In some squids, such as those in the family Loliginidae, upon copulation, females receive and store male-delivered sperm capsules, spermatangia, at two different body locations: the buccal membrane and the distal end of the oviduct. This insemination site dimorphism is associated with alternative reproductive strategies. However, in Loliolus sumatrensis, a species of Loliginidae, the females possess three insemination sites: buccal membrane (BM), basal left IV arm (ARM) and lateral head behind the left eye (EYE), therefore we studied such the unusual phenomena. We developed microsatellite markers and genotyped the paternity of each spermatangium on three sites. We found multiple paternity at every single site and simultaneous usage of all three sites by a few males. The seasonal dynamics of a population in the Seto Inland Sea revealed a set priority for the initial use of insemination sites as BM, followed by ARM and then EYE, whereas the maximum number of stored spermatangia was greater in EYE > ARM > BM. Female maturity status was correlated with the usage pattern of insemination sites but not with the number of stored spermatangia at any insemination site. These results suggest that a male squid inseminates at different locations according to female mating history and female maturity status.

The processed forms of milk, branded liquid, and power milk available in Dhaka city, the capital of Bangladesh, were investigated for essential and trace metal/metalloids regarding nutritional and human health risk aspects. For this, the potential nutritional contribution, estimated daily intake (EDI) and non-carcinogenic risk for six different life stages with male and female categories, as well as the carcinogenic risk for children and adults of both genders, were addressed. In total, 46 branded liquid and powder milk samples were considered for this analysis employing atomic absorption spectroscopy. The concentration of essential elements showed the trends of K > Ca > Na > Mg > Fe > Zn > Mn > Cu and K > Ca > Na > Mg > Fe > Cu > Mn > Zn for liquid and powder milk samples, respectively, but the potentially hazardous one showed the same trends (Cr > Pb > Hg > As > Cd) for both items. Except for Cr, Hg, and Fe, the elemental compositions of both milk categories differed considerably (< 0.05). Compared to the threshold values for milk samples (liquid and powder), Fe (19% and 27%), Mn (100% and 63%), Cu (0% and 23%), Zn (94% and 0%), Pb (25% and 13%), and Cr (0% and 3%) showed above the permissible limits. The nutrient input was the highest for Ca (27.2% and 18.7%), followed by Mg, K, and Na. The EDI of studied elements was within the daily permissible limit in both the milk category (except age group (≤ 3) and the female category). The non-carcinogenic risk assessment showed that the age groups ≤ 3 for liquid milk and ≤ 3 and 3 < X ≤ 14 with female categories for powder milk exceeded the threshold level (> 1) in the case of Cr, Cd, As, Zn, and Mn. The probable carcinogenic risks indicated an unacceptable risk level (< 1.00E-04) for the ingestion of Cr through powder milk samples for children in male and female categories. Finally, it believes that green cow farming practices and green milk processing technology, as well as continuous monitoring of toxic metals, can limit the ultimate risk worldwide.

The space-time fractional Fokas-Lenells (STFFL) equation serves as a fundamental mathematical model employed in telecommunications and transmission technology, elucidating the intricate dynamics of nonlinear pulse propagation in optical fibers. This study employs the Sardar sub-equation (SSE) approach within the STFFL equation framework to explore uncharted territories, uncovering a myriad of optical soliton solutions (OSSs) and conducting a thorough analysis of their bifurcations. The discovered OSSs encompass a diverse array, including bright-dark, periodic, multiple bright-dark solitons, and various other types, forming a captivating spectrum. These solutions reveal an intricate interplay among bright-dark solitons, complex periodic sequences, rhythmic breathers, coexistence of multiple bright-dark solitons, alongside intriguing phenomena like kinks, anti-kinks, and dark-bell solitons. This exploration, built upon meticulous literature review, unveils previously undiscovered wave patterns within the dynamic framework of the STFFL equation, significantly expanding the theoretical understanding and paving the way for innovative applications. Utilizing 2D, contour, and 3D diagrams, we illustrate the influence of fractional and temporal parameters on these solutions. Furthermore, comprehensive 2D, 3D, contour, and bifurcation analysis diagrams scrutinize the nonlinear effects inherent in the STFFL equation. Employing a Hamiltonian function (HF) enables detailed phase-plane dynamics analysis, complemented by simulations conducted using Python and MAPLE software. The practical implications of the discovered OSS solutions extend to real-world physical events, underlining the efficacy and applicability of the SSE scheme in solving time-space nonlinear fractional differential equations (TSNLFDEs). Hence, it is crucial to acknowledge the SSE technique as a direct, efficient, and reliable numerical tool, illuminating precise outcomes in nonlinear comparisons.

