RESUMO

Cutaneous leishmaniasis caused by Leishmania parasites exhibits a wide range of clinical manifestations. Although parasites influence disease severity, cytolytic CD8 T cell responses mediate disease. While these responses originate in the lymph node, we found that expression of the cytolytic effector molecule granzyme B was restricted to lesional CD8 T cells in Leishmania-infected mice, suggesting that local cues within inflamed skin induced cytolytic function. Expression of Blimp-1 (Prdm1), a transcription factor necessary for cytolytic CD8 T cell differentiation, was driven by hypoxia within the inflamed skin. Hypoxia was further enhanced by the recruitment of neutrophils that consumed oxygen to produce reactive oxygen species and ultimately increased the hypoxic state and granzyme B expression in CD8 T cells. Importantly, lesions from cutaneous leishmaniasis patients exhibited hypoxia transcription signatures that correlated with the presence of neutrophils. Thus, targeting hypoxia-driven signals that support local differentiation of cytolytic CD8 T cells may improve the prognosis for patients with cutaneous leishmaniasis, as well as other inflammatory skin diseases where cytolytic CD8 T cells contribute to pathogenesis.

Cytolytic CD8+ T cells mediate immunopathology in cutaneous leishmaniasis without controlling parasites. Here, we identify factors involved in CD8+ T cell migration to the lesion that could be targeted to ameliorate disease severity. CCR5 was the most highly expressed chemokine receptor in patient lesions, and the high expression of CCL3 and CCL4, CCR5 ligands, was associated with delayed healing of lesions. To test the requirement for CCR5, Leishmania-infected Rag1-/- mice were reconstituted with CCR5-/- CD8+ T cells. We found that these mice developed smaller lesions accompanied by a reduction in CD8+ T cell numbers compared to controls. We confirmed these findings by showing that the inhibition of CCR5 with maraviroc, a selective inhibitor of CCR5, reduced lesion development without affecting the parasite burden. Together, these results reveal that CD8+ T cells migrate to leishmanial lesions in a CCR5-dependent manner and that blocking CCR5 prevents CD8+ T cell-mediated pathology.

As global effects of water scarcity raise concerns and environmental regulations evolve, contemporary wastewater treatment plants (WWTPs) face the challenge of effectively removing a diverse range of contaminants of emerging concern (CECs) from municipal effluents. This study focuses on the assessment of advanced oxidation processes (AOPs), specifically UV-C/H2O2 and UV-C/Chlorine, for the removal of 14 target CECs in municipal secondary effluent (MSE, spiked with 10 µg L-1 of each CEC) or in the subsequent MSE nanofiltration retentate (NFR, no spiking). Phototreatments were carried out in continuous mode operation, with a hydraulic retention time of 3.4 min, using a tube-in-tube membrane photoreactor. For both wastewater matrices, UV-C photolysis (3.3 kJ L-1) exhibited high efficacy in removing CECs susceptible to photolysis, although lower treatment performance was observed for NFR. In MSE, adding 10 mg L-1 of H2O2 or Cl2 enhanced treatment efficiency, with UV-C/H2O2 outperforming UV-C/Chlorine. Both UV-C/AOPs eliminated the chronic toxicity of MSE toward Chlorella vulgaris. In the NFR, not only was the degradation of target CECs diminished, but chronic toxicity to C. vulgaris persisted after both UV-C/AOPs, with UV-C/Chlorine increasing toxicity due to potential toxic by-products. Nanofiltration permeate (NFP) exhibited low CECs and microbial content. A single chlorine addition effectively controlled Escherichia coli regrowth for 3 days, proving NFP potential for safe reuse in crop irrigation (<1 CFU/100 mL for E. coli; <1 mg L-1 for free chlorine). These findings provide valuable insights into the applications and limitations of UV-C/H2O2 and UV-C/Chlorine for distinct wastewater treatment scenarios.

