Background: Ascariasis is a common disease in humans and pigs. In previous reports, it has been known that ascariasis in humans is not only caused by Ascaris lumbricoides but also by Ascaris suum which is known to originate from pigs. Aims and Objectives: This research aims to identify the species of Ascaris that infect traditional pig farmers in Bali, Indonesia. Materials and Methods: To achieve this, an observational study was conducted, in which 239 feces samples were collected from traditional pig farmers. The collected samples were then divided into two parts, with one part stored in a pot containing 10% formalin for microscopic testing, and the other part stored in 70% ethanol for molecular testing. In cases where Ascaris infection was observed during the microscopic examination, a further test was conducted using the polymerase chain reaction (PCR) method for molecular examination. The DNA derived from the PCR was then subjected to DNA sequencing. Results: The result was compared to A. suum gene data previously published in the Gene Bank. The results of the microscopic examination showed that 22 farmers were infected with Ascaris. However, PCR testing and DNA sequencing indicated that three infections were caused by A. suum. Conclusion: Therefore, it can be concluded that A. suum is a zoonosis in Bali. Thus, efforts to control ascariasis must pay attention to the presence of pigs around it. Handling of ascariasis is not only carried out in humans but also in pigs and the environment.

BACKGROUND: The international Dog10K project aims to sequence and analyze several thousand canine genomes. Incorporating 20 × data from 1987 individuals, including 1611 dogs (321 breeds), 309 village dogs, 63 wolves, and four coyotes, we identify genomic variation across the canid family, setting the stage for detailed studies of domestication, behavior, morphology, disease susceptibility, and genome architecture and function. RESULTS: We report the analysis of > 48 M single-nucleotide, indel, and structural variants spanning the autosomes, X chromosome, and mitochondria. We discover more than 75% of variation for 239 sampled breeds. Allele sharing analysis indicates that 94.9% of breeds form monophyletic clusters and 25 major clades. German Shepherd Dogs and related breeds show the highest allele sharing with independent breeds from multiple clades. On average, each breed dog differs from the UU_Cfam_GSD_1.0 reference at 26,960 deletions and 14,034 insertions greater than 50 bp, with wolves having 14% more variants. Discovered variants include retrogene insertions from 926 parent genes. To aid functional prioritization, single-nucleotide variants were annotated with SnpEff and Zoonomia phyloP constraint scores. Constrained positions were negatively correlated with allele frequency. Finally, the utility of the Dog10K data as an imputation reference panel is assessed, generating high-confidence calls across varied genotyping platform densities including for breeds not included in the Dog10K collection. CONCLUSIONS: We have developed a dense dataset of 1987 sequenced canids that reveals patterns of allele sharing, identifies likely functional variants, informs breed structure, and enables accurate imputation. Dog10K data are publicly available.

Background and Aims: African swine fever (ASF) is a highly contagious viral disease that causes major economic losses due to morbidity and fatality rates of up to 100% in wild boar and domestic pigs. The disease emerged in Africa in 1921 and then entered several European countries by 1957. In Indonesia, the first outbreak of ASF in 2019 in North Sumatra killed thousands of pigs and quickly spread to 10 out of 34 pig-producing provinces, including Bali and Eastern Nusa Tenggara. As no commercial ASF vaccine is available, the disease has become endemic and continues killing pigs. This study aimed to investigate the epidemiological and virological studies of ASF virus (ASFV) conducted in 2020 and 2021 by the Disease Investigation Center Regional VI of Denpasar Bali, which covers three provinces in Indonesia, including Bali, Western Nusa Tenggara, and Eastern Nusa Tenggara. Materials and Methods: A total of 5402 blood samples were sent to the laboratory to detect ASFV infection using quantitative polymerase chain reaction (qPCR) and enzyme-linked immunosorbent assay tests. The virological studies were performed by culturing local ASFV isolates obtained from field cases in primary macrophages and confirmation of viral growth by qPCR. Results: The qPCR results show that only 156/4528 (3.4%) of samples originating from Bali and Eastern Nusa Tenggara were ASFV-positive with cycle threshold value of 18 to 23, while the virus was not detected in Western Nusa Tenggara. Of 874 serum samples tested, 114 (13%) were antibody positive and were all collected from the two ASFV-affected provinces in 2020. A Bali ASFV isolate (BL21) was isolated and characterized molecularly. Conclusion: These findings suggest that during the time of sampling, ASFV was detected only in Bali and East Nusa Tenggara but not in Western Nusa Tenggara. These findings support the symptomology of ASFV reported in the two regions. Moreover, BL21 may be useful for developing subculture-attenuated vaccines using commercial cell lines. However, the current study has some limitations namely the investigation was not performed during the initial outbreak and no pathological examination of internal organs was conducted.