This study is numerically executed to investigate the influence of heat generation or absorption on free convective flow and temperature transport within a wavy triangular enclosure filled by the nanofluid taking the Brownian effect of nanoparticles. The water (H2O) is employed as base fluid and copper (Cu) as nanoparticles for making effective Cu-H2O nanofluids. The perpendicular sinusoidally wavy wall is cooled at low temperature while the horizontal bottom sidewall is heated non-uniformly (sinusoidal). The inclined wall of the enclosure is insulated. The governing dimensionless non-linear PDEs are executed numerically with the help of the Galerkin weighted residual type finite element technique. The numerically simulated results are displayed through average Nusselt number, isothermal contours, and streamlines for the various model parameters such as Hartmann number, Rayleigh number, heat generation or absorption parameter, nanoparticles volume fraction, and undulation parameter. The outcomes illustrate that the temperature transport rate augments significantly for the enhancement of Rayleigh number as well as nanoparticles volume fraction whereas reduces for the increment of Hartman number. The heat transfer is significantly influenced by the size, shape, and Brownian motion of the nanoparticles. The rate of heat transport increases by 20.43% considering the Brownian effect for 1% nanoparticle volume. The thermal performance increases by 8.66% for the blade shape instead of the spherical shape of nanoparticles. In addition, heat transfer is impacted by the small size of nanoparticles. The thermal transport rate increases by 35.87% when the size of the nanoparticles reduces from 100 to 10 nm. Moreover, the rate of heat transmission increases efficiently as the undulation parameter rises. It is also seen that a crucial factor in the flow of nanofluids and heat transmission is the heat generation/absorption parameter that influences temperature distribution, heat transfer rates, and overall thermal performance.

This study investigates the influence of a rectangular heat source on magnetohydrodynamic hybrid convection flow in a lid-driven cavity. The effects of various parameters, such as the heat source size, magnetic field strength, and heat absorption/generation, are analyzed. The results show that increasing the heat source size decreases the average Nusselt number along the heated wall. The average Nusselt number also decreases with higher magnetic field strength and heat generation, while it increases with heat absorption. The major finding is to apply an important technique the Galerkin weighted residual technique of the finite element (FE) method to solve the non-dimensional equations and the associated boundary conditions. The isotherms are used to show the temperature distribution in a domain. Streamline present the flow field in the enclosure. However, it is easy to realize the direction and intensity of the heat transfer particularly in convection problems which the path of heat flux is perpendicular and the isotherm due to convection effect. Thus, the purpose of this research is to study the results of mixed convection. The effects of location and height of the partitions are considered for the various Richardson numbers. Fluid flow field, thermal field and heat transfer are presented through the streamlines and isotherms, respectively. Results are substantiated relating to the published work.

Titanium dioxide (TiO2) nanoparticles have been extensively studied for catalyzing the photo-degradation of organic pollutants, the photocatalyst being nonselective to the substrate. We, however, found that TiO2 nanoparticles prepared via the sol-gel and hydrothermal synthetic routes each possess a definite specificity to the charge of the substrate for photodegradation. The nanoparticles were characterized by SEM, FTIR, XRD, TGA, and UV-visible spectra, and the photocatalytic degradation under UV-B (285 nm) irradiation of two model compounds, anionic methyl Orange (MO) and cationic methylene blue (MB) was monitored by a UV-visible spectrophotometer. Untreated sol-gel TiO2 nanoparticles (Tsg) preferentially degraded MO over MB (90% versus 40% in two hours), while after calcination at 400 °C for two hours (Tsgc) they showed reversed specificity (50% MO versus 90% MB in one hour). The as-prepared hydrothermal TiO2 nanoparticles (Tht) behaved in the opposite sense of Tsg (41% MO versus 91% MB degraded in one and a half hours); calcination at 400 °C (Thtc) did not reverse the trend but enhanced the efficiency of degradation. The study indicates that TiO2 nanoparticles can be made to degrade a specific class of organic pollutants from an effluent facilitating the recycling of a specific class of pollutants for cost-effective effluent management.