This study assessed the occurrence of five antibiotics, three hormones, caffeine, and long and short-chain perfluoroalkyl and polyfluoroalkyl substances (PFASs) in surface water and feedstuff samples obtained from aquaculture cages in Três Marias reservoir in Brazil. This is the first work to evaluate the presence of PFAS in surface water used for aquaculture in Brazil. Solid-phase extraction and low temperature partitioning extraction followed by liquid chromatography coupled to mass spectrometry (LC-MS) were performed to process and analyze surface water samples and feedstuff, respectively. The ecotoxicological risk quotient was calculated for target compounds detected in water. Ciprofloxacin and caffeine were detected in all surface water samples. Pharmaceutical drugs ranged from 0.7 ng L-1 (trimethoprim) to 389.2 ng L -1 (ß-estradiol). Estrone (10.24 ng g-1) and ß-estradiol (66.20 ng g-1) were also found in feedstuff. Four PFASs (PFOA, PFDoA, PFTeDA, and PFBS) were detected (9.40-15.2 µg L-1) at levels higher than reported in studies conducted worldwide. Ecotoxicological risk assessment indicated high risks for caffeine and PFOA, PFDoA, and PFTeDA with RQ values from 10 to 103. These findings reveal risks to biodiversity, ecosystem integrity and human health considering possible intake of these contaminants by fish consumption due to potential bioaccumulation of these substances. Hence, it is critical to conduct more studies in this direction in Brazil and other low and middle-low-income countries.

OBJECTIVE: This study aimed to evaluate, qualify, and summarize evidence presented in systematic reviews (SR) on treatments for bruxism. METHODS: The overview was conducted using the PICO strategy: children, adolescents, and adults with bruxism (P) were submitted to different treatments (I) compared to other treatments, placebo, or no treatment (C) in order to evaluate incidence, prevalence, and number of episodes of bruxism (O). The search was carried out in six databases and gray literature up to July 2023. Data were extracted, and the ROBS tool was used, followed by a descriptive synthesis of the results. RESULTS: A total of 31 SR were included. Sixteen showed a positive effect on episodes of bruxism (BE), while two had negative, one had neutral, and nine had inconclusive effects. Using the risk of bias in systematic reviews tool (ROBIS), risk of bias varied from low (n = 23) to high (n = 5) among the SR. Pharmacological treatment, oral rehabilitation, and other therapeutic approaches presented inconclusive or negative effects on BE, while oral appliances showed controversial effects. Biofeedback, physical therapy, laser therapy, and botulinum toxin showed positive effects on the reduction of BE. CONCLUSION: Biofeedback, physical therapy, laser therapy, and botulinum toxin showed positive effects on the reduction of BE; there is still a lack of studies to support the safe and long-term use of these therapies. Hydroelectric power is the main source of electrical energy in Brazil. Electrical energy providers have the duty to monitor water quality in reservoirs to preserve water quality and support best management practices that enable multiple water uses, including fish production. In this context, the objectives of this study were (i) to perform a historical evaluation of water quality in Três Marias Reservoir, (ii) to present an optimization of the water quality monitoring network, and (iii) to evaluate the evolution and impact of fish farming upon surface water quality by using secondary data measured in situ and remote sensing. A systematic approach was applied to analyze historical water quality data. Principal component analysis (PCA) and cluster analysis (CA) were applied to identify the most important parameters and monitoring points. Images obtained from Sentinel 2 were treated by contrast to quantify simple and weighted densities of fish farming activities in the region while regression analysis was performed to verify correlations between these densities and water quality parameters. Results showed that the pH and total suspended solids were the most important parameters for characterizing water quality, especially near tributaries, and that monitoring points could be grouped into three clusters (upstream, central, and downstream regions) with distinct water quality conditions. The PCA indicated that there is no redundance among parameters nor monitoring stations and that areas near tributaries must be prioritized for monitoring as these are important sources of suspended solids. Remote sensing images showed that the area occupied by fish farms has increased in the reservoir from 2016 to 2022 and the methodology used for this purpose in this study may be applied to other bodies of water. Chlorophyll-a showed a direct relationship with the density of fish farms indicating a possible influence of nutrient input to the reservoir by this activity. These results provide valuable information to support decision-making related to water management in the reservoir.