Background and Aim: Rabies has been endemic in Bali since 2009, and cases has recently increased. Unfortunately, there is a lack of available vaccines, which hinders the eradication program. This study aimed to investigate the epidemiological and virological aspects of rabies infection in Bali. Materials and Methods: A total of 24 brain samples were collected from rabid dogs in all districts of Bali. The samples were tested using the direct fluorescent antibody (DFA) test and polymerase chain reaction (PCR) to confirm the presence of rabies virus in the samples. Samples with the highest virus content were propagated in vivo and then inoculated into BALB/c mice. The brains of dead mice were used to prepare an inoculate cultured in murine neuroblastoma cells. Supernatant-positive viruses representing each district were then reinoculated into eight groups of five BALB/c mice. A brain sample from each dead mouse was tested using DFA and PCR and detected under a fluorescence microscope. Results: All rabies virus-positive samples collected from rabid dogs in all districts of Bali were positive. Rabies virus was detected by DFA test and PCR and was consistently confirmed in the in vivo and in vitro studies. BALB/c mice inoculated with the highest viral dilution (105 cells/mL) of culture supernatant showed typical signs of rabies, indicating that the virus could be properly investigated. Conclusion: This study demonstrated a wide epidemiological distribution of rabies in Bali. The obtained virus can be adapted for in vitro and in vivo studies and can be used to develop a homologous vaccine.

Background and Aim: Rabies remains a public health concern in Indonesia, and the coronavirus disease (COVID-19) pandemic has stymied rabies prevention and control efforts. There is a need to transform the rabies program to be adaptable to pandemic situations to improve program coverage on dog vaccination and rabies surveillance. This study aimed to create a rabies control (RaCon) mobile application for a community-based rabies surveillance system during COVID-19 in Bali, Indonesia. Materials and Methods: We employ the Design Science Research methodology. Surveillance officers, veterinarians, community leaders, outreach workers, and dog owners participated in a series of offline in-depth interviews and focus group discussions. The RaCon prototype was evaluated using the Post-Study System Usability Questionnaire (PSSUQ) framework, which included the system's usefulness, information quality, and interface quality. In this study, we used both a qualitative (n=50) and quantitative (n=342) approach. Results: According to the findings of this study, integrating public health and animal health into the rabies surveillance system are critical to supporting the One Health approach and encouraging community engagement in rabies programs. The RaCon prototype is expected to include features such as pet ownership, case report, news and announcements, nearest vet, health information, outbreak radar, emergency call, and app feedback. The RaCon prototype passed both qualitative and quantitative evaluations, indicating that it could be used to support the rabies surveillance system, particularly in the COVID-19 situation. Conclusion: The RaCon prototype was accepted by the users and got positive feedback in terms of the system's usefulness, information quality, and interface quality dimension. As a result, this prototype has the potential to be integrated into the rabies surveillance system in Bali, particularly to strengthen the community-based rabies surveillance system. Even though this prototype received positive feedback, this study focuses solely on the design development and evaluation of its user interface. As a result, further development is required before incorporating RaCon into the rabies prevention and control program.

