RESUMO

Some soil microbes, with their diverse inhabitance, biologically active metabolites, and endospore formation, gave them characteristic predominance and recognition among other microbial communities. The present study collected ten soil samples from green land, agricultural and marshy soil sites of Khyber Pakhtunkhwa, Pakistan. After culturing on described media, the bacterial isolates were identified through phenotypic, biochemical and phylogenetic analysis. Our phylogenetic analysis revealed three bacterial isolates, A6S7, A1S6, and A1S10, showing 99% nucleotides sequence similarity with Brevibacillus formosus, Bacillus Subtilis and Paenibacillus dendritiformis. The crude extract was prepared from bacterial isolates to assess the anti-bacterial potential against various targeted multidrug-resistant strains (MDRS), including Acinetobacter baumannii (ATCC 19606), Methicillin-resistant Staphylococcus aureus (MRSA) (BAA-1683), Klebsiella pneumoniae (ATCC 13883), Pseudomonas aeruginosa (BAA-2108), Staphylococcus aureus (ATCC 292013), Escherichia coli (ATCC25922) and Salmonella typhi (ATCC 14028). Our analysis revealed that all bacterial extracts possess activity against Gram-negative and Gram-positive bacteria at a concentration of 5 mg/mL, efficiently restricting the growth of E. coli compared with positive control ciprofloxacin. The study concluded that the identified species have the potential to produce antimicrobial compounds which can be used to control different microbial infections, especially MDRS. Moreover, the analysis of the bacterial extracts through GC-MS indicated the presence of different antimicrobial compounds such as propanoic acid, oxalic acid, phenol and hexadecanoic acid.

UTIs are majorly caused by species of bacteria in patients of almost all ages. The study was aimed to determine the prevalence rate of uropathogens, its antibiotic susceptibility pattern and associated risk factors. Urine samples were collected from n=470 participants using sterilized containers and were inoculated on culture media. The isolates were identified via gram-staining and biochemical characterization. A total of 43.20% samples were positive. Female contributed the highest prevalence rate, 78.82% as compared to male, 21.18%. The highest prevalence 40.90% was observed in the age-group 31-45, followed by 16-30 with 36.90%. Escherichia coli (47.80%) was the most prevalent, followed by Klebsiella pneumoniae (18.2%), Enterococcus faecalis (12.80%), Pseudomonas aeruginosa (10.30%) and Proteus mirabilis (7.40%). Staphylococcus aureus showed high sensitivity (100%) to amikacin, meropenem, imipenem, fosfomycin, vancomycin, clindamycin and linezolid while in case of E. faecalis, vancomycin and linezolid were highly potent. Amikacin and meropenem showed the highest (100%) potency followed by imipenem While Fosfomycin was highly potent to E. coli, K. pneumoniae, P. mirabilis and P. aeruginosa with potency rate 89.97%, 92.31%, 100% and 100% respectively. In the current study, the positivity rate was highly observed in female. E. coli and K. pneumoniae were found the most ubiquitous for UTI.

Escherichia coli is the most studied among those bacteria causing urinary tract infections. This study was aimed to find out antibacterial activity and minimum inhibitory concentration of selected antibacterial agents against E. coli isolates of hospitalized UTI patients. The specimens were inoculated on Eosin Methylene Blue medium. E. coli isolates were identified via colonial morphology, biochemical testing and API-20 kit. The susceptibility pattern of antibacterial agents was determined applying disc diffusion method (Kirby-Bauer) and dilution tube method. Among all, 38.82% (n=158/407) specimens were positive for E. coli, while the rest showed either no growth or exhibited colonies other than E. coli. while observing the susceptibility pattern, Imipenem was found the most effective (73.42%) antibacterial agent, followed by nitrofurantoin (52.53%), cefpirome (44.94%) and tazobactam/ piperacillin (44.94%), whereas the E. coli isolates were highly resistant to sulfamethoxazole/trimethoprim (71.52%), followed by Amoxicillin-clavulanic acid (67.72%), nalidixic acid (66.46%) and Tobramycin (62.03%), when tested by disc diffusion method. The isolates were susceptible to cefpirome (39.87%) and tobramycin (39.87%) and resistant to sulfamethoxazole/trimethoprim (75.32%), followed by levofloxacin (61.39%), when tested by tube dilution method. The study concluded high degree of resistance against Sulfamethoxazole/trimethoprim, in contrast, cephalosporin and Imipenem exhibited good potency which can be recommended for UTI.