Este estudo teve por objetivo avaliar e analisar as informações acerca do tema bráquetes autoligados publicados na plataforma YouTube. Realizou-se busca na plataforma entre os dias 10 e 15 de janeiro de 2022 com o intuito de identificar os vídeos mais relevantes utilizando-se o termo "bráquete autoligado". Atribuiu-se um critério de 7 pontos para classificar os vídeos como satisfatórios e insatisfatórios. Uma escala global de qualidade (QGS) foi utilizada para examinar a qualidade dos vídeos. Para avaliar a confiabilidade, foi utilizado um questionário adaptado do DISCERN de 5 questões. Foram convidados dois ortodontistas experientes na área para avaliarem os vídeos, correlacionar e, posteriormente, comparar as informações. Inicialmente foram encontrados 100 vídeos e, após aplicação dos critérios de exclusão, chegou--se a 27 vídeos. A maioria das mídias foram criadas por ortodontistas. Os vídeos criados por generalistas obtiveram melhor escore e também houve diferença significativa no número de comentários e taxa de interação p<0,05. Não foi observada diferença entre os escores e a origem dos vídeos p>0,05. A média do escore GQS foi de 2,25, do DISCERN 1,18, e 19 vídeos foram classificados como suficientes e 8 vídeos como insuficientes. Concluiu-se que a maioria dos vídeos não exibiu uma fonte confiável de informação e a origem das mídias não influenciou os resultados dos critérios de conteúdos nem de confiabilidade.(AU)


This study aimed to evaluate the information on the subject of self-ligating brackets published on the YouTube platform. A search was carried out on the platform between January 10 and 15, 2022 in order to identify the most relevant videos using the term "self-ligating bracket". A criterion of 7 points was assigned to classify the videos as satisfactory and unsatisfactory. A global quality scale (QGS) was used to examine the quality of the videos. To assess reliability, an adapted 5-question DISCERN questionnaire was used. Two experienced orthodontists in the field were invited to evaluate the videos, correlate and later compare the information. Initially, 100 videos were found and, after applying the exclusion criteria, 27 videos were reached. Most media were created by orthodontists. Videos created by generalists had a better score and there was also a significant difference in the number of comments and interaction rate p<0.05. No difference was observed between the scores and the origin of the videos p>0.05. The average GQS score was 2.25, from DISCERN 1.18, and 19 videos were classified as sufficient and 8 videos as insufficient. It was concluded that most videos did not show a reliable source of information and the origin of the media did not influence the results of the content or reliability criteria(AU)

This manuscript critically reviews the state of the art on the application of photo-Fenton processes irradiated by light-emitting diode arrays (LED) with a focus on the removal of contaminants of emerging concern (CEC) from aqueous matrices. LEDs are clean, low-cost radiation sources with longer lifespan compared to mercury lamps. This study covers the influence of LED sources, wavelengths, and dose upon CEC removal, and the potential for disinfection, abatement of antibiotic-resistant bacteria (ARB), and genes (ARG). The bibliographic search was performed in Scopus database using keyword combinations and resulted in a portfolio containing 52 relevant articles published between 2010-2023. According to reviewed papers, LED photoreactor design has evolved in the past decade aiming to improve CEC degradation in aqueous matrices while reducing construction and operation costs, and energy consumption. Among several reactors (annular, fluidized bed, parallel plate, wireless, pathway systems, and microreactor) surveyed for their performance and scalability, LED chips and strips are particularly suitable for application due to their wide emission angle (≈120°) and small size (mm2), which allow for, respectively, efficient illumination coverage and flexible arrangement and design. LED microreactors are very efficient in the degradation of contaminants and scalable with reduced area requirements. Although most studies were performed in synthetic solutions and at laboratory scale, the externally LED irradiated cylindrical reactor was successful for application in full-scale municipal water treatment plants. Future studies should focus on evaluating CEC removal in wastewater using scalable devices for continuous operation of solar photo-Fenton at night.

Leishmania braziliensis is a parasitic infection that can result in inflammation and skin injury with highly variable and unpredictable clinical outcomes. Here, we investigated the potential impact of microbiota on infection-induced inflammatory responses and disease resolution by conducting an integrated analysis of the skin microbiome and host transcriptome on a cohort of 62 patients infected with L. braziliensis. We found that overall bacterial burden and microbiome configurations dominated with Staphylococcus spp. were associated with delayed healing and enhanced inflammatory responses, especially by IL-1 family members. Quantification of host and bacterial transcripts on human lesions revealed that high lesional S. aureus transcript abundance was associated with delayed healing and increased expression of IL-1ß. This cytokine was critical for modulating disease outcomes in L. braziliensis-infected mice colonized with S. aureus, given that its neutralization reduced pathology and inflammation. These results highlight how the human microbiome can shape disease outcomes in cutaneous leishmaniasis and suggest pathways toward host-directed therapies to mitigate the inflammatory consequences.

