RESUMO

Gulf War Illness (GWI) describes a series of symptoms suffered by veterans of the Gulf war, consisting of cognitive, neurological and gastrointestinal dysfunctions. Two chemicals associated with GWI are the insecticide permethrin (PER) and the nerve gas prophylactic pyridostigmine-bromide (PB). In this study we assessed the effects of PER and PB exposure on the pathology and subsequent alcohol (EtOH)-induced liver injury, and the influence of a macrophage depletor, PLX3397, on EtOH-induced liver damage in PER/PB-treated mice. Male C57BL/6 mice were injected daily with vehicle or PER/PB for 10 days, followed by 4 months recovery, then treatment with PLX3397 and a chronic-plus-single-binge EtOH challenge for 10 days. PER/PB exposure resulted in the protracted increase in liver transaminases in the serum and induced chronic low-level microvesicular steatosis and inflammation in GWI vs Naïve mice up to 4 months after cessation of exposure. Furthermore, prior exposure to PER/PB also resulted in exacerbated response to EtOH-induced liver injury, with enhanced steatosis, ductular reaction and fibrosis. The enhanced EtOH-induced liver damage in GWI-mice was attenuated by strategies designed to deplete macrophages in the liver. Taken together, these data suggest that exposure to GWI-related chemicals may alter the liver's response to subsequent ethanol exposure.

The aim of the present experiment was to determine the standardized ileal digestible (SID) methionine (Met) requirement during the grower phase (22 to 29 days). A total of 192 Ross 308 male broilers (745 ± 50.5 g) were assigned to six dietary treatments in a randomized complete block design, with eight replicate cages per treatment. The experimental diets were formulated based on corn, soybean meal, and synthetic amino acids (AA), with different levels of dietary SID Met ranging from 0.40% to 0.65%, in increments of 0.05%. Individual body weight and feed intake per cage were measured at the beginning and end of the experimental period. Weight gain (g/bird), feed intake (g/bird), gain-to-feed ratio (g/kg), feather weight (g), and relative feather percentage (% of body weight) were analyzed for linear or quadratic responses using orthogonal polynomial contrasts. Final body weight, weight gain, and feed intake increased quadratically as dietary SID Met concentration increased (p < 0.05). However, the composition of AA in the feathers showed no quadratic or linear effect based on dietary SID Met concentrations, except for Ile (p = 0.05; quadratic). The SID Met requirements for optimum and maximum weight gain of 22 to 29-days-old male broilers were estimated using the broken-line and quadratic-line models to 0.45% and 0.55%, respectively.

Objective: The objective was to investigate growth performance, antioxidant enzyme activity, intestinal morphology, immune cell distribution, short chain fatty acid (SCFA) profile, and microbiota in broiler chickens fed a diet containing Lacticaseibacillus paracasei NSMJ15. Methods: A total of 120-day-old Ross 308 male broilers were allocated to 2 dietary treatments in a randomized complete block design. A control group was fed a corn-soybean meal control diet, and an NSMJ15-supplemented group was fed a control diet supplemented with 1 g/kg L. paracasei NSMJ15 at the expense of cornstarch. Each dietary treatment had 6 replicates with 10 birds per cage. Growth performance was recorded on day 9. On day 10, one bird representing median body weight was selected to collect serum for antioxidant enzyme activity, jejunal tissue for immune cell isolation and morphometric analysis, and cecal digesta for 16S rRNA gene sequencing and SCFA analysis. Results: Supplementation of L. paracasei NSMJ15 did not affect growth performance, serum antioxidant enzyme activity, and jejunal histomorphology compared to the control group. In the NSMJ15-supplemented group, the population of CD3+CD4+CD8- T cells increased (p=0.010), while the population of CD3+CD8+TCRγδ+ T cells decreased (p=0.022) compared to the control group. The L. paracasei NSMJ15 supplementation decreased (p=0.022) acetate concentration in the cecal digesta compared to the control group. The 16S rRNA gene sequencing analysis showed that NSMJ15-supplemented group differentially expressed (p<0.05) 10 more amplicon sequence variants compared to control group without affecting alpha and beta diversity indices of the cecal microbiota. Genera Mediterraneibacter and Negativibacillus were positively (p<0.05) correlated with CD4+ T cells, while genera Gemmiger, Coprococcus, Sellimonas, Massilimicrobiota, and Blautia were negatively (p<0.05) correlated with SCFA concentration. Conclusion: The results of the present study suggest dietary L. paracasei NSMJ15 supplementation may increase percentage of CD4+ T cells and decrease acetate concentration in broiler chickens by increasing the differential expression of specific microbial genera.