The combination of nano fluid and changing cross-section mini-channel heat sink effects have become a remarkable choice for the use of thermal devices such as miniature electronic devices to be effectively cooled. In this paper, the comparison of three dimensional straight and wavy channel configuration with using different types nano fluids are numerically investigated. The effects of wave amplitude and A particular type of volume fraction of (Copper Oxide CuO, Dimond Al2O3, Iron Oxide Fe3O4, Titanium Oxide TiO2 and Silver Ag-nano fluids are offered. Three amplitudes of waves (0.15 mm, 0.2 mm and 0.25 mm) and Reynold's number from 200 to 1000 and concentration volume varieties from 0 to 0.075 are used. The effect on thermal resistance, pressures drop, factor of friction of the mini channel is displayed. It is observed that the mini-channel sink's heat transfer efficiency is greatly enhanced compared to the straight channel in an event of adding distilled water as accoolant. The results indicate that nano fluid and wavy mini-channel can boost the heat sink's hydrothermal efficiency and Ag- water nano fluid in term of heat transfer, it outperforms other nanofluids an enhancement in the Nusselt number reached to 54% at concentration volume 0.075.

In this study, Galerkin Finite Element Method or GFEM is used for the modeling of mixed convection with the entropy generation in wavy lid-driven porous enclosure filled by the CNT-water nanofluid under the magnetic field. Two different cases of boundary conditions for hot and cold walls are considered to study the fluid flow (streamlines) and heat transfer (local and average Nusselt numbers) as well as the entropy generation parameters. Richardson (Ri), Darcy (Da), Hartmann angle (γ), Amplitude (A), Number of peaks (N), Volume fraction (φ), Heat generation factor (λ), Hartmann number (Ha) and Reynolds number (Re) are studied parameters in this study which results indicated that at low Richardson numbers (< 1) increasing the inclined angle of magnetic field, decreases the Nu numbers, but at larger Richardson numbers (> 1) it improves the Nu numbers.

Continuously increasing human population demands increased food production, which needs greater fertilizer's input in agricultural lands to enhance crop yield. In this respect, different fertilization practices gained acceptance among farmers. We reviewed effect of three main fertilization practices (Conventional-, Organic-, and Bio-fertilization) on soil microbial diversity, activity, and community composition. Studies reported that over application of inorganic fertilizers decline soil pH, change soil osmolarity, cause soil degradation, disturb taxonomic diversity and metabolism of soil microbes and cause accumulation of extra nutrients into the soil such as phosphorous (P) accumulation. On the contrary, organic fertilizers increase organic carbon (OC) input in the soil, which strongly encourage growth of heterotrophic microbes. Organic fertilizer vermicompost application provides readily available nutrients to both plants as well as microbes and encourage overall microbial number in the soil. Most recently, role of beneficial bacteria in long-term sustainable agriculture attracted attention of scientists towards their use as biofertilizer in the soil. Studies documented favorable effect of biofertilization on microbial Shannon, Chao and ACE diversity indices in the soil. It is concluded from intensive review of literature that all the three fertilization practices have their own way to benefit the soil with nutrients, but biofertilization provides long-term sustainability to crop lands. When it is used in integration with organic fertilizers, it makes the soil best for microbial growth and activity and increase microbial diversity, providing nutrients to soil for a longer time, thus improving crop productivity.

Diabetic retinopathy (DR) is an eye disease that alters the blood vessels of a person suffering from diabetes. Diabetic macular edema (DME) occurs when DR affects the macula, which causes fluid accumulation in the macula. Efficient screening systems require experts to manually analyze images to recognize diseases. However, due to the challenging nature of the screening method and lack of trained human resources, devising effective screening-oriented treatment is an expensive task. Automated systems are trying to cope with these challenges; however, these methods do not generalize well to multiple diseases and real-world scenarios. To solve the aforementioned issues, we propose a new method comprising two main steps. The first involves dataset preparation and feature extraction and the other relates to improving a custom deep learning based CenterNet model trained for eye disease classification. Initially, we generate annotations for suspected samples to locate the precise region of interest, while the other part of the proposed solution trains the Center Net model over annotated images. Specifically, we use DenseNet-100 as a feature extraction method on which the one-stage detector, CenterNet, is employed to localize and classify the disease lesions. We evaluated our method over challenging datasets, namely, APTOS-2019 and IDRiD, and attained average accuracy of 97.93% and 98.10%, respectively. We also performed cross-dataset validation with benchmark EYEPACS and Diaretdb1 datasets. Both qualitative and quantitative results demonstrate that our proposed approach outperforms state-of-the-art methods due to more effective localization power of CenterNet, as it can easily recognize small lesions and deal with over-fitted training data. Our proposed framework is proficient in correctly locating and classifying disease lesions. In comparison to existing DR and DME classification approaches, our method can extract representative key points from low-intensity and noisy images and accurately classify them. Hence our approach can play an important role in automated detection and recognition of DR and DME lesions.

