The use of plants for medicinal purposes has a long history, however it is desirable a continuous evaluation seeking for complementary scientific evidences for their safe application. Species within the Kalanchoe genus are often referred to as "miracle leaf" due to their remarkable healing properties. Traditionally, these plants have been used to treat infections, inflammation, and cancer. Despite their widespread use, the identification of their active components remains incomplete. This study aimed to differentiate K. crenata (KC), K. marmorata (KM), and K. pinnata (KP) by conducting detailed histochemical and phytochemical analyses, and to assess their antioxidant capabilities. The investigation revealed significant differences between the species, highlighting the variability in phenolic (PC) and flavonoid contents (FC) and their distinct antioxidant effects. The KM demonstrated the greatest results (PC: 59.26±1.53 mgEqGA/g; FC: 12.63±0.91 mgEqCQ/g; DPPH⋅ (IC50): 110.66 ug/mL; ABTS⋅+ (IC50): 26.81 ug/mL; ORAC: 9.65±0.75 mmolTE) when compared to KC and KP. These findings underscore a new reference for research within the Kalanchoe genus.

Euphorbia umbellata is used for its anti-inflammatory properties; however, there are limited data available regarding its effects on vascular function. Its bark is rich in polyphenolic compounds, which potentially improve endothelial dysfunction (ED). This study proposes to investigate the effects of E. umbellata bark extracts and its polyphenolic compounds on arginase (ARG) activity and nitric oxide (NO)-related targets. Chromatographic procedures were used for the chemical characterisation of the extracts. Furthermore, in silico (molecular docking), in vitro (ARG inhibition), in vivo (streptozotocin-induced hyperglycemia model), and ex vivo (l-arginine metabolism, vascular reactivity, western blot, and biochemical) techniques were carried out. Quercetin, gallic acid, and ellagic acid were identified in the extracts. In silico screening predicted that gallic acid and quercetin would have the most promising interactions with ARG -identified cavities. This was confirmed in vitro as both compounds had a direct inhibitory effect on ARG, as was the case regarding the extracts. Oral treatment preserved endothelium-dependent vasodilation through ARG inhibition together with an increase in l-arginine bioavailability and endothelial NO synthase expression. Biochemical parameters determined the lack of toxicity for sub-chronic treatment. E. umbellata bark extracts and its compounds can contribute to ED treatment, at least partly, through the inhibition of vascular ARG.

The current study was carried out by a bioguided fractionation of a hexane extract of the latex of Euphorbia umbellata against leukemic cells. Samples were analyzed by NMR, GC/MS, triterpenes quantification, and MTT reduction assay. Morphological, cell cycle, mitochondrial membrane potential and caspases 3/7 analyses were performed for the dichloromethane and ethanol fractions, and selectivity index for the dichloromethane fraction. NMR analysis presented characteristic signals of terpenes and steroids, data were confirmed by the quantification of triterpenes and GC/MS analysis. MTT reduction assay demonstrated that HL-60 was the most sensitive cell lineage against dichloromethane and ethanol fractions. Compounds of these matrices caused morphological changes compatible with apoptosis induction, altered cell cycle, increment of depolarized population cells and activation of caspases 3/7. Selectivity indices were higher than 22.44. Bioguided-fractionation study showed that samples of the latex of E. umbellata raised the activity of the phytocomplex against leukemic cells, and the cytotoxicity can be associated with an apoptosis pathway.

Abstract Sapium belongs to Euphorbiaceae family and comprises 23 species. Sapium glandulosum (L.) Morong is popularly known in Brazil as "pau-leiteiro" and "leitosinha" and it is used in traditional medicine to cicatrisation. Its leaf extracts have shown analgesic, anti-inflammatory and antibacterial activities. The preliminary set of pharmacognostic tools used for quality assessment of medicinal plant parts is macro- and micro-anatomy and S. glandulosum has not anatomical and histochemical description. Thus the aim of this study was to investigate the anatomical and histochemical characteristics of the leaf and stem of S. glandulosum as a means of providing information for quality assessment of herbal industry. The leaves and stems were investigated by employing field emission scanning electron microscopy, light microscopy, and histochemistry techniques. The analysis showed that S. glandulosum had the following anatomical features: dorsiventral and amphistomatic leaves; paracytic stomata; tabular crystal druses; non-articulated and branched laticifers; midrib's biconvex shape with vascular systems in open arc with invaginated ends; petiole with a round shape and slight concavity on the adaxial side; six collateral vascular bundles in U-shaped organisation; a circular stem shape and a sclerenchymatous ring. In the histochemical tests lipophilic components were found in cuticle and in the latex; phenolic compounds were met in the mesophyll and in the latex; starch grains were found in the parenchymatous sheath; lignified elements were met in the sclerenchymatous ring in the cortex and in the perivascular sclerenchymatous caps, beyond in the vessel elements. These features are helpful when conducting a quality control process.