Importance: In vivo imaging studies of reactive astrocytes are crucial for understanding the pathophysiology of schizophrenia because astrocytes play a critical role in glutamate imbalance and neuroinflammation. Objective: To investigate in vivo reactive astrocytes in patients with schizophrenia associated with positive symptoms using monoamine oxidase B (MAO-B)-binding fluorine 18 ([18F])-labeled THK5351 positron emission tomography (PET). Design, Setting, and Participants: In this case-control study, data were collected from October 1, 2021, to January 31, 2023, from the internet advertisement for the healthy control group and from the outpatient clinics of Seoul National University Hospital in Seoul, South Korea, for the schizophrenia group. Participants included patients with schizophrenia and age- and sex-matched healthy control individuals. Main Outcomes and Measures: Standardized uptake value ratios (SUVrs) of [18F]THK5351 in the anterior cingulate cortex (ACC) and hippocampus as primary regions of interest (ROIs), with other limbic regions as secondary ROIs, and the correlation between altered SUVrs and Positive and Negative Syndrome Scale (PANSS) positive symptom scores. Results: A total of 68 participants (mean [SD] age, 32.0 [7.0] years; 41 men [60.3%]) included 33 patients with schizophrenia (mean [SD] age, 32.3 [6.3] years; 22 men [66.7%]) and 35 healthy controls (mean [SD] age, 31.8 [7.6] years; 19 men [54.3%]) who underwent [18F]THK5351 PET scanning. Patients with schizophrenia showed significantly higher SUVrs in the bilateral ACC (left, F = 5.767 [false discovery rate (FDR)-corrected P = .04]; right, F = 5.977 [FDR-corrected P = .04]) and left hippocampus (F = 4.834 [FDR-corrected P = .04]) than healthy controls. Trend-level group differences between the groups in the SUVrs were found in the secondary ROIs (eg, right parahippocampal gyrus, F = 3.387 [P = .07]). There were positive correlations between the SUVrs in the bilateral ACC and the PANSS positive symptom scores (left, r = 0.423 [FDR-corrected P = .03]; right, r = 0.406 [FDR-corrected P = .03]) in patients with schizophrenia. Conclusions and Relevance: This case-control study provides novel in vivo imaging evidence of reactive astrocyte involvement in the pathophysiology of schizophrenia. Reactive astrocytes in the ACC may be a future target for the treatment of symptoms of schizophrenia, especially positive symptoms.

This study was conducted to determine the effects of amino acid (AA) supplementation in low-protein (LP) diets on growth performance and nitrogen (N) excretion. A total of 175 7-day-old Ross 308 male broilers, with a mean body weight (BW) of 165 g (standard deviation = 11.2 g), were grouped into five blocks by BW and allocated to seven treatments according to a randomized complete block design with five replicate cages at five birds per cage. Dietary treatments comprised a control diet containing 20.0% crude protein (CP) and six LP diets containing either 18.5% or 17.0% CP. These LP diets were supplemented with either no AA supplementation, indispensable AA, or both indispensable and dispensable AA (glutamic acid and glycine). Birds were fed experimental grower diets from day 7 to 21 and then commercial finisher diets until day 28. During the grower period (day 7 to 21), birds fed LP diets supplemented with indispensable AA exhibited greater (p < 0.05) BW, body weight gain (BWG), feed intake (FI), and gain-to-feed ratio (G:F) than birds fed LP diets without crystalline AA and were comparable to birds fed the control diet. During the finisher period (day 21 to 28), birds fed LP diets supplemented with indispensable AA showed greater (p < 0.05) BW than birds fed LP diets without crystalline AA, and their growth performance was comparable to birds fed the control diet. Throughout the overall period, supplementing indispensable AA in LP diets resulted in elevated (p < 0.05) BWG, FI, and G:F more than those of LP diets without crystalline AA and were comparable to those of the control diet. Supplementing indispensable AA in LP diets decreased amount and coefficient of N excretion as much as the control diet. Dispensable AA supplementation in LP diets did not influence growth performance and N excretion. In conclusion, supplementing indispensable AA in LP diets maintains growth performance and N excretion until the dietary CP lowers from 20.0% to 17.0% during the grower period. As long as dietary CP is above 17.0%, dispensable AA may not be deficient in LP diets during the grower period.

