RESUMO

The effect of R-plasmid on the qualitative and quantitative compositions of phospholipids and their fatty acids in bacterial (S. derby) cells has been studied. It has been found that the lack of R-plasmid in bacterial cells leads to a sharp decrease in the phospholipid content, in the phosphatidylcholine content in particular, and causes an increase in the per cent content of lysophosphatidylcholines. The per cent content of saturated and nonsaturated fatty acids of phospholipids in plasmid-free cells does not change appreciably. In the plasmid-free strain of S. derby the intensity of lipid peroxidation is associated with a relatively high content of alpha-tocopherol.

RESUMO

An experimental alcohol intoxication was accompanied by an increase in content of total phospholipids and phosphatide acid in rat brain with simultaneous decrease in content of free ethanolamine. The phenomenon demonstrate the activation of phosphatidogenesis. In the liver tissue an opposite alterations were observed; the lipolytic reactions were activated.

RESUMO

A distinct increase in activity of glycerokinase and glycerophosphate dehydrogenase and in content of L-alpha-glycerophosphate were observed in brain and liver tissues of rats treated with ethanol for 40 days. The similar alterations were observed after treatment with ehtanol for 80 days but in these conditions the content of L-alpha-glycerophosphate in liver tissue was decreased.

RESUMO

A simple modification of the method of Rossiter and Palmer for the quantitative determination of phosphoinositides (PI) in animal tissues (brain and liver) is worked out: Phospholipids are mineralyzed (together with a paper) in 57% perchloric acid, avoiding the time consuming procedure of the tenfold elution of inositids spots from chromatogramms and the subsequent evaporation of the substantial volume of the eluate obtained. The time of the analysis is twice as short as that of the original method, the data obtained being reliable. The quantity of mono-, di- and triphosphoinositides in rat brain is as follows: 85.08, 11.33 and 32.85 mkg P/g wet tissue, but in liver it is 94.5, 1.88 and 2.5 mkg P/g wet tissue respectively.

RESUMO

Alcohol intoxication induced in the rat brain significant decrease in the monophosphoinositides level, but the di- and triphosphoinositides quantity did not change; the content of all the three studied fractions of the inositol-containing phospholipids in the liver decreased. The tendency to retain the normal level of di- and triphosphoinositides in the brain following alcohol intoxication may be considered as an important factor providing physiological condition of the brain metabolism.

Phosphatidic acid, total and individual phospholipids and ethanolamine levels of rat brain and liver are changed considerably following 40 and even more so after 80, days of alcohol intoxication. In brain the content of phosphatidic acid and total and individual phospholipids is increased and that of ethanolamine reduced. In liver exactly the opposite changes take place with the exception of inositol containing phospholipids, which are considerably reduced both in brain and liver. The study of the first stages of phospholipid synthesis showed that during 40 and 80 days of alcohol intoxication activity of enzymes participating in the metabolism of L-alpha-glycerophosphate (glycerokinase, glycerophosphate dehydrogenase) is increased several fold. This brings to the increase of L-alpha-glycerophosphate, the main product of phosphatidogenesis, through activation of its formation by glycerokinase and glycerophosphate dehydrogenase. Inspite of the fact that in liver activity of enzymes taking part in the metabolism of L-alpha-glycerophosphate is considerably increased during chronic alcohol intoxication its level is not raised. This indicates an activation of phosphatidic acid formation from L-alpha-glycerophosphate and the immediate inclusion of phosphatidic acid in the biosynthesis of neutral fats. The data obtained indicate that chronic alcohol intoxication activates biosynthetic processes of phospholipids in brain and accelerates their break down in liver, bringing to the fatty infiltration of liver.
