RESUMO

This study endeavoured to capitalize on prickly pear by-products for the optimization of molasses formulation, targeting elevated antioxidant capacities and superior sugar contents. Through robust statistical modelling, the optimal cooking parameters-temperature (70-80 °C) and duration (60-90 min)-were determined, guided by responses of antioxidant activity and Brix value. A D-Optimal mixture design further delineated the ideal proportions of molasses components (pulp, peel, and seeds). Characterization revealed that peel harboured the highest concentrations of total polyphenols (396.41 mg GAE/100g FW) and flavonoids (234.26 mg CE/100g FW), emphasizing its antioxidant potential (DPPH inhibition IC50: 12.72 µg/ml). The optimal cooking conditions were established at 78.35 °C for 79.70 min, with predictive equations guiding ingredient proportions (0.265 g pulp, 0.710 g peel, 0.025 g seed). Intriguingly, while peel inclusion enhanced total sugar content and antioxidant activity, seed incorporation exerted a contrasting effect by reducing total sugar content and limiting antioxidant activity.

The ethyl acetate, ethanol, methanol, and water extracts of cypress from three Tunisian regions (Bizerte, Ben Arous, and Nabeul) were reported for their phenolic content, antioxidant, and antibacterial activities. Cypress extract had an important antiradical and antibacterial potential which was strongly related to the organoleptic quality of the extract which appeared strongly region dependent. The highest contents of polyphenols were obtained in the methanol extract at the region of Bizerte (315.75 mg GAE/g DW). Cupressuflavone was the major compound with high proportions in methanol extract of Bizerte (68%). Amentoflavone also constituted an important flavonoid compound presented in higher proportion in the ethyl acetate extract of Ben Arous (21.2%). For Gram-positive bacteria strains, Staphylococcus aureus was sensitive to the ethanol extract of Nabeul with IZ = 40 mm. This study suggests cypress as potential natural source of antioxidants and antibacterial agents.

The aerial parts of cypress (Cupressus sempervirens L.) of three collect regions (Bizerte, Ben-Arous and Nabeul) were reported for their essential oil (EO) compositions, antioxidant, antimicrobial and insecticidal activities. Results showed that the higher EO yields were observed in Bizerte and Ben Arous (0.56%), followed by Nabeul (0.49%). The EO composition showed the predominance of α-pinene with 36.72% in Bizerte, 30.22% in Nabeul and 30% in Ben-Arous. Cypress EO of Bizerte showed higher antiradical capacity (IC50 = 55 µg/mL) than Ben-Arous (IC50 = 97.50 µg/mL) and Nabeul (IC50 = 155 µg/mL). E. faecalis was the most sensitive strain to cypress EO of Bizerte with the largest inhibition zone (IZ = 65 mm). Regarding the insecticidal activity, cypress EO of Bizerte had the highest mortality of Tribolium castaneum with a lethal concentration of LC50 = 164.3 µL/L air after 24 h exposure.

This study examined the antioxidant, anticancer and antiviral properties of the methanolic extracts from bigarade (Citrus aurantium L.) leaves at two development stages. Ferulic acid, naringin and naringenin were the principal phenolic components of young and old leaves. The highest total antioxidant capacity was obtained in young leaf extracts (YLE). These latter also exhibited the highest antiradical DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) activities, while the highest iron chelating and reducing power activities were observed in old leaf extracts (OLE). The potent anticancer activity was observed in YLE for human lung carcinoma (A-549) and in OLE for colon adenocarcinoma (DLD-1) cells. YLE showed the highest virucidal effects as compared to OLE and the positive control acyclovir against herpes simplex virus type-1 (HSV-1) propagation in Vero cells during the absorption and replication periods. The young and old leaves might be a source of natural antioxidants and protective agents against oxidative damage.

