RESUMO

Fosmanogepix, a prodrug of Manogepix (MGX), is a groundbreaking antifungal agent with broad-spectrum activity against yeasts, including Cryptococcus and Candida, as well as molds. It exhibits effectiveness against drug-resistant strains, such as Candida strains resistant to echinocandins and Aspergillus strains resistant to azoles. Furthermore, fosmanogepix shows activity against pathogens that typically resist other classes of drugs, such as Scedosporium, Lomentospora prolificans, and Fusarium, although its efficacy against Mucorales varies. In animal models, fosmanogepix has demonstrated notable effectiveness against disseminated infections caused by various Candida species, Coccidioides immitis, and Fusarium solani. It has also shown efficacy in pulmonary infection models involving Aspergillus fumigatus, Aspergillus flavus, Scedosporium prolificans, Scedosporium apiospermum, and Rhizopus arrhizus. Clinical trials have revealed excellent oral bioavailability (>90%), enabling a seamless transition between intravenous and oral formulations without compromising blood concentrations. Fosmanogepix exhibits favorable profiles in terms of drug interactions, tolerability, and extensive distribution in various tissues, making it an appealing choice for treating invasive fungal infections. This comprehensive review aims to examine the outcomes of published data on fosmanogepix, encompassing in vitro, in vivo, and clinical investigations.

Background Lumbar puncture, a common diagnostic and therapeutic procedure, is performed regardless of individual spinal alignment variations. However, the impact of kyphosis, scoliosis, and kyphoscoliosis on spinal cord termination level and lumbar puncture safety remains unclear. Objectives This study aimed to determine if the termination level of the spinal cord is different in individuals with spinal deformities and to assess the necessity of routine neuroimaging for safe lumbar puncture localization. Study design and settings This single-center retrospective study was conducted at a university hospital using patients' electronic medical records. The study was focused on patients diagnosed with kyphosis, scoliosis, or kyphoscoliosis using spinal magnetic resonance imaging from January 2010 to December 2022. Participants We evaluated 240 patients: 120 with diagnosed spinal deformities (kyphosis, scoliosis, or kyphoscoliosis) and 120 without deformities, categorized by sex (deformed: 92 females, 28 males; non-deformed: 72 females, 48 males). Patients with spinal trauma, bleeding, or tumors were excluded. Results No statistically significant correlation was found between spinal deformities and spinal cord termination, with L1 remaining the most common endpoint in all groups. Conclusion Routine neuroimaging prior to lumbar puncture in patients with spinal deformities was not associated with a safer procedure due to no observed impact on the termination level of the spinal cord.

The fatigue crack growth rate (FCGR) of aluminium alloys under the combined influence of temperature and humidity remains a relatively unexplored area, receiving limited attention due to its intricate nature and challenges in predicting the combined impact of these factors. The challenge was to investigate and address the specific mechanisms and interactions between temperature and humidity, as in coastal environment conditions, on the FCGR of aluminium alloy. The present study conducts a comprehensive investigation into the combined influence of temperature and humidity on the FCGR of the Al6082 alloy. The fatigue pre-cracked compact tension specimens were corroded for 7 days and then subjected to various temperature and humidity conditions in a thermal chamber for 3 days to simulate coastal environments. The obtained data were analysed to determine the influence of temperature and humidity on the FCGR of the Al6082 alloy. An empirical model was also established to precisely predict fatigue life cycle values under these environmental conditions. The correlation between FCGR and fracture toughness models was also examined. The Al6082 alloy exhibits a 34% increase in the Paris constant C, indicating reduced FCGR resistance due to elevated temperature and humidity levels. At the same time, fatigue, corrosion, moisture-assisted crack propagation, and hydrogen embrittlement lead to a 27% decrease in threshold fracture toughness. The developed model exhibited accurate predictions for fatigue life cycles, and the correlation between fracture toughness and FCGR showed an error of less than 10%, indicating a strong relationship between these parameters.