The effect of air pollution on public health is severely detrimental. In humans; the physiological response against pollutants is mainly elicited via the activation of aryl hydrocarbon receptor (AhR). It acts as a prime sensor of xenobiotic chemicals, also functioning as a transcription factor regulating a variety of gene expressions. Along with AhR, another pivotal element of the pollution stress pathway is Xenobiotic Response Elements (XREs). XRE, as studied are some conserved sequences in the DNA, responsible for the physiological response against pollutants. XRE is present at the upstream of the inducible target genes of AhR and it regulates the function of the AhR. XRE(s) are highly conserved in species as it has only eight specific sequences found so far in humans, mice, and rats. Inhalation of toxicants like dioxins, gaseous industrial effluents, and smoke from burning fuel and tobacco leads to predominant damage to the lungs. However, scientists are exploring the involvement of AhR in chronic diseases for example chronic obstructive pulmonary disease (COPD) and also other lethal diseases like lung cancer. In this review, we summarise what is known at this time about the roles played by the XRE and AhR in our molecular systems that have a defined control in the normal maintenance of homeostasis as well as dysfunctions.

Perfluorooctanoic acid (PFOA) is a persistent organic pollutant, which has endocrine-disrupting properties and can interfere with the synthesis and secretion of testicular steroid hormones, but the underlying molecular mechanisms are still not fully understood. In this study, we investigated the effects of low doses of PFOA exposure on testicular steroidogenesis in rats and revealed the role of histone modifications. It was found that the serum levels of progesterone, testosterone, and estradiol were significantly increased after 0.015 and 0.15 mg/kg of PFOA exposure, and the expression of Star, a key rate-limiting gene, was up-regulated, while other steroidogenic genes Cyp11a1, Hsd3b, Cyp17a1, and Hsd17b were down-regulated. In addition, the levels of multiple histone modifications (H3K9me1/2/3 and H3K9/18/23ac) were all significantly reduced by PFOA in rat testis. Histone H3K9 methylation is associated with gene silencing, while histone acetylation leads to gene activation. ChIP analysis further showed that H3K9me1/3 was significantly decreased in the promoter region of Star, while H3K18ac levels were down-regulated in other gene promoters. Accordingly, we suggest that low-level PFOA enhances StAR expression through the repression of H3K9me1/3, which stimulates steroid hormone production in rat testis. These results are expected to shed new light on the molecular mechanisms by which low-dose PFOA disturbs male reproductive endocrine from an epigenetic aspect and may be useful for human health risk assessment regarding environmental PFOA exposure.

Stroke is a medical disorder in which the blood arteries in the brain are ruptured, causing damage to the brain. When the supply of blood and other nutrients to the brain is interrupted, symptoms might develop. According to the World Health Organization (WHO), stroke is the greatest cause of death and disability globally. Early recognition of the various warning signs of a stroke can help reduce the severity of the stroke. Different machine learning (ML) models have been developed to predict the likelihood of a stroke occurring in the brain. This research uses a range of physiological parameters and machine learning algorithms, such as Logistic Regression (LR), Decision Tree (DT) Classification, Random Forest (RF) Classification, and Voting Classifier, to train four different models for reliable prediction. Random Forest was the best performing algorithm for this task with an accuracy of approximately 96 percent. The dataset used in the development of the method was the open-access Stroke Prediction dataset. The accuracy percentage of the models used in this investigation is significantly higher than that of previous studies, indicating that the models used in this investigation are more reliable. Numerous model comparisons have established their robustness, and the scheme can be deduced from the study analysis.