Background and Aim: Pigs are the main livestock commodity in Bali Province, Indonesia, where traditional farming practices are widely used. Traditional pig farmers are often closely associated with poverty and a perceived lack of knowledge regarding health and hygiene. Data on soil-transmitted helminthiasis (STH) and risk factors associated with STH worm infection among traditional pig farmers in Bali were previously unavailable. This study aimed to analyze the prevalence and risk factors for STH infections among traditional pig farmers in Bali Province, Indonesia. Materials and Methods: This study involved the fecal examination of 238 traditional pig farmers from all areas of Bali Province in Indonesia. In addition, several pig feces samples were combined into one pooled sample belonging to each farm. All fresh fecal samples were stored in a 5% formaldehyde solution before being analyzed using concentration flotation techniques. Subsequently, risk factors were determined through an interview and a questionnaire. The odds ratio (OR) and Chi-square tests were used to determine the risk factors associated with STH infections. Results: The result showed that there was a high prevalence of STH infections among traditional pig farmers and pig farms in Bali, with rates of 21.8% and 76.5%, respectively. This could be due to risk factors, such as personal hygiene (OR: 5.756; confidence interval [CI]: 2.96-11.193; p=0.00), sanitation (OR: 1.914; CI: 1.024-3.576; p=0.042), education level (OR: 7.579; CI: 2.621-21.915; p=0.00), household income (OR: 2.447; CI: 1.122-5.338; p=0.025), and occupation (OR: 2.95; CI: 1.356-6.415; p=0.006). Conclusion: The infections seen in farmers were distributed among hookworm, Ascaris spp., and Trichuris spp., at 15.1%, 9.2%, and 4.2%, respectively. The risk factors associated with infections of STH and Ascaris spp. were personal hygiene, home sanitation, education level, household income, and having a primary occupation as a traditional pig farmer. In contrast, personal hygiene, education level, and primary occupation were the only risk factors for hookworm infection, while personal hygiene and home sanitation were the risk factors associated with Trichuris spp. infection. The limitation of this study was that the number of samples was relatively small due to the difficulty of obtaining stool samples from traditional pig farmers, with many individuals refusing to provide their stool for inspection. We suggest that future research focus on identifying the species of worms that infect traditional pig farmers and to better identify the zoonotic link of STH transmission from pigs to humans.

BACKGROUND AND AIM: Several free-roaming dogs can be easily found in the public areas of Bali. They go out in search of foods and friends and defecate everywhere. In general, these groups of dogs do not receive good healthcare from their owners and are generally threatened by some disease-causing organisms, especially helminths. This study was conducted to identify and measure the prevalence of soil-transmitted helminths (STHs) that cause infection in free-roaming dogs in Bali Province, Indonesia. MATERIALS AND METHODS: A total of 1611 fresh dog fecal samples were collected from all areas of Bali Province and subjected to qualitative fecal examination using flotation techniques to obtain STH eggs. RESULTS: The incidence of STHs in free-roaming dogs was 38.36%. However, three types of STHs were identified, including Ancylostoma spp., Ascaris spp., and Trichuris spp. Ancylostoma spp. had the highest prevalence of 37.8%, followed by Ascaris spp. and Trichuris spp. at 6.02% and 0.87%, respectively. Multiple infections of these worms were also recorded. The polyparasitism prevalence of Ancylostoma spp. and Ascaris spp. was 3.85%, followed by that of Ancylostoma spp. and Trichuris spp. at 0.5% and that of Ascaris spp. and Trichuris spp. at 0.06%. There were no altitude-wise differences in the prevalence of STH infection. CONCLUSION: The prevalence of STHs was high in free-roaming dogs. This finding necessitates more serious attention as it affects both animal and public health.

BACKGROUND AND AIM: Helicobacter pylori is a well-known zoonotic agent with worldwide distribution. In Indonesia, only one report regarding the variation within the cytotoxic associated antigen A (CagA) protein of H. pylori has been described in the literature, which was conducted in Manado, South Sulawesi. There remains no report concerning the structure of this protein, particularly for the Bali and Lombok isolates. The objective of this study was to investigate the diversity of H. pylori CagA amino acid sequences of Bali and Lombok isolates, to predict their molecular structures and conduct toxicity examination of CagA on gastric cells. MATERIALS AND METHODS: A total of 36 samples were used in equal proportions for each pathologic condition. DNA extraction was performed to subculture H. pylori Bali isolates. The amplification of the CagA 3' variable region was carried out using the primers P1 (5'-GATAACAGGCAAGCTTTTTGAGG-3') and P2 (5'-CTGCAAAAGATTGTTTGGCAG-3'). The W2, W9, and W35 fragments were selected as a representation of H. pylori Bali isolates, which were modeled through the threading modeling approach using I-TASSER. RESULTS: According to the 12 CagA sequences obtained and phylogenetic analyses, the H. pylori strain originating from Bali can be grouped within the East Asian genotypes and is identical to the Lombok strain. In addition, the Bali isolates are phylogenetically more closely related to Southeast Asian strains, particularly the Filipino strain. The relationship between degree of inflammation induced and CagA-positive infection was not statistically significant. CONCLUSION: The structure of the H. pylori Bali isolate is identical to that of Lombok isolate, which belongs to the same group of East Asian genotypes, and bacterial virulence is not related to structure.

