RESUMO

The utilization of novel compounds as cancer treatments offers enormous potential in this field. The advantages of nanomedicine-based therapy include efficient cellular uptake and selective cell targeting. In this study, we employ selenium nanoparticles' green-synthesized by apigenin (SeNPs-apigenin) to treat breast cancer. We used various assays to show that SeNPs-apigenin can reduce MCF-7 cell viability and trigger apoptosis in vitro. Flow cytometry and PCR methods were used to detect apoptosis, while cell migration and invasion methods were used to quantify the possible effect of SeNPs-apigenin therapy on cell migration and invasion. According to cytotoxicity testing, the SeNPs-apigenin treatment can successfully limit MCF-7 cell proliferation and viability in a concentration-dependent manner. Flow cytometric and PCR analyses revealed that SeNPs-apigenin treatment induced apoptosis in MCF-7 cells, demonstrating that SeNPs-apigenin treatment could directly target Bcl-2, Bax, and caspase-3 and result in the discharge of cytochrome C from mitochondria into the cytosol, accompanied by the initiation of cell death, leading to permanent DNA damage and killing of MCF-7 cells. Furthermore, treatment with SeNPs-apigenin increased reactive oxygen species production and oxidative stress in MCF-7 cells. Our findings indicate that SeNPs-apigenin has cytotoxic potential in the treatment of breast cancer.

Bone marrow derived-mesenchymal stem cells (BMSCs)-based therapy is an outstanding candidate for cutaneous wound healing. Melatonin (MEL) has been reported for its anti-inflammatory as well as tissue regenerative properties. Existing work aimed to explore the potential healing power of BMSCs pre-treated with MEL in a skin wound model. Adult rats were allocated into control, PIO, BMSCs (1 × 105 cells), and MEL/BMSCs groups. On the 21 days post-wounding, tissues were sampled for analysis. The results demonstrated that compared to the control group, MEL/BMSCs therapy induced noticeable decline in wound area and elevated rate of wound retraction. Furthermore, marked increases in tissue hydroxyproline, as well as tissue content and gene expression level of vascular endothelial growth factor in MEL/BMSCs treated-wounded animals. Compared to the untreated control group, marked increases were found in antioxidant enzymatic activities together with elevated GSH levels in wounded tissues after MEL/BMSCs treatment. Moreover, therapeutically handled wounds with MEL/BMSCs revealed low levels of MDA, NO and protein carbonyls. Combined therapy with MEL/BMSCs relieved the inflammation witnessed by decreasing IL-1ß, TNF-α and NF-κB levels in wounded tissues. Furthermore, noteworthy rises in levels of TGF-ß and gene expression of α-SMA were noticed after MEL/BMSCs application that reveals their anti-scarring properties. Histologically, noticeable improvement in histopathological skin lesions in wound area and elevated the collagen synthesis and deposition. Collectively, the obtained data depict that the pre-treatment of BMSCs with MEL could potentially be a successful strategy for scaling-up the wound healing outcomes more than using BMSCs monotherapy in rat models.

BACKGROUND: Although there is an emergent increase in the epidemiology of skin cancer in Saudi Arabia, yet knowledge, attitude, and awareness towards skin cancer prevention measures is still poor. Therefore, the present study aimed to assess the knowledge and attitudes and practice towards skin cancer among the Saudi population, as well as, to evaluate the level of awareness relating to exposure to sunlight. METHODS: This cross-sectional survey involved 438 participants who were randomly selected from Riyadh city, Saudi Arabia. A standard questionnaire was used to collect data regarding skin cancer. The questionnaire focused on three main aspects knowledge, attitude, and practice. The skin cancer quality of life impact tool (SCQOLIT) was employed. RESULTS: The present study included 438 participants, aged 18 to 55 years old. The response in the present study was 81.9%. Regarding the causes and effects of skin cancer, 61.2% of the respondents have prior knowledge about it. The positive attitude about skin cancer was exhibited by 68.9%, and only 31.1% showed a negative attitude towards it. CONCLUSION: In conclusion, Knowledge, attitude, and practice towards skin cancer still under the desired level to prevent skin cancer and its related conditions in Saudi Arabia. Greater emphasis should be made through awareness campaigns and available media to raise the knowledge about implications related to prolonged exposure to sunlight.