Glutathione peroxidase 4 (GPX4) is a promising anticancer therapeutic target; however, the application of GPX4 inhibitors (GPX4i) is limited owing to intrinsic or acquired drug resistance. Hence, understanding the mechanisms underlying drug resistance and discovering molecules that can overcome drug resistance are crucial. Herein, we demonstrated that GPX4i killed bladder cancer cells by inducing lipid reactive oxygen species-mediated ferroptosis and apoptosis, and cisplatin-resistant bladder cancer cells were also resistant to GPX4i, representing a higher half-maximal inhibitory concentration value than that of parent bladder cancer cells. In addition, thioredoxin reductase 1 (TrxR1) overexpression was responsible for GPX4i resistance in cisplatin-resistant bladder cancer cells, and inhibiting TrxR1 restored the sensitivity of these cells to GPX4i. In vitro and in vivo studies revealed that Jolkinolide B (JB), a natural diterpenoid and previously identified as a TrxR1 inhibitor, potentiated the antiproliferative efficacy of GPX4i (RSL3 and ML162) against cisplatin-resistant bladder cancer cells. Furthermore, GPX4 knockdown and inhibition could augment JB-induced paraptosis and apoptosis. Our results suggest that inhibiting TrxR1 can effectively improve GPX4 inhibition-based anticancer therapy. A combination of JB and GPX4i, which is well-tolerated and has several anticancer mechanisms, may serve as a promising therapy for treating bladder cancer.

OBJECTIVE: We aimed to investigate electroencephalographic (EEG) markers of aberrant hyperfocusing, a novel framework of impaired selective attention, in schizophrenia patients by using theta phase-gamma amplitude coupling (TGC). METHODS: Fifty-four schizophrenia patients and 73 healthy controls (HCs) underwent EEG recording during an auditory oddball paradigm. For the standard and target conditions, TGC was calculated using the source signals from 25 brain regions of interest (ROIs) related to attention networks and sensory processing; TGC values were then compared across groups and conditions using two-way analysis of covariance. Correlations of altered TGC with performance on the Trail Making Test Parts A and B (TMT-A/B), were explored. RESULTS: Compared to HCs, schizophrenia patients showed elevated TGC in the left inferior frontal gyrus (IFG) and superior temporal gyrus in the standard condition but not in the target condition. Correlation analyses revealed that the TGC in the left IFG was positively correlated with the TMT-A/B completion times. CONCLUSIONS: Aberrant hyperfocusing, as reflected by elevated TGC in attention-related brain regions, was related to behavioral performance on the TMT-A/B in schizophrenia patients. SIGNIFICANCE: This study suggests that TGC is a electrophysiological marker for aberrant hyperfocusing of attentional processes that may result in cognitive impairments in schizophrenia patients.

OBJECTIVES: Gout, as the most prevalent form of inflammatory arthritis, necessitates the use of animal models to investigate the molecular mechanisms involved in its development. Therefore, our objective was to develop a novel chronic mouse model of gout that more closely mimics the progression of gout in humans. METHODS: A novel chronic mouse model of gout was established by a simple method, which does not require high technical proficiency, predominantly involves daily intraperitoneal injections of potassium oxonate for approximately 4 months, combined with a high fat-diet and injections of acetic acid into the hind paws to facilitate the formation of monosodium urate (MSU). Arthritis scores and paw oedema were assessed, behavioural tests were conducted, and histopathological and imaging evaluations of the arthritic paw joints were performed. RESULTS: After 4 months of induction, mice in the model group exhibited noticeable increases in arthritis severity, joint and cartilage damage, as well as bone erosion. Gomori's methenamine silver stain revealed the presence of MSU crystal deposition or tophi in the paw joints or ankle joints of up to 37.9% of the model mice (11 out of 29 mice). Moreover, treatment with benzbromarone effectively prevented the further development of gout or tophi formation in model mice. CONCLUSIONS: Our model more accurately replicates the pathological features of gouty arthritis compared with gout induced by MSU crystal injections. Therefore, it is particularly suitable for further investigations into the pathogenesis of gout and also serves as a valuable platform for screening potential antigout agents.