Exposure to ambient particulate matters (PMs) has been associated with a variety of lung diseases, and high-fat diet (HFD) was reported to exacerbate PM-induced lung dysfunction. However, the underlying mechanisms for the combined effects of HFD and PM on lung functions remain poorly unraveled. By performing a comparative proteomic analysis, the current study investigated the global changes of histone post-translational modifications (PTMs) in rat lung exposed to long-term, real-world PMs. In result, after PM exposure the abundance of four individual histone PTMs (1 down-regulated and 3 up-regulated) and six combinatorial PTMs (1 down-regulated and 5 up-regulated) were significantly altered in HFD-fed rats while only one individual PTM was changed in rats with normal diet (ND) feeding. Histones H3K18ac, H4K8ac and H4K12ac were reported to be associated with DNA damage response, and we found that these PTMs were enhanced by PM in HFD-fed rats. Together with the elevated DNA damage levels in rat lungs following PM and HFD co-exposure, we demonstrate that PM exposure combined with HFD could induce lung injury through altering more histone modifications accompanied by DNA damage. Overall, these findings will augment our knowledge of the epigenetic mechanisms for pulmonary toxicity caused by ambient PM and HFD exposure.

Sperm morphology is generally uniform within a species due to selective pressures that act to achieve better fertilization outcomes under postcopulatory competitive circumstances. Therefore, polyandry that intensifies post-mating sperm competition should constrain intraspecific sperm polymorphism. Contrary to this paradigm, we previously found that a polyandrous squid, Heterololigo bleekeri, produces dimorphic eusperm (flagellum length dimorphism; FLD), which is closely associated with alternative reproductive tactics (ARTs); large males (consorts) transfer their spermatophores inside the female's mantle cavity, while small males (sneakers) do so outside the mantle. Thus, FLD was considered as the consequence of different insemination strategies that arise from different modes of sperm competition, sperm storage and the fertilization environment. However, in other squid species showing ARTs, the choice of mating behaviour is rather conditional (i.e., switching mating tactic between consorts and sneakers), which poses the question of whether sperm FLD could have evolved. Here, we investigated five species in the family Loliginidae that exhibit ARTs and found that all species showed sneaker-biased FLD. However, in a species with conditional ARTs, we found FLD rather ambiguous and the testicular somatic index to be nearly continuous among individuals at transitional state, suggesting that plasticity in mating behaviour compromises the disruptive selection on a sperm morphological trait.

Due to similar coordination chemistry of palladium and platinum, a large number of palladium compounds as well have been investigated for their anticancer activity. In the present study, we describe synthesis, characterization, and anticancer activity of palladium complex [Bis(1,8-quinolato)palladium (II)], coded as NH3 against seven different cancer cell lines. NH3 is found to have higher antitumor activity than cisplatin against both parent ovarian A2780 cell line and cisplatin-resistant cell lines. Also, NH3 has the lower IC50 value in HT-29 colorectal cancer cell line. The higher antitumor activity of NH3 is due to the presence of bulky 8-Hydroxyquinoline ligand, thus reducing its reactivity. Proteomic study has identified significantly expressed proteins which have been validated through bioinformatics. NH3 has been found to be less toxic than cisplatin at 2.5 mg/kg and 5 mg/kg dosages on mice models. Binary combinations of NH3 with curcumin and epigallocatechin gallate (EGCG) have demonstrated dose and sequence-dependent synergism in ovarian and colorectal cancer models. All of the preclinical studies indicate promising therapeutic potential of NH3 [Bis(1,8-quinolato)palladium (II)] as an anticancer drug.