In this paper, numerical Galerkin Finite Element Method (GFEM) is applied for conjugate heat-transfer of a rotating cylinder immersed in Fe3O4-water nanofluid under the heat-flux and magnetic field. The outer boundaries of the cavity were maintained at low temperatures while beside the cylinder were insulated. It is assumed that the cylinder rotates in both clockwise and counter-clockwise directions. The dimensionless governing equations such as velocity, pressure, and temperature formulation were analyzed by the GFEM. The results were evaluated using the governing parameters such as nanoparticles (NPs) volume fraction, Hartmann and Rayleigh numbers, magnetic field angle and NPs shapes. As a main result, the average Nusselt number increases by increasing the NPs volume fraction, inclination angle and thermal conductivity ratios, while increasing the Hartmann number decreased the Nusselt number. Furthermore, platelet NPs had the maximum average Nusselt number and spherical NPs made the minimum values of Nusselt numbers among examined NPs shapes.

OBJECTIVE: To investigate the frequency rate and sensitivity pattern of extended-spectrum beta-lactamase and metallobeta- lactamase producing Pseudomonas aeruginosa isolated from major hospitals. METHODS: The cross-sectional study was conducted in the Microbiology section of the Pathology Department of Ayub Medical College, Abbottabad, Pakistan, from September 2017 to April 2018, and comprised clinical samples collected from different medical wards of major hospitals in the study area. For the selective growth of Pseudomonas aeruginosa, Cetrimide agar was used, and different antibiotics were evaluated for the sensitivity pattern following Kirby-Bauer diffusion method. Pseudomonas aeruginosa producing extended-spectrum beta lactamase and metallo-beta-lactamase were identified through double disk synergy test and imipenem ethylenediaminetetraacetic acid tests respectively. Patient's demographic and medical history was noted on a proforma. Data was analysed using SPSS 22.0. RESULTS: Of the 242 samples screened, 46 (19%) were positive for Pseudomonas aeruginosa. These samples were highly sensitive to levofloxacin, amikacin, imipenem, meropenem and ciprofloxacin (p<0.05). Of the positive cases, 11 (23.91%) were detected for extended-spectrum beta-lactamase production, while 3 (6.52%) samples were detected for metallo-beta-lactamase production. CONCLUSIONS: Pseudomonas aeruginosa samples were widely resistant to most antibiotics, but were sensitive for some antibiotics which may be recommended by physicians when treating Pseudomonas aeruginosa infection.

The fluid flow and mixed convection heat transfer of a non-Newtonian (Cu-water) nanofluid-filled circular annulus enclosure in a magnetic field are investigated numerically for a two-dimensional, steady-state, incompressible, laminar flow using the Galerkin finite element method (GFEM). The Prandtl number (Pr = 6.2) and Grashof number (Gr = 100) are assumed to be constants, whereas the Richardson number varies within a range of 0 ≤ Ri ≤ 1, the Hartman number within a range of 0 ≤ Ha ≤60, the Power law index within a range of 0.2 ≤ n ≤ 1.4, and the volume fraction within a range of 0 ≤ φ ≤ 1. The enclosure consists of an outer rotating cylinder that is kept at a cold temperature (Tc) and an inner non-rotating cylinder kept at a hot temperature (Th). The ratio of the inner circular diameter to the annulus space length is kept constant at 2. The results depict that the stream function increases with increasing power law index, even up to n = 1, which causes the fluid to behave as a Newtonian fluid. The magnetic field has a critical impact on the fluid flow pattern. The average Nusselt number increases with decreasing Richardson number, owing to the improved heat transfer by forced convection.

All of the anastomotic techniques used for jejuno-ileal atresia aim to overcome the size discrepancy between the resected bowel ends, lessen anastomotic complication and prevent development of short bowel syndrome. Aim of study was to evaluate the outcomes of end-to-side oblique anastomosis for jejuno-ileal atresia and verifying the risk factors for complications. A prospective study was conducted on patients with jejuno-ileal atresia managed by end-to-side oblique anastomotic technique over a 7-year period. Data were collected and analyzed to evaluate the outcomes of this surgical technique as morbidity and mortality. The study included 40 patients, 26 males and 14 females. The mean operation time was 105 minutes, the mean time for starting oral feeding was 5.7 days and the mean duration of hospital stay was 9.13 day. Postoperative complications occurred in 11 patients (27.5%). Death was recorded in six cases; survival rate was 85%. End-to-side oblique anastomosis is simple and effective surgical procedure for most types of jejuno-ileal atresia. The technique results in wide and early functioning anastomosis, relatively low morbidity and mortality. As with any neonatal surgery, the overall prognosis and outcomes is primarily dependent on prematurity, birth weight and associated anomalies in addition to the amount of residual bowel function that exist after surgery.