The study investigated the effects of feeding duration on the length and weight of the ileum, and basal endogenous loss (BEL) of phosphorus (P) on broiler chickens fed a P-free diet (PFD). A standard starter diet was fed to 384 birds for 15 days. On day 15, they were weighed and randomly allocated to one of three treatment groups in a randomized complete block design, with eight replicate cages per treatment and 16 birds per cage. The birds in each treatment group were fed the PFD for 2, 3, or 4 days. There was an increase in growth performance (p < 0.05) as the PFD feeding duration increased from 2 to 4 days. Although the relative dry weight of the ileum (mg/100 g of body weight) decreased linearly (p < 0.05) as the PFD feeding duration increased, the amount of dried ileal digesta (g/bird) was not affected. The BEL of P was 110.2, 128.2, and 133.6 mg/kg of dry matter intake in birds fed the PFD for 2, 3, and 4 days, respectively. Feeding a PFD to broiler chickens for 2, 3, or 4 days did not change P BEL.

Decubitus ulcers are a common spinal cord injury (SCI) complication that puts patients' lives in danger and has emerged as a more prevalent issue in modern clinical rehabilitation and care. Decubitus ulcers in humans can currently be treated in a number of different ways, but there are fewer studies on how to treat and care for decubitus ulcers in macaques. To treat a 20-year-old adult male macaque monkey with SCI and decubitus ulcers after a quarter transection of the thoracic spinal cord, a number of scientific care procedures and pharmaceutical treatments, such as dietary changes and topical or intravenous administration of medication, were carried out and continuously monitored in real-time. In comparison to the untreated group, we observed a significant improvement in decubitus wound healing in the macaques. In this article, we provide a good protocol for decubitus ulcer care after SCI and suggest that future experimental animal modeling needs to focus on issues such as care for postoperative complications.

This study aimed to evaluate the effects of dietary Chlorella vulgaris (CV) on the distribution of immune cells, intestinal morphology, intestinal barrier function, antioxidant markers, and the cecal microbiome in 10-day-old broiler chickens. A total of 120 day-old Ross 308 male broiler chicks were assigned to two dietary treatments using a randomized complete block design, with body weight as the blocking factor. Birds fed a diet containing CV showed an increase in CD4+ T cells (p < 0.05) compared to those fed the control diet. The relative mRNA expression of intestinal epithelial barrier function-related markers (occludin and avian ß-defensin 5) was elevated (p < 0.05) in the CV-supplemented group compared to the control group. The alpha diversity indices (Chao1 and observed features) of the cecal microbiome in 10-day-old birds increased (p < 0.05), indicating higher richness within the cecal bacterial community. In the microbiome analysis, enriched genera abundance of Clostridium ASF356 and Coriobacteriaceae CHKCI002 was observed in birds fed the diet containing CV compared to those fed the control diet. Taken together, dietary CV supplementation might alter intestinal barrier function, immunity, and microbiomes in 10-day-old broiler chickens.

