on exposure to epoxy resins (ER) have been shown to reveal several variants of immunogrammes of blood, expression of Fas-ligands and oxygen-dependent metabolism of phagocytes. These are as follows: the immune deficiency variant (38.1%), the autoimmune one (33.3%), the state of norm (28.6%). The blood phagocyte oxygen-dependent metabolism tends towards activation but the phagocyte functional reserve is reduced which facts suggest depression of metabolism and phagocytic defence in the organism. Identified in the study are several variants of kinetic parameters of the blood serum chemiluminescence suggesting activation/inhibition of process of free radical oxidation, strengthening/weakening of the blood antioxidant defence, which events can be regarded as early diagnostic and prognostic criteria of adverse effects of epoxy resins on the organism of those handling them. These data suggest to us an insufficient reliability of the existing maximum permissible concentration of epoxy resins in the air of the working zone and attest to the need for its reconsideration.

The paper focuses on the membrane lipid complex status and lymphocyte metabolic activity in IHD patients to elaborate new approaches toward treatment and prevention of immune disorders in the above pathology. The study was made in blood specimens from 53 coronary patients presenting with progressive unstable angina. The diagnosis was made on the basis of a complex clinical-and-laboratory and electrocardiographic evaluation. The composition of the lymphocyte membrane lipid complex and blood serum was studied with the aid of the gas chromatography techniques. Three variants were identified of change in the lipid composition of lymphocyte membranes in IHD patients having undergone the above evaluation. We consider it expediant to use criteria for the state of lymphocyte membranes in IHD patients in order that we might properly deal with issues concerning diagnosis, prognosis and selection of optimum pathogenetic therapy of the medical condition in question.

RESUMO

Lipopolysaccharide (LPS) of the Pseudomonas fluorescens strain IMV 7769 (biovar I) was isolated and investigated. Fractions of the structural parts of the LPS macromolecule, lipid A, the core oligosaccharide, and the O-specific polysaccharide (O-PS), were obtained in a homogeneous state. 2-Hydroxydecanoic, 3-hydroxydecanoic, dodecanoic, 2-hydroxydodecanoic, 3-hydroxydodecanoic, hexadecanoic, octadecanoic, hexadecenoic, and octadecenoic fatty acids were identified in lipid A. In the hydrophilic moiety of lipid A, after acid hydrolysis, several amino acids, phosphoethanolamine, glucosamine, and three unidentified peaks forming a separate cluster together with glucosamine were found. Lipid A was shown to be phosphorylated. Glucose, fucose, rhamnose, glucosamine, galactosamine, two unidentified amino sugars, 2-keto-3-deoxyoctulonic acid (KDO), heptose, ethanolamine, phosphoethanolamine, and alanine were identified in the core oligosaccharide. O-PS of the LPS consisted of repeating trisaccharide fragments that included residues of amino sugars: 4-acetamido-4,6-dideoxy-D-galactose, 2-acetamido-2,6-dideoxy-D-glucose, and 2-acetamido-2,6-dideoxy-L-glucose. During growth, the strain under study excreted exocellular LPS (ELPS) into the medium. The LPS studied was similar to the LPS of the earlier investigated strains P. fluorescens (biovar I) IMV 1152 and IMV 1433 in the structure of O-PS, but differed from them in the composition of both lipid A and the core oligosaccharide. The LPS of the strain studied differed from LPS of the type strain P. fluorescens IMV 4125 (ATCC 13525) in all characteristics determined.

RESUMO

Lipid fatty-acid composition was determined as was the level of free cholesterol in serum and red cells of venous and arterial blood in patients with myocardial infarction presenting with certain complications. The most significant changes in fatty-acid composition of blood serum lipids were in pulmonary edema and pneumonia, with those in the cell lipid complex being recordable in cardiogenic shock and ventricular fibrillation. Fatty-acid composition of the arterial blood lipids is more balanced compared to the venous blood in pulmonary edema, cardiogenic shock, ventricular fibrillation; the opposite tendency is observed during the development of pneumonia.

Presents a gas chromatographic analysis of lipid fatty acid composition and levels of free cholesterol in high-density and low-density lipoproteins (HDLP and LDLP) of patients with myocardial infarction with and without Q wave. The most expressed changes in the lipid fatty acid composition both in HDLP and LDLP were revealed in the patients without Q wave.

RESUMO

Overall thirty patients with acute Q-wave myocardial infarction (AMI) were studied. There were ten cases among them of uncomplicated AMI, eight ones of AMI presenting with arrhythmias, 12 patients suffered from AMI complicated by acute left ventricular failure. The functional state of T-lymphocytes was studied with the aid of the fluorescent nucleus probing with acridine orange. At day 1 to 3 Q-wave AMI the synthetic activity of T-lymphocytes was found out to be on the decrease. By day 14 to 16, patients with AMI running a complicated course had high values for metabolic activity of T-lymphocytes, which fact might be indicative of poor prognosis.