Pineapple leaf fiber (PALF), striped sunflower seed fiber (SFSF), and watermelon seed (WMS) are considered natural waste polymer materials, which are biodegradable and sustainable. This study presents new novel thermal insulation and sound absorption materials using such waste as raw materials. PALF, SFSF, and WMS were used as loose, bound, and hybrid samples with different compositions to develop promising thermal insulation and sound-absorbing materials. Eleven sample boards were prepared: three were loose, three were bound, and five were hybrid between PALF with either SFSF or WMS. Wood adhesive was used as a binder for both the bound and hybrid sample boards. Laboratory scale sample boards of size 30 cm × 30 cm with variable thicknesses were prepared. The results show that the average thermal conductivity coefficient for the loose samples at the temperature range 20-80 °C is 0.04694 W/(m.K), 0.05611 W/(m.K), and 0.05976 W/m.K for PALF, SFSF, and WMS, respectively. Those for bound sample boards are 0.06344 W/(m.K), 0.07113 W/(m.K), and 0.08344 W/m.K for PALF, SFSF, and WMS, respectively. The hybrid ones between PALF and SFSF have 0.05921 W/m.K and 0.06845 W/(m.K) for two different compositions. The other hybrid between PALF and WMS has 0.06577 W/(m.K) and 0.07007 for two different compassions. The sound absorption coefficient for most of the bound and hybrid boards is above 0.5 and reaches higher values at some different frequencies. The thermogravimetric analysis for both SFSF and WMS shows that they are thermally stable up to 261 °C and 270 °C, respectively. The three-point bending moment test was also performed to test the mechanical properties of the bound and hybrid sample boards. It should be mentioned that using such waste materials as new sources of thermal insulation and sound absorption materials in buildings and other applications would lead the world to utilize the waste until zero agrowaste is reached, which will lower the environmental impact.

The emergence of Coronavrus Disease 2019 (COVID-19) has resulted in significant impacts on individuals and the whole world. It can lead to severe health outcomes that may lead to death. It also affects the psychological status and mental health of the individuals due to fear of infection, isolation, and quarantine. To assess the prevalence of depression, anxiety, and stress among the general population during the COVID-19 pandemic by reviewing the previous studies and original articles published on this subject. Both PubMed and Google Scholar databases were searched for scientific articles that reported the prevalence of depression, anxiety, and/or stress among the general population during the COVID-19 pandemic. The included terms used for the search process were "Prevalence, Anxiety, Depression, Stress, COVID-19, Pandemic, Psychological impact." The inclusion criteria were original articles written in the English language and conducted on the general population and reported the prevalence of depression, anxiety, and/or stress during COVID-19. A total of 2204 articles were obtained; only nine articles were eligible for the inclusion criteria. The included studies involved a total number of 19,277 participants and covered eight countries. The age range of participants was less than 18 years to 90 years, and female participants were more dominant compared to male participants in the majority of the studies. The prevalence of depression, anxiety, and stress was varied based on the region of studies and affected factors; however, the COVID-19 pandemic increased the prevalence of the three items.

The combined effect of temperature and humidity on the fracture toughness of aluminium alloys has not been extensively studied, and little attention has been paid due to its complexity, understanding of its behaviour, and difficulty in predicting the effect of the combined factors. Therefore, the present study aims to address this knowledge gap and improve the understanding of the interdependencies between the coupled effects of temperature and humidity on the fracture toughness of Al-Mg-Si-Mn alloy, which can have practical implications for the selection and design of materials in coastal environments. Fracture toughness experiments were carried out by simulating the coastal environments, such as localised corrosion, temperature, and humidity, using compact tension specimens. The fracture toughness increased with varying temperatures from 20 to 80 °C and decreased with variable humidity levels between 40% and 90%, revealing Al-Mg-Si-Mn alloy is susceptible to corrosive environments. Using a curve-fitting approach that mapped the micrographs to temperature and humidity conditions, an empirical model was developed, which revealed that the interaction between temperature and humidity was complex and followed a nonlinear interaction supported by microstructure images of SEM and collected empirical data.