Recent studies have shown a direct projection of nociceptive trigeminal afferents into the lateral parabrachial nucleus (LPBN). Information about the synaptic connectivity of these afferents may help understand how orofacial nociception is processed in the LPBN, which is known to be involved primarily in the affective aspect of pain. To address this issue, we investigated the synapses of the transient receptor potential vanilloid 1-positive (TRPV1+) trigeminal afferent terminals in the LPBN by immunostaining and serial section electron microscopy. TRPV1 + afferents arising from the ascending trigeminal tract issued axons and terminals (boutons) in the LPBN. TRPV1+ boutons formed synapses of asymmetric type with dendritic shafts and spines. Almost all (98.3%) TRPV1+ boutons formed synapses with one (82.6%) or two postsynaptic dendrites, suggesting that, at a single bouton level, the orofacial nociceptive information is predominantly transmitted to a single postsynaptic neuron with a small degree of synaptic divergence. A small fraction (14.9%) of the TRPV1+ boutons formed synapses with dendritic spines. None of the TRPV1+ boutons were involved in axoaxonic synapses. Conversely, in the trigeminal caudal nucleus (Vc), TRPV1+ boutons often formed synapses with multiple postsynaptic dendrites and were involved in axoaxonic synapses. Number of dendritic spine and total number of postsynaptic dendrites per TRPV1+ bouton were significantly fewer in the LPBN than Vc. Thus, the synaptic connectivity of the TRPV1+ boutons in the LPBN differed significantly from that in the Vc, suggesting that the TRPV1-mediated orofacial nociception is relayed to the LPBN in a distinctively different manner than in the Vc.

BACKGROUND: The Bcl2-associated athanogene4 (BAG4/SODD) protein could be identified as a tumor marker for several malignancies and plays a major role in the occurrence, development, and drug resistance of tumors. However, the role of Silencer of death domains (SODD) in lung carcinogenesis is still elusive. OBJECTIVE: To illuminate the effect of SODD on the proliferation, migration, invasion, and apoptosis of lung cancer cells and tumor growth in vivo and explore the corresponding mechanism. METHODS: The expression of SODD in tumor and normal tissues was determined and compared via western blot. SODD gene knockout lung cancer cells (H1299 cells) were established through a CRISPR/Cas9 gene deleting system, and a transient SODD overexpression of H1299 cells was also constructed. Then, cell proliferation and invasion were assessed through colony formation and cell counting kit-8 assays, transwell migration assays, and wound healing assays. Cell drug sensitivity is also analyzed by Cell Counting Kit-8 assay. The flow cytometer was used to perform cell circle and apoptosis analysis. The interaction of SODD and RAF-1 was confirmed by co-immunoprecipitation, and the phosphorylated level of Phosphatidylinositol 3-kinase (PI3K), Serine/threonine-protein kinase (AKT), Rapidly accelerated fibrosarcoma (RAF)-1,and extracellular signal regulated kinase (ERK) in cells was examined by western blot to evaluate the activation of PI3K/PDK1/AKT and RAF/MEK/ERK pathways. In vivo, Xenograft tumor assay of SODD knockout H1299 cells was used to evaluate further the role of SODD on the proliferation of H1299 cells. RESULTS: SODD binds to RAF-1 and is over-expressed in lung tissues, and promotes the proliferation, migration, invasion, and drug sensitivity of H1299 cells. The reduced cells in the S phase and increased cells arrested in the G2/M phase were found in SODD knockout H1299 cells, and more cells got apoptosis. The expression of 3-phosphoinositide-dependent protein kinase 1(PDK1) protein in SODD knockout H1299 cells decreases distinctively, and the phosphorylated level of AKT, RAF-1, and ERK-1 kinase in SODD knockout H1299 cells is also less than that in normal H1299 cells. In contrast, SODD overexpression significantly increases the phosphorylation of AKT. In vivo, SODD promotes the tumorigenicity of H1299 cells in nude mice. CONCLUSIONS: SODD is overexpressed in lung tissues and plays a considerable role in the development and progression of lung cancer by regulating the PI3K/PDK1/AKT and RAF/MEK/ERK pathways.

RESUMO

Chinese medicine (CM) is an important resource for human life understanding and discovery of drugs. However, due to the unclear pharmacological mechanism caused by unclear target, research and international promotion of many active components have made little progress in the past decades of years. CM is mainly composed of multi-ingredients with multi-targets. The identification of targets of multiple active components and the weight analysis of multiple targets in a specific pathological environment, that is, the determination of the most important target is the main obstacle to the mechanism clarification and thus hinders its internationalization. In this review, the main approach to target identification and network pharmacology were summarized. And BIBm (Bayesian inference modeling), a powerful method for drug target identification and key pathway determination was introduced. We aim to provide a new scientific basis and ideas for the development and international promotion of new drugs based on CM.