Free radical processes and some indices of antibacterial defense system have been examined in 29 male residents of Chernobyl area during adaptation to periods of intermittent hypoxia. 18 men (the experimental group) were exposed to normobaric isocapnic progressive hypoxia during 10 days of three daily 5-7 min sessions with 15 min breaks, and 11 men (control group) were exposed to air breathing. All subjects were divided into two subgroups with initial high (1) and low (2) level of blood chemiluminescence (ChL). Patients of the 1 subgroup were characterized with high oxygen-generated activity of neutrophils (OGA) and high malondialdehyde (MDA) concentration. After hypoxic training (HT) there was a decrease of spontaneous and initiated ChL and MDA. Patients of the 2 subgroup were characterised with low level both spontaneous and initiated ChL, low MDA concentration and low phagocytosing activity of neutrophils. After HT there was significant rise of initiated ChL and MDA concentration up to normal level with the increasing of neutrophil phagocytosing activity. We suggest that HT causes the normalizing effect on free radical processes in subjects who were exposed to radiation influences.

RESUMO

Derangement in freeradical processes (FRP) and lipid metabolism is known to occur in ischemic heart disease. Since autoimmune component appear to play an important role in the pathogenesis of IHD, the contributers to the article studied the state of freeradical processes in IHD patients during development of autosensibilization. A total of 42 patients with unstable angina and 8 essentially healthy subjects were examined. The blood level of FRP, antioxidant protection were determined using chemiluminometry technique. The blood serum content of free cholesterol and phospholipid fatty acid composition were evaluated by gas-liquid chromatography, the T- and B-lymphocyte functional status by fluorescent probing with acridine orange. The level of autoantibodies was determined as recommended by Boiden. The lymphocyte circulating population functional activity was found to be related to the state of FRP, antioxidant protection and the blood serum lipidic complex composition.

RESUMO

The relationship between the state of the lipid complex of lymphocyte membranes and their functional activity has been studied in normal mice after immunization and in mice with staphylococcal infection. The study has revealed essential differences in the dynamics of free-radical processes under normal conditions and in the presence of pathology, as well as their relationship to changes in the functional state of lymphocytes and, in particular, to the development of immunodeficiency.

The cellular and humoral immunity was studied in 32 children with primary rheumocarditis and 34 children with nonrheumatic carditis. It is shown that there are no distinct diagnostic criteria of rheumatism form with a paucity of symptoms. This gave an impetus to develop a new method of diagnosis of the rheumatic process by means of preincubation of blood lymphocytes of the patient with a standard streptococcal allergen (dose: 0.1 ml, 30-40 mcg of protein) within 30 min. with subsequent realization of E-rosette formation reaction. Of diagnostic value was an increase of the relative content of E-rosette formation by 10% as compared with the control. The positive effect of this method as compared with determination of the antistreptolysin titer--79%. This allows to recommend wide introduction of the method in practical medicine.

RESUMO

Analysis of some parameters of cell-mediated immunity (lymphocyte blastogenesis test--LBGT, T-rosette formation, B-rosette formation, leukocyte migration inhibition test--LMIT) in 106 patients with odontogenic phlegmons has lead the authors to a conclusion on the direct relationship between LBGT and LMIT depression and the disease severity. The detected relationship will be helpful in early diagnosis of grave forms of odontogenic phlegmons and conductive to timely start of immunocorrective therapy.

Experimental staphylococcal infection was reproduced in rats by the intraperitoneal injection of S. aureus strain 75. The degree of the development of delayed hypersensitivity (DH) to staphylococci, microviscosity, the levels of free radical oxidation and antioxidation resistance were evaluated in the dynamics of the infectious process by the methods of chemiluminometry and fluorescent probing with pyrene. The functional activity of lymphocytes was determined by the inclusion of 3H-thymidine into DNA as the consequence of stimulation with phytohemagglutinin. The development of DH was found to depend on the microviscosity and antioxidation resistance of membrane lipids. The increase of microviscosity and the simultaneous decrease of the induction time of chemifluorescent rapid flash inhibit the development of DH, leading to the aggravation of the infectious process. The increase of fluidity and the accumulation of antioxidants facilitate the development of DH and lead to a milder course of the infectious process.

It was established during the study of blood serum and urine chemiluminescence in 57 children with pyelonephritis and in 38 children with glomerulonephritis that in the active disease stage, the intensity of overfaint luminescence rises as a result of lipid peroxidation (LPO) activation, accumulation of lipid hydroperoxides and oxygen-containing radicals. Four types of the kinetic curves of urine chemiluminescence were identified. They characterize the correlation between LPO activation and the level of antiradical defence in patients suffering from pyelo- and glomerulonephritis. The measurement of urine chemiluminescence in patients afflicted with pyelonephritis and glomerulonephritis may be one of the criteria for administering drugs possessing antiradical and antioxidant activity and can also be used in the control over the treatment efficacy.