BACKGROUND AND AIM: Taenia saginata hazardously affects human and animal health. The distribution of this disease is found almost all over the world. The study aimed to obtain epidemiological information concerning prevalence and the distribution of bovine cysticercosis in Bali and Nusa Tenggara, Indonesia. MATERIALS AND METHODS: A total of 267 community-owned Bali cattle serum samples from the provinces of Bali, West Nusa Tenggara, and East Nusa Tenggara were examined. The study was conducted by examining the serum of Bali cattle using enzyme-linked immunosorbent assay technique. Risk factors related to cysticercosis that analyzed were sex, breeding type, age, physical condition, source of drinking water, pen condition, and latrine availability. RESULTS: Seven of 91 Bali cattle sera from all regencies/cities in Bali showed a positive result. Those positive sera were originated from Buleleng (1), Gianyar (2), Denpasar (2), and Klungkung (2). Meanwhile, four of 92 Bali cattle sera from West Nusa Tenggara and seven of 84 from East Nusa Tenggara occurred antibodies against T. saginata. We identified that two risk factors that influence the incidence of T. saginata infection in Bali cattle in Bali were the sex and the cattle breeding type. CONCLUSION: Through this research can be made a map of bovine cysticercosis in Bali cattle in Bali and Nusa Tenggara region. By mapping the disease, it is recommended that the animal health officers should be more accurate when conducting postmortem examination, especially on cattle from a positive region.

OBJECTIVE: Streptococcus suis (S. suis) is a causative agent for various syndromes in pigs. It can be transmitted to humans with typical symptoms of meningitis and death. Although human infections have been confirmed at Bali Referral Hospital, Indonesia, since 2014, the bacteria have not been isolated from pigs. Here, we provide confirmation of the presence of the bacteria in sick pigs in the province. RESULTS: Streptococcus suis was confirmed in 8 of 30 cases. The final confirmation was made using PCR and sequencing of the glutamate dehydrogenase (GDH) and recombination/repair protein (recN) gene fragments. Upon PCR serotyping, two were confirmed to be serotype 2 or 1/2. Prominent histopathological lesions of confirmed cases were meningitis, endocarditis, pericarditis, bronchopneumonia, enteritis and glomerulonephritis. The dominant inflammatory cells were neutrophils and macrophages. Further research is needed to understand the risk factors for human infection. Community awareness on the risk of contracting S. suis and vaccine development are needed to prevent human infections.

AIMS: The purposes of this study were to determine the anticancer activity of Xestospongia testudinaria sponge isolate and identify the responsible compounds. MATERIALS AND METHODS: The metabolites were extracted using methanol maceration at room temperature. The separation and purification of metabolites were performed using fractionation and column chromatography. The toxicity was examined using the brine shrimp lethality assay, and the toxic isolates were tested for anticancer activity against HeLa cells. Gas chromatography-mass spectrometry analysis was used to identify the compounds in the isolate. RESULTS: When the methanol extract was partitioned with n-hexane, chloroform, and n-butanol, the chloroform fraction was the most toxic, with a concentration that results in 50% lethality (LC50) value of 39.81 ppm. After separation of the chloroform extract, fraction B (FB) was the most toxic, with an LC50 value of 44.67 ppm. The isolate from FB showed anticancer activity with a concentration at which 50% of growth was inhibited (IC50) of 2.273 ppm. In total, 21 compounds were identified in anticancer isolates: Nonanedioic acid; tetradecanoic acid; trans-phytol; 2-pentadecanone-6,10,14-trimethyl; pentadecanoic acid; 2-hexadecen-1-ol, 3,7,11,15-tetramethyl-; pentadecanoic acid; 2-hexadecen-1-ol, 3,7,11,15-tetramethyl-; 2,3,7-trimethyloctanal; palmitic acid; docosanoic acid, ethyl ester; 1,E-11,Z-13-octadecatriene; chloromethyl 4-chlorododecanoate; 1-tricosene; 9,12-octadecadienoic acid; 4,8,12,16-tetramethylheptadecan-4-olide; 1-docosene; heneicosane; phosphonic acid, dioctadecyl ester; dodecane,4,6-dimethyl-; n-tetratriacontane; 1-iodohexadecane; and n-heneicosane. CONCLUSION: These findings indicate that the isolate of X. testudinaria can be used as a natural anticancer toward HeLa cell.