The growth performance and histochemical characteristics of breast muscle fibers were used to estimate the standardized ileal digestible (SID) Lys requirements for 10- to 21-d-old male broilers. Three hundred and sixty 10-d-old Ross 308 broilers (290 ± 16.6 g) were allocated to 6 diets in a randomized complete block design with 6 replicate cages per treatment and 10 birds per cage. The 6 experimental diets were formulated to contain equally spaced increasing levels of SID Lys from 0.86% to 1.36%. The data were analyzed using the MIXED procedure of SAS. The Lys requirements were estimated by the NLIN procedure of SAS. An increase in dietary SID Lys from 0.86% to 1.36% resulted in a quadratic increase (P < 0.05) in body weight gain (BWG), gain to feed ratio (G:F), breast weight, muscle cross-sectional area (MCSA), and fiber area. The SID Lys requirements based on the one-slope broken-line, quadratic line, the first intercept between the plateau of the one-slope broken-line and quadratic-line models and 95% of the upper asymptote of the quadratic-line model were estimated to be 1.01%, 1.19%, 1.08%, and 1.13% for BWG, 1.06%, 1.22%, 1.11%, and 1.16% for G:F, 1.10%, 1.29%, 1.19%, and 1.22% for breast weight, 1.06%, 1.22%, 1.12%, and 1.16% for MCSA, and 1.14%, 1.22%, 1.16%, and 1.16% for breast muscle fiber area, respectively. It was concluded that the SID Lys requirements for broilers at the age of 10 to 21 d depended on the response variables used for estimation, and that histochemical characteristics of breast muscle fibers could be good indicators for estimating SID Lys requirements.

The weight coefficients of appearance traits, extract yield of standard decoction, and total content of honokiol and magnolol were determined by analytic hierarchy process(AHP), criteria importance though intercrieria correlation(CRITIC), and AHP-CRITIC weighting method, and the comprehensive scores were calculated. The effects of ginger juice dosage, moistening time, proces-sing temperature, and processing time on the quality of Magnoliae Officinalis Cortex(MOC) were investigated, and Box-Behnken design was employed to optimize the process parameters. To reveal the processing mechanism, MOC, ginger juice-processed Magnoliae Officinalis Cortex(GMOC), and water-processed Magnoliae Officinalis Cortex(WMOC) were compared. The results showed that the weight coefficients of the appearance traits, extract yield of standard decoction, and total content of honokiol and magnolol determined by AHP-CRITIC weighting method were 0.134, 0.287, and 0.579, respectively. The optimal processing parameters of GMOC were ginger juice dosage of 8%, moistening time of 120 min, and processing at 100 ℃ for 7 min. The content of syringoside and magnolflorine in MOC decreased after processing, and the content of honokiol and magnolol followed the trend of GMOC>MOC>WMOC, which suggested that the change in clinical efficacy of MOC after processing was associated with the changes of chemical composition. The optimized processing technology is stable and feasible and provides references for the modern production and processing of MOC.

Gut health has been attracting attention in the livestock industry as several studies suggest that it is a crucial factor for growth performance and general health status in domestic animals, including broiler chickens. Previously, antibiotics were widely used to improve livestock growth, but their use is now prohibited due to serious problems related to antibiotic resistance. Thus, finding new feed additives to replace antibiotics is drawing attention. Probiotics are representative feed additives and many beneficial effects on broiler chickens have been reported. However, many probiotic studies are focused on productivity only, and there are insufficient studies related to the gut environment, especially gut immunity and gut microbiome. In this study, we conducted an animal experiment using Lacticaseibacillus paracasei NSMJ56 to determine whether it has beneficial effects on gut immunity and microbiome. To evaluate the effects of NSMJ56 supplementation, newly hatched Ross 308 broiler chickens were fed an NSMJ56-containing diet for 10 days, and growth performance, antioxidant indicators, gut morphology, gut immunity-related parameters, and gut microbiome were analyzed. Flow cytometry analysis results revealed that NSMJ56 treatment increased CD4+ T cells and decreased CD8+ T cells in small intestine lamina propria and decreased IL1b and IL10 gene expression in small intestine tissue. In the microbiome analysis, NSMJ56 treatment increased the alpha diversity indices and led to three enriched genera: Massilimicrobiota, Anaerotignum, and Coprococcus. This study suggests that NSMJ56 supplementation has regulatory effects on gut immunity and microbiome in early-age broiler chickens.