Age-ing is involved in gradual breakdown of biological structure and function of body organs. The heart represents the main organ responsible for pumping the main issues of life which involving oxygen, nutrients and bioactive molecules necessary for maintaining the body functions. The present study has been conducted to assess the anti-aging properties of whey syrup collected from fermented milk in 4, 18 and 30-months-old rats. The histopathological and histochemical changes of carbohydrates and proteins were investigated. Immunohistochemical expression of smooth muscle actin and P53 was performed to assess the function of cardiomyocytes. Furthermore, Annexin v and biochemical changes of different cardio-biomarkers were carried out to evaluate the effects of aging. The present result of 30 months-old rats revealed myocardial infarction assessed by widening of myocardial fibers, diffused with numerous blood capillaries and dense leukocytic infiltration. The assessed biochemical markers confirmed myocardial damage. Whey supplementation improved the myocardial structure, but less improvement was observed for the 30-months-old rats. The author recommended supplementation with whey is beneficial in giving a body the demand for amino acids and minerals essential for supporting the myocardium and also provides protection against age-ing.

Aging is a biological phenomenon that involves an increase of oxidative stress associated with gradual degradation of the structure and function of the optic nerve. Gender differences and subsequent deterioration of optic nerve are an interesting topic, especially because there is little published work concerning it. One hundred male and female Wistar albino rats' with ages 1, 6, 18, 24, and 30 months (n = 20 equal for male and female) were used. At the time interval, optic nerve was investigated by light and transmission electron microscopy (TEM), assessments of antioxidant enzymes (catalase, superoxide dismustase, and glutathione-S-transferase), caspase 3 and 7, malondialdhyde, flow cytometry of DNA, annexin v, and CD8, immunochemistry of vascular endothelial growth factor (VEGF), CD31, and CD45, and single-strand DNA fragmentation. Light and TEM observations of the older specimens (24 and 30 months) revealed apparent deterioration of optic nerve axons, abundant oligodendrocytes with pyknotic nuclei, swollen astrocytes, angiogenesis, vacuolar degeneration, and mitochondrial damage. Females were highly susceptible to aging processes. Concomitantly, there was a marked reduction of antioxidant's enzymes and an increase of lipid peroxidation and apoptotic markers. Old age exhibited a marked increase of G1 apoptosis, UR and LR of annexin V and CD8 as well as increased immuno-positive reaction with VEGR, CD31 and CD45. We conclude that aging contributed to an increase of oxidative stress resulting from damage of mitochondria in axons, oligodendrocytes, and astrocytes. Age-related loss of optic nerve axons is associated with multifactorial agents including reduction in antioxidant enzymes, disruption of vasculature, astrocyte, and oligodendrocyte, demyelination, and damage of mitochondria, which enhance the liberation of reactive oxygen species as assessed by an increase of apoptotic markers malondialdhyde and caspase 3 and 7.

Aging is a biological phenomenon that involves gradual degradation of the structure and function of the retina and optic nerve. To our knowledge, little is known about the aging-related ocular cell loss in avian (Falco tinnunculus) and reptilian species (Uromastyx aegyptia). A selected 90 animals of pup, middle, and old age U. aegyptia (reptilian) and F. tinnunculus (avian) were used. The retinae and optic nerves were investigated by light and transmission electron microscopy (TEM) and assessments of neurotransmitters, antioxidant enzymes (catalase, superoxide dismustase and glutathione s transferase), caspase-3 and -7, malonadialdhyde, and DNA fragmentation. Light and TEM observations of the senile specimens revealed apparent deterioration of retinal cell layers, especially the pigmented epithelium and photoreceptor outer segments. Their inclusions of melanin were replaced by lipofuscins. Also, vacuolar degeneration and demyelination of the optic nerve axons were detected. Concomitantly, there was a marked increase of oxidative stress involved reduction of neurotransmitters and antioxidant enzymes and an increase of lipid peroxidation, caspase-3 and -7, subG0/G1 apoptosis, and P53. We conclude that aging showed an inverse relationship with the neurotransmitters and antioxidant enzymes and a linear relationship of caspases, malondialdhyde, DNA apoptosis, and P53 markers of cell death. These markers reflected the retinal cytological alterations and lipofuscin accumulation within inner segments.
