RESUMO

Saccharomyces cerevisiae Sub1 (ScSub1) has been defined as a transcriptional stimulatory protein due to its homology to the ssDNA binding domain (ssDBD) of human PC4 (hPC4). Recently, PC4/Sub1 orthologues have been elucidated in eukaryotes, prokaryotes, and bacteriophages with functions related to DNA metabolism. Additionally, ScSub1 contains a unique carboxyl-terminal region (CT) of unknown function up to date. Specifically, it has been shown that Sub1 is required for transcription activation, as well as other processes, throughout the transcription cycle. Despite the progress that has been made in understanding the mechanism underlying Sub1's functions, some questions remain unanswered. As a case in point: whether Sub1's roles in initiation and elongation are differentially predicated on distinct regions of the protein or how Sub1's functions are regulated. Here, we uncover some residues that are key for DNA-ScSub1 interaction in vivo, localized in the ssDBD, and required for Sub1 recruitment to promoters. Furthermore, using an array of genetic and molecular techniques, we demonstrate that the CT region is required for transcription elongation by RNA polymerase II (RNAPII). Altogether, our data indicate that Sub1 plays a dual role during transcription-in initiation through the ssDBD and in elongation through the CT region.

BACKGROUND: Despite major medical advances, Type 1 Diabetes (T1D) patients still have greater morbimortality than the general population. Our aim was to describe our cohort of T1D patients and identify potential risk factors susceptible to prevention strategies. METHODS: Cross-sectional, observational study, including T1D patients treated at our center, from 1 March 2017 to 31 March 2020. INCLUSION CRITERIA: T1D, age > 14 years and signed informed consent. EXCLUSION CRITERIA: diabetes other than T1D, age < 14 years and/or refusal to participate. RESULTS: Study population n = 2181 (49.8% females, median age at enrollment 41 years, median HbA1c 7.7%; 38.24% had at least one comorbidity). Roughly 7.45% had severe hypoglycemia (SH) within the prior year. Macro/microvascular complications were present in 42.09% (5.83% and 41.14%, respectively). The most frequent microvascular complication was diabetic retinopathy (38.02%), and coronary disease (3.21%) was the most frequent macrovascular complication. The risk of complications was higher in males than in females, mainly macrovascular. Patients with SH had a higher risk of complications (OR 1.42; 1.43 in males versus 1.42 in females). CONCLUSIONS: Our T1D population is similar to other T1D populations. We should minimize the risk of SH, and male patients should perhaps be treated more aggressively regarding cardiovascular risk factors.

Arterial pressure waves have been described in one dimension using several approaches, such as lumped (Windkessel) or distributed (using Navier-Stokes equations) models. An alternative approach consists of modeling blood pressure waves using a Korteweg-de Vries (KdV) equation and representing pressure waves as combinations of solitons. This model captures many key features of wave propagation in the systemic network and, in particular, pulse pressure amplification (PPA), which is a mechanical biomarker of cardiovascular risk. The main objective of this work is to compare the propagation dynamics described by a KdV equation in a human-like arterial tree using acquired pressure waves. Furthermore, we analyzed the ability of our model to reproduce induced elastic changes in PPA due to different pathological conditions. To this end, numerical simulations were performed using acquired central pressure signals from different subject groups (young, adults, and hypertensive) as input and then comparing the output of the model with measured radial artery pressure waveforms. Pathological conditions were modeled as changes in arterial elasticity (E). Numerical results showed that the model was able to propagate acquired pressure waveforms and to reproduce PPA variations as a consequence of elastic changes. Calculated elasticity for each group was in accordance with the existing literature.

RESUMO

This paper presents the development of a fusion strategy to integrate and calibrate signals from magnetometers, gyroscopes and accelerometers to implement a magnetic, angular rate and gravity (MARG) sensor system. The aim of such algorithms is to capture signals from the individual sensors and identify, compensate and reduce external and internal errors such as bias, scale factor and drifts, which highly depend on the noise levels. The necessary calibrations to ensure the reliability of captured data are also presented. The orientation data obtained by the proposed algorithm will be compared with a commercial motion capture system, which are currently being used by researchers in biomechanical analysis and in clinical motor rehabilitation studies.

INTRODUCTION:This study investigates a gait research protocol to assess the impact of a walker model with forearm supports on the kinematic parameters of the lower limb during locomotion. METHODS: Thirteen healthy participants without any history of gait dysfunction were enrolled in the experimental procedure. Spatiotemporal and kinematic gait parameters were calculated by using wireless inertial sensors and analyzed with Principal Component Analysis (PCA). The PCA method was selected to achieve dimension reduction and evaluate the main effects in gait performance during walker-assisted gait. Additionally, the interaction among the variables included in each Principal Component (PCs) derived from PCA is exposed to expand the understanding of the main differences between walker-assisted and unassisted gait conditions. RESULTS:The results of the statistical analysis identified four PCs that retained 65% of the data variability. These components were associated with spatiotemporal information, knee joint, hip joint and ankle joint motion, respectively. CONCLUSION: Assisted gait by a walker model with forearm supports was characterized by slower gait, shorter steps, larger double support phase and lower body vertical acceleration when compared with normal, unassisted walking.

Models define a simplification of reality, which help to understand function. The arterial system has been modeled in many ways: lumped models, tube models and anatomically based distributed models. In this work, arterial segments were modeled as thin nonlinear elastic tubes filled with an incompressible fluid, whose governing dynamics were denoted by the Korteweg and DeVries equation. In order characterize the pressure pulse propagation, a discrete multi-segmented conduit was proposed. Arterial wall mechanical parameters were obtained from existing literature and assigned to each individual segment. The numerical model was developed starting in the aortic arch, and ending at the femoral artery. The main idea of this article was to perform a computational simulation of pressure wave propagation, considered as a solitons combination, along several segments of the arterial tree.

Estudio retrospectivo de 216 pacientes con peritonitis. La mortalidad global de la serie fue de 11,1% y las complicaciones del 35,2%. En el 40,3% de los pacientes de trataba de una peritonitis generalizada y en el 13,9% era de origen colónico. Se concluyó que la edad avanzada, el retardo excesivo en la consulta médica, la existencia de una peritonitis generalizada y/o colónica, fueron los factores que condicionaron directamente las tasas de morbimortalidad.