BACKGROUND: Despite advances in treatment strategies for coronary heart disease, angina pectoris remains a major cardiovascular disease causing death worldwide. For patients with angina pectoris of coronary heart disease, new or adjuvant treatment regimens are needed. The available evidence suggests that Xuefu Zhuyu Granules combined with Western medicine has advantages in the treatment of angina pectoris of coronary heart disease, but whether its efficacy has a placebo effect and whether it can be used as an adjuvant regimen for the treatment of angina pectoris of coronary heart disease remains controversial. METHODS: This is a prospective, randomized, double-blind, placebo-controlled trial to study the efficacy and safety of Xuefu Zhuyu Granules combined with Western medicine in the treatment of angina pectoris of coronary heart disease. Participants will be randomly divided into a treatment group or a control group, and all patients will receive Western medicine treatment based on guideline recommendations. On this basis, the treatment group orally takes Xuefu Zhuyu Granules and the control group orally takes Xuefu Zhuyu Granules mimic, and are followed up for 24 weeks after 12 weeks of continuous treatment. The observation indexes include: cardiac function parameters (left ventricular end-diastolic diameter; left ventricular end-systolic diameter; left ventricular ejection fraction, blood lipid levels (total cholesterol; triacylglycerol; low-density lipoprotein cholesterol; high-density lipoprotein cholesterol), the number of angina attacks per week, total amount of nitroglycerin tablets taken, and adverse reactions. Finally, SPSS22.0 (IBM Company, New York, NY) software will be used for statistical analysis of the data. DISCUSSION: This study will evaluate the efficacy and safety of Xuefu Zhuyu Granules combined with Western medicine in the treatment of angina pectoris of coronary heart disease. The results of this study will verify whether the efficacy of Xuefu Zhuyu Granules in the treatment of angina pectoris of coronary heart disease belongs to the placebo effect, which will also provide a reference for the clinical use of Xuefu Zhuyu Granules as a supplementary scheme for the treatment of angina pectoris of coronary heart disease.

Two experiments were conducted to determine the effects of bird age on apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of amino acids (AA) for 10-d-old Experiment (Exp. 1) and 22-d-old (Exp. 2) male broilers. This study investigated the effects of different broiler ages and feed ingredients on AID and SID of AA in corn and soybean meal (SBM). Four hundred and eighty (age = 7 d; initial body weight [BW] = 173.4 ± 12.65 g) and 192 (age = 18 d; initial BW = 772.2 ± 62.13 g) birds were allocated to three dietary treatments in a randomized complete block design with eight replicate cages per treatment. Two diets were formulated based on corn or SBM as the sole source of AA in the diet. A nitrogen-free diet was also formulated to measure basal endogenous losses of AA. Experimental diets were given for 3 and 4 days in Exps. 1 and 2, respectively. An interaction was observed (p < 0.05) between the age of birds and the type of ingredient for the AID of most AA, except for methionine, valine, cysteine (Cys), and tyrosine; however, the effects of age and type of ingredients were diminished in the SID of AA, except for histidine, isoleucine, leucine (Leu), phenylalanine, alanine (Ala), and glutamic acid (Glu). The AID of AA, except for Leu and Cys and the SID of AA, except for Leu, Ala, Glu, and Pro in SBM were greater (p < 0.05) than in corn. As the age of birds increased from 10 to 22 d, digestibility of all AA increased (p < 0.05), regardless of the expression of AA digestibility (i.e., AID and SID). In conclusion, the AID and SID of AA in both corn and SBM increased with increasing age, and the AID and SID of AA in SBM were greater than in corn.