Perfluorooctane sulfonate (PFOS), an artificial perfluorinated compound, has been associated with male reproductive disorders. Histone modifications are important epigenetic mediators; however, the impact of PFOS exposure on testicular steroidogenesis through histone modification regulations remains to be elucidated. In this study, we examined the roles of histone modifications in regulating steroid hormone production in male rats chronically exposed to low-level PFOS. The results indicate that PFOS exposure significantly up-regulated the expressions of StAR, CYP11A1 and 3ß-HSD, while CYP17A1 and 17ß-HSD were down-regulated, thus contributing to the elevated progesterone and testosterone levels. Furthermore, PFOS significantly increased the histones H3K9me2, H3K9ac and H3K18ac while reduced H3K9me3 in rat testis. It is known that histone modifications are closely involved in gene transcription. Therefore, to investigate the association between histone modifications and steroidogenic gene regulation, the levels of these histone marks were further measured in steroidogenic gene promoter regions by ChIP. It was found that H3K18ac was augmented in Cyp11a1 promoter, and H3K9ac was increased in Hsd3b after PFOS exposure, which is proposed to result in the activation of CYP11A1 and 3ß-HSD, respectively. To sum up, chronic low-level PFOS exposure activated key steroidogenic gene expression through enhancing histone acetylation (H3K9ac and H3K18ac), ultimately stimulating steroid hormone biosynthesis in rat testis.

Temperature transfer by virtue of natural convection for visualizing heat transport characteristics through heatline method within a prismatic cavity filled with Cu-H2O nanofluid considering two different temperature boundary conditions is performed numerically. Two top inclined walls are warmed-up at low temperature whilst the bottom wall is heated two different heated conditions such as uniform temperature condition and linear temperature condition. Two vertical walls are insulated. Finite element technique of Galerkin weighted residual form is employed for solving nonlinear partial differential equations for numerical calculation. Heatlines, isotherm contours, streamline contours, and Nusselt number are employed for displaying numerical simulated results for the model parameters entitled nanoparticles volume fraction, Hartmann number and Rayleigh number. The outcomes indicate that heat transfer rate has a significant impact on thermal boundary condition and shape of the nanoparticles. The temperature transfer value enhances significantly for higher Rayleigh number as well as nanoparticles volume fraction. Hartmann number has a positive impact on fluid flow and temperature transport. The characteristics of heat transport using heatlines method are also performed for predicting the better energy transform compared to isotherm contours. In addition, different types of nanofluids are also employed to examine the best heat transport performance.

Environmental epigenetic findings shed new light on the roles of epigenetic regulations in environmental exposure-induced toxicities or disease susceptibilities. Currently, environmental emerging contaminants (ECs) are in focus for further investigation due to the evidence of human exposure in addition to their environmental occurrences. However, the adverse effects of these environmental ECs on health through epigenetic mechanisms are still poorly addressed in many aspects. This review discusses the epigenetic mechanisms (DNA methylation, histone modifications, and microRNA expressions) linking ECs exposure to health outcomes. We emphasized on the recent literature describing how ECs can dysregulate epigenetic mechanisms and lead to downstream health outcomes. These up-to-date research outputs could provide novel insights into the toxicological mechanisms of ECs. However, the field still faces a demand for further studies on the broad spectrum of health effects, synergistic/antagonistic effects, transgenerational epigenetic effects, and epidemiologic and demographic data of ECs.

In this article, pharmacological management of circumstantial overdose of an anticancer drug, Harmine (HM), under in vitro and in vivo conditions is described and further validated by employing in silico methods. HM, an efficient cancer cell photosensitizer, interacts extensively with nontoxic ß-cyclodextrin (ß-CD). Steady-state fluorescence studies and molecular docking analysis established differential nature of molecular inclusion depending on the relative concentrations of ß-CD. Presently, ß-CD is commonly used as a standard drug-delivery vehicle but its application for controlled drug withdrawal is rarely explored. Flow cytometric results and in vivo investigations on a zebrafish model showed that conditional overdose of preadministered drug molecules can be efficiently removed by encapsulating successfully within nontoxic ß-CDs, albeit by controlled application of the same. This is an approach to manage the cytotoxicity of a drug in a safe way that is already administered. We believe that this ß-CD-mediated withdrawal of drugs may find possible applications in controlled capturing of excess or unused drug inside living systems and reducing the unwanted toxicity associated with chemotherapeutics.