BACKGROUND AND AIM: Helicobacter pylori infection has been identified as a major cause of peptic ulcer diseases, including gastric and duodenal ulcers, gastritis, chronic and gastric carcinoma, and even gastric lymphoma. In vitro studies using Western blotting analysis, hemagglutination test, adherence inhibition assays, and immunocytochemical staining revealed that the 49.6-kDa subunit pili protein of H. pylori was considered an immunogenic protein. This study aimed to develop a serological diagnostic test using 49.6 kDa for detecting antibodies against H. pylori proteins in an early phase of the infection. MATERIALS AND METHODS: An in-house immunochromatographic test (ICT) kit was developed and used to test a panel of sera sample obtained from a randomly selected symptomatic patient, in which 40 sera were H. pylori positive and 40 sera were H. pylori negative. RESULTS: The results showed that ICT with 49.6 kDa as an antigen was highly sensitive and specific for detecting anti-H. pylori immunoglobulin G antibodies in human serum, with a high negative predictive value. CONCLUSION: The developed test could be used to exclude H. pylori infection in symptomatic patients.

The Indonesian island province of Bali experienced its first rabies incursion in 2008. Mass vaccination of the dog population has proven effective and rabies cases in dogs and people have decreased, however the virus is still circulating among the dog population. Vaccination coverage must be maintained until rabies elimination. Increasing efficiency and effectiveness of vaccination campaigns is therefore desired. Community engagement leading to preventative health actions by community members can reduce disease incidence and costs of control. Here we evaluate 2 years of a novel community-based dog welfare and rabies control project (Program Dharma) in the Sanur sub-district. The project engaged the services of people living in the project area with an interest or experience in dogs or community health services. These people spoke with owners within their own community about dog welfare and health, monitored owned and unowned dogs and increased owner and carer efforts to access vaccination and further veterinary services. The evaluation focused on a sample of dogs whose owners had been regularly engaged with project. Vaccination coverage was increased and there were no dog or human rabies cases reported in the project area; the percentage of the dogs that had never been vaccinated was reduced by an average 28.3% (baseline unvaccinated 41-49%, post-project unvaccinated 11-19%). The welfare of dogs improved from an average of 20.7% of dogs with visible welfare problems at baseline to 2.7% after project implementation. Roaming dog density observed on street surveys also decreased in all project areas (24-47% reduction dependent on desa). A participatory evaluation event with a sample of Program Dharma community-based agents highlighted several additional successes, including that the community appeared to welcome and value their services and were beginning to support the cost of project activities. Conversely, challenges included identifying dogs in the database during revisits, sustaining the costs of community member time spent working on Program Dharma activities and the costs of veterinary care, whilst avoiding dependency of owners on free veterinary services. The benefits revealed by the evaluation were judged to be sufficient to extend Program Dharma to new areas, whilst evolving activities to resolve challenges.