The present study aimed to investigate the qualitative and quantitative performance of five green solvents, namely 2-methyltetrahydrofuran (MeTHF), cyclopentyl methyl ether (CPME), p-cymene, d-limonene and ethanol to substitute n-hexane, for sesame seed oil extraction. In fact, both CPME and MeTHF gave higher crude yields than n-hexane (58.82, 54.91 and 50.84%, respectively). The fatty acid profile of the sesame seed oils remained constant across all the solvent systems, with a predominance of oleic acid (39.27-44.35%) and linoleic acid (38.88-43.99%). The total sterols gained the upmost amount with CPME (785 mg/100 g oil) and MeTHF (641 mg/100 g oil). CPME and MeTHF were also characterized by the optimum content of tocopherols (52.3 and 50.6 mg/100 g oil, respectively). The highest contents of total phenols in the sesame seed oils were extracted by CPME (23.51 mg GAE/g) and MeTHF (22.53 mg GAE/g) as compared to the other solvents, especially n-hexane (8 mg GAE/g). Additionally, sesame seed oils extracted by MeTHF and CPME also had the highest antioxidant and anti-inflammatory properties as compared to the other green solvents and n-hexane, encouraging their manufacturing use for sesame seed oil extraction.

The antifungal effects of laurel, myrtle and peppermint essential oils and their combinations were investigated in vitro on two strains of Alternaria alternata mycelial growth and in vivo on detached Citrus leaf disease incidence. Myrtle essential oil was rich in α-pinene and 1,8-cineole while peppermint essential oil in menthol and menthone. 1,8-Cineole was the main component of the essential oils from laurel, laurel + myrtle and peppermint + laurel. The combined peppermint and myrtle essential oil was characterized by the predominance of menthol and 1,8-cineole. All tested essential oils, incorporated in potato-dextrose agar, inhibited A. alternata mycelial growth and had a fungistatic effect at concentration 3 mg/mL of medium. A great synergism was detected between peppermint and laurel essential oils against the two strains of A. alternata. The combined laurel and peppermint essential oil reduced mycelial growth rates of inoculated detached leaves at concentration above 1.5 mg/mL.

Lycium europaeum is used as a medicinal herb in many countries. In this study, cyclohexane, dichloromethane, ethyl acetate, methanol and water were used as solvents in the extraction of L. europaeum fruits. The contents of total phenolics, total flavonoids, total tannins and condensed tannins as well as the biological activities of these extracts were investigated using various in vitro and ex vivo assays. Results showed that all solvent extracts of L. europaeum had no anticancer activity against cancerous (A-549 and DLD-1) and non-cancerous (WS-1) human cells. Methanol and ethyl acetate were the most effective solvent for extraction of phenolic compounds and also exhibited the highest antioxidant and anti-inflammatory activities. The methanol extract of L. europaeum fruits was the richest in phenolic compounds with the predominance of ferulic acid, catechin and narengin. These results supported the use of L. europaeum fruit as natural source of bioactive compound for pharmaceutical applications.

The organic solvent extracts (hexane, chloroform, acetone, methanol and water) of coriander fruits were reported for their antioxidant and repellent activities. The methanol and acetone extracts revealed the highest antiradical activity as compared to the other solvent extracts. HPLC analysis revealed that phenolic acids were the major compounds in chloroform and acetone extracts, whereas flavonoids in methanol and water extracts. The methanol extract of coriander fruits exhibited the highest repellent effect against coleopteran insects. Additionally, the hexane extract of coriander fruits also exhibited potent repellent and toxic activities against rice weevil. It appeared that coriander fruit extract obtained with the apolar solvent had more repellent potential against Sitophilus oryzae as compared to Tribolium castaneum. For Lasioderma serricorne, the methanol extract of coriander fruits reduced in a very significant way the adults followed by the chloroform and hexane extracts.

The methanolic extracts of cypress (Cupressus sempervirens L.) collected at three phenological stages were evaluated for their cytotoxicity on Vero cells by MTT test as well as on Herpes simplex (HSV-2) and coxsackie (CVB-3) viruses by plaque reduction assay. The methanolic extract exhibited the highest cytotoxicity against HSV-2 (IC50 = 20.40 µg/mL) and CVB-3 (IC50 = 47.50 µg/mL) at the flowering stage. This extract also exhibited a virucidal action both during the entry of viruses and the release of newly formed virions. The methanolic extract bioguided purification showed that the ethyl-acetate fraction was responsible for virucidal activity. This fraction was endowed with more important selectivity index of 8.15 for HSV-2 and 4.40 for CVB-3. The ethyl acetate fraction was subjected to thin layer chromatography fractionation and identification by HPLC-DAD-ESI-MSn. Results showed that the condensed tannin procyanidin B2 was identified for the first time responsible of the antiviral activity of cypress.