Cutaneous leishmaniasis causes alterations in the skin microbiota, leading to pathologic immune responses and delayed healing. However, it is not known how these microbiota-driven immune responses are regulated. Here, we report that depletion of Foxp3+ regulatory T cells (Tregs) in Staphylococcus aureus-colonized mice resulted in less IL-17 and an IFN-γ-dependent skin inflammation with impaired S. aureus immunity. Similarly, reducing Tregs in S. aureus-colonized and Leishmania braziliensis-infected mice increased IFN-γ, S. aureus, and disease severity. Importantly, analysis of lesions from L. braziliensis patients revealed that low FOXP3 gene expression is associated with high IFNG expression, S. aureus burden, and delayed lesion resolution compared to patients with high FOXP3 expression. Thus, we found a critical role for Tregs in regulating the balance between IL-17 and IFN-γ in the skin, which influences both bacterial burden and disease. These results have clinical ramifications for cutaneous leishmaniasis and other skin diseases associated with a dysregulated microbiome when Tregs are limited or dysfunctional.

Cutaneous leishmaniasis exhibits a spectrum of clinical presentations dependent upon the parasites' persistence and host immunopathologic responses. Although cytolytic CD8 T cells cannot control the parasites, they significantly contribute to pathologic responses. In a murine model of cutaneous leishmaniasis, we previously found that NKG2D plays a role in the ability of cytolytic CD8 T cells to promote disease in leishmanial lesions. Here, we investigated whether NKG2D plays a role in human disease. We found that NKG2D and its ligands were expressed within lesions from L. braziliensis-infected patients and that IL-15 and IL-1ß were factors driving NKG2D and NKG2D ligand expression, respectively. Blocking NKG2D reduced degranulation by CD8 T cells in a subset of patients. Additionally, our transcriptional analysis of patients' lesions found that patients who failed the first round of treatment exhibited higher expression of KLRK1, the gene coding for NKG2D, than those who responded to treatment. These findings suggest that NKG2D may be a promising therapeutic target for ameliorating disease severity in cutaneous leishmaniasis caused by L. braziliensis infection.

Monitoring water quality in reservoirs is essential for the maintenance of aquatic ecosystems and socioeconomic services. In this scenario, the observation of abrupt elevations of physicochemical parameters, such as turbidity and other indicators, can signal anomalies associated with the occurrence of critical events, requiring operational actions and planning to mitigate negative environmental impacts on water resources. This work aims to integrate Machine Learning methods specialized in anomaly detection with data obtained from remote sensing images to identify with high turbidity events in the surface water of the Três Marias Hydroelectric Reservoir. Four distinct threshold-based scenarios were evaluated, in which the overall performance, based on F1-score, showed decreasing trends as the thresholds became more restrictive. In general, the anomaly identification maps generated through the models ratified the applicability of the methods in the diagnosis of surface water in reservoirs in distinct hydrological contexts (dry and wet), effectively identifying locations with anomalous turbidity values.

In Brazil, Leishmania braziliensis is the main causative agent of the neglected tropical disease, cutaneous leishmaniasis (CL). CL presents on a spectrum of disease severity with a high rate of treatment failure. Yet the parasite factors that contribute to disease presentation and treatment outcome are not well understood, in part because successfully isolating and culturing parasites from patient lesions remains a major technical challenge. Here we describe the development of selective whole genome amplification (SWGA) for Leishmania and show that this method enables culture-independent analysis of parasite genomes obtained directly from primary patient skin samples, allowing us to circumvent artifacts associated with adaptation to culture. We show that SWGA can be applied to multiple Leishmania species residing in different host species, suggesting that this method is broadly useful in both experimental infection models and clinical studies. SWGA carried out directly on skin biopsies collected from patients in Corte de Pedra, Bahia, Brazil, showed extensive genomic diversity. Finally, as a proof-of-concept, we demonstrated that SWGA data can be integrated with published whole genome data from cultured parasite isolates to identify variants unique to specific geographic regions in Brazil where treatment failure rates are known to be high. SWGA provides a relatively simple method to generate Leishmania genomes directly from patient samples, unlocking the potential to link parasite genetics with host clinical phenotypes.

