RESUMO

Serum samples collected from feral swine (Sus scrofa) throughout Florida (USA) from 1980 to 1989 were tested for antibodies to pseudorabies virus (PRV) by the serum neutralization test, the latex agglutination test, or by the enzyme-linked immunosorbent assay. Seropositive swine were detected at 11 of 13 sites with a composite seroprevalence of 34.8% (579 of 1,662 samples; range = 5.9% to 58.2%) for sites with seropositive swine. Data on age and sex of the swine were available from three sites. Seroprevalence in males and females did not differ significantly (P = 0.62 for the combined data). Seroprevalence in adult (> or = 8 mo) and juvenile swine (< 8 mo) was significantly different at all sites (P < 0.05 for the combined data). From these data, PRV infections appear to occur widely in populations of Florida feral swine and may seriously undermine efforts to eradicate this virus from the domestic swine population of the USA.

Sensitivity and specificity of an antigen-capture ELISA vs virus isolation in cell culture were evaluated for the detection of eastern equine encephalomyelitis (EEE) virus in the brain tissue of naturally infected equids. Brain specimens from 16 equids with neurologic disease were examined by ELISA and by inoculation onto baby hamster kidney cell cultures. Of 10 brain samples from which virus was isolated in the cell culture bioassay, all were correctly identified as containing EEE virus antigen by ELISA. None of the remaining 6 specimens, without detectable infectious EEE virus, contained detectable antigen. Sensitivity and specificity of the ELISA were 100% with no false-positive or false-negative results. The antigen-capture ELISA was a rapid, sensitive, specific, and simple alternative to a traditional bioassay for the detection of EEE virus.

The cryptic plasmid pRUT41 from Zymomonas mobilis was examined for its biological properties. This plasmid was found to be conjugally transferred from Z. mobilis CP4 to Escherichia coli BM21 and to carry genes for antibiotic resistance (gentamicin, kanamycin, and streptomycin). Covalently closed circular plasmid DNA was isolated from eight transconjugants of E. coli BM21. These plasmids were identical in mobility on agarose gels and exhibited the same restriction patterns as the native pRUT41 plasmid isolated from Z. mobilis. The plasmid location of the antibiotic resistance genes was further confirmed by transforming E. coli BM21 with isolated pRUT41 plasmid from strain CP4 and with plasmids from the transconjugants of BM21. Resistance to streptomycin, kanamycin, and gentamicin was tightly linked and transferred together in all cases.