This research aimed to study the chemical composition of Aloysia citrodora methanolic extract and its biological activities as an antioxidant, and its antibacterial, antifungal and anti-inflammatory activities based on four bioclimatic collection stages. The contents of total phenols, total flavonoids and total tannins were determined. Nine phenolic compounds were identified by LC-DAD-ESI-MS/MS. The major compound was acteoside, a phenylpropanoid which represented about 80% of the methanolic fraction in the various regions. The antioxidant activities of different locations were measured by different analytical assays, such as DPPH, ABTS and iron reducing power. The results showed that phenolic compounds and antioxidant activities varied with climatic and environmental factors. Moreover, there was a significant dependency between regions and biological activities. The use of a principal component analysis showed that there was a close relationship among phenylpropanoids, phenolic compounds and the studied biological activities.

The aim of this study was to optimize the mechanical and texture properties of edible film improving its antibacterial property after adding rosemary essential oil (REO) using a Doehlert matrix. Films with the highest mechanical properties were acquired using a polymer composition of 65.2% glycerol, 24.3% gelatin, 10.0% chitosan and 0.5% pectin. This composition provided the highest elongation at break, tensile strength and texture values, which were respectively 51.60 ± 6.04%, 8.53 ± 2.36 MPa and 13.67 ± 1.43. The antibacterial activity of REO enriched films against Bacillus subtilis, Staphylococcus aureus, Enterococcus aerogenes, Enterococcus faecalis and Escherichia coli was enhanced when applying a mixture of 1.995 and 1.250 mg/g of two REO extracted from two rosemary different varieties. The structural, optical and barrier properties of the films were evaluated. To conclude, the enriched film showed potential coatings for controlling most common food borne bacteria growth during the food storage.

This study was conducted to investigate potentially protective and curative effects of Curcuma longa root (turmeric) powder on CCl4-induced hepatotoxicity in rats. Turmeric was administered before (preventive effect) or after (curative effect) treatment with CCl4. Total phenolic and flavonoid levels were 26.35 mg GAE/g and 12.35 mg CE/g, respectively. Using HPLC analysis, turmeric powder was rich in curcumin (62.97%), demethoxycurcumin (20.86%) and bisdemethoxycurcumin (16.17%). Curcuma longa powder showed important in vitro antioxidant activities. Results showed that the activities of aspartate aminotransaminase and alanine aminotransaminase, and the levels of bilirubin and serum lipids were increased in CCl4-treated animals. However, total protein and albumin levels and antioxidant enzyme activities were decreased. Turmeric administration, before or after CCl4 treatment, significantly decreased the activities of marker enzymes and lipid levels in serum. Moreover, total protein and albumin contents were restored to nearly normal levels after turmeric administration accompanied with increase of antioxidant enzymes activities.

The purpose of the present investigation is to assess, for the first time, the antidiabetic, antihyperlipidemic and antioxidant activities of Lycium europaeum extract in alloxan-induced diabetic rats. Diabetes was induced in adult male Wistar rats via a single subcutaneous alloxan injection (120 mg/kg). Lycium europaeum aqueous extract was orally administered at a dose of 20 mg/kg for 28 consecutive days. Serum concentrations of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C) and triglycerides (TG) were assayed at the end of the experimental period in all investigated groups. Antioxidant enzymes such as glutathione peroxidase (GPx), catalase (CAT) and superoxide dismutase (SOD) were sought in the serum and pancreas. Lycium europaeum extract significantly increased HDL-C and reduced blood glucose, TC, LDL-C and TG as compared to the alloxan-control group. Lycium europaeum extract was also efficient in reducing oxidative stress in diabetic rats by increasing SOD, CAT and GPx activities both in the pancreas and the plasma of the animals. Moreover, Lycium europaeum extract contained considerable levels of polyphenols and flavonoids. It also exhibited an important antioxidant capacity and a remarkable ability to quench DPPH radicals and reduce irons. The obtained results highlight potentially relevant health beneficial effects of Lycium europaeum extract, reversing hyperglycemic, hyperlipidemic and oxidative stress effects in rats with alloxan-induced diabetes. Therefore, it may be considered as a promising alternative or complementary agent to diabetes treatment.