Dual RNA-seq of human lesions revealed that high lesional S. aureus transcript abundance was associated with delayed healing and increased expression of IL-1ß. This cytokine was critical for modulating disease outcome in L. braziliensis -infected mice colonized with S. aureus , as its neutralization reduced pathology and inflammation. These results implicate the microbiome in cutaneous leishmaniasis disease outcomes in humans and suggest host-directed therapies to mitigate the inflammatory consequences.

This study aimed to characterize the profile of dental anxiety in pediatric patients, identifying the effect exerted by socioeconomic factors using dental data. A cross-sectional study design with a sample of 120 children aged 7-12 years old was used. Data relating to anxiety levels prior to dental care, socioeconomic aspects (family income, education level, child's school type), and child's dental history (previous dental appointments, previous treatment, caries experience) were collected. Additionally, participants completed the Brazilian version of the Children's Fear Survey Schedule- Dental Subscale (B-CFSS-DS) to assess dental anxiety. Descriptive analyses, chi-squared (X 2 ) tests, and Mann-Whitney U tests were performed, with a significance level of 5%. A total of 51 boys (42.5%) and 69 girls (57.5%) were included. There was no significant difference in dental anxiety between them. However, younger children had higher mean B-CFSS-DS scores (p = 0.036, Mann-Whitney). A higher prevalence of dental anxiety was found in participants from low-income families (p = 0.012, X 2 ) and in patients who did not receive endodontic treatment (p=0.034, X 2 ). Higher mean B-CFSS-DS scores were also observed in participants who did not receive endodontic treatment (p=0.001, Mann-Whitney) compared with those that did receive endodontic treatment. No relationship was found between education level, patient school type, first dental appointment, caries experience, and dental anxiety data. Younger children presented a profile of greater dental anxiety. Socioeconomic factors and dental data exerted some effect on dental anxiety, where children from low-income families and those not subjected to endodontic treatment displayed higher rates of dental anxiety.

The application of solar photo-Fenton as post-treatment of municipal secondary effluents (MSE) in developing tropical countries is the main topic of this review. Alternative technologies such as stabilization ponds and upflow anaerobic sludge blanket (UASB) are vastly applied in these countries. However, data related to the application of solar photo-Fenton to improve the quality of effluents from UASB systems are scarce. This review gathered main achievements and limitations associated to the application of solar photo-Fenton at neutral pH and at pilot scale to analyze possible challenges associated to its application as post-treatment of MSE generated by alternative treatments. To this end, the literature review considered studies published in the last decade focusing on CECs removal, toxicity reduction and disinfection via solar photo-Fenton. Physicochemical characteristics of effluents originated after UASB systems alone and followed by a biological post-treatment show significant difference when compared with effluents from conventional activated sludge (CAS) systems. Results obtained for solar photo-Fenton as post-treatment of MSE in developed countries indicate that remaining organic matter and alkalinity present in UASB effluents may pose challenges to the performance of solar advanced oxidation processes (AOPs). This drawback could result in a more toxic effluent. The use of chelating agents such as Fe3+-EDDS to perform solar photo-Fenton at neutral pH was compared to the application of intermittent additions of Fe2+ and both of these strategies were reported as effective to remove CECs from MSE. The latter strategy may be of greater interest in developing countries due to costs associated to complexing agents. In addition, more studies are needed to confirm the efficiency of solar photo-Fenton on the disinfection of effluent from UASB systems to verify reuse possibilities. Finally, future research urges to evaluate the efficiency of solar photo-Fenton at natural pH for the treatment of effluents from UASB systems.