BACKGROUND: Colorectal cancer is the third most commonly diagnosed cancer in the world, causing many deaths every year. Combined chemotherapy has opened a new horizon in treating colorectal cancer. The objective of the present study is to investigate the activity of oxaliplatin in combination with emetine and patulin against colorectal cancer models. METHODS: IC50 values of oxaliplatin, emetine and patulin were determined against human colorectal cancer cell lines (HT-29 and Caco-2) using MTT reduction assay. Synergistic, antagonistic and additive effects from the selected binary combinations were determined as a factor of sequence of administration and added concentrations. Proteomics was carried out to identify the proteins which were accountable for combined drug action applying to the selected drug combination. RESULTS: Oxaliplatin in combination with patulin produced synergism against human colorectal cancer models depending on dose and sequence of drug administration. Bolus administration of oxaliplatin with patulin proved to be the best in terms of synergistic outcome. Altered expressions of nine proteins (ACTG, PROF1, PPIA, PDIA3, COF1, GSTP1, ALDOA, TBA1C and TBB5) were considered for combined drug actions of oxaliplatin with patulin. CONCLUSION: Bolus administration of oxaliplatin with patulin has the potential to be used in the treatment of colorectal cancer, and would warrant further evaluation using suitable animal model.

We show that the non-linear positive capacitance (PC) of ferroelectrics (FE) can explain the steep subthreshold-slope (SS) observed in FE based MOSFETs and often attributed to the existence of a negative capacitance in FE capacitors. Physically attainable and unattainable regions of the S-shape curve used in the negative capacitance theory are investigated by self-consistently solving Landau-Khalatnikov and Maxwell equations and by experimental validation. Finally, the conditions for attaining a steep SS in FE based MOSFETs assuming only positive capacitances are discussed.

Nowadays, most of the commonly used superabsorbent polymers (SAPs) are derived from synthetic polymers, particularly acrylic acid and its copolymers made with acrylamide. Here, we describe a novel and environmentally friendly aqueous-based process for fabrication of a new, natural, cellulose-based SAP (hydrogel). In this two-step process, cellulose was first reacted with sodium monochloroacetate (MCA) to obtain carboxymethyl cellulose (CMC) and then cross-linked with epichlorohydrin (ECH). In distilled water (d-water), the water retention value (WRV) of the newly fabricated hydrogels reached 725 g d-water/g gel, which is significantly greater than any other commercially available superabsorbent cellulose-based material (WRV of 10-100 g/g) and comparable to the commercial synthetic (polyacrylate) SAP gels (WRV of up to 1000 g/g). In saline water (s-water; 0.9% NaCl), the maximum WRV attained was 118 g s-water/g gel, which exceeds more than 2-fold the WRV of commercial gels (40-50 g/g). Compositional analysis was carried out to determine the amount of carboxyl groups and average molecular mass, and the parameters for hydrogel preparation were optimized. The natural SAP was characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD). The hydrogels showed good re-swelling properties losing only 5-10% of their capabilities to reabsorb d-water when reused in four consecutive cycles. Because of their superior swelling properties in physiological saline, the new hydrogels can compete with their synthetic counterparts in applications such as high-value hygiene and biomedical products.

Nowadays, drug resistance in parasites is considered to be one of the foremost concerns in health and disease management. It is interconnected worldwide and undermines the health of millions of people, threatening to grow worse. Unfortunately, it does not receive serious attention from every corner of society. Consequently, drug resistance in parasites is gradually complicating and challenging the treatment of parasitic diseases. In this context, we have dedicated ourselves to review the incidence of drug resistance in the protozoan parasites Plasmodium, Leishmania, Trypanosoma, Entamoeba and Toxoplasma gondii. Moreover, understanding the role of ATP-binding cassette (ABC) transporters in drug resistance is essential in the control of parasitic diseases. Therefore, we also focused on the involvement of ABC transporters in drug resistance, which will be a superior approach to find ways for better regulation of diseases caused by parasitic infections.