Polymethyl methacrylate (PMMA) is often used in restorative dentistry for its easy fabrication, aesthetics, and low cost for interim restorations. However, poor mechanical properties to withstand complex masticatory forces are a concern for clinicians. Therefore, this study aimed to modify a commercially available PMMA-based temporary restorative material by adding TiO2 and ZrO2 nanoparticles in different percentages as fillers and to investigate its physio-mechanical properties. Different percentages (0, 0.5, 1.5, and 3.0 wt%) of TiO2 and ZrO2 nanoparticles were mixed with the pristine PMMA resin (powder to liquid ratio: 1 : 1) and homogenized using high-speed mixer. The composites obtained were analyzed for their flexural strength (F.S.), elastic modulus (E.M.), Vickers hardness (H.V.), surface roughness Ra, morphology and water contact angle (WCA). The mean average was determined with standard deviation (SD) to analyze the results, and a basic comparison test was conducted. The results inferred that adding a small amount (0.5 wt%) of TiO2 and ZrO2 nanoparticles (NPs) could significantly enhance the physio-mechanical and morphological characteristics of PMMA interim restorations. EM and surface hardness increased with increasing filler content, with 3.0 wt.% ZrO2 exhibiting the highest EM (3851.28 MPa), followed by 3.0 wt.% TiO2 (3632.34 MPa). The WCA was significantly reduced from 91.32 ± 4.21° (control) to 66.30 ± 4.23° for 3.0 wt.% ZrO2 and 69.88 ± 3.55° for 3.0 wt.% TiO2. Therefore, TiO2 and ZrO2 NPs could potentially be used as fillers to improve the performance of PMMA and similar interim restorations.

AIM: To investigate push-out bond strength (PBS) of fiber post to radicular dentin after using different cementation techniques. MATERIAL AND METHODS: Sixty single-rooted premolars were disinfected and cleaned by mechanical instrumentation. S1, S2, and SX were used for canal shaping and finishing of the canal was carried out using F1 and F2. This was followed by constant irrigation and smear layer removal using 17%EDTA. The canal was dried filled with gutta-percha and canal space was prepared using a peso reamer. Based on cementation techniques, samples were randomly allocated into six groups. Group 1: One-step Monoblock MC; Group 2: One-step, Monoblock MC-NA (no adhesive) GFP; Group 3: One-step, RX-MC-Monoblock; Group 4: Two-step, RX-MC; Group 5: Two-step, RX-FZ; and Group 6: Two-step RX-FZ-custom post. All specimens were mounted in polyvinyl pipes using acrylic resin up to cement enamel junction. All specimens were mounted in a universal testing machine subjected to push-out forces at a speed of 0.5 mm/min. Five samples from each group were sputter-coated with 6 nm gold thickness for 300 s at 250 mA. The coated specimens were assessed under the scanning electron microscope (SEM). Analysis of variance (ANOVA) and Tukey Kramer multiple comparisons tests were performed to compare means among groups maintaining the level of significance at (p < 0.001). RESULTS: The highest PBS was displayed in RX-MC-Monoblock (199.020 ± 21.432 MPa). Whereas, lowest PBS was found in Monoblock MC-NA (no adhesive)-GFP (76.440 ± 9.468 MPa). Among one-step groups, RX-MC Monoblock exhibited the highest PBS (199.020 ± 21.432 MPa) comparable to one-step Monoblock MC (134.28 ± 19.37 MPa) (p > 0.05). Similarly, among two-steps groups, two-step RX-MC demonstrated significantly higher PBS values than two-step RX-FZ (143.340 ± 23.68 MPa) and RX-FZ-custom post (86.90 ± 7.41 MPa) (p < 0.05). CONCLUSION: One-step RX-MC-Monoblock technique using self-adhesive cement and core foundation composite resin material multicore flow when cured simultaneously exhibited the highest bond integrity of post retention compared to other cementation technique.