RESUMO

Cancer is a major public health problem in the world. The use of the medicinal plants in cancer prevention and management is frequent in Africa, especially in Tunisia, and it is transmitted from generation to generation within cultures. Many previous studies showed that a wide range of Tunisian medicinal plants exerted cytotoxic and anticancer activity. A comprehensive review was conducted to collect information from scientific journal articles, including indigenous knowledge researches, about Tunisian medicinal plants used for the prevention and management of cancer. The aim of this review article is to provide the reader with information concerning the importance of Tunisian medicinal plants in the prevention and management of cancer and to open the door for the health professionals and scientists working in the field of pharmacology and therapeutics to produce new drug formulations to treat different types of cancer.

The present investigation focuses on the methanolic extract obtained from Geranium robertianum L. (Geraniaceae) (Herb Robert), a herbal plant used in traditional medicine for the treatment of human and animal diseases. The antioxidant capacities of the extract were evaluated using 1,1-diphenyl-2-picrylhydrazyl radical, ß-carotene/linoleic acid and reducing power and metal chelating activity assays. The amount of total phenolic content, flavonoids and condensed tannins was very high, and the correlation between the antioxidant activity potential and total phenolic level of the extract was pointed out.

This study was designed to examine the chemical composition and antioxidant activity of the essential oils and methanol extracts of Myrtus communis var. italica L. leaf, stem and flower. Myrtle leaf and flower were the valuable organs for the essential oil production representing a yield of 0.61% and 0.30% (w/w), respectively. The essential oil composition of myrtle leaf and flower was characterized by high proportions of alpha-pinene, the main compound of monoterpene hydrocarbon class, with 58.05% for leaf and 17.53% for flower. Stem was rich in oxygenated monoterpenes, largely due to 1,8-cineole with 32.84%. The total phenol contents varied between different myrtle parts; leaf extract had higher total phenol content (33.67 mg GAE/g) than flower (15.70 mg GAE/g) and stem (11.11 mg GAE/g) extracts. Significant differences were also found in total tannin contents among different myrtle parts, representing 26.55 mg GAE/g in leaf, 11.95 mg GAE/g in flower, 3.33 mg GAE/g in stem. The highest contents of total flavonoids and condensed tannins were observed in stem (5.17 and 1.99 mg CE/g, respectively) and leaf (3 and 1.22 mg CE/g, respectively) extracts. The HPLC analysis indicated that the main phenolic class was hydrolysable tannins (gallotannins) in leaf (79.39%, 8.90 mg/g) and flower (60.00%, 3.50mg/g) while the stem was characterized by the predominance of flavonoid class (61.38%, 1.86 mg/g) due to the high presence of catechin (36.91%, 1.12 mg/g). Antioxidant activities of the essential oil and the methanolic extract from different myrtle parts were evaluated by using DPPH radical scavenging, beta-carotene-linoleic acid bleaching, reducing power and metal chelating activity assays. In all tests, methanolic extracts of different myrtle parts showed better antioxidant activity than essential oils.

Borago officinalis rosette leaves were sampled in the region of Amdoun (Tunisia) during different stages of their development. Essential oil contents varied from 0.01% to 0.13% respectively in young and adult leaves. Twenty three volatile compounds were identified. Hydrocarbons, mainly represented by nonadecane (29.8%), tetracosane (11.3%) and heptacosane (4.7%), constituted the major class in the young leaves (45.8%), followed by aldehydes (22.4%). The percentages of these two classes decreased to reach respectively 15% and 8.1% in adult leaves in favour of alcohols (57.9%) where cis-3-hexenol (29.6%) and hexanol (14.5%) were the main compounds. Total fatty acids amounts increased from 5.03 mg/g DW in young leaves to 32.23 mg/g DW in adult ones. The predominant fatty acids were alpha-linolenic (C18:3 n-3), stearidonic (C18:4 n-3), gamma-linolenic (C18:3 n-6), palmitic (C16: 0) and linoleic (C18:2 n-6) acids.

Two Myrtus communis varieties (var. italica and baetica) were studied in order to investigate their essential oil yield and composition. Essential oil yield varied in leaves, fruits and stems. So, in leaves, it was 0.5% for italica and 0.3% for baetica and was higher than in fruits and stems with respectively 0.1% and 0.04% for italica and 0.07% and 0.03% for baetica. The essential oil analysis performed by GC and GC/MS showed a composition characterized by a high percentage of monoterpene hydrocarbons in leaves, largely due to alpha-pinene with 51.3% for italica and 27.7% for baetica; 1,8-cineole, the alone compound of ether class, was predominant in fruits and stems with respectively 31.6% and 34.7% for italica and 19.8% and 25.8% for baetica.