This work presents data regarding the occurrence and treatment of Contaminants of Emerging Concern (CECs) in Brazil in the past decade. The literature review (2011-2021) revealed the detection of 87 pharmaceutical drugs and personal care products, 58 pesticides, 8 hormones, 2 illicit drugs, caffeine and bisphenol A in distinct matrices (i.e.: wastewater, groundwater, sea water, rainwater, surface water, drinking water and hospital effluent). Concentrations of CECs varied from ng-µg L-1 depending on the location, compound and matrix. The inefficiency of conventional wastewater treatment methods on the removal of CECs and lack of basic sanitation in some regions in the country aggravates contamination of Brazilian aquatic environments and poses potential environmental and health risks. Advanced oxidation processes (AOPs) are pointed out as viable and efficient alternatives to degrade CECs and prevent environmental contamination. A total of 375 studies involving the use of AOPs in Brazilian aqueous matrices were published in the last decade. Fenton and photo-Fenton processes, photo-peroxidation, ozonation, electrochemical advanced oxidation and heterogeneous photocatalysis are some of the AOPs applied by Brazilian research groups. Although many works discuss the importance of applying these technologies for CECs removal in real treatment plants, most of these studies assess the treatment of distilled water or simulated effluent. Therefore, the conduction of studies applying AOPs in real matrices are critical to drive the implementation of these processes coupled to conventional water and wastewater treatment in real plants in order to prevent the contamination of environmental matrices by CECs in Brazil.

Oobjetivo deste estudo foi desenvolver, verificar a validação do conteúdo e avaliar a eficácia de umfolhetoeducativo sobre protetores bucais (PBs)paracrianças e adolescentes (C/A)com idadeentre oito equinze anos. As etapas de desenvolvimento e validação do conteúdo foram realizadas após pesquisa bibliográfica na literatura baseada em evidências sobre o assunto, e avaliação de estudantesde pós-graduação e/ou professores de odontologia, especialistas em educação, bem como porC/A (público-alvo). Foramcalculadoso índice de validade do conteúdo (IVC) para cada item (I-IVC) e o índice de validade total do conteúdo do folheto(F-IVC). Um questionário foi elaborado com questões relacionadas ao conteúdo do folhetoe a eficácia da aquisição de conhecimentos foi avaliada por meiode testes para dados pareados, considerando as respostasdasC/A antes e depois da leitura dofolheto. Modelos lineares generalizados e mistosforam utilizadospara avaliar associações não ajustadas e ajustadas, respectivamente(p<0,05). O poder da amostratambém foi calculado. Os itens foram considerados relevantes, representativos e com boa operacionalidade, e válidos comI-IVCe F-IVCde 1.0. Quarenta e quatro C/A com 10,9±2,2 anos foram incluídos e, apósaleitura do folheto, a média de respostas corretas teve um aumento significativo (p<0,001). No modelo ajustado, a "leitura do folheto" influenciou positivamente no conhecimento sobre PB (p<0,001). Um poder de amostragem de 100%foi obtido. Ofolhetoapresentou um conteúdo de validade satisfatória e deve ser consideradoum instrumento capaz de aumentar o conhecimento de C/A sobre PB (AU).


The aim of this study was to develop, verify the content validity and evaluate the effectiveness of an educational folder about MG for children and adolescents (C/A)between the ages of eight and fifteen. The steps of development and content validity were carried out after a bibliographic search for an evidence-based literature on the subject, and the evaluation ofMSc or PhD dentistry students and/or professors, education experts, as well as C/A (target group).The content validity index (CVI) for each item (I-CVI) and the total folder content validity index (F-CVI) after the PhD professors (judges) analysis was calculated. A questionnaire with assertions related to the content of the folder (indication, types and storage of MG, myths and truths about the device) was created, and the effectiveness of knowledge acquisition was evaluated by applying tests for paired data, considering C/Aresponses before and after reading the folder. Generalized and mixed linear model were used to evaluate unadjusted and adjusted associations, respectively. The sample power was also calculated. The I-CVI and F-CVI were 1.0, showing that the items were considered relevant, representative, showed good operability and were valid. Forty-four C/Aof 10.9±2.24 yearswere included and, after reading the folder, the mean of correct answers had a significant increase (p<0.001). In the adjusted model, "reading the folder" influenced positively on the knowledge about MG (p<0.001).Sample power of 100% was obtained. The folder presented satisfactory content validity and should be considered as an instrument capable of increasing the knowledge of C/Aabout MG (AU).