AIM: The study was aimed to prepare and examine the potential and safety concerns of egg drop syndrome (EDS) vaccine candidate seed. The potential and safety trials of EDS Medan isolate vaccine need to be done before commercial scale of EDS vaccines are made. MATERIALS AND METHODS: The safety test of EDS candidate vaccine was tested on 4-week-old specified pathogen-free chickens in an experimentally isolated enclosure. RESULTS: The result of the safety test obtained 27.3 hemagglutination inhibition (HI) unit of geometric mean titer antibody post-vaccination. However, the potency test of the EDS candidate vaccine was conducted on 17-week-old laying hens. Test results of the EDS potency vaccine in layer obtained antibody titer increased in every week of blood taking with average titer of antibody: Before vaccinated was 22.9 HI unit, 1 week after vaccination was 23.7 HI unit, 2 weeks post-vaccination was 25 HI unit, and 3 weeks after vaccination was 27.3 HI units. In contrast, decreasing trend was observed in control group (unvaccinated chicken). CONCLUSION: Serologically, the seed vaccine of EDS virus isolates from Medan was able to produce protective antibody titers starting in the 2nd and 3rd weeks post-vaccination.

AIM: The aim of this research was to identify the seroprevalence of Neospora caninum in local Bali dogs. MATERIALS AND METHODS: A total of 147 local Bali dog serum samples have been examined for antibodies of N. caninum using enzyme-linked immunosorbent assay test. RESULTS: The results confirmed that 5 (3.4%) local Bali dogs have antibody for N. caninum. There were no significant differences in seroprevalence of Neospora infection in local Bali dogs between different genders, ages, and take care methods (p>0.05). CONCLUSION: The results provided evidence for the presence of N. caninum infection in local Bali dogs and thus the risk to Bali cattle and dog health.

This study gained an understanding of dog demographics, owner behaviour, and knowledge, attitudes and practices relating to rabies in three villages in Sanur, Bali, providing insights for an intervention to improve responsible dog ownership. A combination of a census of all dogs, street surveys of roaming dogs, and a Knowledge, Attitude and Practices (KAP) survey was used to study both dogs and people. A total of 6009 dogs were recorded, of which only 3.3% appeared to be unowned; unowned dogs had poorer welfare and were more likely to be wary of attempted approaches by people. The source of dogs, method of confinement used by owners, and whether dogs were sterilised differed between three breed types; purebred dogs, mixed breed, and Bali dogs (native breed). Three variables were found to have significant effects on the chances of not being vaccinated: age, dog type, and confinement. A mean of 3.81 roaming dogs per km of street surveyed was recorded along 28 sub-village routes. Responses to attitude statements showed that most people had a positive attitude towards dogs and vaccination and did not agree with culling. Knowledge of appropriate bite treatment and symptoms of rabies in dogs was good.

AIM: This study was designed to validate the effectiveness of the pig confinement system (PCS) in reducing the prevalence of zoonotic and internal parasite burdens in pigs. MATERIALS AND METHODS: Ten PCS households were selected together with 10 households practising traditional scavenging systems. Five pigs were monitored per household every 3 months for 15 months and blood and feces collected. Pigs received a single dose of oxfendazole at 30 mg/kg at baseline. Qualitative fecal examinations for intestinal parasite stages were performed, and serum was tested for antibodies to cysticercus of Taenia solium, Trichinella spp., and Toxoplasma gondii. RESULTS: Based on fecal examination, the prevalence of pigs positive for parasite eggs was reduced in PCS pigs over consecutive samplings (Ascaris suum [14.3% to 0%], Trichuris suis [46.9% to 8.3%], Strongyle-type eggs [81.6% to 8.3%], Physocephalus spp. [6.1% to 0%], and Metastrongylus apri [20.8% to 0%]) compared with increases in the number of pigs positive for parasite eggs in non-PCS pigs (T. suis [20-61.5%], Strongyle-type [60.4-80.8%], Physocephalus spp. [8.3-15.4%], and M. apri [20.8-34.6%]) and little change in pigs positive for A. suum (18.8-19.2%). While the prevalence of pigs with antibodies against to cysticerci of T. solium reduced in PCS pigs from 18% to 14%, the prevalence in non-PCS pigs increased from 42% to 52%. Antibodies to Trichinella were not detected, but the prevalence of T. gondii antibodies increased from 6% to 10% in PCS pigs and from 7% to 24% in non-PCS pigs. CONCLUSION: These data demonstrate the potential of a PCS to reduce the prevalence of pigs infected with zoonotic and internal parasites and thus the risk to human and pig health.