RESUMO

The presence of the vermiform appendix in an inguinal hernial sac is called "Amyand" Hernia, which is a rare condition especially in children. The authors are reporting two cases of left-sided Amyand hernia in infants presented with symptoms and signs of obstructed inguinal hernia. Emergency appendectomy and herniotomy were performed. The patients were followed up for 4 to 6 months without complications. The case histories are presented, and the conditions discussed with review of the literature.

The Ce(IV) atom in the title salt, {(H(3)NCH(2)CH(2)CH(2)NH(3))(2)[Ce(SO(4))(4)(H(2)O)]·2H(2)O}(n), exists in a monocapped square-anti-prismatic coordination geometry. The water-coordinated metal atom is bonded to four sulfate ions; one of them is monodentate and two function in a chelating mode. The fourth is also chelating but it uses one of the other two O atoms to bind to an adjacent metal atom, generating a polyanionic chain. The cations are linked to the polyanionic chain as well as to the uncoordinated water mol-ecules, resulting in an O-H⋯O and N-H⋯O hydrogen-bonded three-dimensional network.

The main aim of the current study was to explore molecular insights for potentially new dual cholinesterase inhibitor(s) from Asparagus adscendens via molecular docking. This medicinal plant is traditionally used as a nerve tonic and remedy for memory impairments. Conypododiol was isolated from the chloroform fraction of methanolic extract of A. adscendens, based on bioactivity guided isolation. Conypododiol exhibited significant inhibition of both acetylcholinesterase and butyrylcholinesterase, having the IC(50) values 2.17 ± 0.1 µM and 11.21 ± 0.1 µM, respectively. IC(50) values of the standard compound Galanthamine for both the enzymes were 0.537 ± 0.018 µM and 8.6 ± 0.27 µM, respectively. Based on MTT cytotoxicity assay, Conypododiol was found safe against LCMK-2 monkey kidney epithelial cells and mice hepatocytes. Molecular docking studies revealed the hydrogen bonding interactions of Conypododiol with His440 and Ser200 at esteratic site (ES), and also with Tyr334 at peripheral anionic site (PAS) of the aromatic gorge of acetylcholinesterase. Simultaneous contacts of Conypododiol with PAS and ES shows its significance as a bivalent ligand. This preliminary study highlighted the potential of Conypododiol to be further developed and modified as new lead compound identified by its folk use.

In the cerate(IV) anion of the title salt, (C(2)H(10)N(2))(5)[Ce(2)(SO(4))(9)]·3H(2)O, the two metal atoms are bridged by three sulfate units; each metal atom is itself chelated by other three sulfate units so that the metal atoms exist in nine-coordinate tricapped trigonal-prismatic geometries. The anions, cations and uncoordinated water mol-ecules are linked by O-H⋯O and N-H⋯O hydrogen bonds, forming a three-dimensional network. One of the five cations is disordered with respect to the ethyl-ene portion in a 1:1 ratio.

Most neuronal Kv1 channels contain Kv1.1, Kv1.2 alpha, and Kvbeta2.1 subunits, yet the influences of their stoichiometries on properties of the (alpha)(4)(beta)(4) variants remain undefined. cDNAs were engineered to contain 0, 1, 2, or 4 copies of Kv1.1 with the requisite number of Kv1.2 and co-expressed in mammalian cells with Kvbeta2.1 to achieve "native-like" hetero-oligomers. The monomeric (Kv1.1 or 1.2), dimeric (Kv1.1-1.2 or 1.2-1.2), and tetrameric (Kv1.1-(1.2)(3)) constructs produced proteins of M(r) approximately 62,000, 120,000, and 240,000, which assembled into (alpha)(4)(beta)(4) complexes. Each alpha cRNA yielded a distinct K(+) current in oocytes, with voltage dependence of activation being shifted negatively as the Kv1.1 content in tetramers was increased. Channels containing 1, 2, or 4 copies of Kv1.1 were blocked by dendrotoxin k (DTX)(k) with similarly high potencies, whereas Kv(1.2)(4) proved nonsusceptible. Accordingly, Kv1.2/beta2.1 expressed in baby hamster kidney cells failed to bind DTX(k); in contrast, oligomers containing only one Kv1.1 subunit in a tetramer exhibited high affinity, with additional copies causing modest increases. Thus, one Kv1.1 subunit largely confers high affinity for DTX(k), whereas channel electrophysiological properties are tailored by the content of Kv1.1 relative to Kv1.2. This notable advance could explain the diversity of symptoms of human episodic ataxia I, which is often accompanied by myokymia, due to mutated Kv1.1 being assembled in different combinations with wild-type and Kv1.2.
